Metabolic reprogramming is a hallmark of cancer. Somatic mutations in genes involved in oncogenic signaling pathways, including KRAS and TP53, rewire the metabolic machinery in cancer cells. We here set out to determine, at the single cell level, metabolic signatures in human colon cancer cells engineered to express combinations of activating KRAS gene mutations and TP53 gene deletions. Specifically, we explored how somatic mutations in these genes and substrate availability (lactate, glucose, substrate deprivation) from the extracellular microenvironment affect bioenergetic parameters, includ

Cold exposure induces adaptive molecular and mitochondrial remodeling in brown adipose tissue (BAT) of mice to sustain thermogenesis. We previously showed that the lipid droplet protein Perilipin 5 (PLIN5) is strongly induced in BAT by cold exposure and promotes thermogenic gene expression, mitochondrial remodeling, and uncoupled respiration. However, constitutive BAT-specific PLIN5 deletion impaired mitochondrial function without causing cold intolerance, suggesting physiological compensation, likely via induction of thermogenic genes in white adipose tissue (WAT). To test this possibility, w

Acute exposure to high-altitude hypobaric hypoxia (HH) disrupts physiological homeostasis. While gastrointestinal and neurological disturbances are its main manifestations, muscle function is also affected. However, its short-term effects on the gut-muscle axis remain poorly understood. The present study investigated the impact of a 4-hour exposure to simulated altitude (5,000 m) on intestinal morphology, gut microbiota composition, short-chain fatty acid (SCFA) levels, and skeletal muscle mitochondrial function in male Sprague-Dawley rats. Despite the severity of the hypoxic stimulus, biometr

Objective: The mechanisms of structured exercise therapy (SET) in peripheral artery disease (PAD) are not clear. We have developed an SET module for our large animal model of ischemic myopathy. We hypothesized that SET would increase muscle strength and walking distance in this model. The objective was to discover the SET-dependent mechanisms involved in this process. Approach: After induction of unilateral hind limb ischemia, three animals were exposed to standard environmental enrichment (sedentary or SED) and four animals underwent SET thrice weekly for 4 weeks postoperatively. Walking, hin

BACKGROUND: Asprosin is a novel fasting glucogenic adipokine discovered in 2016. Asprosin induces rapid glucose releases from the liver. However, its molecular mechanisms and function are still unclear. Adaptation of energy substrates from fatty acid to glucose is recently considered a novel therapeutic target in heart failure treatment. We hypothesized that the asprosin is able to modulate cardiac mitochondrial functions and has important prognostic implications in dilated cardiomyopathy (DCM) patients.
METHODS: We prospectively enrolled 50 patients (86% male, mean age 55 ± 13 years) with DC

Autism spectrum disorder (ASD) is associated with physiological abnormalities, including abnormal redox and mitochondrial metabolism. Lymphoblastoid cell lines (LCLs) from some children with ASD exhibit increased oxidative stress, decreased glutathione redox capacity, and highly active mitochondria with increased vulnerability to reactive oxygen species (ROS). Because unaffected siblings (Sibs) of individuals with ASD share some redox abnormalities, we sought to determine whether LCLs from Sibs share ASD-associated mitochondrial abnormalities. We evaluated mitochondrial bioenergetics in 10 set

AIMS: Cancer cachexia is a syndrome which results in severe loss of muscle mass and marked fatigue. Conditioned media from cachexia-inducing cancer cells triggers metabolic dysfunction in skeletal muscle, including decreased mitochondrial respiration, which may contribute to fatigue. We hypothesized that Lewis lung carcinoma conditioned medium (LCM) would impair the mitochondrial electron transport chain (ETC) and increase production of reactive oxygen species, ultimately leading to decreased mitochondrial respiration. We incubated C2C12 myotubes with LCM for 30 min, 2, 4, 24 or 48 h. We measu

In aerobic conditions, the proton-motive force drives oxidative phosphorylation (OXPHOS) and the conversion of ADP to ATP. In hypoxic environments, OXPHOS is impaired, resulting in energy shortfalls and the accumulation of protons and lactate. This results in cellular acidification, which may impact the activity and/or integrity of mitochondrial enzymes and in turn negatively impact mitochondrial respiration and thus aerobic ATP production. Naked mole-rats (NMRs) are among the most hypoxia-tolerant mammals and putatively experience intermittent hypoxia in their underground burrows. However, if

From birth, animals should possess functional machinery to appropriately regulate its respiration. This machinery has to detect the available oxygen quantity in order to efficiently modulate breathing movements in accordance with body requirements. The chemosensitivity process responsible for this detection is known to be mainly performed by carotid bodies. However, pulmonary neuroendocrine cells, which are mainly gathered in neuroepithelial bodies, also present the capability to exert chemosensitivity. The goal of this article is to put in perspective the potential complementarity in the acti

Respiratory cryptosporidiosis was diagnosed in a 2-week-old peacock chick, whose symptoms were a gurgling respiration, coughing and sneezing, and a serous oculonasal discharge. Light microscopy detected cryptosporidial-like organisms attached to the surface of conjunctival, nasal-sinus, and tracheal-epithelial cells, and electron microscopy demonstrated stages typical of cryptosporidial trophozoites, schizonts, and oocysts. About 30 other in-contact peacock chicks also developed the same clinical symptoms, and all died despite treatment with oxytetracycline HCl at 200 ppm in the drinking water

Perinatal asphyxia (PA) is a primary cause of neonatal morbidity, with persistent neurological sequelae, associated with mitochondrial dysfunction and redox imbalance. The lack of experimental approaches for region-specific evaluation of mitochondrial bioenergetics in structurally preserved neural tissues has limited the identification of selective vulnerabilities and redox-based therapeutic targets. Here, we introduce a real-time metabolic profiling strategy that integrates Seahorse XFe96 respirometry with Cytation 5 image-based quality control to quantify mitochondrial respiration and glycol

CTRP10, also known as C1QL2, is a secreted protein of the C1q family. In the central nervous system, CTRP10 is predominantly expressed by neurons and oligodendrocytes. Changes in brain expression of CTRP10 are associated with addiction, depression, and psychiatric disorder. CTRP10 also serves as a specific molecular marker for the excitatory neurons in the anterior thalamic nuclei complex, as well as for neurons that control proprioception and fine motor skills. Whether CTRP10 is required for nervous system control of behaviors is unknown. Here, we determine whether and how CTRP10 deficiency a

INTRODUCTION: Clinically widely used acetazolamide is a potent carbonic anhydrase (CA) inhibitor. CA accelerates CO2(g)⇌HCO3 -(aq) bidirectional conversion. CAII, the major functional isoform, primarily resides inside red blood cells (RBCs). Erythrocyte-specific anion transporter or anion exchanger-1 (AE1) determines HCO3 - permeability across the cell membrane and is considered the rate-limiting factor for intraerythrocytic CO2(g)/HCO3(aq) conversion facilitated by CAII. This study aimed to use CA-inhibiting acetazolamide to find out whether AE1 could modulate intraerythrocytic CA catalysis.

Smith-Lemli-Opitz syndrome (SLOS) is a rare, autosomal recessive disorder characterized by congenital malformations, intellectual disability, and behavioral abnormalities. SLOS results from mutations in the DHCR7 gene, leading to impaired cholesterol biosynthesis due to dysregulation of 7-dehydrocholesterol reductase. Cholesterol plays crucial roles in neurophysiology, including synaptic formation and neurotransmitter receptor regulation, which likely contribute to neurological manifestations in SLOS patients. While astrocytes are the main cholesterol producing cells in the brain, their specif

Ischemic stroke is a leading cause of mortality and disability, with cerebellar strokes posing severe complications such as herniation and brainstem compression. Edaravone, a radical scavenger known for reducing oxidative stress, has shown neuroprotective effects in cerebral strokes, but its impact on cerebellar strokes remains unclear. This study investigates Edaravone's protective properties in organotypic slice cultures of rat cerebellum and hippocampus, employing an oxygen-glucose deprivation (OGD) model to simulate ischemic stroke. The hippocampus served as comparative structure due to it

The function and cell surface phenotype of lung macrophages vary within the respiratory tract. Alterations in the bioenergetic profile of macrophages may also be influenced by their location within the respiratory tract. This study sought to characterize the bioenergetic profile of macrophages sampled from different locations within the respiratory tract at baseline and in response to ex vivo xenobiotic challenge. Surface macrophages recovered from healthy volunteers by induced sputum and by bronchial and bronchoalveolar lavage were profiled using extracellular flux analyses. Oxygen consumptio

Eukaryotic translation initiation factor 2B (eIF2B) is a master regulator of protein synthesis under normal and stress conditions. Mutations in any of the five genes encoding its subunits lead to vanishing white matter (VWM) disease, a recessive genetic deadly illness caused by progressive loss of white matter in the brain. In this study we used fibroblasts, which are not involved in the disease, to demonstrate the involvement of eIF2B in mitochondrial function and abundance. Mass spectrometry of total proteome of mouse embryonic fibroblasts (MEFs) isolated from Eif2b5R132H/R132H mice revealed
