West Nile Virus and Usutu Virus Monitoring of Wild Birds in Germany.

By systematically setting up a unique nation-wide wild bird surveillance network, we monitored migratory and resident birds for zoonotic arthropod-borne virus infections, such as the flaviviruses West Nile virus (WNV) and Usutu virus (USUV). More than 1900 wild bird blood samples, from 20 orders and 136 different bird species, were collected between 2014 and 2016. Samples were investigated by WNV and USUV-specific real-time polymerase chain reactions as well as by differentiating virus neutralization tests. Dead bird surveillance data, obtained from organ investigations in 2016, were also incl

Molecular detection of Hepatozoon spp. and Cytauxzoon felis in wild carnivorans and domestic dogs in southern Pantanal wetlands of Brazil with new host records.

In this study, we investigated the occurrence of Hepatozoon spp. and Cytauxzoon felis in spleen samples from 26 wild carnivorans and blood samples from 28 semi-domiciled dogs collected in the Pantanal biome of Brazil. Partial 18S rRNA gene sequences of Hepatozoon spp. were detected in 15 (57.7 %) wild carnivorans and 3 (10.7 %) domestic dogs. A sequence obtained from a domestic dog was similar (99.04 %) to that of Hepatozoon canis from a crab-eating fox (Cerdocyon thous) in the same area of Brazil. Moreover, we report the neotropical otter (Lontra longicaudis) and the bush dog (Speothos venati

Quantitative proteomic analysis of salivary glands in female Ornithodoros lahorensis (syn. Alveonasus lahorensis) after blood feeding via DIA-coupled LC-MS/MS.

Ornithodoros lahorensis (syn. Alveonasus lahorensis) is a species of soft tick within the Argasidae family, known for causing debilitating symptoms, as well as transmitting pathogens that pose significant health risks and economic losses to affected livestock and potentially humans. The salivary gland is the key component in the pathogenic mechanisms of ticks, as it not only facilitates feeding but also serves as a medium for pathogen transmission. Here, data-independent acquisition (DIA) coupled to liquid chromatography-tandem mass spectrometry (LC-MS/MS) was used to quantitatively identify d

Complement in human disease: approved and up-and-coming therapeutics.

The complement system is recognised as a protector against blood-borne pathogens and a controller of immune system and tissue homoeostasis. However, dysregulated complement activity is associated with unwanted or non-resolving immune responses and inflammation, which induce or exacerbate the pathogenesis of a broad range of inflammatory and autoimmune diseases. Although the merit of targeting complement clinically has long been acknowledged, the overall complement drug approval rate has been modest. However, the success of the humanised anti-C5 antibody eculizumab in effectively treating parox…

A framework for signaling throughout the life cycle of Babesia species.

Babesia species are tick-borne intracellular parasites that infect the red blood cells of their mammalian host, leading to severe or fatal disease. Babesia spp. infect a wide range of mammalian species and cause a significant economic burden globally, predominantly through disease in cattle. Several Babesia spp. are increasingly being recognized as zoonotic pathogens of humans. Babesia spp. have complex life cycles involving multiple stages in the tick and the mammalian host. The parasite utilizes complex signaling pathways during replication, egress, and invasion in each of these stages. They

Mucosal adjuvants: Opportunities and challenges.

Most pathogens access the body via mucosal surfaces. Mucosal vaccination is a highly effective and recommended method to prevent mucosally transmitted infections. Compared with immunization via intramuscular injection, mucosal immunization offers remarkable advantages, including non-invasiveness, low costs and reduced risk of transmission of blood-borne diseases, which make it more acceptable to human beings, especially to young children. However, only few mucosal vaccines are licensed for human, which is mainly due to the deficiency of safe and effective mucosal adjuvants. Adjuvants, as impor

Seroprevalence and distribution of arboviral infections among rural Kenyan adults: a cross-sectional study.

BACKGROUND: Arthorpod-borne viruses (arboviruses) cause wide-spread morbidity in sub-Saharan Africa, but little research has documented the burden and distribution of these pathogens. METHODS: Using a population-based, cross-sectional study design, we administered a detailed questionnaire and used ELISA to test the blood of 1,141 healthy Kenyan adults from three districts for the presence of anti-viral Immunoglobulin G (IgG) antibodies to the following viruses: dengue (DENV), West Nile (WNV), yellow fever (YFV), Chikungunya (CHIKV), and Rift Valley fever (RVFV). RESULTS: Of these, 14.4% were

Infection control in jails and prisons.

At the end of 2005, approximately 7 million people (or 1 of every 33 American adults) were either in jail, in prison, or on parole. Compared with the general public, newly incarcerated inmates have an increased prevalence of human immunodeficiency virus infection, hepatitis B virus infection, hepatitis C virus infection, syphilis, gonorrhea, chlamydia, and Mycobacterium tuberculosis infection. While incarcerated, inmates are at an increased risk for the acquisition of blood-borne pathogens, sexually transmitted diseases, methicillin-resistant Staphylococcus aureus infection, and infection with…

Cells of the marginal zone--origins, function and neoplasia.

Splenic marginal zone B cells of humans and mice are anatomically positioned with specialized macrophages, dendritic and endothelial cells. Together, they function as the first line of defense against blood borne pathogens with a low triggering threshold for B cells providing a rapid proliferative and antibody response to infections. In humans, B cells with similar cytology and physical relations to follicles are found in lymph nodes and Peyer's patches. However, they also develop in mucosa-associated lymphoid tissue (MALT) and other sites, such as the thyroid and salivary gland, that normally

Development of blood-brain barrier-penetrating antibodies for neutralizing tick-borne encephalitis virus in the brain.

UNLABELLED: Tick-borne encephalitis virus (TBEV) belongs to the genus Flavivirus and causes tick-borne encephalitis (TBE), a disease characterized by severe neurological symptoms with a high mortality rate. Currently, no specific antiviral treatments have been developed for TBE. The blood-brain barrier (BBB) restricts drug delivery to the central nervous system, posing a major challenge in developing effective therapies targeting TBEV in the brain. In this study, we developed recombinant anti-TBEV antibodies fused with BBB-penetrating rabies virus glycoprotein (RVG) peptides to facilitate thei

Avian Flavivirus Infection of Monocytes/Macrophages by Extensive Subversion of Host Antiviral Innate Immune Responses.

Avian Tembusu virus (TMUV) is a newly emerging avian pathogenic flavivirus in China and Southeast Asia with features of rapid spread, an expanding host range, and cross-species transmission. The mechanisms of its infection and pathogenesis remain largely unclear. Here, we investigated the tropism of this arbovirus in peripheral blood mononuclear cells of specific-pathogen-free (SPF) ducks and SPF chickens and identified monocytes/macrophages as the key targets of TMUV infection. In vivo studies in SPF ducks and SPF chickens with monocyte/macrophage clearance demonstrated that the infection of

Diagnostic Value of IgA Antibody Measurement in Tick-Borne Spotted Fever (Astrakhan Rickettsial Fever).

Tick-borne spotted fevers caused by Rickettsia occur worldwide. The symptoms of this bacterial infection are similar to those of viral infection, and thus, diagnostic accuracy has special clinical importance. One of the commonly used methods for the diagnosis of tick-borne spotted fever is enzyme-linked immunosorbent assay (ELISA), which is based on estimation of the presence of specific IgM antibodies in blood. However, IgA analysis has not been used for the diagnosis of rickettsial diseases thus far. We investigated the diagnostic value of IgA antibody determination using patient sera collec

Wild canids as reservoirs of zoonotic Bartonella species in Israel.

BACKGROUND: Bartonella species are Gram-negative bacteria transmitted by blood-feeding arthropods and maintained in a wide range of mammalian hosts worldwide. They are emerging and re-emerging pathogens at the wildlife-domestic, animal-human interface. Wild canids are considered important reservoirs and sentinels for vector-borne pathogens, including zoonotic species of Bartonella. This study evaluated the eco-epidemiological traits and diversity of Bartonella infections among wild canids from Israel. METHODS: Between 2018 and 2025, blood samples from wild canids were collected across 158 sam

Ticks threatening lineage of Anatolian wild sheep (Ovis gmelinii anatolica) and determination of their tick-borne pathogens.

We aimed to determine the ticks of the Anatolian wild sheep and to define their tick-borne pathogens while molecularly studying their relationships with those of the domestic sheep. Furthermore, another aim of this study is to investigate tick paralysis resulting in the death of the Anatolian wild sheep. Ticks and blood samples were collected from the wild sheep whilst tick samples were also collected from hares, guinea fowls, chickens, and a turkey living in the Anatolian wild sheep breeding area. While PCR amplification was carried out for the detection of Babesia spp., Theileria spp. and An

Serological Evidence of Akabane, Bluetongue, and Bovine Ephemeral Fever Virus Exposure in Feral Water Buffaloes from Northern Australia.

Water buffaloes in northern Australia occupy tropical wetlands where conditions favour the proliferation of arthropod vectors and the transmission of vector-borne livestock diseases. However, their role in maintaining economically important arboviruses such as Akabane virus (AKAV), bluetongue virus (BTV), and bovine ephemeral fever virus (BEFV) remains poorly understood. These three viruses cause significant production losses in cattle and pose ongoing surveillance challenges in remote areas. To assess exposure to these viruses, a convenience sample of feral water buffaloes from the Northern T
