Melanocortin receptor 4 is induced in nerve-injured motor and sensory neurons of mouse.

We previously identified melanocortin receptor 4 (MC4R) in a search for genes associated with hypoglossal nerve regeneration. As melanocortins promote nerve regeneration after axonal injury, we investigated whether MC4R functions as a key receptor for peripheral nerve regeneration. In situ hybridization revealed that MC4R mRNA is induced in mouse hypoglossal motor neurons after axonal injury, whereas mRNAs for MC1R, MC2R, MC3R, and MC5R are not expressed either before or after nerve injury. This result was confirmed by RT-PCR. The level of MC4R mRNA expression increased significantly from day

Increased expression and localization of the RNA-binding protein HuD and GAP-43 mRNA to cytoplasmic granules in DRG neurons during nerve regeneration.

The neuronal-specific RNA-binding protein, HuD, binds to a U-rich regulatory element of the 3' untranslated region (3' UTR) of the GAP-43 mRNA and delays the onset of its degradation. We have recently shown that overexpression of HuD in embryonic rat cortical cells accelerated the time course of normal neurite outgrowth and resulted in a twofold increase in GAP-43 mRNA levels. Given this evidence, we sought to investigate the involvement of HuD during nerve regeneration. It is known that HuD protein and GAP-43 mRNA are expressed in the dorsal root ganglia (DRG) of adult rat and that GAP-43 is

An Optimized and Detailed Step-by-Step Protocol for the Analysis of Neuronal Morphology in Golgi-Stained Fetal Sheep Brain.

Antenatal brain development during the final trimester of human pregnancy is a time when mature neurons become increasingly complex in morphology, through axonal and dendritic outgrowth, dendritic branching, and synaptogenesis, together with myelin production. Characterizing neuronal morphological development over time is of interest to developmental neuroscience and provides the framework to measure gray matter pathology in pregnancy compromise. Neuronal microstructure can be assessed with Golgi staining, which selectively stains a small percentage (1-3%) of neurons and their entire dendritic

Biallelic Mutations in TSC2 Lead to Abnormalities Associated with Cortical Tubers in Human iPSC-Derived Neurons.

Tuberous sclerosis complex (TSC) is a genetic disorder caused by mutations in TSC1 or TSC2 Patients frequently have epilepsy, autism spectrum disorder, and/or intellectual disability, as well as other systemic manifestations. In this study, we differentiated human induced pluripotent stem cells (iPSCs) from a female patient with TSC with one or two mutations in TSC2 into neurons using induced expression of NGN2 to examine neuronal dysregulation associated with the neurological symptoms in TSC. Using this method, neuronal differentiation was comparable between the three genotypes of iPSCs. We o

A new assay for nerve fiber repulsion.

In inflammatory lesions, sympathetic nerve fibers get lost soon after the start of inflammation. To be able to identify and examine the factors that are responsible for this repulsion of the sympathetic nerve fibers we established an in vitro assay. Sympathetic trunk ganglia of postnatal mice were used for the outgrowth of axons. They were plated on poly-D-lysine-coated culture slides. Axons were encouraged to grow out by the addition of nerve growth factor-7S (NGF) from murine submaxillary gland. Using live imaging it was clearly shown that SEMA3F is a nerve-repellent factor. Two different ty

Human pathologies associated with defective RNA transport and localization in the nervous system.

RNA localization is emerging as an important process to restrict certain proteins to specific subcellular domains and thus spatially control the expression of genes within cells. It is used, for instance, to compartmentalize the developing embryo during early embryogenesis. The localization of RNA also plays important roles later during development, such as in asymmetric cell divisions, cell migration and the outgrowth and pathfinding of axons and dendrites. In differentiated cells, it serves to subdivide the cell into functionally distinct compartments. For example, in mature neurons it is be

NRN1 inhibits neuronal apoptosis and improves motor recovery after spinal cord injury via binding to FZD2 to activate the GSK-3β/Nrf2 pathway.

Neuritin 1 (NRN1) is a neurotrophic factor known to protect neurons from damage and cell death; however, its role and mechanism in cervical spinal cord injury (CSCI) remain unclear. In this study, we investigated the function of NRN1 using oxygen-glucose deprivation (OGD)-treated PC12 cells and a rat model of CSCI established by anterior cervical screw compression. In vitro, NRN1 overexpression promoted neurite outgrowth, attenuated oxidative stress, and inhibited apoptosis in OGD‑exposed PC12 cells, while NRN1 knockdown showed opposite results. Mechanistically, co-immunoprecipitation and immu

A lesion-responsive ZIF-8/zinc-myricetin nano-biointerface remodels mitochondrial-immune crosstalk for spinal cord repair.

Spinal cord injury (SCI) creates a hostile lesion microenvironment characterized by oxidative stress, mitochondrial dysfunction, inflammatory metabolic remodeling, and pyroptotic cell death, which collectively restrict neural repair. Here, we engineered a lesion-responsive ZIF-8/zinc-myricetin nano-biointerface, AST@ZIF-8@Zn-MYR, to coordinate redox regulation, mitochondrial protection, and immune microenvironment remodeling after SCI. Astaxanthin was encapsulated within a ZIF-8 core to improve cargo protection and enable pH/ROS-responsive release, while a zinc-myricetin coordination shell was

miR-221 activates Sox11 to reduce brain injury after intracerebral hemorrhage via inhibiting neuroinflammation.

As a global public health issue, intracerebral hemorrhage (ICH) is characterized by high morbidity and mortality. Brain injury following ICH is composed of primary and secondary injury, with the latter being more severe and resulting in increased apoptosis. Sox11 (sex-determining region Y-related high-mobility-group 11), a vital member of the Sox gene family, is broadly discovered in the developing nervous system and may have a vital impact on neurogenesis, neuronal survival, and neurite outgrowth. The level and impacts of Sox11 in brain with ICH remain indistinct. The major objective of the c

A Carboxy-terminal Smarcb1 Point Mutation Induces Hydrocephalus Formation and Affects AP-1 and Neuronal Signalling Pathways in Mice.

The BAF (BRG1/BRM-associated factor) chromatin remodelling complex is essential for the regulation of DNA accessibility and gene expression during neuronal differentiation. Mutations of its core subunit SMARCB1 result in a broad spectrum of pathologies, including aggressive rhabdoid tumours or neurodevelopmental disorders. Other mouse models have addressed the influence of a homo- or heterozygous loss of Smarcb1, yet the impact of specific non-truncating mutations remains poorly understood. Here, we have established a new mouse model for the carboxy-terminal Smarcb1 c.1148del point mutation, w

Low-Intensity Pulsed Ultrasound Enhanced Neurite Guidance Growth through Netrin-1/DCC Signal Pathway in Primary Cultured Cortical Neurons of Rats.

Low-intensity pulsed ultrasound is found to be effective in axonal regeneration, while the role of ultrasound in axonal growth guidance is still unclear. This study was performed to explore the neuroprotective role of low-intensity pulsed ultrasound (US) both in vitro and in vivo. Primary cultured rat cortical neurons were subjected to 1.0 MHz ultrasound for 5 min every day at intensity of 0, 0.008, 0.12, and 0.21 W/cm2. Our results demonstrated that low-intensity pulsed ultrasound significantly increased neuronal cell viability and inhibited neuronal apoptosis in vitro as determined by fluore

T-Brain-1--A Potential Master Regulator in Autism Spectrum Disorders.

T-Brain-1 (TBR1), a causative gene in autism spectrum disorders (ASDs), encodes a brain-specific T-box transcription factor. It is therefore possible that TBR1 controls the expression of other autism risk factors. The downstream genes of TBR1 have been identified using microarray and promoter analyses. In this study, we annotated individual genes downstream of TBR1 and investigated any associations with ASDs through extensive literature searches. Of 124 TBR1 target genes, 23 were reported to be associated with ASDs. In addition, one gene, Kiaa0319, is a known causative gene for dyslexia, a dis

WT1 and its transcriptional cofactor BASP1 redirect the differentiation pathway of an established blood cell line.

The Wilms' tumour suppressor WT1 (Wilms' tumour 1) is a transcriptional regulator that plays a central role in organogenesis, and is mutated or aberrantly expressed in several childhood and adult malignancies. We previously identified BASP1 (brain acid-soluble protein 1) as a WT1 cofactor that suppresses the transcriptional activation function of WT1. In the present study we have analysed the dynamic between WT1 and BASP1 in the regulation of gene expression in myelogenous leukaemia K562 cells. Our findings reveal that BASP1 is a significant regulator of WT1 that is recruited to WT1-binding si

Axonal growth and guidance defects in Frizzled3 knock-out mice: a comparison of diffusion tensor magnetic resonance imaging, neurofilament staining, and genetically directed cell labeling.

Previous work has identified axonal outgrowth and/or guidance defects in the brain and spinal cord of prenatal Frizzled3 (Fz3)(-/-) mice. To systematically explore the axonal defects in Fz3(-/-) mice and to compare techniques for the global assessment of axon tracts in the developing mouse, we have analyzed wild-type and Fz3(-/-) brains using (1) diffusion tensor magnetic resonance imaging (muDTI), (2) neurofilament staining, and (3) two genetically directed neuronal labeling methods. Confirming and extending the previous work of Wang et al. (2002), we find that the following structures/tracts

Sciatic conditioned medium increases survival, proliferation and differentiation of retinal cells in culture.

Many evidences clearly demonstrate that Schwann cells provide trophic support for neurons. Different cytokines, including neurotrophins (NTs), are produced and released by Schwann cells. These trophic molecules play an important role on neuronal survival either during the development or during adult life. Cytokines have also a pivotal role on neuronal regeneration after lesions occurring during pathological conditions. The aim of this work was to study the effect of sciatic conditioned medium (SCM) on rat retinal cells maintained in culture. Our results show that treatment with SCM obtained af

MiR-126 Contributes to Human Umbilical Cord Blood Cell-Induced Neurorestorative Effects After Stroke in Type-2 Diabetic Mice.

Diabetes mellitus (DM) is a high risk factor for stroke and leads to more severe vascular and white-matter injury than stroke in non-DM. We tested the neurorestorative effects of delayed human umbilical cord blood cell (HUCBC) treatment of stroke in type-2 diabetes (T2DM). db/db-T2DM and db/+-non-DM mice were subjected to distal middle cerebral artery occlusion (dMCAo) and were treated 3 days after dMCAo with: (a) non-DM + Phosphate buffered saline (PBS); (b) T2DM + PBS; (c) T2DM + naïve-HUCBC; (d) T2DM + miR-126(-/-) HUCBC. Antioxidant suppression of hyperoxia-induced reactive oxygen species in cultured rat hippocampal neurons.

Central nervous system oxygen toxicity (CNS-OT) is driven, in part, by excess reactive oxygen species (ROS) generated during hyperoxic exposure. Herein, we developed an in vitro screening platform using embryonic rat hippocampal neurons to evaluate antioxidant candidates under hyperoxic stress (1.34 ATA) and quantify ROS by Dihydrorhodamine (DHR)123 fluorescence imaging and flow cytometry. Neuronal cultures remained viable throughout the assay, with approximately 87% anti-βIII tubulin staining efficiency and 88% neurite outgrowth-associated viability. Among the compounds tested, sulforaphane a

Fibroblast growth factor 21 affects the colocalization of post-synaptic density 95 and Kalirin7 through CYLD-expression regulation.

Endocrine Fibroblast Growth Factor21 (FGF21) is mainly produced in the liver and regulates energy metabolism. Although FGF21 is a systemic signaling factor that functions in the central nervous system (CNS), its precise role remains unknown. We have previously reported that FGF21 regulates the expression of genes associated with neurite outgrowth and is involved in the induction of growth hormone-releasing hormone (GHRH) secretion in the hypothalami of neonatal mice. In this study, we aimed to clarify the role of FGF21 in the neonatal mouse hypothalamus by identifying the regulatory mechanism

Loss of Zmiz1 in mice leads to impaired cortical development and autistic-like behaviors.

De novo mutations in transcriptional regulators are emerging as key risk factors contributing to the etiology of neurodevelopmental disorders. Human genetic studies have recently identified ZMIZ1 and its de novo mutations as causal of a neurodevelopmental syndrome strongly associated with intellectual disability, autism, ADHD, microcephaly, and other developmental anomalies. However, the role of ZMIZ in brain development or how ZMIZ1 mutations cause neurological phenotypes is unknown. Here, we generated a forebrain-specific Zmiz1 mutant mouse model that develops brain abnormalities, including

Highly specific σ2R/TMEM97 ligand FEM-1689 alleviates neuropathic pain and inhibits the integrated stress response.

The sigma 2 receptor (σ2R) was described pharmacologically more than three decades ago, but its molecular identity remained obscure until recently when it was identified as transmembrane protein 97 (TMEM97). We and others have shown that σ2R/TMEM97 ligands alleviate mechanical hypersensitivity in mouse neuropathic pain models with a time course wherein maximal antinociceptive effect is approximately 24 h following dosing. We sought to understand this unique antineuropathic pain effect by addressing two key questions: do these σ2R/TMEM97 compounds act selectively via the receptor, and what is t
