Looking under the hood of the dot-probe task: an fMRI study in anxious youth.

BACKGROUND: Anxious youth have shown altered behavioral performance on the dot-probe task, but neural activation patterns provoked by the task remain poorly understood. In particular, neural mechanisms of threat disengagement, a clinically relevant construct, have been inadequately explored. METHOD: During fMRI acquisition, 121 youth (ages 9-13; 90 with Generalized Anxiety Disorder, Separation Anxiety Disorder, and/or Social Phobia; 31 nonanxious controls) completed a dot-probe task, which required participants to identify the location of a dot replacing either a neutral or fearful face in a

Structural brain differences in major depressive disorder patients with suicidal risk: Evidence from the DIRECT consortium.

OBJECTIVE: Although previous studies have reported structural brain alterations in major depressive disorder (MDD) patients at risk of suicide, no wide consensus has been reached. This study aimed to elucidate structural brain differences between MDD patients with and without suicide risk using data from the DIRECT Consortium, advancing our understanding of the neurophysiological mechanisms underlying suicide risk in MDD patients. METHODS: A total of 203 healthy controls (HCs), 208 MDD patients without suicide risk (MDD-NSR), and 376 MDD patients with suicide risk (MDD-SR) were included. T1-w

Long-term outcome of subgenual cingulum and uncinate fasciculus deep brain stimulation for the treatment of post-traumatic stress disorder.

BACKGROUND: Deep brain stimulation (DBS) has been investigated as a treatment for various neuropsychiatric disorders. We recently conducted a feasibility trial to assess the safety and efficacy of DBS in the subgenual cingulate region (SCG) and uncinate fasciculus (UF) for post-traumatic stress disorder (PTSD). In our study, we observed a 55 % reduction in Clinician-Administered PTSD Scale (CAPS) scores at six months. Here, we present long-term follow-up data on the patients included. METHODS: Four patients with PTSD were treated with SCG/UF DBS at our center. Long-term outcome was measured u

Being praised and criticized in adolescents and adults: Age-related changes in neural responses to social evaluations.

Adolescence is a developmental period marked by heightened sensitivity to social evaluation due to the ongoing maturation of brain regions involved in emotion regulation and social cognition. Adolescents may be more vulnerable to socially evaluative feedback, particularly criticism, than adults. Although previous studies have documented age-related differences in neural responses to social evaluation, it remains unclear how the dynamic interactions among brain regions differ between these two age groups during social evaluation. In this study, sixty-four adolescents (14-17 years) and fifty-nin

Regulation of short-term declarative memory by selective activation of the locus coeruleus-retrosplenial cortex dopaminergic pathway and its pathological alterations in Parkinson's disease.

Short-term declarative memory impairment, encompassing deficits in spatial working memory and object recognition, represents a prevalent non-motor symptom of Parkinson's disease (PD). Despite its clinical significance, the neural circuit mechanisms underlying this cognitive dysfunction remain poorly understood. Here, we identify a previously underappreciated dopaminergic projection from the locus coeruleus (LC) to the retrosplenial cortex (RSC) as a critical regulator of short-term declarative memory. Using optogenetic activation of LC dopaminergic neurons, we evoked dopamine release in the RS

Effect of cognitive enhancement therapy for early course schizophrenia on gray matter volume: A confirmatory multisite randomized clinical trial.

BACKGROUND: Cognitive Enhancement Therapy (CET) is an evidence-based cognitive remediation intervention for early course schizophrenia with established benefits for cognition. CET may protect against broad temporolimbic gray matter volume loss associated with cognitive improvement, but this finding has yet to be replicated. This research reexamined if CET protects against temporolimbic gray matter volume loss in an independent and larger multisite early course sample, and if this neuroprotective effect predicts cognitive and social adjustment improvement. METHODS: Ninety-nine participants wit

The Salience of Psychotic Experiences: Neural Correlates in the General Population.

BACKGROUND AND HYPOTHESIS: Psychotic experiences (PEs) frequently occur at a subclinical level in the general population, including symptoms like hallucination and delusion. An important hypothesis is the aberrant salience hypothesis, according to which the salience network (SN) plays a decisive role coordinating between inner and outer world, anchored in the dorsal anterior cingulate cortex (dACC) and anterior insula (AI). Given SN's central role in PE development and maintenance, investigating its functional connectivity (FC) in a population-based sample is critical for understanding its eti

Subclinical anxiety is associated with reduced self-distancing and enhanced self-blame-related connectivity between anterior temporal and subgenual cingulate cortices.

Excessive self-blaming emotions are commonly observed in anxiety disorders, with qualitatively similar symptomatology reported in subclinical populations. Interpretation of moral information requires assessing the social conceptual information, a process overseen by the superior anterior temporal lobe (sATL). Feelings of self-blame evoke interactions of sATL and socio-affective regions, and previous research shows that subclinical anxiety modulates the organisation of the self-blame circuitry. This study aimed to extend these findings by exploring links of trait-anxiety with (i) self-blaming e

Metabotropic glutamate receptor 5 in the anterior cingulate cortex predicts individual differences in motor impulsivity but not in risky decision-making.

Impulsivity is a multidimensional construct implicated in various psychiatric disorders, yet its neurobiological underpinnings are insufficiently understood. The metabotropic glutamate receptor type 5 (mGluR5) has emerged as a promising target for modulating impulsive behavior, but whether impulsivity is associated with alterations in mGluR5 availability is unknown. Here, we investigated mGluR5 availability in relation to different impulsivity constructs in male Roman high- (RHA) and low-avoidance (RLA) rat lines, which exhibit divergent impulsive profiles. Motor impulsivity and risky decision

Functional inputs to the subgenual cingulate cortex differentiate levels of depressive symptom severity in neurotypical young adults: granger causality and clustering analyses.

INTRODUCTION: Extant research has implicated functional connectivity of the subgenual anterior cingulate cortex (sgACC) in major depressive disorders or depressive traits in neurotypical populations. However, prior studies have not distinguished the inputs and outputs of the sgACC, and the "diagnostic" accuracy of these connectivity metrics remains elusive. METHODS: Here, we analyzed data of 890 neurotypical young adults (459 women, age 22-35) from the Human Connectome Project using Granger causality analyses (GCA) with the sgACC as the seed and 268 regions of interest from the Shen's atlas a

Reproducibility assessment of magnetic resonance spectroscopy of pregenual anterior cingulate cortex across sessions and vendors via the cloud computing platform CloudBrain-MRS.

Proton magnetic resonance spectroscopy (1H-MRS) has potential in clinical diagnosis and understanding the mechanism of illnesses. However, its application is limited by the lack of standardization in data acquisition and processing across time points and between different magnetic resonance imaging (MRI) system vendors. This study examines whether metabolite concentrations obtained from different sessions, scanner models, and vendors can be reliably reproduced and combined for diagnostic analysis-an important consideration for rare disease research. Participants underwent magnetic resonance sc

Glutamate, NAA, and energy metabolism in clinical high risk and first episode psychosis.

Regulation of brain glutamate is closely related to brain energy metabolism. Changes in both central glutamatergic function and peripheral energy metabolism have been implicated in psychosis risk, onset and long-term illness, but there is a lack of empirical evidence to link these processes. We investigated the relationships between glutamate and N-acetyl-aspartate (NAA, a potential marker of neuronal metabolic integrity) in the anterior cingulate cortex (ACC), measured using proton magnetic resonance spectroscopy (1H-MRS), and peripheral markers of energy metabolism (mitochondrial complex I-V

Transcriptome-wide alternative splicing and transcript-level differential expression analysis of post-mortem Lewy body dementia brains.

Lewy body dementias (LBD) are the second most common dementia. Several genes have been associated with LBD, but little is known about their contributions to LBD pathophysiology. Each gene may transcribe multiple RNA, and LBD brains have extensive RNA splicing dysregulation. Hence, we completed the first transcriptome-wide transcript-level differential expression analysis of post-mortem LBD brains for gaining more insights into LBD molecular pathology that are essential for facilitating discovery of novel therapeutic targets and biomarkers for LBD. We completed transcript-level quantification o

Neurobiological differences in early-onset obsessive-compulsive disorder: A study of the glutamatergic system based on functional magnetic resonance spectroscopy.

In this study, the combination of functional state magnetic resonance spectroscopy (fMRS) and cognitive tasks was used to conduct subgroup analyses on early-onset OCD (EO) and non-early-onset OCD (non-EO) and explore differences in the glutamatergic system and cognitive function among OCD subtypes. A total of 70 OCD and 30 healthy controls (HCs) underwent clinical evaluation and were subsequently divided into the EO or non-EO groups. Next, both resting and functional state MRS data were collected, with the anterior cingulate cortex (ACC) serving as the region of interest. Quantitative analysis

White matter microstructure of the cingulum bundle and the uncinate fasciculus in primary and secondary callous-unemotional traits.

Callous-unemotional (CU) traits appear to be linked to differences in neural networks, including the uncinate fasciculus (UF) and cingulum bundle (CB). It has recently been suggested that CU traits are comprised of primary and secondary subtypes based on the relative co-occurrence of internalising symptoms. Here, we explored the white matter microstructure of these subtypes. White matter microstructure of the UF and CB was analysed in youths (aged 8–18; n = 121; females = 31) recruited through Europe-based multisite studies. Our sample comprised a control (n = 53) and a case group with disrupt

Association of grey matter cerebral blood flow with white matter integrity in relation to youth bipolar disorder.

BACKGROUND: Both cerebral blood flow (CBF) and fractional anisotropy (FA) are altered in bipolar disorder (BD). We sought to investigate the unexplored CBF-FA association in relation to youth BD. METHODS: We recruited youth with BD, high-risk youth with a family history of BD, and healthy controls. Using 3 T magnetic resonance imaging, we measured global and regional grey matter CBF using arterial spin labelling and we measured regional FA using diffusion tensor imaging. We explored the CBF-FA association across groups, between groups, and by sex. Analyses of the BD group further examined moo

Altered Morphological Characteristics Associated With Spatial Working Memory Performance in Children With ADHD.

BACKGROUND: Spatial working memory (SWM) deficit is a common problem in attention deficit disorder with hyperactivity (ADHD), often correlating with the severity of ADHD symptoms and academic difficulties. Although previous studies have broadly described abnormal brain structural changes in ADHD, the potential changes in brain morphology in children with ADHD with SWM dysfunction are still uncharacterized. This cross-sectional study was used to elucidate the brain morphological alterations associated with SWM performance in boys with ADHD. METHODS: Data for this investigation were retrieved f

Comparative lipidomic alterations in the anterior cingulate cortex in schizophrenia and major depression.

Schizophrenia and major depressive disorder are typically classified as distinct psychiatric conditions; however, they share overlapping symptoms and biological risk factors. Identifying molecular features that distinguish or unify these disorders may provide insights into their underlying mechanisms. Lipid metabolism is increasingly recognized as an important factor influencing brain function and the development of mental disorders, yet direct comparative lipidomic analyses of these two conditions remain limited. In this study, we examined the lipidomic profiles of the anterior cingulate cort

Dopaminergic changes in the subgenual cingulate cortex in dementia with lewy bodies associates with presence of depression.

In addition to the core clinical features of fluctuating cognition, visual hallucinations, and parkinsonism, individuals with dementia with Lewy bodies (DLB) frequently experience chronic and debilitating major depression. Treatment of depression in DLB is hampered by a lack of available effective therapies and standard serotonergic medication for major depressive disorder (MDD) is typically ineffective. Dysfunction of dopaminergic neurotransmission contributing to anhedonia and loss of motivation has been described in MDD. The subgenual anterior cingulate cortex (sgACC) is important in mood r

Activity-dependent modulation of glutamate, glutamine and glutathione in first-episode schizophrenia: A 7T functional MRS study.

BACKGROUND AND HYPOTHESIS: Glutamatergic abnormalities have been implicated as an important component of schizophrenia symptoms. Functional magnetic resonance spectroscopy (fMRS) has emerged as a strong candidate for making dynamic observations of neurometabolites, overcoming the challenges of single-state observations in traditional magnetic resonance spectroscopy studies. To date, dynamic measurements of glutathione have not been presented in patients in the early stages of schizophrenia. STUDY DESIGN: Dynamic glutamate, glutamine, and glutathione concentrations were quantified from the dor
