Toxicities of and inflammatory responses to moxifloxacin, cefuroxime, and vancomycin on retinal vascular cells.

Prophylactic intracameral injection of antibiotics is commonly used to prevent endophthalmitis after cataract surgery. However, devastating visual complications have been reported including hemorrhagic occlusive retinal vasculitis (HORV).To determine the toxic and inflammatory effects of moxifloxacin, cefuroxime, and vancomycin on human retinal vascular cells, human retinal vascular endothelial cells (RVEC) and pericytes were exposed to three antibiotics, and the adverse effects were assessed by membrane damage, loss of intrinsic esterase activity, kinetic cell viability, and inflammatory cyto…

Calorie restriction reverses age-related alteration of cavernous neurovascular structure in the rat.

Calorie restriction (CR) refers to a reduction of calorie intake without compromising essential nutrients to avoid malnutrition. CR has been … surement of serum total testosterone level. The contents of endothelial cells, smooth muscle cells, pericytes, and neuronal cells as well as serum testosterone levels were significantly lower in the penis of aged rats than in their young littermates. CR significantly restored cavernous endothelial cells, smooth muscle cells, pericytes, and neuronal cell con …

Generation of 5 hiPSC lines from pediatric patients with Heritable pulmonary arterial hypertension (HPAH) caused by heterozygous mutations in the TBX4 gene.

Heritable pulmonary arterial hypertension (HPAH) and underlying pulmonary vascular disease (PVD) are often caused by TBX4 mutations-either loss- or gain-of-function-which are a leading cause of childhood-onset PAH. The clinically heterogeneous TBX4 syndrome can include skeletal anomalies (e.g., small patella syndrome) and developmental lung disease (DEVLD) (Galambos, 2019). TBX4 is expressed in lung mesenchymal cells such as matrix fibroblasts, pericytes, and smooth muscle cells, all contributing to PAH pathogenesis (Karolak, 2023; Maldonado, 2025). Our five patient-derived TBX4-mutant hiPSC l…

Neurovascular pathways to neurodegeneration in Alzheimer's disease and other disorders.

The neurovascular unit (NVU) comprises brain endothelial cells, pericytes or vascular smooth muscle cells, glia and neurons. The NVU controls blood-brain barrier (BBB) permeability and cerebral blood flow, and maintains the chemical composition of the neuronal 'milieu', which is required for proper functioning of neuronal circuits. Recent evidence indicates that BBB dysfunction is associated with the accumulation of several vasculotoxic and neurotoxic molecules within brain parenchyma, a reduction in cerebral blood flow, and hypoxia. Together, these vascular-derived insults might initiate and/…

Cytopathological changes in human and animals brains in prion diseases.

Studies of histological preparations of the brains of humans and animals with prion diseases showed that neuron vacuolization often starts and develops intensely in the distal segments of dendrites. Two types of neuron body death were noted - cytolysis and pyknosis, the latter usually being associated with vacuolization of dendrites. The involvement of the main types of neuroglia in the pathogenesis of prion diseases was established, which provides grounds for referring to gliosis rather than astrocytosis. Accumulation of phagocytes (microgliocytes, blood macrophages, pericytes) and release of…

Skeletal muscle microvascular alterations in euthyroid and hypothyroid patients with autoimmune thyroid disease.

An electron microscopic study of skeletal muscle biopsies from euthyroid and hypothyroid patients with autoimmune thyroid disease (ATD) showed the existence of capillary alterations and a mononuclear cell infiltrate. Microvascular abnormalities included thickening and lamination of basement membrane, endothelial proliferation with progressive lumen occlusion, and hypertrophied pericytes. Capillary degeneration was also observed. Macrophages, and scarce lymphocytes and mast cells formed the cell infiltrate. This pathological picture is similar to that found in the muscular compromise of other a…

Intracranial hemangiopericytoma in a child.

Hemangiopericytoma in the central nervous system is a rare tumor occurring predominantly in supratentorial sites. Classification may be confusing because the tumor may mimic the histological pattern of other vascular neoplasms. The particular feature of hemangiopericytoma are pericytes of blood vessels; they distinguish it as a separate and distinct tumor. The case of a 3 3/4-year-old child with a 3 3/4-year course is reported with a tumor in the middle fossa, the diencephalon, and the brainstem. The cystic tumor was resected twice. The child is presently alive with neurological deficit. The c…

Single-cell and spatial transcriptomic insights into fibrotic mechanisms and therapeutic opportunities.

Fibrosis is a conserved response to chronic injury in which excessive extracellular matrix (ECM) deposition disrupts tissue architecture and organ function. Single-cell RNA sequencing and spatial transcriptomics now show that fibrosis is driven by multicellular niches rather than a uniform fibroblast population. We propose an ECM-associated mesenchymal cell (ECM-MC) continuum that links stromal progenitors, pericytes, fibroblasts and myofibroblasts across organs. This framework distinguishes conserved pathogenic states from tissue-specific adaptations and connects epithelial injury, immune-str…

Sinonasal hemangiopericytoma: A rare sinonasal tumor.

Hemangiopericytoma is a rare vascular tumor arising from pericytes and occurs very rarely in sinonasal region. A 48-year-old man with sinonasal mass presented with nasal obstruction and occasional epistaxis. Nasal endoscopy showed a readily bleeding mass in the left nasal cavity. Endoscopic removal of the mass was done. The histopathology came out to be hemangiopericytoma. The patient was kept on follow-up, and no metastasis or recurrence was seen in last 1 year. Hemangiopericytoma is a very rare vascular tumor. Surgery is the mainstay treatment of choice. A long-term follow-up is needed after…

Ultrastructure of the conjunctiva, skin, and gingiva: a case of Sandhoff's disease in a Jewish patient.

Pleomorphic membranous cytoplasmic bodies that indicated glycolipid storage were found in the conjunctiva, skin, and gingiva of a Jewish patient with Sandhoff's disease. The clinical symptoms were typical of GM2 gangliosidosis. Both hexosaminidase A and hexosaminidase B activities were deficient in the leukocytes and serum. Glycosaminoglycan levels in cultured fibroblasts were elevated. Membranous cytoplasmic bodies were observed in high concentrations in a large proportion of the vascular endothelial cells, pericytes, and Schwann cells and to a somewhat lesser extent in the fibrocytes of all …

Dexamethasone restores blood-brain barrier integrity in an in vitro heatstroke model.

Heat-related diseases and their treatments are becoming the center of focus due to global warming resulting in rising global temperatures. Heatstroke is the most hazardous condition of heat-related diseases, which when left untreated leads to death. One of the main characteristics of heatstroke is the dysfunction of the central nervous system. In this study, we established an in vitro heatstroke model of the blood-brain barrier (BBB) consisting of endothelial cells and pericytes. Following heat exposure at 43°C for 3 h, the model failed to recover during the subsequent 24 h regeneration period…

A Long-Lived Human Neurovascular PENTA Culture Model Captures Incomplete Vascular Repair and Glia-Associated Signaling After Traumatic Brain Injury.

Traumatic brain injury (TBI) frequently leads to chronic neurovascular dysfunction, yet mechanistic insights into human-specific responses have been limited by the absence of long-term, multicellular in vitro models. Here, we report a five-cell-type human neurovascular culture system, comprising endothelial cells, astrocytes, pericytes, microglia, and neurons, engineered within a 3D scaffold to study injury-induced remodeling over multiple weeks. This PENTA-culture platform captures key structural and molecular features of the neurovascular unit and supports compartment-resolved profiling of v…

Single-cell RNA sequencing of cultured human endometrial CD140b+CD146+ perivascular cells highlights the importance of in vivo microenvironment.

BACKGROUND: Endometrial mesenchymal-like stromal/stem cells (eMSCs) have been proposed as adult stem cells contributing to endometrial regeneration. One set of perivascular markers (CD140b&CD146) has been widely used to enrich eMSCs. Although eMSCs are easily accessible for regenerative medicine and have long been studied, their cellular heterogeneity, relationship to primary counterpart, remains largely unclear. METHODS: In this study, we applied 10X genomics single-cell RNA sequencing (scRNA-seq) to cultured human CD140b+CD146+ endometrial perivascular cells (ePCs) from menstrual and secret

Beyond EMT: Mesenchymal drift as an emerging driver of stromal-immune reprogramming in prostate cancer.

Cellular plasticity within the tumor microenvironment (TME) extends far beyond classical epithelial-mesenchymal transition (EMT). Emerging evidence indicates that diverse non-epithelial cell populations, including macrophages, endothelial cells, pericytes, adipocytes, and fibroblasts, undergo a progressive and often partial reprogramming toward mesenchymal-like states during tumor progression. We conceptualize this broader phenomenon as mesenchymal drift (MD), a trans-lineage adaptive process characterized by erosion of lineage-specific identity, acquisition of extracellular matrix-remodeling …
