Differences in acute hepatotoxicity of carbon tetrachloride (CCl4) and dimethylnitrosamine (DMN) have been tested in normal foetal, newborn and adult rats, foetal, newborn and adult rats pretreated with phenobarbitone and partially hepatectomized adult rats. While CCl4 is non-toxic to the foetal and newborn liver, DMN induces identical necrosis at all ages. Prior dosing with phenobarbitone augments CCl4 toxicity only in the adult and the newborn but the foetus continues to be resistant. The carcinogenic effects of dimethylnitrosamine (DMN) and nitrosomethylurea (NMU) injected subcutaneously at 3 different dose levels in European hamsters were studied. DMN induced malignant haemangioendotheliomata of the liver and kidney, hepatocellular carcinomata and, in one animal, a cholangiocellular carcinoma. The effect of NMU was localized at the site of administration and resulted in subcutaneous fibrosarcomata, carcinosarcomata or epidermal carcinomata.

The purpose of this paper is to review the epidemiologic evidence for the effects of tobacco use and tobacco use cessation on a variety of oral diseases and conditions. Exposures considered include cigarette and bidi smoking, pipe and cigar smoking, and smokeless tobacco use. To study growth regulation in the beginning of carcinogenesis, we established a novel ex vivo model for co-cultivation of normal and putatively initiated hepatocytes. Rats received the genotoxic hepatocarcinogen N-nitrosomorpholine (NNM). This led to the appearance of hepatocytes expressing placental glutathione S-transferase (G(+) cells).
