Morphological variability and molecular identification of Uncinaria spp. (Nematoda: Ancylostomatidae) from grizzly and black bears: new species or phenotypic plasticity?

The hookworms Uncinaria rauschi Olsen, 1968 and Uncinaria yukonensis ( Wolfgang, 1956 ) were formally described from grizzly ( Ursus arctos horribilis) and black bears ( Ursus americanus ) of North America. We analyzed the intestinal tracts of 4 grizzly and 9 black bears from Alberta and British Columbia, Canada and isolated Uncinaria specimens with anatomical traits never previously documented. We applied morphological and molecular techniques to investigate the taxonomy and phylogeny of these Uncinaria parasites. The morphological analysis supported polymorphism at the vulvar region for fema

Molecular characterization of anisakid nematode larvae from 13 species of fish from Western Australia.

This study characterized anisakid nematodes in estuarine and near-shore species of fish in southern Western Australia. A total of 108 fish representing 13 species were examined for anisakid larvae. For the molecular characterization of anisakid larvae (n=218), we used PCR-coupled mutation scanning-sequencing-phylogenetic analyses of sequence variation in the internal transcribed spacers of nuclear ribosomal DNA. With the exception of Sillaginoides punctatus and Sillago schomburgkii, all the fish species examined (Aldrichetta forsteri, Arripis georgianus, Hyporhamphus regularis, Mugil cephalus,

Investigations of peritoneal and intestinal infections of adult hookworms (Uncinaria spp.) in northern fur seal (Callorhinus ursinus) and California sea lion (Zalophus californianus) pups on San Miguel Island, California (2003).

The peritoneal cavity (PNC) and intestine of northern fur seal (Callorhinus ursinus) pups and California sea lion (Zalophus californianus) pups that died in late July and early August, 2003, on San Miguel Island, California, were examined for hookworms. Prevalence and morphometric studies were done with the hookworms in addition to molecular characterization. Based on this and previous molecular studies, hookworms from fur seals are designated as Uncinaria lucasi and the species from sea lions as Uncinaria species A. Adult hookworms were found in the PNC of 35 of 57 (61.4%) fur seal pups and o

ITS1 intra-individual variability of Ascaris isolates from Brazil.

The zoonotic potential of Ascaris infecting pigs has stimulated studies of molecular epidemiology with internal transcribed spacer 1 (ITS1) as the target. The aim of this study was to determine the value of Ascaris ITS1 as a molecular marker through assessing the intra-individual genetic diversity of Ascaris isolates from two geographical areas of Brazil. DNA was extracted from single isolated eggs, ITS1 PCR was performed, and the PCR products were cloned and sequenced. Clone analysis showed high ITS1 intra-individual variability revealed by 2-4 ITS1 genotypes/haplotypes per sample (egg). Two

Two new blood flukes (Digenea: Sanguinicolidae) from Epinephelinae (Perciformes: Serranidae) of the Pacific Ocean.

Pearsonellum pygmaeus n. sp. is described from Cromileptes altivelis (Serranidae), the Barramundi Cod, from Heron Island (southern Great Barrier Reef) and Lizard Island (northern Great Barrier Reef). This new species differs from Pearsonellum corventum (type and only species) in the combination of smaller overall body size, the relative distance of the brain from the anterior end, the relative lengths of both the oesophagus and the testis, the degree to which the testis extends outside the intercaecal field, the shape of the testis, the shape and size of the ovary and the extent to which the u

CK2 phosphorylation of Schistosoma mansoni HMGB1 protein regulates its cellular traffic and secretion but not its DNA transactions.

BACKGROUND: The helminth Schistosoma mansoni parasite resides in mesenteric veins where fecundated female worms lay hundred of eggs daily. Some of the egg antigens are trapped in the liver and induce a vigorous granulomatous response. High Mobility Group Box 1 (HMGB1), a nuclear factor, can also be secreted and act as a cytokine. Schistosome HMGB1 (SmHMGB1) is secreted by the eggs and stimulate the production of key cytokines involved in the pathology of schistosomiasis. Thus, understanding the mechanism of SmHMGB1 release becomes mandatory. Here, we addressed the question of how the nuclear S

Endoparasite survey of free-swimming baleen whales (Balaenoptera musculus, B. physalus, B. borealis) and sperm whales (Physeter macrocephalus) using non/minimally invasive methods.

A number of parasitic diseases have gained importance as neozoan opportunistic infections in the marine environment. Here, we report on the gastrointestinal endoparasite fauna of three baleen whale species and one toothed whale: blue (Balaenoptera musculus), fin (Balaenoptera physalus), and sei whales (Balaenoptera borealis) and sperm whales (Physeter macrocephalus) from the Azores Islands, Portugal. In total, 17 individual whale fecal samples [n = 10 (B. physalus); n = 4 (P. macrocephalus); n = 2 (B. musculus); n = 1 (B. borealis)] were collected from free-swimming animals as part of ongoing

A recombinant protein from Schistosoma mansoni useful for the detection of S. mansoni and Schistosoma haematobium antibodies.

A recombinant Schistosoma mansoni protein has been identified as a useful antigen for the detection of S. mansoni and Schistosoma haematobium antibodies. The purified recombinant protein, Sm22.3, was assayed using an enzyme-linked immunosorbent assay format against a battery of 491 well defined sera, including S. mansoni, S. haematobium, and Schistosoma japonicum infection sera, normal human sera, sera from 9 other parasitic infections, and sera from 2 additional infections. The sensitivity for detecting S. mansoni and S. haematobium infections with this single recombinant protein is 80.1%. Th

Genetic and molecular analysis of spe-27, a gene required for spermiogenesis in Caenorhabditis elegans hermaphrodites.

Hermaphrodites with mutations in the spe-27 gene are self-sterile, laying only unfertilized eggs; mutant males are fertile. Hermaphrodites make spermatids that fail to activate to crawling spermatozoa so passing oocytes sweep them out of the spermatheca. These spermatids do activate and produce self-progeny if young mutant hermaphrodites are mated by fertile (or sterile) males. Spermatids isolated from either mutant males or hermaphrodites initiate activation in vitro when treated with proteases, but then arrest with spiky membrane projections that resemble those of a normal intermediate in ps

Morphology and molecular characterisation of two new species of Mymarothecioides (Monopisthocotyla: Dactylogyridae) parasitising the gills of anostomid fishes from Brazil.

Using an integrative taxonomic approach, we describe two new species of Monopisthocotyla, Mymarothecioides leporini n. sp. and Mymarothecioides paranaensis n. sp., parasitising the gills of anostomid fishes from the Jaguaribe River and Paraná River basins, Brazil. These descriptions represent the first record of species of Mymarothecioides in hosts of the family Anostomidae. Mymarothecioides leporini n. sp. is distinguished by a male copulatory organ (MCO) with a spoon-shaped distal portion and a ventral bar with an ornamented anteromedial process. Mymarothecioides paranaensis n. sp. differs f

Tapeworm bolus expelled from New Zealand variable oystercatchers (Haematopus unicolor) during handling: first record of this phenomenon in wild birds, and a global checklist of Haematopus cestode parasites.

On two occasions in November and December 2009, whilst being captured and handled for banding in Tasman Bay, New Zealand, two variable oystercatchers (Haematopus unicolor Forster, 1844) voided a number of cestode strobilae from their cloaca. Their morphology indicates that they belong to the family Hymenolepididae, confirmed by BLASTn searches of large and small subunits of ribosomal DNA partial sequences (18S and 28S, respectively). However, they cannot currently be assigned to any species reported for any oystercatcher species from New Zealand nor from oystercatchers worldwide. We present a

Redescription and resurrection of Bertiella satyri (Cestoda, Anoplocephalidae) parasitizing the orangutan (Pongo abelii) in Indonesia.

The tapeworm species Bertiella satyri from a semi-wild Sumatran orangutan (Pongo abelii: Ponginae) is redescribed and the sequence of its 18S rDNA is presented. The tapeworms parasitizing the genera Pan, Pongo, Homo and Hylobates from Muséum National d'Histoire Naturelle, Paris and from Muséum d´Histoire Naturelle, Genève are also presented. The validity of B. satyri is confirmed. B. satyri (BSA) differs from the most similar species Bertiella studeri (BSTU) in the following characteristics: (1) testes number, BSTU 300-400; BSA 116-124, (2) genital opening, BSTU regularly alternate; BSA irregu

Comparison of conventional techniques to differentiate between Taenia solium and Taenia saginata and an improved polymerase chain reaction-restriction fragment length polymorphism assay using a mitochondrial 12S rDNA fragment.

Given the constraints of classical diagnostic methods, i.e., morphological and isoenzymatic studies of proglottids, a polymerase chain reaction test complemented with restriction enzyme analysis has been modified by redesigning one of the primers to reduce nonspecific amplifications experienced when using field samples. The use of these new, highly cestode-specific primers and the restriction enzyme Ddel led to the development of a diagnostic assay that clearly distinguishes between Taenia saginata and T. solium proglottids in field samples. This assay confirms the presence of T. saginata in E

Accession-level reassessment of cox1 identity in the first Korean case of Echinococcus multilocularis.

The first Korean case of Echinococcus multilocularis interpreted the probable origin of the isolate on the basis of cox1 sequence similarity, originally reporting 99.8%-99.9% similarity to Asian isolates and 99.5%-99.6% similarity to European isolates. To examine whether this similarity-based interpretation remains epidemiologically informative, we re-evaluated currently available cox1 sequences with high sequence identity at the accession level. Using AB780998.1 as the query, BLAST hits with ≥99.5% identity were collected, and 110 non-self accessions were manually annotated according to clade

First confirmation of the chicken eyeworm, Oxyspirura mansoni, as a causative pathogen of human cutaneous larva migrans by morphological and molecular evidence.

Human larva migrans is a zoonotic disease caused by larvae of various nematode species, with all previously confirmed pathogens originating from mammalian reservoirs. In 2020, a case series in Vietnam reported instances of cutaneous larva migrans caused by avian eyeworm larvae of the Oxyspirura genus. This nematode genus, belonging to the Thelaziidae family, includes 84 species that mainly parasitize the eyes of various bird species. They utilize different arthropod intermediate hosts, such as cockroaches, grasshoppers, and crickets, in their life cycle. Although the 18S sequences of the larva

A QUARTER CENTURY OF SAMPLING TWO OCEAN BASINS TO DISAMBIGUATE THE IDENTITY OF A MULLET PARASITE, SACCOCOELIOIDES BEAUFORTI (DIGENEA: HAPLOPORIDAE).

Saccocoelioides beauforti (Hunter and Thomas, 1961) Overstreet, 1971, an intestinal parasite of flathead grey mullet, Mugil cephalus Linnaeus, was originally described from off North Carolina (northwestern Atlantic Ocean) and purportedly ranges south and west to Louisiana (north-central Gulf of America). We surveyed the parasites of hundreds of flathead grey mullet captured from coastal habitats of Georgia, Alabama, and Mississippi over a 25 yr period, from 1998 through 2023. Examination of whole-mounted specimens of "S. beaufort" revealed substantial morphological variation between specimens

Molecular detection of filarioid nematodes (Nematoda: Onchocercidae) in wild mammals from different Brazilian biomes.

Parasitic nematodes within Onchocercidae are a diverse group transmitted by hematophagous arthropods. This study investigated the molecular occurrence of filarioid nematodes in 93 wild mammals from the Amazon, Cerrado and Pantanal biomes in Brazil, based on the analysis of the mitochondrial genes 12S ribosomal DNA gene (12S rDNA) and cytochrome c oxidase subunit I gene (COI). Conventional polymerase chain reaction (cPCR) targeting the 12S rDNA gene yielded positive results in 14·44% (13/93) of the samples, including 9·86% of jaguars (7/71), 50% of pumas (1/2), 12·5% of giant anteaters (1/8), 5
