FTY720-induced blockage of autophagy enhances anticancer efficacy of milatuzumab in mantle cell lymphoma: is FTY720 the next autophagy-blocking agent in lymphoma treatment?

Anti-melanoma differentiation-associated protein 5 (MDA5) antibody have been found in dermatomyositis (DM)-associated interstitial lung disease (DM-ILD) and DM-associated rapidly progressive ILD (DM-RPILD). Due to the conflicting results regarding the association between anti-MDA5 antibody and DM-ILD or DM-RPILD and the diagnostic value of this antibody for DM-ILD and DM-RPILD, we performed this meta-analysis. A systematic search was performed to identify studies published to January 14, 2017. Sixteen publications with 491 DM with ILD versus 605 DM without ILD, as well as eighteen publications

N6-methyladenosine-modified USP13 induces pro-survival autophagy and imatinib resistance via regulating the stabilization of autophagy-related protein 5 in gastrointestinal stromal tumors.

Secondary resistance to imatinib (IM) represents a major challenge for therapy of gastrointestinal stromal tumors (GISTs). Aberrations in oncogenic pathways, including autophagy, correlate with IM resistance. Regulation of autophagy-related protein 5 (ATG5) by the ubiquitin-proteasome system is critical for autophagic activity, although the molecular mechanisms that underpin reversible deubiquitination of ATG5 have not been deciphered fully. Here, we identified USP13 as an essential deubiquitinase that stabilizes ATG5 in a process that depends on the PAK1 serine/threonine-protein kinase and wh…

Differential autophagy cargo engagement in amyloid-β-induced neuronal toxicity.

Neurodegenerative diseases are characterized by progressive neuronal loss and the accumulation of misfolded proteins. In Alzheimer's disease, several pathogenic protein species, including amyloid precursor protein, amyloid beta, tau, phosphorylated tau, and alpha-synuclein, are known substrates of macroautophagy. However, the extent to which macroautophagy selectively engages these proteins for degradation under conditions of proteotoxic stress remains poorly understood. Here, the contribution of specifically macroautophagy to the degradation of amyloid precursor protein, amyloid beta, tau, ph…

Regulation of STK38 by autophagy governs YAP1 activity during paligenosis.

Paligenosis is a conserved cellular plasticity program that allows mature cells to reenter the cell cycle in response to tissue injury. Paligenosis progresses via three stages: autodegradation (with dramatic increase in autophagy and lysosomes), induction of metaplastic or fetal-like genes, and cell cycle entry. Hippo signaling, particularly the downstream effector YAP1, regulates cellular plasticity, but its role in paligenosis has not been studied. Here, we examine YAP1 dynamics during paligenosis in digestive-enzyme-secreting chief cells from the mouse stomach. We identified Serine/Threonin…

Autophagy-regulated myocardial fibrosis: from mechanistic exploration to therapeutic prospects.

Cardiovascular diseases (CVDs) represent a leading cause of global mortality. Myocardial fibrosis (MF), a common pathological hallmark in CVDs, signifies disease progression to severe stages. Its pathogenesis is highly complex, with autophagy playing a pivotal regulatory role. This narrative review synthesizes and critically evaluates the current literature to elucidate the interplay between MF and autophagy. The focus lies on systematically examining the molecular mechanisms and key signaling pathways involved in autophagy-mediated regulation of MF. We systematically examine key molecular pat…

Small bites for big problems: stepwise aggregate degradation by autophagy.

Protein aggregates are a pathological hallmark of diverse disorders, including many neurodegenerative diseases, but also cardiometabolic disease and cancer. While the ubiquitin-proteasome system efficiently removes many soluble misfolded proteins, large or persistent assemblies often require the autophagy-lysosome pathway for their degradation. In the present mini-review, we summarize our knowledge of aggrephagy, the selective clearance of protein aggregates by autophagy, and discuss two recent manuscripts that argue that some aggregates must be primed for autophagosomal degradation, through c…

Omipalisib reduces hyperphosphorylated tau protein by modulating mTOR-autophagy pathway.

Tauopathies are neurodegenerative diseases characterized by the presence of hyperphosphorylated tau (p-tau) and neurofibrillary tangles. Autophagy is a critical self-degradation mechanism that preserves cellular homeostasis and function, including the clearance of misfolded proteins. Autophagy is impaired in tauopathies, resulting in excessive accumulation of p-tau. Omipalisib, a dual phosphatidylinositol 3-kinase/mammalian target of rapamycin (PI3K/mTOR) inhibitor, was explored in a phase I clinical trial involving solid tumors and lymphoma. In this study, we aimed to investigate the effects …

Natural Combatants: How Herbal Monomers Regulate Autophagy in Ovarian Cancer.

INTRODUCTION: Ovarian cancer (OC) is a lethal gynecological malignancy, with current therapies constrained by drug resistance and side effects. Autophagy plays a dual role in OC, while Traditional Chinese Medicine (TCM) monomers, as natural bioactive compounds, demonstrate significant potential in regulating autophagy and combating tumors. This review aims to summarize the mechanisms by which TCM monomers regulate autophagy in OC, providing a theoretical basis for TCM-based drug development and clinical applications. METHODS: Relevant literature was retrieved from databases including PubMed, …

Chaperone-mediated autophagy is required for regulatory T cell function.

Chaperone-mediated autophagy (CMA) is a selective form of protein degradation in lysosomes that declines with age. Besides protein quality control, CMA also regulates several cellular processes through timely proteome remodeling. We previously demonstrated the importance of CMA in the activation of helper T cells. In this work, we analyzed the role of CMA in the generation and function of regulatory T cells (Tregs), a specialized type of T cells that suppress immune responses. We found that the basal CMA activity of Tregs further increases upon their activation. Using a Treg-specific CMA-defic…

Targeting LAPTM5 enhances AML sensitivity to cytarabine through autophagy inhibition.

Upregulation of autophagy in acute myeloid leukemia (AML) cells contributes to the development of resistance to cytarabine (AraC). LAPTM5 is mainly expressed in hematopoietic and immune cells, and has been associated with the progression of multiple cancers; however, its role in AML drug resistance remains uncharacterized. Here, we reanalyzed publicly available single-cell RNA sequencing (scRNA-seq) data from AML patients and found distinct gene expression profiles between AraC-resistant AML cells and untreated controls. Differentially expressed genes were significantly enriched in lysosome-re…

Nrf1 coordinates proteasome activity and autophagy to maintain cardiac proteostasis.

Proteolytic stress frequently arises during disease and aging, particularly in long-lived, post-mitotic cells such as cardiomyocytes. To maintain proteostasis, cardiomyocytes depend on coordinated protein quality control pathways, including the ubiquitin-proteasome system and autophagy. Mechanisms that activate these pathways hold therapeutic potential for heart disease. Here, we demonstrate that transient activation of nuclear factor erythroid 2-like 1 (Nfe2l1, also known as Nrf1), a transcriptional regulator of proteasome activity, in cardiomyocytes during ischemia/reperfusion injury improve…

A dendritic cell autophagy-neutrophil axis limits intratumoral STING immunotherapy.

The cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) pathway promotes tumor immunogenicity, but intratumoral STING agon … erferon signaling was dispensable for cGAMP-VLP-mediated tumor control. In contrast, dendritic cell autophagy was required for generating circulating antitumor CD8 T cells and for regulating baseline neutrophil levels in lymph nodes. cGAMP-VLP overrode this regulation, inducing neutrophil accumulation in tumors and draining lymph nodes that limited efficacy …

Glutathione Primes Mitochondrial Autophagy to Guide Pulp-Dentine Complex Repair.

AIM: Pulp-dentine complex repair following injury is fundamental to preserving tooth vitality and function. Despite advances in understanding reparative dentinogenesis, the molecular mechanisms orchestrating odontogenic differentiation of human dental pulp stem cells (hDPSCs) remain fragmented, lacking systematic integration across transcriptomic, proteomic and metabolomic levels. This study employs integrated multi-omics approaches to identify key regulatory pathways during odontogenic differentiation and elucidate the underlying mechanisms controlling pulp-dentine complex repair. Autophagy selectively clears ER in TNF-α-induced muscle atrophy.

Skeletal muscle atrophy is a pathological condition characterized by the progressive loss of muscle mass and function, driven by factors such as disuse, inflammation, and aging. While the ubiquitin-proteasome system is established as the central mediator of myofibrillar protein degradation, the role of selective autophagy and the degradation of organelles remains underexplored in this context. To address this, we employed a quantitative, time-resolved in vitro analysis of protein synthesis and degradation in C2C12 myotubes undergoing TNF-α-induced atrophy, using dynamic Stable Isotope Labeling…

CCT7 Regulates TRAF6-Mediated Autophagy to Promote Posttraumatic Joint Contracture.

Posttraumatic joint contracture (PTJC) is a prevalent complication of joint injury, characterised by marked reductions in both active and pa … e expression of α-SMA and COL-I and suppressed fibroblast migration, while simultaneously promoting autophagy, as indicated by increased LC3-II and Beclin-1 levels and decreased p62 expression, thereby alleviating joint fibrosis. Interestingly, CCT7 interacted with TRAF6 to facilitate its ubiquitination and degradation. Knockdown of TRAF6 reversed the promo …

Tank-binding kinase 1 regulates inflammation and autophagy in glaucoma.

TANK-binding kinase (TBK1) copy number variations have been identified as a monogenic cause of familial normal tension glaucoma (NTG). While elevated intraocular pressure (IOP) is a common risk factor in primary open angle glaucoma (POAG), NTG develops in the absence of IOP elevation. The current available standard of care for glaucoma is lowering IOP, highlighting the need to explore alternative therapeutic mechanisms. The established roles of TBK1 in nuclear factor kappa B (NF-κB) and interferon regulatory factor (IRF) signaling, as well as its involvement in autophagy, suggest it may contri…
