Infection of mice with lactate dehydrogenase-elevating virus destroys the subpopulation of Kupffer cells involved in receptor-mediated endocytosis of lactate dehydrogenase and other enzymes.

In previous experiments in rats, we have shown that the rapid plasma clearance of a number of clinically important enzymes is due to receptor-mediated endocytosis by Kupffer cells and other resident macrophages. Others have shown that infection of mice with lactate dehydrogenase-elevating virus, a virus that proliferates in macrophages, leads to reduced plasma elimination of these enzymes. This paper integrates these two sets of experiments. Plasma elimination of intravenously injected, radioactively labeled lactate dehydrogenase M4 and mitochondrial malate dehydrogenase in mice was shown to b

Studies on the effects of copper deficiency on rat liver mitochondria. II. Effects on oxidative phosphorylation.

Effects of dietary copper deficiency in rats on respiratory enzymes of isolated rat liver mitochondria have been studied. After 2 weeks of Cu-depletion, cytochrome c oxidase (EC 1.9.3.1) activity had declined by 42% and between 4 and 8 weeks exhibited between 20 and 25% of the activity of control mitochondria. Activities of NADH cytochrome c reductase (EC 1.6.99.3) and succinate cytochrome c reductase (EC 1.3.99.1), were unaffected initially but declined by 32 and 46%, respectively, after 8 weeks of Cu-depletion. After 4 weeks there was a significant (34%) decline in succinate supported state

Renal tubular biochemistry during acute and chronic metabolic alkalosis in the dog.

Acute metabolic alkalosis was induced in dogs by the infusion of sodium bicarbonate, 0.3 M. Chronic alkalosis was induced by chloride restriction and the administration of sodium bicarbonate and furosemide. In a third group of dogs, potassium was added to the regimen to prevent frank potassium depletion. Plasma bicarbonate ranged from 29.0 to 32.9 mM. In all three dog groups, renal ammoniagenesis fell by over 30%, which was consistent with a decrease in the renal uptake of glutamine. Glutamate was released in the renal vein and alanine production was decreased. Total production of ammonia was

Alkaline phosphatase synthesis and properties of subcellular organelles during in vitro culture of jejunal biopsies from control subjects and patients with coeliac disease.

Portions of jejunal biopsies from control subjects and from patients with coeliac disease were cultured for 24 hours using an in vitro organ culture technique. Alkaline phosphatase activity was measured in the tissue and medium before and after culture; enzyme activities were expressed per microgram tissue DNA. The increase in enzyme activity during the culture period was taken to represent net enzyme synthesis. Alkaline phosphatase synthesis by mucosa from patients with untreated gluten-sensitive coeliac disease and by mucosa from patients with non-responsive coeliac disease was significantly

Effect of maternal canine starvation on fetal and neonatal liver metabolism.

Heptic glycolytic and gluconeogenic intermediates from fasted newborns of five control and five 3-day starved canine mothers (MCS) were studied at 0, 3, 6, 9, and 24 h of age. MCS did not affect fetal hepatic glycogen concentration; however, a significant increase in uridine diphosphate glucose (UDPG) (0.186 vs. 0.106 mumol/g), fructose 6-phosphate (0.084 vs. 0.034), pyruvate (0.321 vs. 0.126), and citrate (0.190 vs. 0.140) concentrations occurred. At 3 h, the intrahepatic glucose concentration among the MCS newborns declined (3.09 vs. 6.34) and remained lower than the controls for up to 9 h.

MDH2 S246 phosphorylation protects mitochondria against hypoxia/reoxygenation injury due to acetate/lactate metabolism.

Ischemic cardiomyopathy (ICM) is characterized by impaired myocardial function resulting from reduced coronary blood flow, resulting in heart failure. Emerging evidence from proteomic studies indicates that ischemic preconditioning confers cardioprotection against ICM through the regulation of mitochondrial proteins. Proteomic analyses have identified phosphorylation in mitochondrial proteins, including malate dehydrogenase 2 (MDH2), as mediators of cardioprotective mechanisms. However, the mechanism by which MDH2 phosphorylation contributes to cardioprotection in cardiovascular diseases remai

A metabolomics study unravels the hepatotoxic mechanism of diuron in zebrafish: Disruption of glutathione synthesis and mitochondrial energy metabolism.

The liver plays a central role in xenobiotic metabolism and is consequently highly vulnerable to chemical-induced injury. Nevertheless, the mechanisms underlying diuron-induced hepatotoxicity remain poorly understood. Zebrafish (Danio rerio) were exposed to diuron at concentrations of 50 and 500 μg/L for 21 days, with subsequent analysis of the induced hepatotoxicity employing a combination of physiological, biochemical, and metabolomic techniques. Results showed that diuron significantly bioaccumulated in zebrafish, with bioconcentration factor (BCF) values ranging from 14.0 to 40.49 L/kg. Ti

Speciation, Protein Binding, Biotransformation, and Cytotoxicity of a VV-Lactate Complex.

We studied the speciation, protein binding, biotransformation, and cytotoxicity of the dioxidovanadium(V) lactate complex Cs2[VV2O4(lact)2]·2H2O and compared the results with those obtained for the analogous malate compound. 51V NMR and ESI-MS results show that Cs2[VV2O4(lact)2]·2H2O forms [VVO2]+, [H2VVO4]-, [H2VV2O7]2-, [VV2O4(lact)2]2-, [VV3O7(lact)2]3-, [VV4O12]4-, [VV5O15]5-, [VVO2(lact)(H2O)]-, [VVO2(lact)(OH)]2-, and [VV10O28]6- species in aqueous solution. Glycogen deficiency impairs diurnal energy metabolism and cell division in Synechocystis.

Diurnal changes in light availability are a defining feature of life on Earth. Photoautotrophic organisms therefore store reduced carbon during the day to sustain energy metabolism at night. In cyanobacteria, glycogen is the primary carbon storage compound and supports both energy homeostasis and stress responses. Although glycogen-deficient Synechocystis strains have been studied previously, how these mutants cope with the loss of the major daytime carbon sink and can sustain themselves during the night remains unclear. Using single-cell microfluidics, transcriptomics, and metabolomics, we sh

Dominant Effect of Ecological Restoration on Microbial Carbon Cycle in Plant Rhizosphere of Mining Areas.

Rhizosphere microorganisms play critical roles in biogeochemical processes including carbon cycling; however, their linkages to carbon-cycling functions under distinct mine restoration approaches remain unclear. In this study, we compared natural and artificial vegetation restoration via metagenomic sequencing to characterize rhizosphere microbial communities and carbon-cycling functional genes. Artificial restoration (AR) decreased soil electrical conductivity (EC) and salt content (SC) while increasing total phosphorus and available phosphorus by 12.33% and 16.44%, respectively. AR also elev

S-Nitrosylation-mediated differential regulation of STOP1 and STAR1 coordinates external and internal aluminum resistance in Arabidopsis.

Aluminum (Al) toxicity severely limits crop productivity on acidic soils. In Arabidopsis, STOP1 is a key transcription factor that promotes external Al detoxification primarily by inducing ALMT1-mediated malate secretion, whereas the ATP-binding cassette transporter complex STAR1/ALS3 mediates internal detoxification through vacuolar sequestration of Al. However, the regulatory mechanism that coordinates these external and internal detoxification pathways remains poorly understood, particularly with respect to the role of nitric oxide (NO) signaling. In this study, using a forward genetic scre

The PuNAC2-PutDT regulatory module drives late-stage vacuolar citrate accumulation in pear fruit.

In most fleshy fruits, including apple, cherry, plum, and grape, citric acid content declines during the late stages of fruit development, a process that largely determines fruit flavor. In contrast, pear (Pyrus spp.) exhibits a less common developmental pattern characterized by continued citric acid accumulation during late fruit development. Although similar patterns have been reported in a limited number of other fruit species and germplasms, the molecular mechanisms underlying this developmental trajectory remain largely unknown. Here, comparative analyses of organic acid dynamics among cu

Ginkgolide B drives TCA cycle to accelerate Streptococcus suis biofilm dissociation.

Streptococcus suis (S. suis) is an important zoonotic pathogen that often forms biofilm, leading to persistent clinical infections and exacerbation of antibiotic resistance. There is an urgent need to develop novel antibiofilm strategies. In this study, for the first time, lactate dehydrogenase (LDH) was targeted with the aim of reactivating the metabolic activity of bacteria embedded in biofilm using natural product molecules, thereby disrupting mature biofilm. Through virtual screening based on the TCMSP database, molecular docking, and molecular dynamics simulations, two potential molecules

Optimization of permeabilized brain tissue preparation to improve the analysis of mitochondrial oxidative capacities in specific subregions of the rat brain.

BACKGROUND: As the major energy producer of cerebral tissue, mitochondria play key roles in brain physiology and physiopathology. Yet, the fine details of the functioning of mitochondrial oxidative phosphorylation in this organ are still scattered with grey area. This is partly due to the heterogeneity of this tissue that challenges our abilities to study specific cerebral subregions. In the last decades, cerebral mitochondria have largely been studied as a single entity by isolating mitochondria from large sections of brain. Given the evidence that these organelles must adapt to brain areas f

Spatial metabolic gradients in the liver and small intestine.

The properties of mammalian cells depend on their location within organs. Gene expression in the liver varies between periportal and pericentral hepatocytes1-3, and in the intestine from crypts to villus tips4,5. A key element of tissue spatial organization is probably metabolic, but direct assessments of spatial metabolism remain limited. Here we map spatial metabolic gradients in the mouse liver and intestine. We develop an integrated experimental-computational workflow using matrix-assisted laser desorption/ionization (MALDI) imaging mass spectrometry (IMS), isotope tracing and deep-learnin

A heteropolysaccharide from Hypsizygus ulmarius fruit bodies attenuates D-galactose-induced cognitive decline by enhancing mitochondrial function and improving antioxidant activity in mice.

The study aimed to examine the antioxidant properties in vitro and the in vivo D-galactose-induced oxidative damage model from the fruiting bodies of Hypsizygus ulmarius Polysaccharide (HUP), which have an average molecular weight of 2.076 × 103 Da and are primarily composed of glucose and galactose, with minor sugars including xylose, rhamnose, fucose, and mannose. Through in vitro antioxidant tests, HUP exhibited iron-reducing ability, β-carotene bleaching inhibition, and hydroxyl radical scavenging action. Additionally, in vivo antioxidant assays demonstrated that HUP at 200 or 400 mg/kg BW

TCA cycle mode switch determines the fate of pirtobrutinib-tolerant persister cells in mantle cell lymphoma.

Bruton tyrosine kinase inhibitors (BTKis) and cell therapy have successfully been used to treat mantle cell lymphoma (MCL). However, therapy resistance inevitably emerges. Cancer cells can progressively develop stable resistance by traversing through a transient drug-tolerant persister (DTP) state. The mechanisms enabling DTP cells to reversibly adapt to therapies and evolve to acquire heterogeneity remain poorly understood, and characterizing DTP cells in MCL continues to pose a challenge for clinic translation. Here, using pirtobrutinib, a recently US Food and Drug Administration-approved no

Disturbances in mitochondrial quality control and mitochondria-lysosome contact underlie the cerebral cortex and heart damage of mucopolysaccharidosis type II mice.

Mucopolysaccharidosis type II (or Hunter syndrome) is a lysosomal disease caused by mutations in the IDS gene, which encodes the enzyme iduronate 2-sulfatase. MPS II patients present with systemic clinical manifestations and, in the most severe cases, with severe central nervous system abnormalities. Cardiac alterations are also commonly observed. In this study, we evaluated the communication between mitochondria and lysosomes, as well as mitochondrial dynamics and bioenergetics, mitophagy/autophagy, and redox homeostasis in the cerebral cortex and heart of 6-month-old MPS II mice. Our finding

Characterization and Inhibition of the Chaperone Function of Plasmodium falciparum Glucose-Regulated Protein 94 kDa (Pf Grp94).

Plasmodium falciparum expresses four heat shock protein 90 (Hsp90) members. Among these, one, glucose-regulated protein 94 (PfGrp94), is localized in the endoplasmic reticulum (ER). Both the cytosolic and ER-based Hsp90s are essential for parasite survival under all growth conditions. The cytosolic version has been extensively studied and has been targeted in several efforts through the repurposing of anticancer therapeutics as antimalarial drugs. However, PfGrp94 has not been fully characterized and some of its functions related to the ER stress response are not fully understood. Structural a
