Innate Immunity to Intracellular Pathogens: Lessons Learned from Legionella pneumophila.

Intracellular bacterial pathogens have the remarkable ability to manipulate host cell processes in order to establish a replicative niche within the host cell. In response, the host can initiate immune defenses that lead to the eventual restriction and clearance of intracellular infection. The bacterial pathogen Legionella pneumophila has evolved elaborate virulence mechanisms that allow for its survival inside protozoa within a specialized membrane-bound organelle. These strategies also enable L. pneumophila to survive and replicate within alveolar macrophages, and can result in the severe pn

Phosphoproteomic analysis of Salmonella-infected cells identifies key kinase regulators and SopB-dependent host phosphorylation events.

Salmonella enterica is a bacterial pathogen that causes gastroenteritis and typhoid fever. Virulence is achieved by two type III secretion systems that translocate effector proteins into host cells, where they mimic or block host protein function. Effectors translocated into host cells by the first type III secretion system facilitate invasion and stimulate intracellular signaling cascades leading to inflammation. Here, we performed global temporal analysis of host signaling events induced during the initial stages of Salmonella infection and identified the dynamics of host protein phosphoryla

Mycobacterial Membrane Proteins QcrB and AtpE: Roles in Energetics, Antibiotic Targets, and Associated Mechanisms of Resistance.

Infections caused by mycobacteria are difficult to treat due to their inherent physiology, cellular structure, and intracellular lifestyle. Mycobacterium tuberculosis is a pathogen of global concern as it causes tuberculosis (TB) in humans, which requires 6-9 months of chemotherapy. The situation is further exacerbated in the case of infections caused by drug-resistant strains, which necessitate the prolonged use of agents associated with increased host toxicities. Great effort has been invested into the development of new agents for the treatment of drug-resistant infections, in addition to n

Listeria monocytogenes PrsA2 is required for virulence factor secretion and bacterial viability within the host cell cytosol.

In the course of establishing its replication niche within the cytosol of infected host cells, the facultative intracellular bacterial pathogen Listeria monocytogenes must efficiently regulate the secretion and activity of multiple virulence factors. L. monocytogenes encodes two predicted posttranslocation secretion chaperones, PrsA1 and PrsA2, and evidence suggests that PrsA2 has been specifically adapted for bacterial pathogenesis. PrsA-like chaperones have been identified in a number of Gram-positive bacteria, where they are reported to function at the bacterial membrane-cell wall interface

The Brucella type IV secretion system and effector proteins.

Brucella spp. are Gram-negative, facultative intracellular bacteria responsible for brucellosis, a globally prevalent zoonosis affecting both humans and animals. The genus includes several pathogenic species which primarily infect mammals but can cause chronic infections in humans through accidental transmission. As for most intracellular pathogens, Brucella pathogenicity relies on its capacity to invade host cells, evade immune defenses, and establish a replicative niche within a specialized organelle, the Brucella-containing vacuole (BCV). Central to this process is the VirB Type IV secretio

Immune Activation and Microbial Translocation as Prognostic Biomarkers for AIDS-Related Non-Hodgkin Lymphoma in the AMC-034 Study.

PURPOSE: AIDS-related non-Hodgkin lymphoma (ARL) is the most common cancer in HIV-infected individuals in the United States and other countries in which HIV-positive persons have access to effective combination antiretroviral therapy (cART). Our prior work showed that pretreatment/postdiagnosis plasma levels of some cytokines, such as IL6, IL10, and CXCL13, have the potential to serve as indicators of clinical response to treatment and survival in ARL. The aims of this study were to identify novel prognostic biomarkers for response to treatment and/or survival in persons with ARL, including bi

HIV and liver disease: a comprehensive update.

Despite substantial advances in the field, liver disease morbidity and mortality remain serious issues among people with HIV. The causes of liver disease are often multifactorial and include hepatitis viruses, hepatic steatosis and oxidative stress, bacterial translocation with activation of hepatic macrophages and stellate cells, and direct toxicities from alcohol and drugs of abuse. Biopsychosocial factors including a high prevalence of psychiatric disorders, food insecurity, insufficient access to care and medications, and social stigma all play roles in the persistence of liver injury and …

Health evaluation of black-faced impala (Aepyceros melampus petersi) using blood chemistry and serology.

As part of ongoing ecological studies of black-faced impala (Aepyceros melampus petersi) in northern Namibia, blood samples were collected and analyzed from 26 animals captured for translocation in 1992. All animals appeared to be in good condition and no abnormality was noted during physical examination. Serum chemistry and mineral levels were measured and correlated with the results of bacterial and viral serology and were within the normal ranges for domestic ruminants. Antibody titers for infectious bovine rhinotracheitis and bovine viral diarrhea were detected. Serological tests for bluet

Barrier breakdown: insights into the skin-gut axis in psoriatic arthritis.

Psoriatic arthritis (PsA) is a chronic inflammatory disease that primarily affects the skin, joints, and entheses. Growing evidence implicates alterations at the gut and skin barriers in PsA pathogenesis. These sites play a crucial role in tissue homeostasis, and their disruption can contribute to the translocation of immune cells and bacterial products, resulting in systemic inflammation. Crosstalk between barrier sites and joints can promote or exacerbate disease development, with barriers also being a potential target of disease. This review article provides a barrier-centered perspective a

The role of bacterial outer membrane vesicles in inflammatory response of acute-on-chronic liver failure.

Acute-on-chronic liver failure (ACLF) is a clinical syndrome that manifests as acute deterioration of liver function due to a series of etiologies and triggers in patients with pre-existing chronic liver diseases. Systemic inflammatory response is the major feature of ACLF. Gut microbiota dysbiosis impairs the intestinal barrier, facilitating the translocation of microorganisms and their metabolites into the liver and thereby exacerbating liver inflammation and disease progression. Recent studies have revealed that bacterial outer membrane vesicles (OMVs) derived from gut microbiota act as key

Clostridium ramosum bacteremia: A case series at a general acute care hospital.

Clostridium ramosum infections have been rarely reported, probably due to underestimating in clinical practice. Seven patients with bacteremia from gastrointestinal sources and skin and soft tissue were recognized between 2009 and 2020. Most of them were older and in compromised status, and they had risk factors including cancer, diabetes, liver cirrhosis, gangrene, and pressure ulcers. The source of infections was considered bacterial translocation from the gastrointestine and the skin and soft tissue infections. All patients were treated with antimicrobials, and two received surgical interve…

CSN-associated c-Cbl control alternative NF-κB/RelB activity in Helicobacter pylori infection.

TheThe colonization of the gastric epithelium with Helicobacter pylori elicits a robust nuclear factor kappa-light chain enhancer of activated B cells (NF-κB)-dependent inflammatory response. The bacterial effector molecule adenosine diphosphate-glycero-β-D-manno-heptose (ADP-heptose) triggers both the classical and alternative NF-κB pathways almost simultaneously leading to activation of the transcription factor heterodimers p50/RelA and p52/RelB. Previous studies have demonstrated that the COP9 signalosome complex (CSN) plays an important role in regulating the classical NF-κB pathway throug
