Nasal Lavage IL-4 and IL-13 in Cystic Fibrosis-Related Chronic Rhinosinusitis: Associations With Disease Severity and Potential Implications for Targeted Therapies.

BACKGROUND: Chronic rhinosinusitis (CRS) is highly prevalent in patients with cystic fibrosis (CF) and has traditionally been considered a predominantly neutrophilic condition. Emerging evidence, however, suggests that type-2 inflammatory pathways may contribute to disease heterogeneity. Data linking local inflammatory mediators with objective sinonasal disease severity and olfactory dysfunction in CF-associated CRS remain limited. METHODS: In this cross-sectional study, 42 adults with CF-associated CRS (22 with nasal polyps and 20 without) underwent assessment of disease severity using the n

Changes in olfactory function and serum interleukin-6 levels in the acute phase of bipolar I disorder: a longitudinal study.

OBJECTIVES: In this study, we horizontally compared olfactory function and interleukin-6 levels in bipolar disorder (BD) patients with manic and depressive episodes with those in healthy controls. We also compared these variables longitudinally between patients in the acute versus euthymic phase, and assessed the correlation between olfactory function and interleukin-6 in BD patients overall. The purpose of this study was to search potential biomarkers for early diagnosis and efficacy evaluation of BD, and to provide more clinical evidence for exploring the hypothesis of neuroimmune pathways i

The Role of BDNF as a Biomarker in Cognitive and Sensory Neurodegeneration.

Brain-derived neurotrophic factor (BDNF) has a crucial function in the central nervous system and in sensory structures including olfactory and auditory systems. Many studies have highlighted the protective effects of BDNF in the brain, showing how it can promote neuronal growth and survival and modulate synaptic plasticity. On the other hand, conflicting data about BDNF expression and functions in the cochlear and in olfactory structures have been reported. Several clinical and experimental research studies showed alterations in BDNF levels in neurodegenerative diseases affecting the central

[Studies on chemotactic behaviors of Strongyloides ratti infective larvae in vitro and migration routes of the larvae in rat head].

For the purpose of clarifying host-finding mechanisms, the infective larvae of Strongyloides ratti were run on an agarose plate which had been placed various attractants around the larvae. They were accumulated at the sites of sodium compounds, serum proteins and albumins. At 20, 30, 40 hours after infection, rats were killed and heads were separated. They were then processed for thin section specimens. Larvae were found in brains, olfactory bulbs and its nerves. These larvae seemed to move along olfactory nerves during descending from the cranial cavity. Another migration study was done by wh

The interleukin-4/interleukin-13 pathway in type 2 inflammation in chronic rhinosinusitis with nasal polyps.

Chronic rhinosinusitis with nasal polyps (CRSwNP) is predominantly a type 2 inflammatory disease associated with type 2 (T2) cell responses and epithelial barrier, mucociliary, and olfactory dysfunction. The inflammatory cytokines interleukin (IL)-4, IL-13, and IL-5 are key mediators driving and perpetuating type 2 inflammation. The inflammatory responses driven by these cytokines include the recruitment and activation of eosinophils, basophils, mast cells, goblet cells, M2 macrophages, and B cells. The activation of these immune cells results in a range of pathologic effects including immunog

radish encodes a phospholipase-A2 and defines a neural circuit involved in anesthesia-resistant memory.

Источник пометил публикацию как отозванную

BACKGROUND: In both vertebrate and invertebrate animals, anesthetic agents cause retrograde amnesia for recently experienced events. In contrast, older memories are resistant to the same treatments. In Drosophila, anesthesia-resistant memory (ARM) and long-term memory (LTM) are genetically distinct forms of long-lasting memory that exist in parallel for at least a day after training. ARM is disrupted in radish mutants but is normal in transgenic flies overexpressing a CREB repressor transgene. In contrast, LTM is normal in radish mutants but is disrupted in CREB repressor transgenic flies. To

Early olfactory involvement in Alzheimer's disease.

BACKGROUND: In Alzheimer's disease (AD) the olfactory system, including the olfactory bulb, a limbic paleocortex is severely damaged. The occurrence of early olfactory deficits and the presence of senile plaques and neurofibrillary tangles in olfactory bulb were reported previously by a few authors. The goal of the present study was to analyze the occurrence of AD-type degenerative changes in the peripheral part of the olfactory system and to answer the question whether the frequency and severity of changes in the olfactory bulb and tract are associated with those of the cerebral cortex in AD.

Development of P2 olfactory glomeruli in P2-internal ribosome entry site-tau-LacZ transgenic mice.

Primary olfactory neurons project their axons to the olfactory bulb, where they terminate in discrete loci called glomeruli. All neurons expressing the same odorant receptor appear to terminate in a few glomeruli in each olfactory bulb. In the P2-IRES-tau-LacZ line of transgenic mice, LacZ is expressed in the perikarya and axons of primary olfactory neurons that express the P2 odorant receptor. In the present study, we examined the developmental appearance of P2 neurons, the topographical targeting of P2 axons, as well as the formation of P2 glomeruli in the olfactory bulb. P2 axons were first

Inhibition of olfactory cyclic nucleotide-activated current by calmodulin antagonists.

1. In amphibian olfactory receptor neurones, much of the depolarizing current in response to odours is carried by cationic channels that are directly gated by cyclic AMP. The effects of four calmodulin antagonists on the cyclic AMP-activated receptor current were studied in single olfactory cilia of the frog. 2. Two antagonists, W-7 and trifluoperazine, were potent and reversible inhibitors of the cyclic AMP-activated current. IC50 values were 5 microM for W-7 and 13 microM for trifluoperazine. A third antagonist, calmidazolium, irreversibly blocked the current. The fourth, mastoparan, had lit

SARS-CoV-2 infects olfactory neurons and basal stem cells and induces axonal degeneration through TRPV1 activation.

Neurological complications such as anosmia are among the most frequent and persistent symptoms of COVID-19; yet, the mechanisms linking SARS-CoV-2 infection to sensory neuronal injury remain unclear. We demonstrate that SARS-CoV-2 directly infects olfactory sensory neurons through ACE2 upregulation and stimulates basal stem cell proliferation, as shown in human iPSC-derived sensory neurons and validated in golden Syrian hamsters. Exposure to live viruses or its S1 spike protein induces TRPV1 channel redistribution from the nucleus to the plasma membrane, resulting in axonal degeneration. Singl

Dissociated Biologic Responses Across the United Airway: A Case Report of Severe Asthma With Chronic Rhinosinusitis With Nasal Polyps and Eosinophilic Otitis Media.

Asthma, chronic rhinosinusitis with nasal polyps (CRSwNP), and eosinophilic otitis media (EOM) share type 2 inflammatory pathways. Although type 2-targeted biologic therapies have shown efficacy for these diseases, some patients exhibit dissociated responses to a single biologic. A case of severe asthma with CRSwNP and EOM that showed dissociated responses to biologic therapies and was successfully managed by biologic switching is reported. A 55-year-old man with asthma, recurrent CRSwNP requiring multiple surgeries and oral corticosteroid-dependent EOM was referred to our hospital. Nineteen m

Breastfeeding, bonding, and olfaction: unlocking the potential of mother-infant odour exchange.

Breastfeeding is crucial for infant survival, growth, and health, and it enhances maternal-infant bonding and well-being. However, breastfeeding rates typically fall below international targets, partly due to a high prevalence of latching difficulties, intermittent sucking, refusing the breast, or poor milk supply. Here, we propose that such uniquely human difficulties might be ameliorated by recognising, understanding, and facilitating the olfactory mechanisms that, in other mammals, regulate breastfeeding initiation and maternal-infant relationships in the first weeks of life. We briefly rev

Sufficient Gas and Solvated Ion Transport Assisted Rapid NH3 Detection at Room Temperature Through Bionic Olfactory Fibres.

The human olfactory sensing system, based on ionic signal transmission, is featured with fastness, high efficiency and low energy consumption. However, bionic gas sensing materials exhibit performance limitations compared to materials based on electronic signal transmission. Herein, bionic olfactory fibres are prepared by electrospinning for rapid gas sensing at the ppb level, which consist of confined ionic liquids (ILs) within nano spacing in a polymer matrix. The fibres showed a high response (69.29%) to 500 ppb NH3, ultrafast response (4 s) and a low theoretical limit of detection (45 ppb)

Worms like catnip too! Identification of a new odor attractant in C. elegans.

Organisms across the phyla are capable of sensing an array of sensory cues to control, or shape behavioral responses in order to survive in a complex environment consisting of an array of attractive and repulsive dangerous cues. Mammalian systems extensively use olfactory and gustatory behavior to fine tune sensory-dependent decision-making behaviors. Despite understanding the importance of behavioral responses to cues in the form of odors in shaping decision-making behavior. The underlying mechanisms that mediate these responses at the level of sensation, processing, integration, and modulati

beta-amyloid deposition and neurofibrillary tangle formation in the olfactory bulb in ageing and Alzheimer's disease.

Impaired olfaction, hyposmia or anosmia are part of the clinical phenotype in neurodegenerative disorders including Alzheimer's disease (AD). It has been proposed that the most severely affected areas are interconnected with the central olfactory system in contrast to the relative sparing of other sensory areas which lack olfactory connections. The pathology of the first synaptic relay in the olfactory pathway, the olfactory bulb (OB), has been studied in AD, but the results have been inconsistent. In order to define more fully the pathology of the OB, we analysed 15 AD and 15 control cases, u

The Neural and Perceptual Effects of Stevia During Retronasal Occlusion.

We have recently shown that occluding retronasal pathways with a nose clip reduces both the subjective and neural responses to sucrose, suggesting the involvement of retronasal pathways in sucrose perception. However, how other sweet tastes such as stevia might also be affected by retronasal occlusion at the subjective and neural level is unknown. We examined the neural activity to stevia with a nose clip on (blocking retronasal pathways) and nose clip off, in a robust sample of healthy adults (N = 34, mean 25 years). Neural activity to stevia was reduced with the nose clip on in the olfactory

Potential pharmacologic treatments for COVID-19 smell and taste loss: A comprehensive review.

The acute loss of taste and smell following COVID-19 are hallmark symptoms that affect 20-85% of patients. However, the pathophysiology and potential treatments of COVID-19 smell and taste loss are not fully understood. We searched the literature to review the potential pathologic pathways and treatment options for COVID-19 smell and taste loss. The interaction of novel coronavirus with ACE-2 receptors expressed on sustentacular cells and taste buds results in direct damage to the olfactory and gustatory systems. Also, the invasion of the virus to the olfactory neurons and consequent local inf

A Labeled-Line Neural Circuit for Pheromone-Mediated Sexual Behaviors in Mice.

In mice, various instinctive behaviors can be triggered by olfactory input. Despite growing knowledge of the brain regions involved in such behaviors, the organization of the neural circuits that convert olfactory input into stereotyped behavioral output remains poorly understood. Here, we mapped the neural circuit responsible for enhancing sexual receptivity of female mice by a male pheromone, exocrine gland-secreting peptide 1 (ESP1). We revealed specific neural types and pathways by which ESP1 information is conveyed from the peripheral receptive organ to the motor-regulating midbrain via t

Olfactory Adaptation is Dependent on Route of Delivery.

Odorants are perceived orthonasally (nostrils) or retronasally (oral cavity). Prior research indicates route of delivery impacts odorant perception, pleasantness, and directed behaviors thus suggesting differential processing of olfactory information. Adaptation is a form of neural processing resulting in decreased perceived intensity of a stimulus following prolonged and continuous exposure. The present study objective was to determine whether route of delivery differentially impacts olfactory adaptation and whether cross-adaptation occurs between orthonasal and retronasal pathways.
