Treatment of pediatric pulmonary arterial hypertension: A focus on the NO-sGC-cGMP pathway.

OBJECTIVE: While pulmonary arterial hypertension (PAH) is rare in infants and children, it results in substantial morbidity and mortality. In recent years, prognosis has improved, coinciding with the introduction of new PAH-targeted therapies, although much of their use in children is off-label. Evidence to guide the treatment of children with PAH is less extensive than for adults. The goal of this review is to discuss the treatment recommendations for children with PAH, as well as the evidence supporting the use of prostanoids, endothelin receptor antagonists (ERAs), and phosphodiesterase typ

Ca2+ spikes in the flagellum control chemotactic behavior of sperm.

The events that occur during chemotaxis of sperm are only partly known. As an essential step toward determining the underlying mechanism, we have recorded Ca2+ dynamics in swimming sperm of marine invertebrates. Stimulation of the sea urchin Arbacia punctulata by the chemoattractant or by intracellular cGMP evokes Ca2+ spikes in the flagellum. A Ca2+ spike elicits a turn in the trajectory followed by a period of straight swimming ('turn-and-run'). The train of Ca2+ spikes gives rise to repetitive loop-like movements. When sperm swim in a concentration gradient of the attractant, the Ca2+ spike

cGAS-STING pathway in systemic lupus erythematosus: biological implications and therapeutic opportunities.

The cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) signaling pathway has been identified as a significant modulator of inflammation in various clinical contexts, including infection, cellular stress, and tissue injury. The extensive participation of the cGAS-STING pathway can be attributed to its ability to detect and control the cellular reaction to DNAs originating from both microorganisms and hosts. These DNAs are well recognized as molecules linked with potential risks. At physiological levels, the STING signaling system exhibits protective effects. However, prolonge…

[The neuro-ophthalmological complications in SLE (author's transl)].

In a 44 years old woman, a flare up of polyarthritis becoming positive for sero-reaction at the time of hospitalization, is referred to an earlier unknown SLE. The bilateral optic neuritis which began 15 years earlier, while she was pregnant, simultaneously with a bilateral carpal tunnel syndrome is retrospectively related to the SLE. Such an optic neuritis is rare, its clinical features are specified among the neuro-ophthalmological complications in SLE. Very often optic neuritis is associated with myelopathy. Hypercytosis with moderate elevation of albumin C.S.F., a high level of cyclic GMP,…

Docosahexaenoic acid reduces both cyclic nucleotide and melatonin synthesis in rat pinealocytes.

The effect of docosahexaenoic acid (22:6n-3) on melatonin (MT) production in rat pinealocytes was measured. In pinealocytes, supplementation with 50 microM 22:6n-3 for 48 h decreased MT production after a norepinephrine (NE)-stimulation (1 and 10 microM). Using co-treatment with IBMX (1 mM), a cyclic nucleotide phosphodiesterase inhibitor known to increase NE-stimulated cAMP and prazosin (1 microM), an alpha 1 antagonist, the results indicate that a) 22:6n-3 did not act on cyclic nucleotide phosphodiesterase activities to decrease both cAMP (-64%) and cGMP (-45%) levels and b) the inhibition o

Roles of cGMP-dependent protein kinase I (cGKI) and PDE5 in the regulation of Ang II-induced cardiac hypertrophy and fibrosis.

Conflicting results have been reported for the roles of cGMP and cGMP-dependent protein kinase I (cGKI) in various pathological conditions leading to cardiac hypertrophy and fibrosis. A cardioprotective effect of cGMP/cGKI has been reported in whole animals and isolated cardiomyocytes, but recent evidence from a mouse model expressing cGKIβ only in smooth muscle (βRM) but not in cardiomyocytes, endothelial cells, or fibroblasts has forced a reevaluation of the requirement for cGKI activity in the cardiomyocyte antihypertrophic effects of cGMP. In particular, βRM mice developed the same hypertr

Augmentation of cGMP/PKG pathway and colonic motility by hydrogen sulfide.

Hydrogen sulfide (H2S), like nitric oxide (NO), causes smooth muscle relaxation, but unlike NO, does not stimulate soluble guanylyl cyclase (sGC) activity and generate cyclic guanosine 5'-monophosphate (cGMP). The aim of this study was to investigate the interplay between NO and H2S in colonic smooth muscle. In colonic smooth muscle from rabbit, mouse, and human, l-cysteine, substrate of cystathionine-γ-lyase (CSE), or NaHS, an H2S donor, inhibited phosphodiesterase 5 (PDE5) activity and augmented the increase in cGMP levels, IP3 receptor phosphorylation at Ser1756 (measured as a proxy for PKG

The mechanism of action and role of hydrogen sulfide in the control of vascular tone.

Our knowledge about hydrogen sulfide (H2S) significantly changed over the last two decades. Today it is considered as not only a toxic gas but also as a gasotransmitter with diverse roles in different physiological and pathophysiological processes. H2S has pleiotropic effects and its possible mechanisms of action involve (1) a reversible protein sulfhydration which can alter the function of the modified proteins similar to nitrosylation or phosphorylation; (2) direct antioxidant effects and (3) interaction with metalloproteins. Its effects on the human cardiovascular system are especially impo

The vasodilatory effect of sulfur dioxide via SGC/cGMP/PKG pathway in association with sulfhydryl-dependent dimerization.

The present study was designed to explore the role of soluble guanylate cyclase (sGC)/cyclic guanosine monophosphate (cGMP)/PKG pathway in sulfur dioxide (SO2)-induced vasodilation. We showed that SO2 induced a concentration-dependent relaxation of phenylephrine (PE)-precontracted rat aortic rings in association with an increase in cGMP concentration, whereas l-aspartic acid β-hydroxamate (HDX), an inhibitor of SO2 synthase, contracted rings in a dose-dependent manner. Pretreatment of aortic rings with the sGC inhibitor ODQ (30 μM) attenuated the vasodilatory effects of SO2, suggesting the inv

Regulation of intracellular Ca2+ and calcineurin by NO/PKG in proliferation of vascular smooth muscle cells.

AIM: To determine whether Ca2+/calcineurin mediated the inhibitory effects of nitric oxide /cGMP-dependent protein kinase (NO/PKG) on the proliferation of vascular smooth muscle cells (VSMC). METHODS: Proliferation and viability of primary VSMC from rat aorta were measured using [3-(4,5-dimethyl thiazol-2-yl)-2,5-diphenyl tetrazolium bromide] (MTT) assay and acridine orange and ethidium bromide staining, respectively. Cytosolic Ca2+ was determined by Fluo-3/AM. Calcineurin protein and its activity were assayed using immunoblotting and free inorganic phosphate analysis, respectively. RESULTS:

Defects in cGMP-PKG pathway contribute to impaired NO-dependent responses in hepatic stellate cells upon activation.

NO antagonizes hepatic stellate cell (HSC) contraction, although activated HSC in cirrhosis demonstrate impaired responses to NO. Decreased NO responses in activated HSC and mechanisms by which NO affects activated HSC remain incompletely understood. In normal rat HSC, the NO donor diethylamine NONOate (DEAN) significantly increased cGMP production and reduced serum-induced contraction by 25%. The guanylate cyclase (sGC) inhibitor 1H-[1,2,4]oxadiazolo-[4,3-a]quinoxalin-1-one (ODQ) abolished 50% of DEAN effects, whereas the cGMP analog 8-bromoguanosine 3',5'-cyclic monophosphate (8-BrcGMP) reit

Opening of rod photoreceptor CNG channels by photodynamically generated singlet oxygen.

Molecular oxygen exists in three electronic states: the triplet ground state and two singlet (1O2) excited states. Under physiological conditions, 1O2 can be produced either through photodynamic processes, which require light, photosensitizer, and oxygen, or via metabolic reactions involving enzymes and other reactive oxygen species (ROS). 1O2 readily reacts with biomacromolecules, however, its volatile chemical nature and the lack of precise working models hamper the study of its molecular mechanism and physiological significance. Here we report that human CNG channels from rod photoreceptors

Multi-Compartment, Early Disruption of cGMP and cAMP Signalling in Cardiac Myocytes from the mdx Model of Duchenne Muscular Dystrophy.

Duchenne muscular dystrophy (DMD) is the most frequent and severe form of muscular dystrophy. The disease presents with progressive body-wide muscle deterioration and, with recent advances in respiratory care, cardiac involvement is an important cause of morbidity and mortality. DMD is caused by mutations in the dystrophin gene resulting in the absence of dystrophin and, consequently, disturbance of other proteins that form the dystrophin-associated protein complex (DAPC), including neuronal nitric oxide synthase (nNOS). The molecular mechanisms that link the absence of dystrophin with the alt

[Duodenal ulcer: clinical significance of adaptation defects in the system of neurohumoral regulation].

Radioimmunoassay was used to determine prostaglandin (PGE-2) and cyclic nucleotides (cAMP and cGMP) concentrations in the serum before, 1 and 2 hours after standard test breakfast (5066 kJ) in 65 patients with duodenal ulcer. Basal hyperproduction of prostaglandins and cyclic nucleotides were interpreted as an adaptation-compensatory mechanism. The unbalance of their aftermeal reactions persistent even in ulcer healing may belong to factors of the disease progression.

A cyclic AMP phosphodiesterase inhibitor, 8'-pivaloyloxymethyl ester (POM-ester) of griseolic acid, lowers rabbit intraocular pressure.

The effects of griseolic acid (GA), a cyclic-AMP phosphodiesterase (PDE) inhibitor, and its 8'-pivaloyloxymethyl (POM) ester on intraocular pressure (IOP) in rabbits were investigated. When 50 microliters of 1 and 2% GA POM ester solutions were topically applied to one eye in normal rabbits, significant IOP decreases were detected at 2 hrs and at 1 to 5 hrs, respectively. Other than ocular hypotension, no other ocular effects were detected locally even after administration of 2% GA POM ester. A more marked reduction in IOP occurred after the intravitreal injection of the GA POM ester. IOP was

[Drug-induced changes in chemotaxis under normal conditions and in rheumatoid arthritis].

The effects of agents used in RA treatment, various drugs, RF, rheumatoid nodule and synovial fluid was studied on chemotaxis of PMNs. NSAIDS, corticosteroids, theophylline, colchicine, SOD, RF, rheumatoid nodule and synovial fluid were found to inhibit the chemotactic responsiveness while AMPc, GMPc, PGEI and immunodulator drugs enhanced chemotaxis. The results support the hypothesis that drugs tested may modulate chemotactic function by affecting cellular microtubules assembly and/or GMPc accumulation.

Islet contents of cyclic 3',5'-guanosine monophosphate under conditions which affect the cyclic 3',5'-adenosine monophosphate.

The sensitivity of the radioimmunoassay for cGMP was considerably increased by previous 2'-O-succinylation of the nucleotide. The basal content of cGMP in beta-cell-rich pancreatic islets isolated from ob/ob-mice was similar to that of cAMP, i.e. about 3 mumoles per kg dry weight. Extra-cellular Ca2+ was a prerequisite for maintaining this amount of cGMP. The islet cGMP differed from cAMP in being only slightly enhanced or not affected at all when the islets were exposed to high concentrations of glucose, the sulphydryl reagents chloromercuribenzene-p-sulphonic acid and iodoacetamide, or the p

The DNA damage response and cancer immunotherapy.

The DNA damage response (DDR) and immune system serve as critical defence mechanisms, safeguarding genomic integrity and providing host protection, respectively. Emerging research reveals their interconnection in dictating the response to cancer immunotherapy. DDR-deficient tumours or those exposed to DDR inhibitors display genomic instability that generates two key immunogenic signals: neoantigens that activate tumour-specific T cells; and cytosolic DNA that stimulates the cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) pathway, triggering type I interferon production an…

The emerging cellular metal ions-immune axis in cancer.

Cancer immunotherapy is frequently hindered by the immunosuppressive tumor microenvironment. Although metal ions have been viewed as essential nutrients or cytotoxic payloads, emerging evidence reveals that they are potent immune modulators that orchestrate the cancer-immunity cycle. Here, we synthesize how ionic competition dictates immune cell plasticity and metabolic fate. We detail how metal ions regulate cell death pathways and immune signaling cascades, including the cyclic GMP-AMP synthase-stimulator of interferon genes and nuclear factor kappa B pathways, as well as immunological synap…

The cGAS-STING/MITA pathway in innate antiviral immunity and beyond.

Upon binding double-stranded DNA (dsDNA), the cyclic GMP-AMP (cGAMP) synthase (cGAS) catalyzes the synthesis of 2',3'-cGAMP, which subsequently binds to the endoplasmic reticulum (ER)-resident adaptor STING/MITA and triggers STING trafficking to the Golgi apparatus and recruits TANK-binding kinase 1 (TBK1) to induce type I interferons (IFNs) and inflammatory cytokines. Aberrant activation of this pathway drives severe autoinflammatory diseases, such as STING-associated vasculopathy with onset in infancy (SAVI) and Coatomer subunit alpha (COPA) syndrome. Furthermore, recent studies have reveale…
