The long-term safety of donor eye for 180 degrees limbal transplantation.

BACKGROUND: Little information is currently available on the long-term safety of eyes following limbal tissue donation. We have therefore evaluated long-term refractive changes, corneal surface stability, and visual outcome following limbal tissue donation of 180 degrees with permanent amniotic membrane transplantation (AMT). METHODS: Four healthy eyes of four patients underwent limbal keratectomy involving half of the limbal circumference (half limbal) for the purpose of limbal donation with permanent AMT. The best corrected visual acuity (BCVA), astigmatic changes by manifest refraction, an

[Cause and repair of a corneoscleral fistula in two patients].

BACKGROUND: A slowly progressing bulbar hypotension with maculopathy in a male patient as well as an endophthalmitis in a female patient both required surgical repair of posttraumatic leaks. PATIENTS AND METHODS: In a 32-year-old male patient, a filtration bleb leak occurred 4 months after a penetrating stab wound in the corneoscleral region. Persistent bulbar hypotension demanded microsurgical revision, revealing a 1 x 1 mm scleral defect. The leakage area was close to the limbus. In a 75-year-old female patient, signs of endophthalmitis occurred 4 years after extracapsular cataract extracti

Corneal stromal changes following reconstruction by ex vivo expanded limbal epithelial cells in rabbits with total limbal stem cell deficiency.

AIM: To study corneal stromal changes and the presence of myofibroblasts after transplantation of ex vivo expanded limbal epithelium. METHODS: A state of limbal deficiency was induced in 16 rabbits. After transplantation with autologous ex vivo expanded limbal epithelium on amniotic membrane (AM), their clinical outcomes were classified as success, partial success or failure according to surface smoothness, stromal clarity, and vascularisation. Clinical outcomes were correlated with phenotypic outcomes of corneal, conjunctival, or mixed epithelium, defined by expression of K3 keratin or MUC5A

Losartan Alleviates Chemical Burn-Induced Limbal Stem Cell Deficiency: Repurposing a Venerable Anti-Hypertension Drug.

PURPOSE: Currently, treatment for limbal stem cell deficiency (LSCD) involves limbal stem cell transplantation using either autologous or allogeneic tissue to restore the corneal surface. However, limitations such as donor shortages, immune rejection, and variable outcomes highlight a crucial need for more effective and safer therapeutic options. METHODS: We utilized a nitrogen mustard (NM)-induced corneal injury model to create LSCD, characterized by conjunctivalization. Mice exposed to NM were intraperitoneally injected with losartan. Single cell RNA sequencing (scRNA-seq) was conducted usi

miRNA Expression Profile in Primary Limbal Epithelial Cells of Aniridia Patients.

PURPOSE: This study evaluates the microRNA (miRNA) expression profile in primary limbal epithelial cells (pLECs) of patients with aniridia. METHODS: Primary human LECs were sampled and isolated from 10 patients with aniridia and 10 healthy donors. The miRNA profile was analyzed using miRNA microarrays. The biological roles of miRNA-validated target genes were delineated in silico by the enrichment analyses of the gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway. The expression of the most deregulated miRNAs was analyzed using quantitative real-time PCR (qRT-PCR).

Zonular fibre Insertion-to-Limbus Distance (ZLD): normative data to assess lens position and diagnose ectopia lentis.

PURPOSE: Subluxation of the crystalline lens (Ectopia Lentis, EL) can lead to significant visual impairment and serves as a diagnostic criterion for genetic disorders such as the Marfan syndrome. There is no established criterion to diagnose and quantify EL. We prospectively investigated the distance between the zonular fibre insertion and the limbus (ZLD) in healthy subjects as a parameter to assess the position of the lens, quantify EL and provide normative data. METHODS: This prospective, observational, cross-sectional study includes one-hundred-fifty eyes of 150 healthy participants (mean

[Modified mini-SLET for treatment of complex cases in pterygium surgery].

BACKGROUND: The major problem associated with the benign but destructive growing pterygium is the high recurrence rate. A new surgical technique to lower recurrence rates is minor ipsilateral simple limbal epithelial transplantation (mini-SLET), where the regeneration potential of limbal stem cells is used in combination with amniotic membrane transplantation (AMT) for surgical reconstruction. The aim of this study is to assess the surgical outcome of the mini-SLET technique with tenonectomy, mitomycin C, and AMT as used in the authors' hospital. MATERIALS AND METHODS: A total of 16 eyes from
