The polyamine inhibitor alpha-difluoromethylornithine modulates hippocampus-dependent function after single and combined injuries.

Exposure to uncontrolled irradiation in a radiologic terrorism scenario, a natural disaster or a nuclear battlefield, will likely be concomi … pha-difluoromethylornithine (DFMO) would reduce the adverse effects of single or combined injury on hippocampus structure and function. Hippocampal dependent cognitive impairments were quantified with the Morris water maze and showed that DFMO effectively reversed cognitive impairments after all injuries, particularly traumatic brain injury. Similar results w …

Grafted neuronal precursor cells differentiate and integrate in injured hippocampus in experimental pneumococcal meningitis.

Bacterial meningitis (BM) frequently causes persisting neurofunctional sequelae. Autopsy studies in patients dying from BM show characteristic apoptotic brain injury to the stem cell niche in the subgranular zone of the hippocampal dentate gyrus (DG), and this form of brain damage is associated with learning and memory deficits in experimental BM. With an eye to potential regenerative therapies, the survival, migration, and differentiation of neuronal precursor cells (NPCs) were evaluated after engraftment into the injured hippocampus in vitro and in vivo in an infant rat model of pneumococcal…

Dimebon enhances hippocampus-dependent learning in both appetitive and inhibitory memory tasks in mice.

Pre-clinical and clinical studies on dimebon (dimebolin or latrepirdine) have demonstrated its use as a cognitive enhancer. Here, we show th … ed mice with dimebon also did not affect their water intake. Our data suggest that dimebon enhances hippocampus-dependent learning in both appetitive and inhibitory tasks in mice.

ROS-GC subfamily membrane guanylate cyclase-linked transduction systems: taste, pineal gland and hippocampus.

In the continuous efforts to test the validity of the theme that the Ca(2+)-modulated ROS-GC subfamily system is a universal transduction component of the sensory and sensory-linked network of neurons, this article focuses on the presence and variant biochemical forms of this transduction system in the gustatory epithelium, the site of gustatory transduction; in the pineal, a light-sensitive gland; and in the hippocampus neurons, linked with the perception of all SENSES.

Selective disruption of hippocampus-mediated recognition memory processes after episodes of transient global amnesia.

Transient global amnesia (TGA) is characterized by the abrupt onset of severe amnesia without concomitant focal neurological symptoms. Recent studies revealed that small and punctate MR-signal diffusion-weighted imaging (DWI) lesions can be found within the hippocampus of TGA patients during the post-acute phase. On the basis of dual-process models of recognition memory, the present study examined the hypothesis that hippocampal dysfunction as suggested by these DWI lesions disrupts hippocampus-mediated recollection in patients with TGA, whereas familiarity-based recognition memory that is ass…

SO(2) inhalation induces protein oxidation, DNA-protein crosslinks and apoptosis in rat hippocampus.

Previous studies provide evidence for the possible neurotoxicity of SO(2), but little information is available about its mechanisms. In the present study, SO(2) inhalation-induced effects on the protein oxidation, DNA-protein crosslinks and apoptosis in rat hippocampus were studied, by exposing Wistar rats to SO(2) at 14, 28 and 56mg/m(3). The results indicate that the protein carbonyl content, an indicator of protein oxidation, and DNA-protein crosslink coefficient were significantly augmented with concentration-dependent properties. In addition, SO(2) inhalation at all concentrations tested …

Prion disease causes less severe lesions in human hippocampus than other parts of brain.

AIM: The hippocampus can be very sensitive to damage in the scrapie-infected mouse, a well-established animal model of prion diseases. Terminally ill scrapie-infected animals exhibit nearly complete loss of cornu ammonis (CA) 1 pyramidal neurons, but few studies have focused on the neuropathological lesions of the human hippocampus in autopsied brain tissue; in particular, few findings on differences in severity of pathology between the hippocampal and parahippocampal formations have been obtained. The aim of the present paper is to evaluate the human hippocampus of prion disease through neuro…

TAG-1-deficient mice have marked elevation of adenosine A1 receptors in the hippocampus.

TAG-1 is a neural recognition molecule in the immunoglobulin superfamily that is predominantly expressed in the developing brain. Several lines of evidence suggest that TAG-1 is involved in the outgrowth, guidance, and fasciculation of neurites. To directly assess the function of TAG-1 in vivo, we have generated mice with a deletion in the gene encoding TAG-1 using homologous recombination in embryonic stem cells. Gross morphological analysis of the cerebellum, the spinal cord, and the hippocampus appeared normal in TAG-1-deficient mice. However, TAG-1 (-/-) mice showed the upregulation of the…

Hypocapnia under hypotension induces apoptotic neuronal cell death in the hippocampus of newborn rabbits.

We investigated the adverse effect of hypocapnia on the neonatal rabbit brain. Two-week-old Japanese white rabbits were assigned to three gr … d. These results suggested that hypocapnia under hypotension might cause neuronal cell death in the hippocampus of neonatal rabbit. Not only ischemia but also a metabolic change induced by hypocapnia might contribute to this apoptotic neuronal cell damage.

Corticosterone and phenytoin reduce neuronal nitric oxide synthase messenger RNA expression in rat hippocampus.

The production and release of the corticosteroids, namely the glucocorticoids and the mineralocorticoids, are regulated by various stimuli, including stress. Previous studies from our laboratory have shown that chronic exposure to stress or to stress levels of glucocorticoids produces atrophy of the apical dendrites of CA3 pyramidal neurons in the hippocampus. This stress-induced dendritic remodeling is blocked by the anti-epileptic drug phenytoin, which suppresses glutamate release, and also by N-methyl-D-aspartate receptor antagonists. These results suggest an interaction between glucocortic…

In contrast, c-Fos expression following generalised convulsions induced by non-anaesthetic convulsants was very marked in the hippocampal region, piriform cortex and amygdala, and extended to the thalamus and neocortex. Increases in cerebral blood flow in rat hippocampus after medial septal injection of naloxone.

BACKGROUND AND PURPOSE: In a previous study, we occasionally found that the rat given naloxone in the preoptic region develops behavioral seizures. In view of knowledge that the forebrain including the medial septal nucleus provides cholinergic projections to the hippocampal formation, the present study examined the effects of naloxone injected into the medial septal nucleus on the local blood flow in the hippocampus. METHODS: A polyurethane-coated platinum electrode with a 1-mm bare tip for measurement of blood flow and a guide cannula made of stainless steel tube for naloxone injection were…

APP family in inhibitory neurons controls inhibitory recruitment and short-term plasticity in the hippocampus.

Amyloid precursor protein (APP) is associated with both familial and sporadic forms of Alzheimer's disease. We previously reported that APP and its family members, amyloid precursor-like proteins 1 and 2 (APLP1 and APLP2), regulate intrinsic neuronal excitability and synaptic plasticity in excitatory principal neurons, though APP family is dispensable for neuronal survival. However, the physiological role of APP family in inhibitory interneurons remains poorly understood. Here, we use our previously characterized floxed APP, APLP1, APLP2 and GAD2-Cre alleles to generate inhibitory neuron-speci

Novel object recognition in the dorsomedial and ventral hippocampus of young domestic chicks (Gallus gallus).

The avian hippocampus is well known for its role in spatial cognition, but its contribution to non-spatial domains such as novelty detection remains poorly understood. Here, we examined whether the ventral hippocampus in domestic chicks (Gallus gallus) is involved in processing non-spatial novelty. On post-hatching day 5, chicks were exposed to either a novel or a familiar object. Chicks of both sexes recognised the novel object, showing delayed approach, greater distance from the object, and increased locomotion, consistent with neophobic behaviour. Neural activation was mapped using c-Fos im…

Androgens and estrogens rapidly facilitate short-term memory in the dorsal hippocampus of male mice.

Sex steroid hormones influence cognition through both classical delayed genomic and rapid mechanisms. While rapid estrogenic modulation of memory in the dorsal hippocampus (dHPC) is well established in females, the extent to which the sex hormones rapidly regulate short-term memory (STM) in males remains unresolved. Here, we investigated the rapid effects of the testosterone metabolites 17β-estradiol (E2) and dihydrotestosterone (DHT) on hippocampal-mediated STM in castrated CD1 male mice. Bilateral infusions into the dHPC were administered prior to testing object placement, object recognition…

Cross-fostering affects microglia and cell death in the hippocampus of female and male degu pups.

Parental care is essential for social, behavioural, and neural development of offspring. Disruption of parental care, for example through parental separation, can negatively affect offspring development in rodents. While previous work has focused on maternal and paternal deprivation, the effects of cross-fostering, another form of parental-offspring instability, on brain development remain poorly understood. Such disruptions are thought to induce stress, which in turn can suppress neurogenesis and increases inflammation and apoptosis in the hippocampus, with effects that often differ between s…

Structural features of the hippocampus covary with memory-guided attention depending on the side of hippocampal sclerosis.

Past experiences stored in long-term memory (LTM) provide a valuable resource for making predictions that shape perception and guide goal-directed behavior. Contents from the high-capacity LTM system guide contextual selective attention to enhance sensory and higher-order processing of memory-predicted targets, in a process known as LTM-guided attention. While this essential cognitive function is believed to depend on the hippocampus, evidence is still scarce. In this study, we used a neuropsychological approach to test LTM-guided attention in the context of isolated hippocampal pathology and …

Neuroprotective impact of nicotinamide riboside and metformin on the hippocampus of male rats with diet-induced obesity.

BACKGROUND: Obesity-induced metabolic disturbances are increasingly recognized as major contributors to hippocampal dysfunction and neurodegeneration. Chronic exposure to a high-fat diet (HFD) has been associated with hippocampal structural alterations, dysregulation of neurotrophic and metabolic signalling, and increased neuronal vulnerability, ultimately impairing cognitive performance. OBJECTIVE: This study investigated the neuroprotective effects of nicotinamide riboside (NR), metformin (MET), and their combination on hippocampal structure, gene expression, and spatial memory in a male ra
