Genetic, Immunological, Dietary, Gut Microbiota, and Environmental Determinants of Osteoporosis in the Course of Celiac Disease: Which Factor Plays the First Violin in This Orchestra?

Celiac disease (CD) is a chronic small intestinal immune-mediated enteropathy precipitated by exposure to dietary gluten in genetically predisposed individuals. The worldwide prevalence of CD is estimated to be 0.7-1.4% of the general population. Etiopathology of this disease is multifactorial, with genetic determinants being a major contributing player to CD susceptibility. Its manifestation embraces different organs, including the musculoskeletal apparat. Patients with CD have increased risk of bone disorders. According to data, bone disorders - osteopenia and osteoporosis - can affect up to

Genetic characterisation and phylogenetic status of whipworms (Trichuris spp.) from captive non-human primates in China, determined by nuclear and mitochondrial sequencing.

BACKGROUND: Whipworms (Nematoda: Trichuridae), among the most common soil-transmitted helminths (STHs), can cause the socioeconomically important disease trichuriasis in various mammalian hosts including humans and non-human primates. For many years, Trichuris from non-human primates has been assigned to the same species as the one infecting humans Trichuris trichiura. More recently, several molecular reports challenged this assumption following recognition of a Trichuris species complex observed in humans and non-human primates. A refined concept for species limits within Trichuris contribute

Incidence and carrier frequency of Sandhoff disease in Saskatchewan determined using a novel substrate with detection by tandem mass spectrometry and molecular genetic analysis.

Sandhoff disease is a rare progressive neurodegenerative genetic disorder with a high incidence among certain isolated communities and ethnic groups around the world. Previous reports have shown a high occurrence of Sandhoff disease in northern Saskatchewan. Newborn screening cards from northern Saskatchewan were retrospectively screened in order to investigate the incidence and determine the carrier frequency of Sandhoff disease in these communities. PCR-based screening was conducted for the c.115delG (p.(Val39fs)) variant in the HEXB gene that was previously found in 4 Sandhoff disease patie

A genetic interaction network of five genes for human polycystic kidney and liver diseases defines polycystin-1 as the central determinant of cyst formation.

Autosomal dominant polycystic liver disease results from mutations in PRKCSH or SEC63. The respective gene products, glucosidase IIβ and SEC63p, function in protein translocation and quality control pathways in the endoplasmic reticulum. Here we show that glucosidase IIβ and Sec63p are required in mice for adequate expression of a functional complex of the polycystic kidney disease gene products, polycystin-1 and polycystin-2. We find that polycystin-1 is the rate-limiting component of this complex and that there is a dose-response relationship between cystic dilation and levels of functional

Genetic diversity of Trichomonas vaginalis clinical isolates determined by EcoRI restriction fragment length polymorphism of heat-shock protein 70 genes.

Restriction fragment length polymorphism (RFLP) analysis using a multilocus heat-inducible cytoplasmic heat-shock protein 70 (Hsp70) hybridization probe with EcoRI-digested genomic DNA was used in molecular typing of 129 Trichomonas vaginalis isolates. Results indicate that Trichomonas organisms exhibit considerable polymorphism in their Hsp70 RFLP patterns. Analysis of seven American Type Culture Collection reference strains and 122 clinical isolates, including 84 isolates from Jackson, Mississippi, 18 isolates from Atlanta, Georgia, and 20 isolates from throughout the United States, showed 1

Genetic alterations determine chemotherapy resistance in childhood T-ALL: modelling in stage-specific cell lines and correlation with diagnostic patient samples.

Acquired drug resistance eventually leads to treatment failure in T-cell acute lymphoblastic leukaemia (T-ALL). Immunophenotypic and cytogenetic heterogeneities within T-ALL influence susceptibility to cytotoxic therapy, and little is known about the mechanisms of drug resistance at specific stages of T-cell ontogeny. We developed tolerance to therapeutic concentrations of daunorubicin (DNR) and L-asparaginase (L-asp) in Jurkat (CD1a(-), sCD3(+)) and Sup T1 (CD1a(+), sCD3(-)) cell lines, having respective 'mature' and 'cortical' stages of developmental arrest. DNR resistant cells acquired mult

Evaluation of nutritional and genetic determinants of total homocysteine, methylmalonic acid and S-adenosylmethionine/S-adenosylhomocysteine values in Brazilian childbearing-age women.

BACKGROUND: Cobalamin (Cbl) and folate deficiencies and gene polymorphism of key enzymes or carriers can impair homocysteine metabolism and may change the serum values of S-adenosylmethionine (SAM) and S-adenosylhomocysteine (SAH). We investigated the nutritional and genetic determinants for total homocysteine (tHcy), methylmalonic acid (MMA) and SAM/SAH in healthy Brazilian childbearing-age women. METHODS: Serum concentrations of Cbl, folate, red blood cell folate, ferritin, tHcy, MMA, SAM, SAH and other metabolites were measured in 102 healthy unrelated women. The genotypes for MTHFR C677T,

Genetic variants of APOE and CCR4 as determinants of myocardial infarction among type 2 diabetes patients: a South Indian case-control analysis.

BACKGROUND: Myocardial infarction (MI) is a leading cause of mortality worldwide, and Type 2 Diabetes Mellitus (T2DM) markedly increases cardiovascular risk. Genetic variation in lipid metabolism pathways, such as APOE, and in immune regulatory genes, such as CCR4, may contribute to myocardial infarction susceptibility among individuals with T2DM. OBJECTIVE: This study aimed to evaluate the individual and combined effects of APOE (rs429358 and rs7412) and CCR4 (rs2228428) polymorphisms on MI risk in a South Indian population. METHODS: In this prospective case–control study, 400 participants we

Risk stacking in peripheral artery disease (PAD): Integrating genetics, cardiometabolic disease and social determinants of health to explain worsening PAD outcomes.

Peripheral artery disease (PAD) contributes disproportionately to disability, limb loss, and premature mortality, and U.S. major amputation rates have risen in the past decade among increasingly younger adults. Traditional risk tools incompletely explain these trends. We propose to examine a hypothesis generating risk-stacked model in which three interacting layers: 1) genetic susceptibility via lipoprotein(a) [Lp(a)]; 2) cardiovascular-kidney-metabolic syndrome (CKM; chronic kidney disease/diabetes mellitus/insulin resistance); and 3) social determinants of health (SDOH) jointly influence PAD

Antimicrobial resistance in Staphylococcus pseudintermedius isolated from asymptomatic and symptomatic dogs in Montevideo, Uruguay: characterization of MRSP strains and genetic determinants of resistance.

Staphylococcus pseudintermedius is a common opportunistic pathogen in dogs and an increasing concern in veterinary medicine due to rising antimicrobial resistance, particularly to methicillin. This study aimed to characterize resistance profiles and genetic mechanisms in isolates from healthy and diseased dogs in Montevideo, Uruguay. A total of 133 isolates was analyzed (83 from clinical infections and 50 from asymptomatic carriers). Antimicrobial susceptibility was assessed by disk diffusion following veterinary guidelines. Resistance genes and SCCmec types were detected by PCR. Ten represent

Discovery of XY Sex Chromosomes in Mauremys mutica Provides Insights Into the Role of KDM6B Gene in Coexistence of Temperature-Dependent and Genetic Sex Determination.

The Asian yellow pond turtle (Mauremys mutica) has long been thought to lack dimorphic sex chromosomes, with prevailing theories suggesting a solely temperature-dependent sex determination (TSD) system. In this study, a male chromosome-level genomic sequence with a contig N50 of ~23.59 Mb was generated using a combination of both Nanopore and Hi-C sequencing technologies. We utilise a combination of bioinformatics and cytogenetic experimental validation to demonstrate that this species indeed possesses XY chromosomes, thereby correcting a longstanding misconception. The results suggest that th

Dissecting the genetic determinants and biological associations between body mass index and female reproductive disorders based on genome-wide association study.

BACKGROUND: While the phenotypic link between body mass index (BMI) and some female reproductive disorders is well established, the genetic architecture and causal relationships have not been systematically studied. We aimed to create an atlas of the shared genetic associations of BMI and 16 female reproductive disorders and to identify their common risk loci, biological pathways, and potential mechanisms. METHODS: We assessed the genetic correlations between BMI and 16 reproductive disorders using summary data from large-scale genome-wide association studies. Cross-trait pleiotropic analysis

Genetic diversity and population structures of Plasmodium falciparum parasites imported to China from Central and West Africa determined using a SNP barcode.

BACKGROUND: Malaria remains a highly threatening infectious disease, with Africa being the main epidemic area. China has achieved the notable feat of eliminating malaria domestically, yet the looming threat of imported malaria cases remains a concern. This study aims to apply the SNP barcode technology to investigate the population structure of P. falciparum imported to China from Central and West Africa. METHODS: We applied a 24-SNP high-resolution melting (HRM) barcode to analyze the diversity of P. falciparum populations imported to China from Central and West Africa. RESULTS: A total of

An Effort to Identify Genetic Determinants in Siblings With Wilson Disease Manifesting Striking Clinical Heterogeneity: An Exome Profiling Study of Two Indian Families.

BACKGROUND: Wilson disease (WD) is a rare autosomal recessive disorder of copper metabolism caused due to mutations in the copper transporter ATP7B. There is often a striking variability of clinical manifestations among patients with ATP7B mutations, including in siblings. This phenomenon may be caused by individual differences in copper accumulation in hepatocytes and intolerance to copper toxicity as governed by genetic variations in copper metabolism genes acting as modifier loci to the disease. OBJECTIVE: To elucidate the genetic basis of striking clinical heterogeneity among two siblings

Evaluating the potential of third generation metagenomic sequencing for the detection of BRD pathogens and genetic determinants of antimicrobial resistance in chronically ill feedlot cattle.

BACKGROUND: Bovine respiratory disease (BRD) is an important cause of morbidity and mortality and is responsible for most of the injectable antimicrobial use in the feedlot industry. Traditional bacterial culture can be used to diagnose BRD by confirming the presence of causative pathogens and to support antimicrobial selection. However, given that bacterial culture takes up to a week and early intervention is critical for treatment success, culture has limited utility for informing rapid therapeutic decision-making. In contrast, metagenomic sequencing has the potential to quickly resolve all

Drug susceptibility profiling and genetic determinants of drug resistance in Mycobacterium simiae isolates obtained from regional tuberculosis reference laboratories of Iran.

BACKGROUND: Among Non-tuberculous mycobacteria (NTM) which generally cause opportunistic infections, especially in immunocompromised hosts, Mycobacterium simiae (M. simiae) is one of the most important NTM, associated with pulmonary disease. The main concern about M. simiae infections is the extreme resistance of this NTM to antibiotics. There are limited studies about drug susceptibility testing (DST) and the causes of drug resistance in M. simiae. Hence, the current study aimed to identify the M. simiae isolates and to assess the drug resistance of the isolates using phenotypic and molecular
