UNLABELLED: The process of extravasation of the high metastatic Lewis lung carcinoma line was examined in different organs. Four of the five organs (liver, lungs, brain and adrenals) represent the most frequent metastatic sites in humans. In the case of each organ 150-350 tumor cells were analysed. Carcinogenic effect of N-ethyl-N-nitrosourethan (ENUR) and N-amyl-N-nitrosourethan (ANUR) was examined by continuous oral administration or topical application to female Donruy rats. The frequency of spontaneously occuring neoplasms in the urogenital system and endocrine organs was examined in two Syrian hamster colonies [Eppley colony (EC); Hannover colony (HC)]. Molecular genetic studies of C. elegans vulval development have helped to define an evolutionarily conserved signaling pathway from an EGF-like ligand through EGF-receptor, Ras and MAP kinase to the nucleus. Further studies have identified novel positive regulators such as KSR-1 and SUR-8/SOC-2 and negative regulators such as cbl/SLI-1. The many negative regulatory proteins might serve to prevent inappropriate signaling, and thus are analogous to tumor suppressor genes.
