Transcriptomic and regulatory landscape of liver tissue associated with meat and carcass quality traits in cattle.

BACKGROUND: The integration of multiple omics strategies represents a transformative paradigm in farm animal genetics and breeding. By capturing molecular complexity across biological layers, integrative omics offers new opportunities to reveal the regulatory mechanisms underlying economically important traits. Here, we characterize the hepatic transcriptomic landscape of Nelore cattle and its relationship with meat and carcass quality phenotypes. For that, we integrated gene expression data, co-expression networks, expression quantitative trait locus (eQTL) mapping, single-nucleotide polymorp

Genotype-Specific Root System Remodeling of Paspalum spp. Induced by Plant Growth-Promoting Bacteria Enhances the Potential for Sustainable Tropical Pastures.

Brazilian cattle production relies predominantly on pasture systems, many of which are degraded by nutrient depletion, particularly nitrogen deficiency. Plant growth-promoting bacteria (PGPB) offer a sustainable strategy to improve pasture productivity, but their effects are often host-specific and remain poorly explored in Paspalum. This study evaluated the effect of inoculation with Azospirillum brasilense strains CNPSo 2083 (=Ab-V5) and CNPSo 2084 (=Ab-V6) and Pseudomonas fluorescens strain CNPSo 2719 (=CCTB 03), applied by seed inoculation or leaf spray, on the root morphology and shoot gr

Ex vivo assay for organ-specific cancer cell invasion.

BACKGROUND: Metastasis is the leading cause of cancer-related mortality, yet experimental models often fail to recapitulate the tissue-specific microenvironments shaping metastatic dissemination. While in vivo systems provide physiological relevance, they remain challenging for mechanistic studies. Conversely, conventional in vitro assays lack the organ-specific extracellular matrix (ECM) that regulates invasive behavior. Accessible models that balance biological relevance with experimental feasibility are thus needed. RESULTS: We developed an ex vivo invasion platform based on mild detergent

Addressing unmet needs in rare diseases: A patient-centred vision through INFORM-RD.

Rare diseases collectively affect approximately 6% of the global population, yet 95% of affected individuals lack access to effective treatments. Beyond this persistent therapeutic gap, patients and caregivers face a broad spectrum of unmet needs, spanning psychosocial challenges, access to reliable diagnostic information, care coordination, and long-term follow-up, which remain poorly characterised in both clinical and policy contexts. Three interrelated challenges underpin this problem. First, validated instruments capable of systematically capturing unmet needs across diverse patient groups

Physical, chemical, and structural properties of human gastric organoid-derived mucus.

The gastric mucus layer protects the epithelium from gastric acid and ingested pathogens. However, studies of human gastric mucus have been limited due to poor accessibility of native human mucus and the abundance of contaminants in these samples. Here, we explored the potential of human gastric organoids as models for mucus production. Immunofluorescence staining confirmed that the organoids produced mucus containing MUC5AC and MUC6. The luminal mucus had viscoelastic properties similar to those of native human gastric mucus, as determined by particle tracking microrheology. To collect organo

Human-Derived Cellular Models in Psychiatry: A Focus on the Olfactory Neuroepithelium.

Severe mental disorders, including schizophrenia (SCZ), bipolar disorder (BD), and major depressive disorder (MDD), are leading causes of global disability, yet current treatments remain largely symptomatic and fail to alter disease trajectories. Converging evidence from genetics, longitudinal studies, and systems neuroscience supports a dimensional and transdiagnostic architecture of psychopathology, involving shared polygenic risk and overlapping neurodevelopmental and circuit-level alterations. Traditional approaches-such as post-mortem brain analysis, neuroimaging, and animal models-have d

Engineering controlled antimicrobial delivery across biofilm matrix barriers: mechanics, transport and nanocarrier design.

Bacterial biofilms are a major challenge for antimicrobial therapy because they function not only as structured microbial communities, but also as complex biological barriers to drug delivery. Biofilm-associated bacteria are embedded within an extracellular polymeric substance (EPS) matrix that provides mechanical stability, regulates antimicrobial transport and generates heterogeneous microenvironments characterised by nutrient limitation, oxygen gradients and metabolically diverse bacterial subpopulations. As a result, therapeutic failure in mature biofilms often reflects a mismatch between

Fabrication and Biocompatibility Testing of Thin-Film Metamaterial Neurostimulation Leads for Magnetic Resonance Imaging.

OBJECTIVE: Chronically implantable neurostimulation leads, such as those used for deep brain stimulation, remain only partially MRI-compatible due to the presence of wire conductors that can induce hazardous radiofrequency (RF) heating. As a result, current systems are limited to MRI-conditional use, restricting patients' access to routine clinical imaging. Here, we propose a novel metamaterial based Resistive Tapered Stripline (RTS) architecture as a candidate solution for next-generation DBS leads. METHODS: The RTS design incorporates impedance transitions along the lead, producing metamate

A novel protein-preserving passive tissue clearing approach using sodium cholate and urea for whole-organ imaging.

The recent advancements in tissue-clearing techniques have opened new possibilities for non-invasive three-dimensional (3D) volumetric imaging of a wide range of biological specimens. Passive tissue-clearing methods use diffusion-based processes to infiltrate clearing reagents into samples without mechanical forces or energy input, aiming to minimize the sample disruption while preserving the tissue architecture and molecular information. Nevertheless, these methods often rely on sodium dodecyl sulfate (SDS) as a delipidating detergent, which has a risk of causing tissue damage and protein dis

Development and Clinical Validation of Lightweight, Multimodal Machine Learning Models for Smartphone-Based Cataract Detection and Classification.

Globally, cataract remains the leading cause of blindness, affecting over 100 million people, with a disproportionate burden in low- and middle-income countries where access to ophthalmologists is limited. Although cataract surgery can restore vision almost immediately, timely diagnosis and referral remains a major barrier to care. We developed and prospectively evaluated lightweight, multimodal machine learning models capable of classifying lens status on a smartphone, enabling accessible screening in low-resource environments. We trained and evaluated both early and late fusion model archite

Colorimetric sensing for transdermal phospho-Tau 181 detection mediated by wearable microneedle functionalized with gold nanoparticle (MN-AuNP).

Microneedles (MNs) have rapidly emerged as powerful tools in wearable biosensing, providing minimally invasive access to interstitial fluid (ISF). Among the neurodegenerative based biomarkers detectable in ISF, phosphorylated Tau at threonine 181 (p-Tau181) is reaching a clinically evaluable significance for Alzheimer's disease (AD) and other tauopathies. Elevated p-Tau181 levels are strongly correlated with abnormal Tau aggregation in the brain and with cognitive decline. Current diagnostic methods rely on invasive cerebrospinal fluid (CSF) sampling or costly laboratory immunoassays and radio

Three-Dimensional Ultrastructure of Arabidopsis Cotyledons Infected with Colletotrichum higginsianum.

We used serial block-face scanning electron microscopy (SBF-SEM) to study the host-pathogen interface between Arabidopsis cotyledons and the hemibiotrophic fungus Colletotrichum higginsianum. By combining high-pressure freezing and freeze-substitution with SBF-SEM, followed by segmentation and reconstruction of the imaging volume using the freely accessible software IMOD, we created 3D models of the series of cytological events that occur during the Colletotrichum-Arabidopsis susceptible interaction. We found that the host cell membranes underwent massive expansion to accommodate the rapidly g

From aid dependency to market transformation: harnessing international trade and innovative business models to revitalize the assistive technology ecosystem in Africa.

Africa faces a widening and largely unaddressed gap in access to assistive technology (AT). More than 240 million people on the continent require at least one assistive product, yet fewer than 3% of those in sub-Saharan Africa currently have access. The principal causes are four interlocking structural market failures: restrictive trade policies that inflate import costs, chronically underdeveloped local manufacturing capacity, inadequate national health financing for rehabilitation, and the absence of sustainable business models capable of maintaining AT services beyond the lifespan of time-l

Resolving Chemically Inequivalent 11B NMR Sites via Interpretable Hybrid Machine Learning.

Organoboron compounds are widely used across pharmaceuticals and materials science, where 11B NMR spectroscopy serves as a valuable tool for structural characterization. However, severe spectral line broadening induced by the quadrupolar nature of the boron nucleus often causes signal overlap, making it exceptionally difficult to experimentally resolve chemically inequivalent sites in complex multiboron architectures. While traditional density functional theory can resolve these ambiguities, it faces prohibitive computational bottlenecks, whereas data-driven alternatives remain constrained by

Intrinsic Nonlinearity Modulation in Two-Dimensional (Cu,Ag)InP2S6 for Selectorless Nonvolatile Memory Array.

Selectorless resistive random-access memory is essential for scaling high-density crossbar arrays, yet suppressing sneak path currents (SPCs) without external selector components remains a major challenge. In this work, we investigated a two-dimensional (2D) van der Waals (vdW) mixed cation crystal Cu0.5Ag0.5InP2S6 (CAIPS) as a switching layer and systematically compared its resistive switching with CuInP2S6 (CIPS) and AgInP2S6 (AIPS). The coexistence of Cu+ and Ag+ ions produces asymmetric out-of-plane diffusion barriers, as confirmed by first-principles density functional theory (DFT) calcul

Sustainable Fragment Peptide Synthesis (SFPS): Leveraging Oxyma as a Dual Resin Cleavage-Peptide Coupling Agent.

Coupling of fully protected peptide fragments (FPPFs) released off acid labile polymer supports such as 2-chlorotrityl chloride (CTC) resin is a powerful peptide synthesis approach used from R&D to manufacturing type 2 diabetes/obesity drugs on a ton scale. Nevertheless, cleavage of acid labile FPPF resins typically entails PFAS-classified TFA as a reagent and suspected carcinogen DCM as a solvent while necessitating laborious work-ups and isolations. Here, in an approach we term sustainable fragment peptide synthesis (SFPS), a cleavage of FPPFs off 2-chlorotrityl (2-CT) resin was carried out

RGCNN-nnUNet: Recurrent group equivariant nnU-Net for robust brain tissue segmentation on stroke NCCT.

Computed tomography (CT), particularly non-contrast CT (NCCT), is crucial in emergency settings for rapid stroke diagnosis due to its affordability and accessibility, yet subtle density variations in early ischemia are often obscured by high noise and low soft-tissue contrast in NCCT. To address this, we present RGCNN-nnUNet, a novel architecture designed for high-fidelity brain tissue segmentation. We introduce a Recurrent Group Convolutional Cell that leverages group equivariance to extract consistent anatomical features and stabilize iterative refinement. Evaluations on multi-center dataset

Space-Time Coding Conformal Metasurfaces for Multifrequency Beam Steering and Shaping.

Space-time coding metasurfaces enable dynamic control of electromagnetic waves by jointly exploiting spatial coding and temporal modulation, unlocking functionalities unattainable with static platforms. While these concepts have been widely explored in planar configurations, their extension to conformal geometries has received comparatively little attention, despite the prevalence of curved platforms in practical applications. Here, space-time coding conformal metasurfaces are examined as a platform for multifrequency beam steering and shaping on nonplanar structures. A representative cylindri
