BACKGROUND: The paraventricular nucleus of the hypothalamus (PVN) orchestrates neuroendocrine and autonomic output to maintain systolic blood pressure (SBP). Emerging evidence suggests that the PVN utilizes paracrine signals to modulate neighboring neurons. Here, we test the hypothesis that OXT (oxytocin)-synthesizing neurons of the PVN (PVNOXT) release paracrine signals that regulate SBP via modulation of AVP (arginine vasopressin)-synthesizing neurons of the PVN.
METHODS: To test the hypothesis, experiments were conducted ex vivo and in vivo in mice with the expression of ChR2 (channelrhodo

Adipose tissue disturbance underlies the pathogenesis of major depressive disorder (MDD); however, the underlying neural and molecular mechanisms remain elusive. Utilizing a social defeat stress mouse model of MDD, we identified a paraventricular hypothalamic (PVH) control of susceptibility to social stress via β3 adrenoceptor (β3AR) signaling in the brown adipose tissue (BAT). Susceptible mice exhibited a significantly elevated expression of BAT β3AR. Pharmacological activation of β3AR increased immobility time in the forced swim test in naïve mice, and decreased social interaction time in mi

BACKGROUND: Many patients undergoing surgery experience accompanying pain symptoms preoperatively. The impact of painful stimuli on general anesthesia remains largely unknown. Corticotrophin-releasing hormone neurons in the paraventricular nucleus of the hypothalamus (PVN CRH neurons) are crucial central stress hubs that respond to painful stimuli. These neurons also participate in regulating processes such as sleep and anesthesia. Natural reward can inhibit PVN CRH neurons to relieve stress-induced behavioral changes, but the effect of natural reward on the anesthesia process in patients with

Limosilactobacillus reuteri has attracted attention for its potential to modulate the gut-brain axis. This study investigates whether L. reuteri administration mitigates autism-related abnormalities in a maternal separation rat model of autism. Male Wistar rats were divided into maternal-separated L. reuteri-treated (MS+LR), maternal-separated placebo-treated (MS), and non-separated control groups. Behavioral assessments evaluated autism-related behaviors, including social interaction, repetitive behavior, anxiety, and hyperactivity. Immunohistochemical analysis of the hypothalamus assessed ox

Catecholamine neurons in the rostral ventrolateral medulla (RVLM) have long been recognized as a crucial neuronal population involved in cardiovascular regulation. However, its function and related circuits in ventricular remodeling after myocardial ischemia/reperfusion injury (MIRI) remain unclear. In this study, we investigated the potential role of RVLM catecholaminergic neurons and the underlying mechanisms that drive MIRI and the development of post-MIRI ventricular remodeling. In vivo electrophysiological recordings revealed that the spontaneous spike rate of RVLM neurons increased throu

BACKGROUND: Sympathetic overactivation and neuroinflammation in the paraventricular nucleus (PVN) are crucial factors in post-myocardial infarction (MI) cardiac remodeling and ventricular arrhythmias (VAs). Prior study has indicated that low-intensity focused ultrasound stimulation could attenuate sympathetic neuroinflammation within the PVN to prevent the occurrence of VAs in an acute MI model. Meanwhile, the cGAS-STING pathway has shown potential to ameliorate the neuroinflammatory response. However, the effect and mechanisms of long-term transcranial ultrasound stimulation (LTUS) for modula

Patients with secondary adrenal insufficiency can present with impaired free water excretion and hyponatremia, which is due to the enhanced secretion of vasopressin (AVP) despite increased total body water. AVP is produced in magnocellular neurons in the paraventricular nucleus of the hypothalamus (PVH) and supraoptic nucleus and in parvocellular corticotropin-releasing factor (CRF) neurons in the PVH. This study aimed to elucidate whether magnocellular AVP neurons or parvocellular CRF neurons coexpressing AVP are responsible for the pathogenesis of hyponatremia in secondary adrenal insufficie

Evidence has mounted that insulin can be synthesized in various brain regions, including the hypothalamus. However, the distribution and functions of insulin-expressing cells in the hypothalamus remain elusive. Herein, we show that in the mouse hypothalamus, the perikarya of insulin-positive neurons are located in the paraventricular nucleus (PVN) and their axons project to the median eminence; these findings define parvocellular neurosecretory PVN insulin neurons. Contrary to corticotropin-releasing hormone expression, insulin expression in the PVN was inhibited by restraint stress (RS) in bo

Perinatal exposure to polychlorinated biphenyls (PCBs) leads to significant alterations of neural and hormonal systems. These alterations have been shown to impair motor and sensory development. Less is known about the influence of PCB exposure on developing emotional and motivational systems involved in social interactions and social learning. The present study examined the impact of perinatal PCB exposure (mixture of congeners 47 and 77) on social recognition in juvenile animals, conspecific-directed investigation in adults and on neural and hormonal systems involved in social functions. BACKGROUND: Studies show that electroacupuncture (EA) has beneficial effects in patients with inflammatory diseases. This study investigated the mechanisms of EA anti-inflammation, using a rat model of complete Freund's adjuvant (CFA)-induced hind paw inflammation and hyperalgesia.
DESIGN: Four experiments were conducted on male Sprague-Dawley rats (n = 6-7/per group). Inflammation was induced by injecting CFA into the plantar surface of one hind paw. Experiment 1 examined whether EA increases plasma adrenocorticotropic hormone (ACTH) levels. Experiments 2 and 3 studied the effects of the ACT

The decline in the release of various anterior pituitary hormones, particularly prolactin (PRL), as a result of exposure to inhibitory, short-day photoperiods in long-day breeders is a well-documented phenomenon. This alteration in the hypothalamic-hypophyseal-gonadal axis is largely controlled by changes in the duration of the nocturnal melatonin pulse secreted by the pineal gland. Increased duration of melatonin secretion serves to increase the inhibitory activity of the tuberoinfundibular dopaminergic (TIDA) neurons, which in turn suppress PRL release and synthesis. However, other hypothala

Medial hypothalamic injections of 5-hydroxytryptamine (5-HT) or its agonists have been reported to inhibit feeding elicited by norepinephrine (NE), suggesting that these two transmitter systems interact antagonistically in the control of ingestive behavior. The present study was designed to directly test the hypothesis that 5-HT inhibits adrenergic feeding, specifically at the onset of the rat's nocturnal eating cycle. Free-feeding animals were injected with 5-HT (5-20 nmol) immediately before NE (20 nmol) and food intake was measured 1.5 h postinjection. In separate groups of rats, the seroto

An overactive endogenous opioid peptide system (EOP) in the hypothalamus of the obese rats could contribute to a subnormal metabolic response to cold stress. Specific mu, delta, kappa opioid receptor antagonists and naloxone were infused into cannulaes aimed at the paraventricular nucleus (PVN) of awake freely moving obese (LA/N-cp corpulent) and lean littermate rats. Metabolic responses were measured by indirect calorimetry during thermoneutrality (30 degrees C) and at 10 degrees C for 60 minutes each. When expressed relative to metabolic body size (kg(-.75)) obese rats had lower values (obes

The dynamics of changes in the thresholds of the origin of ventricular extrasystole, paroxysmal ventricular tachysystole and ventricular fibrillation in long-term stimulation of the ventromedial hypothalamic nucleus were studied in experiments on rabbits. It was found that myocardial resistance to the development of arrhythmias depended on the manifestation of the sympathetico-parasympathic interaction on the heart. In predominance of parasympathetic effects on the heart during hypothalamic stimulation, the threshold of the origin of arrhythmia increased by 40% for ventricular extrasystole, by

Stress is a major risk factor for depression- and anxiety-related disorders and produces neuroplastic adaptations within brain regions that regulate emotion, cognition, and stress responsivity. These adaptations include dendritic retraction in the medial prefrontal cortex (mPFC), suppression of plasticity and neurogenesis in the hippocampus (HIPP), and hypertrophic remodeling of circuits within the amygdala (AMY), promoting emotional dysregulation and maladaptive behavior. Although these alterations are well-documented, the mechanisms that coordinate their emergence across multiple neural syst

Spinal cord injury (SCI) disrupts the integrated neuroendocrine network linking the hypothalamic-pituitary-adrenal (HPA) axis, renin-angiotensin-aldosterone system, and sympathetic nervous system, resulting in endocrine and immune dysfunction. However, the effects of SCI on angiotensin II receptor signaling remain unclear. This study investigated temporal and lesion-dependent changes in angiotensin II type 1 (AT1) and type 2 (AT2) receptor expression following SCI in rats. Low-thoracic SCI (Th9 compression, 40 g/15 min) was evaluated over 28 days, while acute responses to partial (Th9) versus

The hypothalamic-pituitary-adrenal (HPA) axis is a well characterized endocrine response system. Hypothalamic Crh in the paraventricular nucleus of the hypothalamus (PVH) initiates HPA axis signaling to cause the release of cortisol (or corticosterone in rodents) from the adrenal gland. PVH-specific deletion of Crh reduces anxiety-like behaviors in mice. Here we report that manipulation of PVH Crh expression in primary adrenal insufficiency or by dexamethasone (DEX) treatment do not alter mouse anxiety behaviors. In Experiment 1, we compared wildtype (WT) mice to those with primary adrenal ins

Social isolation is a known stressor that negatively impacts the well-being of social species. In rodents, social deprivation experienced either before or after weaning profoundly impacts adult behavioral and neuroendocrine profiles. This study compared the effects of post-natal and post-weaning social deprivation on behavioral profiles and hypothalamic oxytocin (OT) neurons. Male and female Sprague-Dawley rats were assigned to two post-natal groups, maternally separated (MS) or non-MS. MS pups were separated from their mothers for 4h daily during post-natal days 2-21 while non-MS litters rema

OBJECTIVE: Cocaine- and amphetamine-regulated transcript (CART) codes for a hypothalamic neuropeptide, CART (55-102), which inhibits food intake. Intracerebroventricular injection of CART (55-102) reduces appetite, but also results in motor abnormalities. More recently, studies have demonstrated that administration of CART directly into the paraventricular nucleus (PVN) increases food intake. To investigate the role of CART in the regulation of energy balance in the PVN, we used recombinant adeno-associated virus (rAAV) to overexpress CART in the PVN.
