Clinical manifestations, outcomes, and antibody profile of Brazilian adult patients with dermatomyositis: a single-center longitudinal study.

BACKGROUND: Currently, only a few retrospective cohort or cross-sectional studies have described the general characteristics of Brazilian patients with classical dermatomyositis (DM). In contrast, we aimed to longitudinally assess a large sample of these patients, and several myositis autoantibodies. METHODS: This single-center longitudinal study included 91 Brazilian adults with defined DM (EULAR/ACR 2017) who underwent follow-up appointments in our tertiary center from 2012 to 2021. Myositis autoantibody analysis was performed using a commercial kit. RESULTS: The mean age of the patients w

AAV1.NT-3 gene therapy in a CMT2D model: phenotypic improvements in GarsP278KY/+ mice.

Glycyl-tRNA synthetase mutations are associated to the Charcot-Marie-Tooth disease type-2D. The GarsP278KY/+ model for Charcot-Marie-Tooth disease type-2D is known best for its early onset severe neuropathic phenotype with findings including reduced axon size, slow conduction velocities and abnormal neuromuscular junction. Muscle involvement remains largely unexamined. We tested the efficacy of neurotrophin 3 gene transfer therapy in two Gars mutants with severe (GarsP278KY/+ ) and milder (GarsΔETAQ/+ ) phenotypes via intramuscular injection of adeno-associated virus setoype-1, triple tandem m

Increased Protein S-Glutathionylation in Leber's Hereditary Optic Neuropathy (LHON).

Leber's hereditary optic neuropathy (LHON, MIM#535000) is the most common form of inherited optic neuropathies and mitochondrial DNA-related diseases. The pathogenicity of mutations in genes encoding components of mitochondrial Complex I is well established, but the underlying pathomechanisms of the disease are still unclear. Hypothesizing that oxidative stress related to Complex I deficiency may increase protein S-glutathionylation, we investigated the proteome-wide S-glutathionylation profiles in LHON (n = 11) and control (n = 7) fibroblasts, using the GluICAT platform that we recently devel

Evaluation of usefulness in surfactant protein D as a predictor of mortality in myositis-associated interstitial lung disease.

OBJECTIVE: Surfactant protein D (SP-D) is considered a serum biomarker of various forms of interstitial lung disease (ILD). In this study, we examined the utility of SP-D as a predictive biomarker for mortality in patients with ILD associated with polymyositis/dermatomyositis (PM/DM) using large-scale multicentre cohort data. METHODS: We enrolled 381 patients with incident PM/DM-associated ILD in a multicentre retrospective cohort based on the availability of serum SP-D at the baseline. Demographic and clinical characteristics as well as the presence of autoantibodies to melanoma differentiat

Differential network analysis and protein-protein interaction study reveals active protein modules in glucocorticoid resistance for infant acute lymphoblastic leukemia.

BACKGROUND: Acute lymphoblastic leukemia (ALL) is the most common type of cancer diagnosed in children and Glucocorticoids (GCs) form an essential component of the standard chemotherapy in most treatment regimens. The category of infant ALL patients carrying a translocation involving the mixed lineage leukemia (MLL) gene (gene KMT2A) is characterized by resistance to GCs and poor clinical outcome. Although some studies examined GC-resistance in infant ALL patients, the understanding of this phenomenon remains limited and impede the efforts to improve prognosis. METHODS: This study integrates

The proteomic comparison of peripheral circulation-derived exosomes from the epithelial ovarian carcinoma (EOC) patients and non-EOC subjects.

BACKGROUND: Ovarian cancer is the most lethal tumor of the female reproductive system. Establishing a methodology to screen and diagnose ovarian cancer in the early stage is important. Exosomes have been shown to be loaded with tumor-associated molecules. In this study, we compared the proteins loaded in exosomes from the peripheral circulation of epithelial ovarian carcinoma (EOC) patients and controls. METHODS: Exosomes were purified via ultracentrifugation plus 0.22 µm filtration from the blood of EOC patients and patients with pelvic floor dysfunction (PFD). Tumor tissues and normal ovari

An integrated quantitative proteomic and metabolomics approach to reveal the negative regulation mechanism of LamB in antibiotics resistance.

Previously, a maltose-specific channel porin, LamB was found to be associate with multi-drug resistance in a lamB deleted strain, but the exact mechanisms require further elucidation. Herein, differential protein expression between the Escherichia coli mutant strain ΔlamB and the wild type strain BW25113, with and without ciprofloxacin (CFLX), was identified using iTRAQ based liquid chromatography-tandem mass spectrometry (LC-MS/MS); while differential metabolite expression was examined using gas chromatography-mass spectrometry (GC-MS). Angiogenesis and VEGF-expressing cells are identified predominantly in the fascia rather than in the muscle during the early phase of dermatomyositis.

BACKGROUND: We previously demonstrated that fasciitis is a common lesion in dermatomyositis (DM) and that DM-associated fasciitis is detectable, as the result of the increased vascularity in the fascia, by power Doppler ultrasonography. We aimed to investigate whether angiogenesis and vascular endothelial growth factor (VEGF)-expressing cells in the fascia are histologically demonstrated during the early phase of DM, and whether inflammation is involved in angiogenesis and an increased number of VEGF-expressing cells. METHODS: We prospectively evaluated 22 patients with DM and 11 patients wit

Autoantibodies and their clinical significance in idiopathic inflammatory myopathies; polymyositis/dermatomyositis and related conditions.

The inflammatory muscle diseases, polymyositis (PM) and dermatomyositis (DM) are systemic connective tissue disorders characterized by chronic inflammation in skeletal muscle and involvement of various systemic organs. The pathogenesis of these heterogeneous diseases is unknown, but appear to mediate an autoimmune disorder that culminates in the tissue damage. Autoantibodies directed against various cellular constituents have been detected in patients with PM/DM, and 40-50% of patients have autoantibodies (myositis-specific antibodies : MSAs) that are found specifically in myositis patients. Acetylcholine receptor degradation measured by density labeling: effects of cholinergic ligands and evidence against recycling.

The methodology of density labeling of proteins by biosynthetic incorporation of 2H, 13C, 15N-amino acids into newly synthesized polypeptide chains allows the direct measurement of the turnover rate of the acetylcholine receptor in cultured chick skeletal muscle. In this study, receptors synthesized in medium containing 2H, 13C, 15N-amino acids were resolved from 1H, 12C, 14N-receptors by velocity sedimentation in sucrose-deuterium oxide gradients, and their proportions were determined by computer analysis of the gradient profiles. The kinetics of turnover of acetylcholine receptors are identi

Integrative multi-omics analysis reveals systemic and intestinal responses to heat stress in finishing pigs.

Heat stress (HS) is a major environmental threat to swine production that impairs growth performance and health. Because of their limited thermoregulatory capacity, pigs are highly vulnerable to HS, which results in compromised intestinal integrity, systemic inflammation, and metabolic inefficiency. To elucidate the mechanisms underlying HS acclimation in pigs, we conducted a longitudinal multi-omics analysis integrating fecal microbiome, whole-blood transcriptome, and immune cell deconvolution in finishing pigs under thermoneutral (TN) or HS conditions. HS markedly reduced the average daily g

Transcriptome and metabolome integration reveals molecular mechanisms underlying divergent cold tolerance in geographically distinct Procambarus clarkii populations.

Low temperature is a major environmental stressor that constrains the survival and distribution of Procambarus clarkii. To explore the cold adaptation mechanisms of geographically differentiated populations, we exposed P. clarkii from Northeast China, Hubei and Anhui to continuous cold stress at 4 °C, while crayfish maintained at 20 °C serving as the control group. After 10 days of stress exposure, tail-muscle tissues were collected for transcriptomic and metabolomic analyses. Phenotypic observation revealed distinct cold tolerance among populations: the Northeast population had the highest co

Hybrid Dual-Context Prompted Cross-Attention Framework with Language Model Guidance for Multi-Label Prediction of Human Off-Target Ligand-Protein Interactions.

Accurately identifying drug off-targets is essential for reducing toxicity and improving the success rate of pharmaceutical discovery pipelines. However, current deep learning approaches often struggle to fuse chemical structure, protein biology, and multi-target context. Here, we introduce HDPC-LGT (Hybrid Dual-Prompt Cross-Attention Ligand-Protein Graph Transformer), a framework designed to predict ligand binding across sixteen human translation-related proteins clinically associated with antibiotic toxicity. HDPC-LGT combines graph-based chemical reasoning with protein language model embedd

Morphine-induced dysbiosis and metabolic remodeling of the gut microbiota is transmissible via fecal microbiota transplantation in mice.

OBJECTIVE: Opioids such as morphine are widely used analgesics but are highly addictive. Emerging evidence implicates the gut microbiota in modulating opioid-associated pathophysiology. However, the global metabolic impact of morphine on gut microbial function and the causal transmissibility of morphine-induced dysbiosis remain to be fully elucidated. METHODS: Adult C57BL/6 mice received repeated morphine administration for four consecutive days. Fecal metabolomic profiling was performed using integrated gas chromatography-mass spectrometry (GC/MS) and liquid chromatography-mass spectrometry

Effects of Different Selenium Concentrations on Agronomic Traits, Antioxidant Defense, and Leaf Metabolome in Blueberry (Vaccinium corymbosum L. 'Brigitta').

Selenium (Se) is an important micronutrient that is required in very small amounts and plays a significant role in enhancing plant growth, stress resistance, and fruit quality. In this study, we investigated the effects of different sodium selenite concentrations (CK, 0 mg/L; Se1, 0.50 mg/L; Se2, 1.00 mg/L, Se3, 2.00; and Se4, 3.00 mg/L) on the growth, nutrient absorption, antioxidant capacity, and leaf metabolome of blueberry (Vaccinium corymbosum L. 'Brigitta') in hydroponic culture. Our results showed that moderate Se concentration (1.00 mg/L, Se2) had noticeable enhancements in key traits

"Lariocidin and ribosome-targeting lasso peptides as emerging antimicrobial agents against multidrug-resistant bacteria".

Lasso peptides are ribosomally synthesized and post-translationally modified peptides (RiPPs) with mechanically constrained structures that provide high stability and target selectivity. Among them, lariocidin (LAR), produced by Paenibacillus sp. M2, represents a recently discovered peptide antibiotic with a ribosome-targeting mechanism distinct from classical antibacterial agents. lariocidin binds a distinct site within the bacterial 30S ribosomal subunit, involving interactions with 16S rRNA and aminoacyl-tRNA, thereby disrupting protein synthesis through translocation inhibition and miscodi

Prognostic Value of EEF1A1 and Its Correlation with Immune Regulation in Kidney Renal Clear Cell Carcinoma.

BACKGROUND: Eukaryotic translation elongation factor 1 alpha 1 (EEF1A1) primarily participates in protein synthesis by binding aminoacyl-tRNA complexes to facilitate peptide chain elongation on ribosomes. Its expression and functional roles exhibit significant heterogeneity across various malignancies, exerting dual regulatory effects as both an oncogene and a tumor suppressor. This study aims to investigate the potential prognostic value and tumor-suppressive role of EEF1A1 in kidney renal clear cell carcinoma (KIRC). METHODS: We analyzed the differential expression of EEF1A1 in KIRC and its

Differentially abundant proteins, metabolites, and lipid molecules in spaghetti meat compared to normal chicken breast meat: Multiomics analysis.

Spaghetti meat (SM), a recently emerging muscle myopathy in chicken breast meat, is characterized by a loss of muscle fiber integrity, resulting in a spaghetti-like appearance. Understanding the differences in proteins, metabolites, and lipids through a multiomics approach in SM can identify its quality traits and elucidate its exact causes. The purpose of this study was to investigate differentially abundant proteins, metabolites, and lipid molecules in SM compared to normal chicken breast meat (Control). The supernatant from sample homogenates was subjected to ultra-high performance liquid c

AIMP2 accumulation in brain leads to cognitive deficits and blood secretion in Parkinson's disease.

BACKGROUND: Propagation of neuronal α-synuclein aggregate pathology to the cortex and hippocampus correlates with cognitive impairment in Parkinson's disease (PD) dementia and dementia with Lewy body disease. Previously, we showed accumulation of the parkin substrate aminoacyl-tRNA synthetase interacting multifunctional protein-2 (AIMP2) in the temporal lobe of postmortem brains of patients with advanced PD. However, the potential pathological role of AIMP2 accumulation in the cognitive dysfunction of patients with PD remains unknown. METHODS: We performed immunofluorescence imaging to examin
