Identification of a Ste20-like kinase-dependent signaling network orchestrating cytoskeletal and synaptic development of cortical neurons.

BACKGROUND: The Ste20-like kinase (SLK) is a serine/threonine kinase that regulates cytoskeletal organization, apoptosis, and cell polarity in non-neuronal cells. In neurons, SLK guides dendrite morphogenesis and the stability of inhibitory synapses, thereby controlling excitation-inhibition balance through parvalbumin interneuron function. Almost complete loss of SLK in dysmorphic neurons of epileptogenic cortical dysplasias implicates a function of this kinase in neurodevelopmental disorders. However, its neuronal signaling mechanisms remain poorly understood. METHODS: To address this knowl

Neurobiological Correlates of Behavioral Resilience to Chronic Alcohol and Acute Stress in Male and Female Rats.

BACKGROUND: Chronic alcohol use and stress exposure are risk factors for psychiatric disorders, often leading to emotion dysregulation and impaired behavioral inhibition. While stress-related disorders are often comorbid with alcohol use disorder, not all individuals exposed to stress or alcohol develop either condition. One approach to assessing impaired behavioral inhibition of fear and reward behaviors is through conditioned inhibition paradigms. METHODS: Sixty-three male and seventy-five female Long Evans rats were assigned to either an intermittent two-bottle choice paradigm (alcohol ver

DREADD agonist Clozapine-N-oxide, but not Compound 21, impairs spatial memory encoding.

Chemogenetic studies using Designer Receptors Exclusively Activated by Designer Drugs (DREADDs) enable the precise manipulation of neuronal activity in specific brain regions and cell types. DREADDs are widely used to dissect neural circuits underlying animal behavior, including learning and memory. Clozapine-N-Oxide (CNO), a metabolite of clozapine and one of the earliest-developed ligands for muscarinic DREADDs, was initially considered pharmacologically inert. However, CNO is now known to undergo back-metabolism to clozapine, leading to undesired off-target behavioral effects, including alt

Different early-life adversity paradigms have disparate effects on maternal care, neuronal activation, and social behavior in developing mice.

In humans, early-life adversity (ELA) predisposes individuals to neuropsychiatric conditions, with different types of adverse experiences linked more strongly to certain maladaptive outcomes. Maternal separation with early weaning (MSEW) and limited bedding and nesting (LBN) are rodent models that expose litters to very different postnatal manipulations, but the effect of these paradigms on offspring through differences in maternal care and neuronal activation remains incompletely explored. To address this gap, we directly compared behavior of dams in MSEW and LBN conditions to control conditi

Context-dependent effects of microglial MyD88 removal on voluntary ethanol consumption in mice.

Neuroimmune signaling is increasingly implicated in alcohol use disorder (AUD). Microglia, the brain's resident immune cells, signal in part through the adaptor protein myeloid differentiation primary response 88 (MyD88), a key mediator of innate immune responses. Here, we investigated whether microglial-specific MyD88 signaling regulates voluntary alcohol consumption in adulthood, as whole-body loss of MyD88 was previously shown to increase drinking. We further determined if alcohol altered parvalbumin-expressing interneurons (PVIs) and microglia within the pre-frontal cortex, based on our pr

Estrogen receptor β in the prefrontal cortex mediates stress-induced anxiety-like behaviors in female mice.

Females are more likely to develop an anxiety disorder following exposure to stress. We previously identified prefrontal parvalbumin (PV) neurons as a potential cellular mechanism underlying this sex-specific vulnerability. By exposing adult mice to 4 weeks of unpredictable chronic mild stress (UCMS), we revealed a female-specific increase in prefrontal PV neurons activity that we causally related to increased anxiety-like behaviors. Because female prefrontal PV neurons mature during adolescence potentially through an estrogen receptor-beta (ERβ)-dependent mechanism, and because the sex differ

Neuropixels reveal laminar microcircuit organization in monkey V1 in vivo.

The relationship between different cell populations in monkey primary visual cortex and their role in visual function is not fully resolved. We combined high-density Neuropixels recordings across layers of macaque V1, and a state-of-the-art nonlinear dimensionality reduction approach on waveform shape to delineate nine putative cell classes: 4 narrow-spiking (NS), 4 broad-spiking (BS), and 1 triphasic (TP). Then, we performed targeted analyses of laminar organization, spike amplitude, multichannel spatial features, functional properties, and network connectivity of these cell classes. These an

Aversive 22-kHz ultrasonic vocalization playback reveals differences in affective (dys)function following early life adversity in male and female juvenile rats.

Early life adversity (ELA), such as childhood neglect, is a significant risk factor for the development of anxiety and other disorders characterized by affective dysfunction, with worse outcomes in women/girls. Rodent models of ELA, such as maternal separation, show comparable behavioral and neural correlates to patient populations. Despite this, many behavioral assays aimed at understanding affective processing in rodents lack species-specific relevance and are therefore limited in translational application. To address this, we utilized an ethologically relevant rat-specific cue of potential

Interneuron theta phase locking controls seizure susceptibility.

The timing of neuronal activity is highly precise and often organized by brain-wide oscillations. Many neurons modulate their firing rates at specific phases of theta (known as theta phase locking), creating discrete windows for information processing. Disrupted theta phase locking has been found across several neurological and psychiatric disorders (e.g., epilepsy), but gaps in technology have prevented its causal influence from being tested. Here, we developed PhaSER, a closed-loop optogenetic system designed to control the phase locking of specific interneurons, and demonstrate a causal rol

Association between pyramidal neurone spiking in the medial prefrontal cortex and the sedative potency of volatile anaesthetics in mice.

BACKGROUND: The mechanisms underlying volatile anaesthetic-induced unconsciousness remain unclear. Glutamatergic pyramidal neurones and fast-spiking interneurones in cerebral cortex circuits play distinct roles in cortical network dynamics under volatile anaesthesia. We investigated the roles of medial prefrontal cortex (mPFC) pyramidal and fast-spiking interneurones in volatile anaesthetic-induced hypnosis. METHODS: The electrophysiological properties of pyramidal and fast-spiking parvalbumin (PV+) neurones were explored by in vivo multichannel recordings and in vitro patch-clamp electrophys

Responses to conflicting binocular stimuli in mouse primary visual cortex.

Binocular vision requires that the brain integrate input from both eyes to form a unified percept. Small interocular differences support depth perception (stereopsis), while larger disparities can cause diplopia or binocular rivalry. The neural mechanisms by which early visual circuits process concordant versus conflicting binocular signals remain incompletely understood, particularly in the case of large disparities. Here, we used visually evoked potential (VEP) recordings, unit recordings, and 2-photon calcium imaging in the binocular region of mouse primary visual cortex (bV1) to examine ho

Acute Intermittent Theta-Burst Stimulation Produces Antidepressant-Like Effects by Modulating Neuronal Oscillations and Serotonin Levels of the Medial Prefrontal Cortex in Experimental Parkinson's Disease.

Parkinson's disease (PD)-related depression is associated with aberrant neuronal oscillations and 5-hydroxytryptamine (5-HT) neurotransmission in the medial prefrontal cortex (mPFC). Intermittent theta-burst stimulation (iTBS), an updated pattern of high-frequency repetitive transcranial magnetic stimulation, has possible efficacy in PD-related depression. However, whether iTBS alleviates PD-related depression through modulating neuronal oscillations and 5-HT levels in the mPFC has not been determined. In this study, male Sprague-Dawley rats were used to establish a unilateral 6-hydroxydopamin

Early life ACE2 inhibition leads to long term anxiety-like behaviour in rats by disrupting the amygdala.

Angiotensin-converting enzyme 2 (ACE2) is important for cardiac and renal development as well as appropriate perinatal growth. In addition, ACE2 has recently attracted notice as the cellular entry receptor for severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), with evidence that the downregulation of ACE2 associated with SARS-CoV-2 may be linked to its long-lasting neurological and neurocognitive effects. However, the long-term neurological consequences of early life ACE2 suppression remain unclear. Here, we hypothesized that downregulation of ACE2 would predispose juveniles to anxi

How Does Altered Function of the Hippocampus Contribute to the Development of Psychosis?

In this review, we explore the critical role of hippocampal dysfunction in the pathophysiology of psychosis, focusing on translational insights from 22q11.2 deletion syndrome (22q11DS). Converging evidence indicates that individuals with 22q11DS, one of the highest genetic risk factors for schizophrenia, exhibit a shared neurobiological vulnerability with idiopathic psychosis, characterized by cognitive decline and atypical brain development, particularly within the hippocampus. Here, we explore translational evidence for hippocampal dysfunction in humans with 22q11DS and the homologous mouse

Interneuron Loss and Microglia Activation by Transcriptome Analyses in the Basal Ganglia of Tourette Disorder.

BACKGROUND: Tourette disorder (TS) is characterized by motor hyperactivity and tics that are believed to originate in the basal ganglia. Postmortem immunocytochemical analyses has revealed decreases in cholinergic (CH), as well as parvalbumin and somatostatin GABA (gamma-aminobutyric acid) interneurons (INs) within the caudate/putamen of individuals with TS. METHODS: We obtained transcriptome and open chromatin datasets by single-nucleus RNA sequencing and single-nucleus ATAC sequencing, respectively, from caudate/putamen postmortem specimens of 6 adults with TS and 6 matched normal control s

Cortical excitatory and inhibitory neuron deficits may underlie the cognitive and social impairments in a mouse model of schizophrenia with exonic Reln deletion.

Reelin is an essential extracellular matrix glycoprotein that regulates cortical layers formation and has been implicated in several neuropsychiatric conditions, including schizophrenia (SCZ). To explore its role in brain function and its potential involvement in SCZ, we developed a Reln heterozygous deletion (Relndel/+) mouse model that replicates a genetic deletion identified in a Japanese patient with SCZ. In previous studies, we demonstrated that Relndel/+ mice exhibit cognitive impairments in a visual discrimination test. In the present study, we found that Relndel/+ mice displayed impair

Anodal direct current stimulation of the auditory cortex at the onset of presbycusis delays cortical aging.

Presbycusis or age-related hearing loss (ARHL) affects millions of people worldwide, increasing their risk of cognitive decline and poor quality of life. However, ARHL remains an irreversible condition due to our inability to induce inner-ear hair cell regeneration. Nevertheless, multisession epidural stimulation of the auditory cortex (AC) at the onset of ARHL prevents hearing threshold elevation in naturally aging Wistar rats. Accordingly, we hypothesized that anodal direct current (DC) stimulation of the AC may also compensate for age-related maladaptive, activity-dependent changes. Here, w

GABAergic and inflammatory changes in the frontal cortex following neonatal PCP plus isolation rearing, as a dual-hit neurodevelopmental model for schizophrenia.

The pathogenesis of schizophrenia begins in early neurodevelopment and leads to excitatory-inhibitory imbalance. It is therefore essential that preclinical models used to understand disease, select drug targets and evaluate novel therapeutics encompass similar neurochemical deficits. One approach to improved preclinical modelling incorporates dual-hit neurodevelopmental insults, like neonatal administration of phencyclidine (PCP, to disrupt development of glutamatergic circuitry) then post-weaning isolation (Iso, to mimic adolescent social stress). We recently showed that male Lister-hooded ra
