Impaired neutrophil extracellular trap formation: a novel defect in the innate immune system of aged individuals.

Neutrophil extracellular traps (NETs) are a recently discovered addition to the defensive armamentarium of neutrophils, assisting in the immune response against rapidly dividing bacteria. Although older adults are more susceptible to such infections, no study has examined whether aging in humans influences NET formation. We report that TNF-α-primed neutrophils generate significantly more NETs than unprimed neutrophils and that lipopolysaccharide (LPS)- and interleukin-8 (IL-8)-induced NET formation exhibits a significant age-related decline. NET formation requires generation of reactive oxygen…

Hydroxyethyl starch 130/0.4 decreases inflammation, neutrophil recruitment, and neutrophil extracellular trap formation.

BACKGROUND: During systemic inflammation, leucocytes are activated and extravasate into damaged tissue. Activation and recruitment are influenced by different mechanisms, including the interaction of leucocytes with platelets and neutrophil extracellular traps (NET) formation. Here, we investigated the molecular mechanism by which hydroxyethyl starch (HES 130/0.4) dampens systemic inflammation in vivo. METHODS: Systemic inflammation was induced in C57Bl/6 wild-type mice by caecal ligation and puncture and cytokine concentrations in the blood, neutrophil recruitment, platelet-neutrophil aggreg…

Impaired neutrophil extracellular trap (NET) formation: a novel innate immune deficiency of human neonates.

Neutrophils are highly specialized innate effector cells that have evolved for killing of pathogens. Human neonates have a common multifactorial syndrome of neutrophil dysfunction that is incompletely characterized and contributes to sepsis and other severe infectious complications. We identified a novel defect in the antibacterial defenses of neonates: inability to form neutrophil extracellular traps (NETs). NETs are lattices of extracellular DNA, chromatin, and antibacterial proteins that mediate extracellular killing of microorganisms and are thought to form via a unique death pathway signa…

Lipid asymmetry disruption by XKR8 orchestrates neutrophil extracellular trap formation and inhibits fungal infection.

Neutrophil extracellular traps (NETs) constitute a vital antimicrobial defense mechanism of neutrophils, contributing to various physio-pathological processes; however, the role of plasma membrane asymmetry in this process remains unknown. Here we identify Xk-related protein 8 (XKR8), a plasma membrane phospholipid scramblase, as a pivotal regulator of NETs formation. Upon NETs induction, XKR8 is cleaved by caspase-3, thereby disrupting plasma membrane lipid asymmetry via phospholipid scrambling. Mutation of the caspase-3 cleavage site in XKR8 impairs NET formation. Inhibition of calcium signa…

Human serum albumin modified in myeloperoxidase-dependent reactions is a mediator of neutrophil extracellular trap formation.

Activation of neutrophil membrane receptors initiates intracellular signal transduction cascades that orchestrate the cell's effector functions, including phagocytosis, production of reactive oxygen and halogen species, degranulation, and NETosis (formation of neutrophil extracellular traps [NETs]). NETs, which contain antimicrobial compounds such as myeloperoxidase (MPO), represent a strategy to combat infection. However, excessive production of NETs promotes thrombosis, diabetes mellitus, and other diseases. Therefore, investigations into the mechanisms of NETosis and the identification of m…

Scavenging of Isolevuglandins Attenuates Neutrophil Migration and Neutrophil Extracellular Trap Formation in Systemic Lupus Erythematosus.

OBJECTIVE: Systemic lupus erythematosus (SLE) is an autoimmune disease that affects numerous organs. Neutrophil extracellular traps (NETs) contribute to sterile inflammation and autoantibody generation in SLE. Isolevuglandins (isoLGs) are reactive oxygen species that are formed during NETosis and contribute to chromatin expansion. Scavenging of isoLGs attenuates SLE disease activity and hypertension in murine models of SLE. We hypothesized that isoLGs drive NETosis in SLE. METHODS: Neutrophils were isolated from patients with SLE (n = 6), treated with the isoLG scavenger ethyl-2-hydroxybenzyl…

Unraveling the Mechanisms of Neutrophil Extracellular Trap Formation and the Role of Reactive Oxygen Species in Myocardial Bridging.

Myocardial bridging (MB) is a congenital coronary anomaly in which a segment of a coronary artery courses through the myocardium, resulting in systolic compression and alterations in coronary vasoreactivity. Consequently, morphological, functional, and hemodynamic changes may occur, with various clinical implications. Although traditionally considered a benign anatomical variant, accumulating evidence suggests an association between MB and endothelial dysfunction, oxidative stress, local inflammatory activation, and atherosclerotic plaque formation, predominantly in the arterial segment proxim

Attenuation of Neutrophil Extracellular Trap Formation in Lipopolysaccharide-Induced Acute Lung Injury Associated with Cinnamaldehyde.

BACKGROUND: Excessive neutrophil activation and neutrophil extracellular trap (NET) formation play critical roles in the pathogenesis of acute lung injury (ALI). Cinnamaldehyde, a major bioactive component of cinnamon, has been reported to exert anti-inflammatory effects; however, its impact on NET formation in ALI remains unclear. METHODS: A lipopolysaccharide (LPS)-induced ALI rat model and a PMA-stimulated primary neutrophil model were employed to evaluate the effects of cinnamaldehyde in vivo and in vitro. Lung injury severity, pulmonary edema, inflammatory cell infiltration, cytokine pro

Deoxynivalenol induces macrophage extracellular trap formation via the PER2-c-Myc-ROS Axis.

Deoxynivalenol (DON), a Fusarium-derived mycotoxin, disrupts immune homeostasis and induces immunotoxicity. Macrophage extracellular traps (METs) are key effectors in innate immunity and inflammation, yet whether DON triggers METs formation and the underlying mechanisms remain unclear. This study aims to investigate the molecular mechanism by which DON induces METs generation through circadian rhythm immune checkpoints involving the PER2-c-Myc axis. Using RAW264.7 and THP-1 macrophage models, we combined siRNA-mediated Per2 knockdown, and pharmacological inhibitors to dissect the mechanisms of…

Extracellular Traps at the Crossroads of Immune Evasion and Antimicrobial Resistance: The Case of Escherichia coli in the Reproductive Tract.

Extraintestinal pathogenic Escherichia coli (ExPEC) is among the bacterial pathogens associated with infections of the female and male reproductive tracts, where it may contribute to inflammatory processes and impaired reproductive function. In this context, neutrophils constitute a critical component of host defense through the release of neutrophil extracellular traps (NETs), web-like structures composed of decondensed chromatin, histones, and antimicrobial proteins that capture and eliminate invading microorganisms. However, the interaction between E. coli and NETs is highly dynamic, as the…

Quantification of H3.1-nucleosomes using a chemiluminescent immunoassay: A reliable method for neutrophil extracellular trap detection.

Neutrophil extracellular traps (NETs) are chromatin-based web-like structures released by activated neutrophils in response to infectious agents. Overproduction or insufficient clearance of NETs contributes to dysfunction of immune response and disease pathogenesis, underlying the importance of early detection and monitoring of NET levels in clinical samples. While existing methods for NETs detection and quantification face limitations, there is a pressing need for a reliable, sensitive, and clinically applicable assay. Since NETs consist of long strains of decondensed chromatin, with nucleoso…

The Role of Neutrophil Extracellular Traps in Hepatocellular Carcinoma. What Are the Implications of Anesthetic Techniques? A Narrative Review.

Neutrophil extracellular traps (NETs)-webs of DNA and granular proteins expelled by neutrophils-have been implicated in hepatocellular carcinoma (HCC) progression. NETs promote tumor angiogenesis, facilitate invasion/metastasis, and enable immune evasion. Recent data suggest that perioperative factors, including anesthetic techniques, may modulate NET formation (NETosis), thus potentially influencing oncologic outcomes. We conducted a literature review of experimental and clinical studies on NETosis pathophysiology and involvement in HCC and how anesthetic techniques may modulate NET formation…

Neutrophil extracellular trap-borne C3-driven endothelial dysfunction in Klebsiella pneumoniae liver abscess.

Klebsiella pneumoniae (K. pneumoniae) liver abscess (KPLA) is a severe bacterial infection that is frequently complicated by intrahepatic thrombophlebitis and extrahepatic metastatic infections leading to high mortality rates. This study investigates the role of neutrophil extracellular traps (NETs) in endothelial injury and disease progression in KPLA. Our findings reveal that KPLA patients with intrahepatic thrombophlebitis are more prone to developing sepsis and extrahepatic migratory infections. K. pneumoniae induces NET formation via the TLR4-PI3Kα-AKT signaling pathway, and C3 deposition…

The effect of chemical fixation with paraformaldehyde, glutardialdehyde or methanol on immunofluorescence staining of neutrophils and neutrophil extracellular traps.

The formation of neutrophil extracellular traps (NETs) is known as an important part of the innate immune response. Still, some mechanisms regarding their formation and role during a disease are not completely understood yet. To visualize NETs by immunofluorescence microscopy, a chemical fixation is required. Therefore, this study focused on the effect of chemical fixatives on immunofluorescence staining of selected neutrophil and NET-markers, including myeloperoxidase (MPO), DNA/histone-1-complexes and citrullinated histone H3 (H3cit). Neutrophils isolated from fresh human blood were stimulat…

Aberrant formation of the neutrophil extracellular trap and the expression of the PLEKHA1 in systemic lupus erythematosus and ulcerative colitis.

Systemic lupus erythematosus (SLE) and ulcerative colitis (UC) are both chronic autoimmune diseases with unclear shared mechanisms, largely due to limited mechanistic studies and clinical research cohorts. Transcriptome datasets from the Gene Expression Omnibus (GEO) database were analyzed for SLE and UC, identifying differentially expressed genes (DEGs). Weighted gene co-expression network analysis (WGCNA) identified significant module genes, including PLEKHA1. The diagnostic potential of PLEKHA1 was confirmed using machine-learning algorithms and real-time fluorescence quantitative PCR (RT-P

Sinomenine alleviates gouty inflammation by inhibiting macrophage M1 polarization and neutrophil extracellular trap formation.

Gout is a common inflammatory arthropathy characterized by the deposition of monosodium urate (MSU) crystals, leading to severe pain and swelling. Sinomenine (SIN) is the major active component of Sinomenium acutum. SIN has been demonstrated to exert preventive and therapeutic effects on arthritis in cell-based, animal, and clinical studies. The present study focused on the efficacy and role of SIN in relieving symptoms of gouty inflammation in vivo and in vitro. The anti-inflammatory effects of SIN were evaluated in mice with MSU-induced air-pouch via hematoxylin-eosin (HE) staining, and enzy

Inhalable Artificial Polymeric Nucleases Degrading Neutrophil Extracellular Trap-DNAs and Alleviating Pulmonary Fibrosis.

Pulmonary fibrosis resulting from recurrent lung inflammation due to pathogen infection may lead to serious problems and death. Neutrophil extracellular traps (NETs), consisting of DNAs and proteins released by neutrophils in response to infection, are major pathogenesis factors for pathogen-associated pulmonary fibrosis. By mimicking nucleic acid hydrolase, polymeric artificial DNases bearing imidazole units (PEG-PIm) are developed to degrade the DNAs and thus deconstruct NETs, inhibiting pulmonary fibrosis. By tailoring the PIm segments with varied imidazole units, the polymer hydrolase with…
