Protective activities of ellagic acid and urolithins against kidney toxicity of environmental pollutants: A review.

Oxidative stress and inflammation are two possible mechanisms related to nephrotoxicity caused by environmental pollutants. Ellagic acid, a powerful antioxidant phytochemical, may have great relevance in mitigating pollutant-induced nephrotoxicity and preventing the progression of kidney disease. This review discusses the latest findings on the protective effects of ellagic acid, its metabolic derivatives, the urolithins, against kidney toxicity caused by heavy metals, pesticides, mycotoxins, and organic air pollutants. We describe the chelating, antioxidant, anti-inflammatory, antifibrotic, a…

Honey bee toxicology.

Insecticides are chemicals used to kill insects, so it is unsurprising that many insecticides have the potential to harm honey bees (Apis mellifera). However, bees are exposed to a great variety of other potentially toxic chemicals, including flavonoids and alkaloids that are produced by plants; mycotoxins produced by fungi; antimicrobials and acaricides that are introduced by beekeepers; and fungicides, herbicides, and other environmental contaminants. Although often regarded as uniquely sensitive to toxic compounds, honey bees are adapted to tolerate and even thrive in the presence of toxic …

Mulberry Leaf Extracts Ameliorate Aflatoxin B1-Induced Liver Injury by Modulating Nrf2 Pathway and Gut Microbiota.

Mulberry leaf, an edible medicinal plant, is traditionally recognized for its diverse biological functions, including antioxidant and anti-inflammatory effects. Aflatoxin B1 (AFB1), a prevalent foodborne mycotoxin, causes oxidative stress, inflammation, and liver injury. However, the role of functional components of mulberry leaf in AFB1-induced liver injury is still elusive. This study investigated the hepatoprotective potential of mulberry complex extract (MCE) and mulberry leaf polysaccharide (MLP) in a murine model of AFB1-induced liver injury. Twenty-four male mice were randomly divided i

Dietary curcumin alleviates aflatoxin B1-induced liver injury and improves growth performance in broilers via Nrf2 activation and NLRP3 inflammasome inhibition.

BACKGROUND: Aflatoxin B1 (AFB1), a potent mycotoxin frequently detected in improperly stored poultry feed, induces hepatic oxidative stress, inflammation, and growth retardation in broilers. Curcumin (CUR), a polyphenolic compound derived from Curcuma longa, exhibits strong antioxidant and anti-inflammatory activities; however, its efficacy and mechanisms in mitigating AFB1-induced hepatotoxicity in broilers remain unclear. This study aimed to evaluate the protective effects of dietary CUR supplementation on growth performance and liver health of broilers exposed to AFB1. RESULTS: A total of

Estragole Inhibits Growth and Aflatoxin Biosynthesis of Aspergillus flavus by Affecting Reactive Oxygen Species Homeostasis.

A variety of essential oils and edible compounds have been widely recognized for their antifungal activity in recent years. In this study, we explored the antifungal activity of estragole from Pimenta racemosa against Aspergillus flavus and investigated the underlying mechanism of action. The results showed that estragole had significant antifungal activity against A. flavus, with a minimum inhibitory concentration of 0.5 μL/mL against spore germination. Additionally, estragole inhibited the biosynthesis of aflatoxin in a dose-dependent manner, and aflatoxin biosynthesis was significantly inhi

Regulation of gliotoxin biosynthesis and protection in Aspergillus species.

Aspergillus fumigatus causes a range of human and animal diseases collectively known as aspergillosis. A. fumigatus possesses and expresses a range of genetic determinants of virulence, which facilitate colonisation and disease progression, including the secretion of mycotoxins. Gliotoxin (GT) is the best studied A. fumigatus mycotoxin with a wide range of known toxic effects that impair human immune cell function. GT is also highly toxic to A. fumigatus and this fungus has evolved self-protection mechanisms that include (i) the GT efflux pump GliA, (ii) the GT neutralising enzyme GliT, and (i…

Identification and quantification of aflatoxins and aflatoxicol from poultry feed and their recovery in poultry litter.

Aflatoxins (AF) are toxic fungal secondary metabolites and are known mycotoxins pathological to animals and humans. Poultry litter is frequently used as a food supplement for ruminants, and when poultry feed contains AF, the litter becomes contaminated as well, thus having an effect on livestock health. This study identified and quantified AF (AFB(1), AFB(2), AFG(1), and AFG(2)) from poultry feed and their recovery, together with their metabolites (AFM(1), AFM(2), AFP(1), and aflatoxicol) in litter. An experiment with 25 Hy-Line W-36 hens, in their second production stage, 121 wk old, was carr…

Development of a urinary biomarker of human exposure to deoxynivalenol.

Deoxynivalenol (DON) is a mycotoxin frequently found as a contaminant of cereal crops and may be etiologically associated with adverse health effects in developing countries where considerable quantities of contaminated crops are consumed. We investigated the metabolism of DON in rats as a basis to establish methodology for a candidate biomarker of human exposure to this toxin and tested this methodology on urine samples from a potentially highly exposed population. Sprague-Dawley rats received a single dose of [14C]DON (5.0+/-0.1 mg/kg body weight, 5.5+/-0.1 microCi/kg) and the distribution o

A distinct zearalenone detoxification strategy mediated by cytochrome P450.

Developing the bio-detoxification of estrogenic mycotoxin zearalenone (ZEN) and its more potent metabolite α-zearalenol (α-ZOL) represents a research priority in food safety. Here, we report a variant cytochrome P450 enzyme termed T8F3 that consumes 54% ZEN and 63.9% α-ZOL in an initial screening assay. The hydroxylated products of ZEN and α-ZOL, termed ZEN-P and ZOL-P, respectively, were isolated, purified and structurally characterized. The estrogenicity of ZEN-P and ZOL-P is suppressed by 21- and 105-fold, respectively, compared with their parental compounds. Structural elucidation shows th

Probing human tubulins-phomopsins interaction: An in silico approach for a mechanism-based hazard prioritisation of the phomopsin family.

Phomopsins are mycotoxins mainly contaminating lupin-derived food matrices posing safety concerns, with phomopsin A considered the most potent congener. Their toxicity is linked to disruption of microtubule dynamics, yet the shortage of toxicokinetic and toxicodynamic data prevents adequate human risk assessment, representing a critical gap for food safety. To bridge this data gap through New Approach Methodologies (NAMs), a computational 3D modelling pipeline, combining molecular dynamics simulations and binding free energy calculations, was applied to investigate the interactions of phomopsi…

Enniatin B attenuates deoxynivalenol-induced toxicity in liver cells and modulates lipid metabolism-related pathways.

Mycotoxin contamination of food and feed is an increasing global health concern, particularly due to Fusarium toxins such as the regulated deoxynivalenol (DON) and the emerging enniatin B (ENNB). However, the combined mechanisms of action of these toxins remain poorly understood. This study aimed to assess their toxic effects, alone and in combination, using the HepaRG liver cell model. Toxicity, individually and in combination at concentrations equivalent to those used in single exposures (5:1 DON to ENNB ratio), was assessed in HepaRG cells. Several endpoints were investigated, including via

Emerging Alternaria Toxins Drive LX-2 Cells Transdifferentiation into Myofibroblasts for Liver Fibrosis and CotA Detoxification.

Alternariol (AOH), alternariol monomethyl ether (AME), and tenuazonic acid (TeA) are three major emerging Alternaria toxins in food. In this lncRNA-mRNA omics-based study, we first decrypt the blueprint: how emerging Alternaria toxins induce hepatic stellate cell LX-2 transdifferentiation for liver fibrosis. AOH and AME caused the fibrotic marker α-smooth muscle actin, extracellular matrix collagen expression, and cell contraction, while TeA had no significant effect. Mechanistically, AOH, AME, and AAT (their combination) activated the NF-κB pathway and induced ferroptosis and AMPK/AKT/m-TOR-r

T-Toxin Virulence Genes: Unconnected Dots in a Sea of Repeats.

In 1970, the Southern Corn Leaf Blight epidemic ravaged U.S. fields to great economic loss. The outbreak was caused by never-before-seen, supervirulent, Race T of the fungus Cochliobolus heterostrophus. The functional difference between Race T and O, the previously known, far less aggressive strain, is production of T-toxin, a host-selective polyketide. Supervirulence is associated with ~1 Mb of Race T-specific DNA; only a fraction encodes T-toxin biosynthetic genes (Tox1). Tox1 is genetically and physically complex, with unlinked loci (Tox1A, Tox1B) genetically inseparable from breakpoints of

The Dual Function of the Fungal Toxin Candidalysin during Candida albicans-Macrophage Interaction and Virulence.

The dimorphic fungus Candida albicans is both a harmless commensal organism on mucosal surfaces and an opportunistic pathogen. Under certain predisposing conditions, the fungus can overgrow the mucosal microbiome and cause both superficial and life-threatening systemic infections after gaining access to the bloodstream. As the first line of defense of the innate immune response, infecting C. albicans cells face macrophages, which mediate the clearance of invading fungi by intracellular killing. However, the fungus has evolved sophisticated strategies to counteract macrophage antimicrobial acti
