Independence of GIP-induced insulin secretion from sympathetic and parasympathetic innervation in the isolated perfused rat pancreas.

The incretin candidate GIP (gastric inhibitory polypeptide) is released from the gut by nutrients and can stimulate insulin secretion. Metabolic and hormonal factors have been shown to modulate insulin response to GIP. It is unknown, however, whether the autonomic nervous system, which itself controls insulin secretion, can modulate the insulinotropic effect of GIP. In the isolated perfused rat pancreas, we therefore investigated the influence of sympathetic and parasympathetic agonists and antagonists on the insulin response to GIP. As compared to control (6990 +/- 890 microU/10 min), the eff

Changes in the VIPergic, cholinergic and adrenergic innervation of human penile tissue in diabetic and non-diabetic impotent males.

Penile tissue (consisting of corpus cavernosum and tunica albuginea) was obtained from 19 patients undergoing surgery for the implantation of penile prostheses. The tissue was examined for vasoactive intestinal polypeptide-like immunoreactivity in nerves, acetylcholinesterase-positive staining in nerves and noradrenaline content. Impotence was due to a variety of causes; 11 patients were classified as a 'non-neuropathic' group on the basis of their clinical history which included Peyronie's disease, vascular disease, hypertension and psychogenic impotence. Vasoactive intestinal polypeptide-lik

Ocular accommodation, personality, and autonomic balance.

The autonomic nervous system is made up of two subsystems: the parasympathetic (PNS) and the sympathetic (SNS). The balance between these systems regulates bodily functions during routine (PNS-dominant) and crisis (SNS-dominant) situations. It may also control visual accommodation: PNS activation for inward focus, and SNS activation for outward focus. Near- and far-sighted persons have been characterized as introverted and extraverted, respectively. These personality characteristics have been linked to autonomic balance: introversion and PNS-dominance, extraversion and SNS-dominance. Because a

[Change in the thresholds of development of ventricular arrhythmias after stimulation of negative emotiogenic hypothalamic centers].

The dynamics of changes in the thresholds of the origin of ventricular extrasystole, paroxysmal ventricular tachysystole and ventricular fibrillation in long-term stimulation of the ventromedial hypothalamic nucleus were studied in experiments on rabbits. It was found that myocardial resistance to the development of arrhythmias depended on the manifestation of the sympathetico-parasympathic interaction on the heart. In predominance of parasympathetic effects on the heart during hypothalamic stimulation, the threshold of the origin of arrhythmia increased by 40% for ventricular extrasystole, by

The distribution and secretion of kallikrein in some exocrine organs of the rat.

Extractable kallikrein was quantitated in the submandibular, sublingual, and parotid glands and in the pancreas. No kallikrein was detected in the exorbital lacrimal glands and tears. The highest kallikrein concentrations (EU/ml) were in all major salivary gland secretions seen after alpha-adrenergic stimulation, less after beta-adrenergic and least after parasympathetic stimulation. When taking into account the large variations in salivary flow rate, alpha-adrenergic stimulation was in the parotid and particularly in the submandibular gland found to result in the highest kallikrein secretory

Ontogeny of vesicular monoamine transporter mRNAs VMAT1 and VMAT2. II. Expression in neural crest derivatives and their target sites in the rat.

We used in situ hybridization histochemistry to study the expression of the two vesicular monoamine transporters (VMAT1 and VMAT2) during em … 21 days. In addition, VMAT2 (and to some extent VMAT1) mRNA is expressed in ganglionic cells of the parasympathetic nervous system and in cranial ganglia (trigeminal, vestibular and spiral ganglia) between E12 and E21. The sensory neurons of the dorsal root ganglia, which are also neural crest derivatives, express VMAT2 mRNA (E11-E21), exclusively. Both VMAT mRNAs are found in …

Sex differences in monocyte expression of IL-6: role of autonomic mechanisms.

The interstitial nucleus of Cajal (INC) is thought to be the "neural integrator" for torsional/vertical eye position and head posture. Here, we investigated the coordination of eye and head movements after reversible INC inactivation. Three-dimensional (3-D) eye-head movements were recorded in three head-unrestrained monkeys using search coils. INC sites were identified by unit recording/electrical stimulation and then reversibly inactivated by 0.3 mul of 0.05% muscimol injection into 26 INC sites. After muscimol injection, the eye and head 1) began to drift (an inability to maintain stable fi

Immunohistochemical studies on sympathetic and non-sympathetic nerve fibers and neuronal cell bodies in the pineal gland of cotton rats, Sigmodon hispidus.

Immunohistochemistry revealed the presence of tyrosine hydroxylase (TH)-, neuropeptide Y (NPY)-, calcitonin gene-related peptide (CGRP)- and substance P (SP)-immunoreactive nerve fibers and SP-immunoreactive neuronal cell bodies in the pineal gland of the cotton rat (Sigmodon hispidus). Abundant TH- and NPY-immunoreactive fibers were distributed evenly throughout the gland; less numerous CGRP- and SP-immunoreactive fibers were distributed in the superficial pineal and the stalk, but were scarce in the deep pineal. All the immunoreactive fibers were usually found around blood vessels. Since TH-

The Input-Output Relationship of the Cholinergic Basal Forebrain.

Basal forebrain cholinergic neurons influence cortical state, plasticity, learning, and attention. They collectively innervate the entire cerebral cortex, differentially controlling acetylcholine efflux across different cortical areas and timescales. Such control might be achieved by differential inputs driving separable cholinergic outputs, although no input-output relationship on a brain-wide level has ever been demonstrated. Here, we identify input neurons to cholinergic cells projecting to specific cortical regions by infecting cholinergic axon terminals with a monosynaptically restricted

Parasympathetic blockade promotes recovery from atrial electrical remodeling induced by short-term rapid atrial pacing.

The purpose of the present study was to assess the influence of autonomic blockade on shortening of effective refractory period (ERP) induced by short-term rapid atrial pacing (RAP) and its recovery process. Fifteen patients (8 men, 7 women, age 52 +/- 16 years) without structural heart disease and without a history of atrial fibrillation were included in this study. All patients underwent RAP at a cycle length of 300 ms for 5 minutes, after which the ERP was measured in all patients at 1, 3, 5, 7.5, and 10 minutes following cessation of RAP. In ten patients, these RAP and measurements of ERPs

Periodicity of a noninvasive measure of cardiac vagal tone during non-rapid eye movement sleep in non-sleep deprived and sleep-deprived normal subjects.

Cyclic alternating pattern (CAP) of microarousals reported in EEG studies is now regarded as an integrating mechanism for the different parts of the central nervous system including the autonomic system. A recently developed continuous index of cardiac parasympathetic activity (CIPA) can be time locked to the EEG allowing the relationship between EEG and autonomic changes to be studied in sleep. Eleven normal subjects were studied for evidence of periodicity in CIPA in non-REM sleep, five of whom had been sleep deprived. Fast Fourier's transformation of the CIPA data confirmed its periodicity

Modulation of parasympathetic and baroreceptor control of heart rate.

The heart rate is modulated from beat to beat by efferent vagal and sympathetic fibers, the former being the predominant mediators of the chronotropic influence of arterial baroreceptors and respiration and the latter being important in the cardiac responses to physical and mental stress. Cardiac vagal influences are modulated by a number of factors. These can be grouped as: 1) neural factors, such as the wakefulness-sleep cycle, the alerting reaction, and exercise; 2) humoral-pharmacological factors, such as angiotensin II, atrial natriuretic factor, cardiac glycosides; 3) normal aging; 4) a

Postsynaptic inhibitory effects of phenothiazines at cholinergic synapses may not involve calmodulin.

Treatment of frog cutaneous-pectoris nerve-muscle preparations with calmidazolium (R 24571), a calmodulin-inhibitor, at concentrations of 2 X 10(-7) mol/l and 5 X 10(-7) mol/l had no discernable effect on MEPP amplitude. 10(-6) mol/l calmidazolium caused a small (10-35%) increase in MEPP amplitude in most preparations. The phenothiazine calmodulin-inhibitor chlorpromazine (5 X 10(-6) mol/l) caused a clear reduction in MEPP amplitude (20%) after 30 min treatment. Similar experiments carried out with chlorpromazine sulphoxide (a derivative of chlorpromazine that is 60 X less potent in inhibition
