Assessment of the infectivity of malaria parasites from asymptomatic school children to Anopheles gambiae mosquitoes in a high transmission area in Ghana.

Asymptomatic carriage of malaria parasite is a major public health issue in Ghana and sub-Saharan Africa. These infections are mostly sub-microscopic and not detected by routine malaria screening methods. Asymptomatic malaria infection carriers serve as an infectious reservoir for malaria transmission. This study assessed the infectivity of asymptomatic children to Anopheles gambiae mosquitoes in a high transmission area in Ghana. Ninety-eight (98) healthy children were screened for malaria parasites by microscopy and PCR. Sub-microscopic gametocytes carriage was also determined using RT-PCR. …

The Effect of Larval Exposure to Plastic Pollution on the Gut Microbiota of the Major Malaria Vector Anopheles arabiensis Patton (Diptera: Culicidae).

Plastic pollution is prevalent in water bodies. However, most studies on plastic pollution focus on marine environments, with limited knowledge about its impact on freshwater ecosystems. This paucity of information extends to the effect on aquatic insects, with little reported data on the effect of plastic on malaria vectors. This is concerning as microplastics are reported to perturb the gut microbiota of culicine mosquitoes. This study examines how larval exposure to degraded plastic, plastic additives (phthalic acid, Bisphenol-A) and latex beads affects the gut microbiota of adult Anopheles…

Habitat heterogeneity and green filamentous algae influence the larval ecology of Anopheles stephensi during the dry season in Eastern Ethiopia.

BACKGROUND: The recent invasion of Anopheles stephensi, an urban-adapted malaria vector, poses a threat to malaria elimination efforts in Africa. Understanding the larval ecology of this mosquito during the dry season, which represents a potential population bottleneck due to limited larval habitats and harsher environmental conditions, is critical for informing targeted interventions. METHODS: We conducted systematic surveys in three climatically distinct Ethiopian cities-Semera, Logiya and Jigjiga-during the dry season of 2023. A total of 523 water-holding habitats were identified and chara…

The status of insecticide resistance of Anopheles coluzzii on the islands of São Tomé and Príncipe, after 20 years of malaria vector control.

BACKGROUND: Insecticide-based malaria vector control has been implemented on the islands of São Tomé and Príncipe (STP) for more than 20 years. During this period malaria incidence was significantly reduced to pre-elimination levels. While cases remained low since 2015, these have significantly increased in the last year, challenging the commitment of the country to achieve malaria elimination by 2025. To better understand the reasons for increasing malaria cases, levels and underlying mechanisms of insecticide resistance in the local Anopheles coluzzii populations were characterized. METHODS…

A review of applications and limitations of using aquatic macroinvertebrate predators for biocontrol of the African malaria mosquito, Anopheles gambiae sensu lato.

Macroinvertebrate predators such as backswimmers (Heteroptera: Notonectidae), dragonflies (Odonata: Aeshnidae), and predatory diving beetles (Coleoptera: Dytiscidae) naturally inhabit aquatic ecosystems. Some aquatic ecosystems inhabited by these macroinvertebrate predator taxa equally form malaria vector larval habitats. The presence of these predators in malaria vector larval habitats can negatively impact on development, adult body size, fecundity, and longevity of the malaria vectors, which form important determinants of their fitness and future vectorial capacity. These potential negative

In Vitro Culture of Plasmodium falciparum for the Production of Mature Gametocytes for Performing Standard Membrane Feeding Assay and Infection of Anopheles spp.

Plasmodium falciparum is the parasite responsible for the disease malaria. In vitro cultivation of mature gametocytes of P. falciparum plays a central role in evaluating and developing the transmission-blocking drugs and sexual stage vaccines. These types of preventive molecules are crucial for controlling malaria in the future. Among different Plasmodium species that are involved in human malaria, only P. falciparum is cultivable. Therefore, an efficient method is required for in vitro culture of P. falciparum producing mature and infective gametocytes. This chapter describes a reliable and e

Molecular Drivers of Multiple and Elevated Resistance to Insecticides in a Population of the Malaria Vector Anopheles gambiae in Agriculture Hotspot of West Cameroon.

(1) Background: Malaria remains a global public health problem. Unfortunately, the resistance of malaria vectors to commonly used insecticides threatens disease control and elimination efforts. Field mosquitoes have been shown to survive upon exposure to high insecticide concentrations. The molecular mechanisms driving this pronounced resistance remain poorly understood. Here, we elucidated the pattern of resistance escalation in the main malaria vector Anopheles gambiae in a pesticide-driven agricultural hotspot in Cameroon and its impact on vector control tools; (2) Methods: Larval stages an…

Variation in Pyrethroid Resistance Phenotypes in Anopheles darlingi in an Area with Residual Malaria Transmission: Warning of Suspected Resistance in French Guiana.

Anopheles darlingi is the main vector of malaria in South America. In French Guiana, malaria transmission occurs inland and along the rivers with a regular reemergence in the lower Oyapock area. Control against malaria vectors includes indoor residual spraying of deltamethrin and the distribution of long-lasting impregnated bednets. In this context, the level of resistance to pyrethroids was monitored for 4 years using CDC bottle tests in An. darlingi populations. A loss of susceptibility to pyrethroids was recorded with 30-minute knock-down measured as low as 81%. However, no pyrethroid molec…

Nonrandom Selection and Multiple Blood Feeding of Human Hosts by Anopheles Vectors: Implications for Malaria Transmission in Papua New Guinea.

Nonrandom selection and multiple blood feeding of human hosts by Anopheles mosquitoes may exacerbate malaria transmission. Both patterns of blood feeding and their relationship to malaria epidemiology were investigated in Anopheles vectors in Papua New Guinea (PNG). Blood samples from humans and mosquito blood meals were collected in villages and human genetic profiles ("fingerprints") were analyzed by genotyping 23 microsatellites and a sex-specific marker. Frequency of blood meals acquired from different humans, identified by unique genetic profiles, was fitted to Poisson and negative binomi…

A method for distinguishing the important malaria vectors Anopheles dirus and An. cracens (Diptera: Culicidae) based on antennal sensilla of adult females.

Some species of the Anopheles dirus species complex are considered to be highly competent malaria vectors in Southeast Asia. Anopheles dirus is the primary vector of Plasmodium falciparum and P. vivax while An. cracens is the main vector of P. knowlesi. However, these two species are difficult to distinguish and identify based on morphological characters. Hence, the aim of this study was to investigate the potential use of antennal sensilla to distinguish them. Large sensilla coeloconica borne on the antennae of adult females were counted under a compound light microscope and the different typ…

Ultra-Low Volume Application of Spinosad (Natular 2EC) Larvicide as a Residual in a Tropical Environment Against Aedes and Anopheles Species.

We investigated the efficacy of a liquid larvicide, Natular 2EC® (spinosad), applied with ultra-low volume sprayer as a residual application during the dry season in southeastern Thailand against 4 medically important species-Aedes aegypti, Ae. albopictus, Anopheles dirus, and An. minimus. We found that this larvicide could be applied as a residual to dry areas known to collect water and potentially still be effective after rains or irrigation, which could increase the flexibility and efficiency of an integrated vector management program targeting these species. This investigation also demonst…

Genetic diversity and population structure of the primary malaria vector Anopheles sinensis (Diptera: Culicidae) in China inferred by cox1 gene.

BACKGROUND: Anopheles sinensis is a primary vector for Plasmodium vivax malaria in most regions of China. A comprehensive understanding of genetic variation and structure of the mosquito would be of benefit to the vector control and in a further attempt to contribute to malaria elimination in China. However, there is only inadequate population genetic data pertaining to An. sinensis currently. METHODS: Genetic variations and structure among populations of An. sinensis was examined and analyzed based on the nucleotide sequences of a 662 nt variable region of the mitochondrial cox1 gene among 1…

Modulation of transmission success of Plasmodium falciparum gametocytes (sexual stages) in various species of Anopheles by erythrocytic asexual stage parasites.

During malaria infection, a small proportion of erythrocytic asexual stages undergo sexual differentiation. Male and female gametocytes ingested in the blood meal initiate the sexual development of malaria parasites in the mosquito midgut. During blood feeding on a host, a mosquito ingests, in addition to mature gametocytes, host immune factors present in the blood, as well as large excess of erythrocytic asexual stages. In the current study we addressed the impact of the presence of large excess of asexual stages, hitherto not known or even suspected to influence, on the infectivity of gameto

Tissue-specific differences in the spatial interposition of X-chromosome and 3R chromosome regions in the malaria mosquito Anopheles messeae Fall.

Spatial organization of a chromosome in a nucleus is very important in biology but many aspects of it are still generally unresolved. We focused on tissue-specific features of chromosome architecture in closely related malaria mosquitoes, which have essential inter-specific differences in polytene chromosome attachments in nurse cells. We showed that the region responsible for X-chromosome attachment interacts with nuclear lamina stronger in nurse cells, then in salivary glands cells in Anopheles messeae Fall. The inter-tissue differences were demonstrated more convincingly in an experiment of…

Age and prior blood feeding of Anopheles gambiae influences their susceptibility and gene expression patterns to ivermectin-containing blood meals.

BACKGROUND: Ivermectin has been proposed as a novel malaria transmission control tool based on its insecticidal properties and unique route of acquisition through human blood. To maximize ivermectin's effect and identify potential resistance/tolerance mechanisms, it is important to understand its effect on mosquito physiology and potential to shift mosquito population age-structure. We therefore investigated ivermectin susceptibility and gene expression changes in several age groups of female Anopheles gambiae mosquitoes. METHODS: The effect of aging on ivermectin susceptibility was analyzed …

A potential threat to malaria elimination: extensive deltamethrin and DDT resistance to Anopheles sinensis from the malaria-endemic areas in China.

BACKGROUND: Insecticide resistance in malaria vectors is a growing concern in many countries and requires immediate attention because of the limited chemical arsenal available for vector control. There is lack of systematic and standard monitoring data of malaria vector resistance in the endemic areas, which is essential for the ambitious goal of malaria elimination programme of China. METHODS: In 2010, eight provinces from different malaria endemic region were selected for study areas. Bioassays were performed on F1 progeny of Anopheles sinensis reared from wild-caught females using the stan…
