The Mycobacterium tuberculosis membrane is rich in antigens that are potential targets for diagnostics and the development of new vaccines. To better understand the mechanisms underlying MTB virulence and identify new targets for therapeutic intervention, we investigated the differential composition of membrane proteomes between virulent M. tuberculosis H37Rv (MTB) and the Mycobacterium bovis BCG vaccine strain. To compare the membrane proteomes, we used LC-MS/MS analysis in combination with label-free quantitative proteomics, utilizing the area under the curve of the extracted ion chromatogra…

Buruli ulcer, caused by Mycobacterium ulcerans, is a necrotizing skin disease emerging particularly in West Africa. M. bovis BCG vaccine offers only short-term protection against experimental footpad infection of C57BL/6 mice with M. ulcerans, and the duration of this protection cannot be prolonged by a booster vaccination.

Inbred C3H mice were vaccinated intradermally with a single dose of live BCG, whole extracts of mechanically disrupted Mycobacterium lepraemurium (MLM), or a mixture of both these agents. Four weeks later, they were infected in one hind foot pad with freshly harvested MLM. Vaccination with BCG-containing preparations significantly reduced the multiplication of MLM in the infected foot pad and the bacillary dissemination to the draining popliteal lymph node and the spleen, while vaccination with MLM extracts solely limited the growth of MLM in the foot pad. MLM antigens in admixture with BCG di…

Mycobacterium celatum is a slow-growing, non-tuberculous mycobacterium with occasional branching morphology that has been reported mainly in immunocompromised human patients and rarely in other animal species. A 5-y-old female ferret was presented to a veterinary clinic with a history of anorexia and lethargy; peripheral lymphadenopathy, splenomegaly, and pneumonia were detected. Despite treatment, progressive deterioration of the ferret's health led to euthanasia of the animal. Autopsy revealed numerous small 1-4-mm white nodules in the lung, spleen, and kidneys. Histologically, the kidney an…

Mycobacterium tuberculosis primarily causes pulmonary disease but can involve extrapulmonary sites, including bones and joints. Tuberculosis (TB) prosthetic joint infections (TB-PJIs) are rare, often diagnosed late due to low suspicion, indolent progression, and frequent culture negativity. A 73-year-old man from Guyana underwent elective right total knee arthroplasty for osteoarthritis. Postoperatively, he developed a prosthetic joint infection without an identifiable pathogen by routine cultures. Advanced testing detected Mycobacterium tuberculosis complex via next-generation 16S rRNA gene s…

Membrane atg8ylation is a broad homeostatic process of immunological import. It encompasses membrane repair and remodeling pathways, including canonical autophagy, in cells subjected to stress, damage, infection, and immune or metabolic signaling under microbe-induced or sterile inflammatory conditions. The initial reports on autophagy, which is one of membrane atg8ylation outputs, as a defense against Mycobacterium tuberculosis and other intracellular pathogens have ushered a new direction for immunological research but proved to be controversial once the studies have moved from in cellulo to…

This case report describes a rare instance of minimal change disease (MCD) associated with Mycobacterium marinum infection. MCD is typically idiopathic but can occasionally occur secondary to infections. The patient was a 61-year-old fisherman with a history of relapsing MCD who developed a subcutaneous abscess in the lower leg while undergoing rituximab and glucocorticoid therapy. Ziehl-Neelsen staining revealed acid-fast bacilli, and culture at 30 °C isolated M. marinum, a slow-growing nontuberculous mycobacterium (NTM) commonly found in aquatic environments. The infection was attributed to …

Therapies counteracting pathogen-induced immune modulation of the host responses represent a promising improvement for tuberculosis treatments. Despite the documented role of host immune heterogeneity and bacterial genotypic background in determining infection outcomes, their interplay remains largely uncharacterized. We investigated the Mycobacterium tuberculosis (MTB)-human macrophage interaction, considering both macrophage phenotypic variability and MTB genetic diversity. Using single-cell techniques, we show how diverse MTB lineages fine-tune phagosome acidification differently based on m…

Slow growth rates are considered a hallmark of Mycobacterium tuberculosis (Mtb) and have been historically associated with persistence and subsequent drug tolerance. Despite the fundamental role intracellular growth plays in the pathogenesis of tuberculosis (TB), approaches that define intracellular growth rates have remained challenging. Here, we developed a high-throughput, live-cell imaging approach to quantify Mtb replication within human macrophage populations at high spatiotemporal, single-cell resolution. Unexpectedly, we discovered fast-growing intracellular Mtb populations with doubli…

INTRODUCTION: Miliary tuberculosis is a severe form of tuberculosis caused by the hematogenous spread of Mycobacterium tuberculosis (MTB). Nontuberculous mycobacteria (NTM) are environmental pathogens that can become aerosolized and cause infections, particularly in immunocompromised individuals. Mycobacterium Abscessus is a rapidly growing NTM that commonly causes pulmonary disease in individuals with preexisting lung conditions. However, its presentation as miliary nodules in an immunocompetent individual is extremely rare.

Mycobacterium tuberculosis (Mtb) is a successful pathogen that has developed a variety of strategies to survive and replicate within host cells. Among them, resistance mechanisms that allow Mtb to cope with the acidic environment of the endolysosomal network play a crucial role in its host adaptation and pathogenesis. In this Review, we summarize our understanding regarding the molecular factors contributing to Mtb acid resistance and the maintenance of intrabacterial pH homeostasis within in vitro and in cellulo systems. We also discuss some of the recently emerged concepts regarding host ada…

BACKGROUND: New drugs targeting the electron transport chain (ETC) seem to be a promising advance in leprosy treatment. In this study, we evaluated the bactericidal activity of telacebec (TCB), a phase 2 drug candidate for tuberculosis, alongside known ETC-targeting antibiotics, bedaquiline (BDQ) and clofazimine (CFZ), as monotherapy or in combination.
METHODOLOGY/ PRINCIPAL FINDINGS: We used the reference leprosy proportional bactericidal mouse footpad model. Four hundred and ten mice were inoculated in the footpads with 5x104 to 5x100 bacilli of M. leprae strain THAI53 for the untreated con

Mycobacterium tuberculosis, the causative agent of tuberculosis (TB), a leading cause of death by an infectious disease globally, has no efficacious vaccine. Antibodies are implicated in M. tuberculosis control, but the mechanisms of action remain poorly understood. We assembled a library of monoclonal antibodies (mAb) and screened for M. tuberculosis-restrictive activity in mice, identifying protective antibodies targeting diverse antigens. To dissect the mechanism of mAb-mediated M. tuberculosis restriction, we optimized a protective lipoarabinomannan-specific mAb, generating Fc variants. In…

Interferon-induced protein with tetratricopeptide repeat 2 (IFIT2) is known for its antiviral activity and has recently been implicated in the defense against Mycobacterium tuberculosis (M. tb). This study employed global transcriptomics to investigate the downstream effects of IFIT2 induction in THP-1 macrophages infected with R179 M. tb, aiming to elucidate its role and downstream contributing factors that aid it in intracellular M. tb killing. Using a vector-based overexpression approach, IFIT2 expression was induced in THP-1 cells infected with R179 M. tb, followed by RNA extraction 12 hou…
