Targeting TXNIP in endothelial progenitors mitigates IL-8-induced neutrophil recruitment under metabolic stress.

BACKGROUND: This study explores the potential role of Thioredoxin-interacting protein (TXNIP) silencing in endothelial colony-forming cells … il recruitment both in vitro and in vivo. The in vivo effects were examined using a murine model of hindlimb ischemia to observe the physiological relevance of TXNIP modulation under metabolic disorders. RESULTS: TXNIP silencing did not mitigate the adverse effects on cell recruitment, vasculogenic properties, or senescence induced by metabolic stress in …

Remote Ischemic Preconditioning induces Cardioprotective Autophagy and Signals through the IL-6-Dependent JAK-STAT Pathway.

: Autophagy is a cellular process by which mammalian cells degrade and assist in recycling damaged organelles and proteins. This study aimed to ascertain the role of autophagy in remote ischemic preconditioning (RIPC)-induced cardioprotection. Sprague Dawley rats were subjected to RIPC at the hindlimb followed by a 30-min transient blockade of the left coronary artery to simulate ischemia reperfusion (I/R) injury. Hindlimb muscle and the heart were excised 24 h post reperfusion. RIPC prior to I/R upregulated autophagy in the rat heart at 24 h post reperfusion. In vitro, autophagy inhibition or…

Chronic lymphoplasmacytic villonodular proliferative synovitis in a 10-year-old Jack Russell Terrier dog.

We describe a case of chronic lymphoplasmacytic villonodular synovitis (CLPVNS) associated with cranial cruciate ligament (CCL) disease in a 10-year-old spayed Jack Russell Terrier bitch. The bitch was presented to a veterinary clinic with severe, non-weight bearing, acute left hindlimb lameness. The bitch had previously been treated surgically for stifle CCL disease of the same joint, using the lateral fabellar suture (LFS) technique. Since the treatment, the patient had a history of intermittent left hindlimb non-weight bearing lameness that was manageable with nonsteroidal anti-inflammatory…

Treadmill training after spinal cord hemisection in mice promotes axonal sprouting and synapse formation and improves motor recovery.

Treadmill training with weight-support is a therapeutic strategy used in human patients after spinal cord injury (SCI). Exercise leads to locomotor improvement in a variety of animal models; however, the effect of exercise on axonal regrowth has not been directly examined. This study used several locomotor tests, including kinematic gait analysis, to analyze the differences between treadmill-trained and untrained mice in the usage of their paretic hindlimb following a low thoracic hemisection. Analysis of muscle atrophy, anterograde axonal tracing and expression of the synaptic markers synapto…

Mass allometry of the appendicular skeleton in terrestrial mammals.

Most analyses on allometry of long bones in terrestrial mammals have focused on dimensional allometry, relating external bone measurements e … allometry. These include the pectoral and pelvic girdles, humerus, radius+ulna, and forelimb. Total hindlimb mass and the masses of individual hindlimb bones (femur, tibia, and metatarsus) scale isometrically. Amylin influences insulin-stimulated glucose metabolism by two independent mechanisms.

The effects of amylin on fiber type-specific muscle glucose metabolism under hyperglycemic (10 mmol/l) and hyperinsulinemic (2.1 nmol/l) conditions were investigated using a rat hindlimb perfusion system. Amylin concentration ranged from 1 to 100 nM. Efficacy for inhibition of glucose uptake traced with 2-deoxyglucose by amylin was demonstrated in all three fiber types. The incorporation of 2-deoxy-[3H]glucose tracer decreased from control values by 41% in fast oxidative (FO), 36% in fast glycolytic (FG), and 37% in slow oxidative (SO) muscle with 100 nM amylin. Amylin increased intracellular …

The muscles in chronic ligamentous instability of the knee: an experimental study.

Several authors have formulated the idea that insufficiency of the anterior cruciate ligament (ACL) might lead to a biological adaptation, consisting of atrophy of the quadriceps and other extensors (antagonists of the ACL) and relative preservation of the hamstrings and other flexors (agonists of the ACL). We studied the modifications in hindlimb muscles that spontaneously occurred in 36 rabbits with surgically-induced chronic ligamentous instability of the right knee. The unoperated left hindlimb served as a control. Three months after surgery the rabbits were sacrificed and the muscles diss…

Augmentation by 4-aminopyridine of vestibulospinal free fall responses in chronic spinal-injured cats.

This study examines the effect of the potassium channel blocker 4-aminopyridine (4-AP) on free fall responses (FFR) in the hindlimb muscles of chronically spinal injured cats. The thoracic spinal cord of 7 adult female cats was injured by a standardized contusion method. At 3-7 months post-injury the FFR in 6 hindlimb muscles was recorded electromyographically in each animal, under ketamine sedation. The normal short-latency response to a sudden drop was severely attenuated in all injured animals and practically undetectable in 2 cases. Within 15 min following intravenous administration of 1 m…

The efficacy of total parenteral nutrition in malnourished tumor-bearing rats.

The current study was designed to compare the efficacy of total parenteral nutrition (TPN), provided either before or after tumor removal, in replenishing protein stores in the malnourished tumor-bearing (TB) host. Fisher 344 rats bearing a transplantable methylcholanthrene (MCA)-induced sarcoma were used. After the animals were cachectic from their tumor, a central venous catheter was inserted. At the time of catheter insertion, the animals were randomized into two groups. Group I rats had the TB hindlimb amputated whereas Group II rats had the nontumor bearing (NTB) hindlimb amputated. After…

Locomotor capacity attributable to step training versus spontaneous recovery after spinalization in adult cats.

Locomotor performance, hindlimb muscle activity and gait patterns during stepping were studied in step-trained and non-trained female, adult spinal cats. Changes in locomotor characteristics relative to prespinalization bipedal and quadrupedal stepping patterns were used to evaluate the effects of step training on the capacity to execute full weight-bearing stepping after spinalization. Step training consisted of full weight-bearing stepping of the hindlimbs at the greatest range of treadmill speeds possible at any given stage of locomotor recovery. In the initial stages of training the limbs …

Dermoid sinus at the lumbosacral junction in an English springer spaniel.

A dermoid sinus was identified in a springer spaniel that presented with hindlimb neurological deficits. The sinus was continuous with the dura mater at the level of the lumbosacral junction. The presence of hair and debris adjacent to neural tissue had elicited a myelitis. A dorsal laminectomy was required to allow complete surgical resection of the sinus. The dog's neurological status improved after treatment and this improvement was maintained over a five-month follow-up period.

ANALGESIC EFFICACY OF SUBCUTANEOUS MELOXICAM IN RED-EARED SLIDER TURTLES (TRACHEMYS SCRIPTA).

Meloxicam is a commonly used analgesic for reptiles. There are few published studies regarding the efficacy of meloxicam and dosing is typically extrapolated from mammals. We evaluated the analgesic efficacy of two meloxicam doses in red-eared slider turtles (Trachemys scripta). We hypothesized that subcutaneously administered 1 mg/kg meloxicam would significantly increase thermal hindlimb withdrawal latency for a greater time period than 0.4 mg/kg meloxicam. Twenty red-eared sliders were randomly assigned to one of three groups: 1) 0.9% saline (NaCl) (0.5 ml/kg), 2) 0.4 mg/kg meloxicam (Melox…

Brief early-life motor training induces behavioral changes and alters neuromuscular development in mice.

In this study, we aimed to determine the impact of an increase in motor activity during the highly plastic period of development of the motor spinal cord and hindlimb muscles in newborn mice. A swim training regimen, consisting of two sessions per day for two days, was conducted in 1 and 2-day-old (P1, P2) pups. P3-trained pups showed a faster acquisition of a four-limb swimming pattern, accompanied by dysregulated gene expression in the lateral motor column, alterations in the intrinsic membrane properties of motoneurons (MNs) and synaptic plasticity, as well as increased axonal myelination i…

In vivo imaging of axonal transport in peripheral nerves of rodent forelimbs.

Axonal transport is the essential process by which neurons actively traffic a variety of cargoes between the cell soma and axon terminals. Accordingly, dysfunctional axonal transport is linked to many nervous system conditions. Therefore, being able to image and quantify this dynamic process in live neurons of animal disease models is beneficial for understanding neuropathology and testing new therapies at the preclinical level. As such, intravital approaches have been developed to assess cargo movement in the hindlimb sciatic nerves of live, anaesthetised mice. Here, we describe an adapted me…
