A low-cost microfluidic chip for rapid genotyping of malaria-transmitting mosquitoes.

BACKGROUND: Vector control is one of the most effective measures to prevent the transmission of malaria, a disease that causes over 600,000 deaths annually. Around 30-40 Anopheles mosquito species are natural vectors of malaria parasites. Some of these species cannot be morphologically distinguished, but have behavioral and ecological differences. Emblematic of this is the Anopheles gambiae species complex. The correct identification of vector species is fundamental to the development of control strategies and epidemiological studies of disease transmission. METHODOLOGY/PRINCIPAL FINDINGS: An

Predictive spatial models for risk of West Nile virus exposure in eastern and western Colorado.

In the absence of a vaccine for use in humans against West Nile virus (WNV), mosquito control and personal protection against mosquito bites are the only measures available to prevent disease. Improved spatial targeting is desirable for costly mosquito and WNV surveillance and control schemes. We used a multivariate regression modeling approach to develop spatial models predicting high risk of exposure to WNV in western and eastern Colorado based on associations between Geographic Information System-derived environmental data and zip code of residence for 3,659 human WNV disease cases from 200…

Structure of Aedes aegypti procarboxypeptidase B1 and its binding with Dengue virus for controlling infection.

Metallocarboxypeptidases play critical roles in the development of mosquitoes and influence pathogen/parasite infection of the mosquito midgut. Here, we report the crystal structure of Aedes aegypti procarboxypeptidase B1 (PCPBAe1), characterized its substrate specificity and mechanism of binding to and inhibiting Dengue virus (DENV). We show that the activated PCPBAe1 (CPBAe1) hydrolyzes both Arg- and Lys-substrates, which is modulated by residues Asp251 and Ser239 Notably, these residues are conserved in CPBs across mosquito species, possibly required for efficient digestion of basic dietary

The interplay between dose and immune system activation determines fungal infection outcome in the African malaria mosquito, Anopheles gambiae.

The Toll pathway is a central regulator of antifungal immunity in insects. In mosquitoes, the Toll pathway affects infections with the fungal entomopathogen, Beauveria bassiana, which is considered a potential mosquito biopesticide. We report here the use of B. bassiana strain I93-825 in Anopheles gambiae to analyze the impact of Toll pathway modulation on mosquito survival. Exposure to a narrow dose range of conidia by direct contact decreased mosquito longevity and median survival. In addition, fungal exposure dose correlated positively and linearly with hazard ratio. Increased Toll signalin

Laboratory Evaluation of the Rapid Analyte Measurement Platform Assay to Detect Dengue Virus in Mosquito Pools.

We report the results of a laboratory sensitivity and specificity evaluation of the Rapid Analyte Measurement Platform (RAMP®) Dengue Virus (DENV) antigen detection assay, which is designed to detect all serotypes of DENV in mosquito pools. The RAMP DENV assay was able to detect geographically distinct strains of all 4 DENV serotypes in virus-spiked mosquito pools that contained at least 4.3 log10 plaque forming units/ml, although discrete sensitivity limits varied slightly for each serotype. The RAMP DENV assay also detected DENV 1-4 in mosquito pools containing a single infected mosquito and

Engineering resilient gene drives for sustainable malaria control by predicting, testing and overcoming target site resistance in Anopheles gambiae.

CRISPR-based gene drives are selfish genetic elements with the potential to spread through entire insect populations for sustainable vector control. Gene drives designed to disrupt the reproductive capacity of females can suppress laboratory populations of the malaria mosquito, Anopheles gambiae. However, any suppressive intervention will inevitably exert an evolutionary pressure for resistance, and the likelihood of resistance emerging at natural population scales remains poorly defined. Here, we present a pipeline to quantify the evolutionary space for resistance, enabling accelerated discov

Inhibition of butyrylcholinesterase in a population exposed to insecticides during a campaign against Aedes aegypti.

BACKGROUND: Chemical control remains a widely used strategy to reduce morbidity from vector-borne diseases such as dengue. However, the potential health risks to exposed populations necessitates ongoing monitoring, particularly due to the toxicity and volume of insecticides employed. METHODS: This study assessed butyrylcholinesterase activity in residents of a city in southern Mexico during distinct periods of exposure to carbamate and organophosphate insecticides. RESULTS: A statistically significant inhibition of enzymatic activity was observed during the rainy season, which coincided with

Gene drive-based population suppression in the malaria vector Anopheles stephensi.

Gene drives are alleles that can bias the inheritance of specific traits in target populations for the purpose of modification or suppression. Here, we construct a homing suppression drive in the major urban malaria vector Anopheles stephensi targeting the female-specific exon of doublesex, incorporating two gRNAs and a nanos-Cas9 to reduce functional resistance and improve female heterozygote fitness. Our results show that the drive was recessive sterile in both females and males, with various intersex phenotypes in drive homozygotes. Both male and female drive heterozygotes show only moderat

Daytime and public space exposure to Anopheles funestus bites in Western Province, Zambia: implications for malaria surveillance and control.

BACKGROUND: Western Province, Zambia experiences persistent and residual malaria transmission despite high household coverage of core vector control interventions. Standard vector surveillance, conducted overnight at households, indicates that the dominant malaria vector Anopheles funestus sensu stricto (s.s.) bites opportunistically both indoors and outdoors, and remains active throughout the night and into the late morning after the sun rises. This suggests that the full extent of community exposure to An. funestus s.s. bites may not be well characterized in Zambia. This study piloted an exp

Impact of indoor residual spraying on malaria incidence in Ugandan prisons: an interrupted time series analysis.

BACKGROUND: Indoor residual spraying (IRS) is an effective intervention for reducing mosquito vector density and malaria transmission. Uganda Prison Services (UPS) routinely implements IRS for malaria control in main prison facilities; however, no assessment of its impact had been performed. The study assessed the general malaria incidence trends for 5 years and determined the impact of IRS on malaria incidence in the main prison facilities in Uganda. METHODS: This was a cross-sectional study which employed interrupted time series analysis to determine the effect of IRS programme on malaria i

Susceptibility of Aedes aegypti to spinosad larvicide and space spray adulticides in Brazil.

BACKGROUND: Insecticides play a critical role in controlling insect vectors, particularly during epidemics. Effective chemical control relies on the robust monitoring of insecticide resistance to guide evidence-based decision-making in vector control strategies. OBJECTIVES: This study assessed the susceptibility of Aedes aegypti, the primary vector of dengue, Zika, and Chikungunya viruses, to various larvicides and adulticides deployed during Brazil's national campaigns from 2020 to 2023. METHODS: Mosquito collection was performed in 46 Brazilian municipalities using ovitraps. Eggs were tran

Monitoring Dengue Virus in Aedes aegypti to Improve Dengue Surveillance and Control in Puerto Rico.

Aedes aegypti is the primary urban vector for several important arboviruses, including dengue, chikungunya, yellow fever, and Zika viruses. Traditional dengue virus (DENV) surveillance relies on passive reporting of human cases, which often underestimates transmission due to asymptomatic or unreported infections. This study evaluated the utility of monitoring DENV in Ae. aegypti mosquitoes to improve detection of local dengue transmission and inform vector control strategies during the 2024 dengue epidemic in Puerto Rico. Mosquito surveillance was conducted in 15 neighborhoods within the San J

The need for active and integrated involvement of the community and health professionals in the prevention and control of dengue hemorrhagic fever in Indonesia.

Successful control and prevention of dengue fever requires active involvement from all parties. For this reason, three innovative programs are needed, namely: i) increasing knowledge, attitude and practice (KAP) of the community and health professionals as capital in controlling dengue fever in a sustainable manner; ii) application of "3M Plus" to suppress vector breeding in household settings; iii) promotion of the "Jumantik" program as an effective community empowerment approach to prevent and control dengue fever based on community independence. It was concluded that successful control of d

Impacts of ground ultra-low volume adulticide applications on Culex pipiens and Culex restuans (Diptera: Culicidae) abundance, age structure, and West Nile virus infection in Cook County, Illinois.

Since the introduction of West Nile virus (WNV) to the United States over 20 years ago, thousands of cases of human disease and death have been reported. Yearly seasonal outbreaks continue to persist, and the city and suburbs of Chicago, Illinois, is considered a "hot spot" for WNV activity. To interrupt WNV transmission, ground ultra-low volume (ULV) adulticide applications are regularly used to reduce Culex pipiens L. and Culex restuans Theobold (Diptera: Culicidae) abundance and infection. The real-world effectiveness of adulticide applications has not been comprehensively assessed, and pri
