Overcoming host restrictions to enable continuous passaging of GII.3 human norovirus in human intestinal enteroids.

The establishment of human intestinal enteroids (HIEs) as a model for human norovirus (HuNoV) replication has been transformative for studying this leading cause of gastroenteritis. However, indefinite passaging of HuNoVs in HIEs remained a challenge, necessitating the use of patient stool samples as viral inocula. Using RNA-seq, we identified CXCL10, CXCL11, and CCL5 as up-regulated chemokines, suggesting their potential as host restriction factors. TAK-779, a CXCR3/CCR5/CCR2 antagonist, enhanced GII.3 HuNoV replication and viral spread in a dose- and time-dependent manner, enabling successfu

A Double-Edged Sword: Extracellular Serine Proteases as Facilitators of Infection and Mediators of Immunity.

Serine proteases are a class of enzymes that orchestrate an immune response. These proteases can be hijacked by viruses to facilitate entry and spread, while simultaneously supporting the innate immune system in neutralizing pathogens. This review highlights the dual roles of exogenous serine proteases, emphasizing neutrophil serine proteases (NSPs) that facilitate viral entry and promote disease progression while also contributing to antiviral defense by degrading viral glycoproteins. Additionally, the potential to modulate serine protease activity to boost host defenses will be discussed, of

A dual-host regulatory hub integrating biofilm formation and innate immunity resistance.

Vector-borne pathogens must adapt to sharply distinct host environments, yet the regulatory logic that coordinates transitions between opposing host-specific programs remains poorly defined. Here, we identify a minimal regulatory module in the flea-borne pathogen Yersinia pestis that integrates vector transmission with resistance to mammalian innate immunity. Screening of flea-induced genes uncovered the regulator HdfR, which acts primarily through activation of maoP, encoding a nucleoid-associated protein. This HdfR-MaoP module promotes biofilm-dependent foregut blockage in the flea while coo

Isolation of Fusobacterium nucleatum-Induced Host Exosomal sRNAs.

In eukaryotic cells, small RNAs (sRNAs) are a class of noncoding RNAs that are less than 200 nucleotides in length and mainly classified per their biogenesis as microRNA, small interfering RNA, PIWI-interacting RNA (piRNA) or tRNA-derived small RNA (tsRNA). Increasing lines of evidence indicate that these host-derived sRNAs may contribute to the maintenance of host-microbial homeostasis through cross-kingdom gene modulation. Recent studies have shown that when challenged with Fusobacterium nucleatum, a Gram-negative, anaerobic oral pathobiont, host epithelial cells release specific exosome-bor

Lesser mealworm Alphitobius diaperinus Panzer (Coleoptera: Tenebrionidae) larvae are differentially susceptible to Beauveria bassiana (Bals.-Criv.) Vuill. (Hypocreales: Clavicipitaceae) under moderate and extreme high temperature stress.

Lesser mealworm larvae topically treated with Beauveria bassiana, under different temperature regimens demonstrated complex interactions with the pathogen. Larvae treated with B. bassiana and incubated at 32.5 °C were less susceptible to B. bassiana than those treated in the same way but then incubated at 25 °C. Larvae incubated at 35 °C for two days after B. bassiana treatment showed limited susceptibility to disease, but those incubated at 35 °C two days prior to treatment and then incubated at 25 °C immediately after B. bassiana treatment were more susceptible than those maintained at 25 °C

Rethinking vector-borne disease prediction: toward an integrative modeling approach including vector and host data.

Vector-borne diseases significantly impact human and animal health, as well as global economies. Over the past decades, modeling approaches have significantly advanced our ability to understand and predict their dynamics. However, most predictive frameworks still rely predominantly on the abiotic components. This narrow focus fails to capture the ecological reality of transmission, since abiotic features alone do not ensure pathogen maintenance in the absence of appropriate hosts or vectors. The scarcity of models incorporating host-related variables underscores limitations that can over- or u

The polar landscape of bacterial pathogens and predators.

Precise spatial positioning of cellular constituents is critical for bacterial replication, interactions, and behavior. Rod-shaped bacteria may concentrate molecules and machinery at the cell poles, often by leveraging polar scaffolding proteins to recruit other factors. These typically small proteins can self-assemble into higher-order structures that provide platforms to direct fundamental processes like chromosome segregation, division plane selection, and differentiation. Beyond fundamental cellular processes, polarity is implicated in a wide range of bacterial behaviors, including environ

Evolution of virulence of a plant RNA virus in developmental stage-structured host populations.

Natural host populations are age-structured, and developmental stages differ in susceptibility and within-host pathogen dynamics, potentially imposing distinct selective pressures on viruses. However, the evolutionary consequences of host age structure remain poorly understood. We experimentally evolved turnip mosaic potyvirus for 5 passages in Arabidopsis thaliana populations spanning 7 demographic regimes, from juvenile- to mature-dominated cohorts. We quantified disease progression, symptom severity, and viral load, cross-inoculated evolved lineages across host stages to construct infection

The Caenorhabditis elegans microbiome impacts microsporidia infection through nutrient limitation and spore inactivation.

Microsporidia are fungal parasites that can only reproduce inside the cells of a host, and they infect most animal groups, including humans and agriculturally important species. Animals also harbor a microbiome, which can shape resistance to infection, but the mechanisms by which bacteria influence microsporidia infection are poorly understood. To address this, we tested how bacteria associated with the nematode Caenorhabditis elegans affect infection by its natural microsporidian parasite, Nematocida parisii. Here we show that nematodes grown on Chryseobacterium scophthalmum or Sphingobacteri

How elephant host proteins fight-or fail-against EEHV: Insights from a multi-contrast proteomic enrichment study.

Elephant endotheliotropic herpesvirus hemorrhagic disease (EEHV-HD) is a rapidly fatal syndrome of juvenile Asian elephants, but the host-response programs distinguishing progression from survival and the underlying pathophysiology remain poorly defined. The serum proteome of 62 Asian elephants (Elephas maximus) was profiled using a multi-contrast design stratified by age, clinical status, and infection history; protein abundance was analyzed by empirical Bayes linear modeling and Gene Ontology enrichment with semantic similarity reduction. Clinically affected elephants showed enrichment of in

Human airway organoids as a versatile model to study BSL-4 virus replication and pathogenesis.

Research with BSL-4 viruses such as Ebola, Marburg, and Nipah presents significant challenges due to their high virulence and the stringent containment measures required. This study establishes human airway organoids as a robust model for investigating BSL-4 pathogens. In contrast to conventional cell lines, airway organoids enable investigation of virus-host interactions within a human tissue context, providing insights that are more directly translatable to human disease. We generated airway organoids from both clinical donor tissues and commercially available nasal epithelial cells and show

Kiss and spit metabolomics highlight the role of host purine metabolism during pathogen infection.

Intracellular bacteria and protists rely on the host cell to supply many metabolites, but the mechanisms through which pathogens manipulate host metabolism to their benefit are not understood. Here, we demonstrate that when the obligate intracellular parasite Toxoplasma gondii secretes its rhoptry organelle contents into the host cytoplasm before invasion-a process called "kiss and spit"-host cell metabolite abundance is altered in nucleotide synthesis, the pentose phosphate pathway, glycolysis, and amino acid synthesis. U-13C6-labeling metabolomics confirmed that kiss and spit increased the f

Comparative transcriptomic insights into ticks infected by alongshan and tick-borne encephalitis viruses.

Ticks are hematophagous ectoparasites and key vectors of a wide range of pathogens, including viruses, bacteria, and protozoa. Climate change and environmental shifts have contributed to the expansion of tick populations and the pathogens they harbor. Ticks rely solely on their innate immune system to combat infections, utilizing mechanisms such as RNA interference, phagocytosis, and antimicrobial peptide expression. However, pathogens have evolved strategies to evade or manipulate these defenses to facilitate their replication and transmission. This study compares the infection dynamics of tw

Feeding the invaders: how metabolic imbalance shapes infection and biofilm development.

Infectious and metabolic disorders have risen sharply and pose a significant threat to human populations worldwide. Despite being deemed unrelated, mounting evidence indicates that metabolic disorders markedly heightened both the likelihood and severity of infectious disease. The mechanism underlying this association is only beginning to be appreciated and involves a complex interaction between altered metabolic state, pathogen virulence mechanisms, and immune effector pathways. Here, we highlight that a dysregulated metabolic environment disrupts homeostasis, creating a nutrient-rich, immunoc

Shielded by the Dead: How Killed Bacteria Shape the Dynamics and Evolution of Innate Immunity.

AbstractThe outcome of an infection is determined by the dynamic interplay between microbial growth and host immunity. During a bacterial infection, bacteria killed by innate immune effectors can accumulate as corpses in the extracellular space, where they can continue to bind (and thus sequester) immune effectors. The impacts on infection outcomes of continued biochemical activity ("sponginess") by the dead have been generally overlooked in theoretical and empirical studies of within-host disease dynamics. We develop a mechanism-based mathematical model of within-host dynamics that incorporat

[Molecular mechanisms of enolase-mediated bacterial penetration of the blood-brain barrier].

Bacterial meningitis is associated with a high mortality and frequently leads to severe sequelae. Elucidating the molecular mechanisms underlying bacterial traversal of the blood-brain barrier (BBB) holds significant theoretical and practical value for the prevention and control of meningitis in both humans and animals. As a key virulence factor conserved in various pathogenic bacteria, enolase (ENO) plays a pivotal role in the process of BBB penetration. This review focuses on the molecular mechanisms of ENO-mediated BBB disruption. From the perspective of pathogen-host interactions, it syste

Neutrophils and tuberculosis - insights into immunobiology and pathogenesis.

Neutrophils are the most abundant leukocytes in human blood and play a key role in controlling infections. Their high circulating numbers allow rapid recruitment to sites of infection or injury, where they phagocytose pathogens and deploy a multitude of antimicrobial mechanisms. Neutrophil recruitment and clearance regulate infection severity and modulate broader immune response by influencing leukocyte populations. Over the past decade, research has demonstrated that neutrophils are functionally heterogeneous and more complex than previously appreciated with significant implications for immun

Enterococcus and cancer: from mechanistic insights to clinical translation.

Despite advances in traditional therapies, the global burden of malignant tumors remains substantial. Although immunotherapy has achieved remarkable breakthroughs by activating the host immune system, its clinical benefits are limited to a subset of patientsowing to low response rates and drug resistance. Emerging evidence highlights the pivotal role of the gut microbiota in determining immunotherapy outcomes. Within this ecosystem, the genus Enterococcus acts as a "double-edged sword": whereas certain strains can enhance antitumor immunity, other species notably Enterococcus faecalis, drive t

An Introduction to the Influence of Nutritional Factors on the Pathogenesis of Opportunist Fungal Pathogens in Humans.

Fungi such as Aspergillus fumigatus, Candida albicans, and Cryptococcus neoformans are opportunistic pathogens in humans. They usually infect individuals whose immune system is compromised due to either a primary infection, e.g., HIV/AIDS, or as part of treatment for another condition, e.g., stem cell or solid organ transplant. In hosts with a weakened immune system, these fungi can cause life-threatening infections. Unlike true pathogens, opportunistic pathogens do not have specific mechanisms to overcome a healthy host, requiring a different approach to understand how they cause infection. T
