Harnessing transcriptomics for discovery of natural products to overcome acquired cancer resistance.

Targeted cancer therapy is often compromised by the development of acquired drug resistance. Beyond genetic mutations, recent studies underscore the role of non-genetic plasticity and adaptive network rewiring in driving this resistance. Overcoming this challenge requires innovative approaches, including the integration of transcriptomics and natural product research. Natural products are chemically diverse agents that can modulate multiple resistance pathways due to their polypharmacological properties. In parallel, transcriptomic profiling of drug-exposed cells provides genome-wide snapshots

Breaking barriers in antimicrobial therapy: resistance mechanisms and novel antimicrobial strategies.

INTRODUCTION: Modern healthcare systems face significant challenges from antimicrobial resistance (AMR), which threatens our ability to effectively manage infectious diseases. The widespread prevalence of AMR and its constantly evolving patterns define its epidemiological landscape, with multidrug-resistant organisms emerging at an alarming rate. METHODS: A thorough examination of the literature was conducted using multiple databases, including Google Scholar, PubMed, Science Direct, and Springer Link. The search included keywords such as "antimicrobial resistance," "causes and mechanisms of

Antibacterial activity of naphthyl derived bis-(3-hydroxy-4-pyridinonate) copper(II) complexes against multidrug-resistant bacteria.

In this work, we report the synthesis and characterization of three novel copper(II) complexes of naphthyl derived 3-hydroxy-4-pyridinone chelators. Their antibacterial activity against several Gram-positive and Gram-negative reference strains and multidrug-resistant clinical isolates was assessed through determination of the minimum inhibitory concentration (MIC). The complex Cu(naph1pp)2 shows the highest antibacterial activity, including against multidrug-resistant isolates, nonetheless, being more active against Gram-positive than Gram-negative bacteria. Cu(naph1pp)2 was further explored i

[Nosocomial infection caused by multidrug-resistant Staphylococcus aureus with reduced susceptibility to vancomycin and teicoplanin.--Yamagata Prefecture, Japan; May 2004-Jun 2005].

Nosocomial infection caused by methicillin-resistant Staphylococcus aureus (MRSA) occurred at a major in Yamagata prefecture hospital between May 2004 and June 2005. We studied pulsed-field gel electrophoresis patterns, antimicrobial susceptibility patterns, and bacteriological features, such as coaglase type for eight isolates, including two of methicillin-resistant Staphylococcus epidermidis (MRSE). Our results suggest that this case was caused by a single strain of multidrug-resistant S. aureus. These 8 clinical isolates indicated reduced susceptibility to vancomycin and teicoplanin. PCR as

Managing anthelmintic resistance--parasite fitness, drug use strategy and the potential for reversion towards susceptibility.

The rotation of different anthelmintic classes, on an approximately annual basis, has been widely promoted and adopted as a strategy to delay the development of anthelmintic resistance in nematode parasites. Part of the rationale for recommending this practice was the expectation that resistant genotype worms have a lower ecological fitness than susceptible worms, at least in the early stages of selection, and so reversion towards susceptibility could be expected in those years when an alternative class of anthelmintic was used. The routine use of combination anthelmintics might be expected to

Unpacking 'Artemisinin Resistance'.

Artemisinin and its derivatives, in combination with partner drugs, are currently the most effective treatments for malaria parasite infection. Even though artemisinin has been widely used for decades, its mechanism of action had remained controversial until recently. Artemisinin combination therapies (ACTs) have recently been found to be losing efficacy in Southeast Asia. This 'artemisinin resistance', defined by a delayed parasite clearance time, has been associated with several genetic mutations. As with any other drug resistance phenotype, resistance can best be understood based on its mec

Elucidating drug resistance in human fungal pathogens.

Fungal pathogens cause life-threatening infections in immunocompetent and immunocompromised individuals. Millions of people die each year due to fungal infections, comparable to the mortality attributable to tuberculosis or malaria. The three most prevalent fungal pathogens are Candida albicans, Cryptococcus neoformans and Aspergillus fumigatus. Fungi are eukaryotes like their human host, making it challenging to identify fungal-specific therapeutics. There is a limited repertoire of antifungals in clinical use, and drug resistance and host toxicity compromise the clinical utility. The three c

Suppressive drug combinations and their potential to combat antibiotic resistance.

Antibiotic effectiveness often changes when two or more such drugs are administered simultaneously and unearthing antibiotic combinations with enhanced efficacy (synergy) has been a longstanding clinical goal. However, antibiotic resistance, which undermines individual drugs, threatens such combined treatments. Remarkably, it has emerged that antibiotic combinations whose combined effect is lower than that of at least one of the individual drugs can slow or even reverse the evolution of resistance. We synthesize and review studies of such so-called 'suppressive interactions' in the literature.

[Combination therapy against multidrug-resistant bacteria].

The purpose of antibiotic combination therapy includes two points, as follows; 1) expectation of synergistic effects, 2) preventing the emergence of drug-resistant bacteria following exposure to antibiotics. Synergism has been shown in vitro for many antibiotic combinations and bacterial species, but it is uncertain whether synergistic effects in vitro are applicable to the clinical setting and lead to a better prognosis. beta-lactam and aminoglycoside combinations have synergistic effects against Gram-negative bacteria, including Pseudomonas aeruginosa, Klebsiella, Escherichia coli, and other
