Open channel block and open channel destabilization: contrasting effects of phenol, TEA+ and local anaesthetics on Kv1.1 K+ channels.

(1) Voltage-gated K+ channels are inhibited by a variety of clinical and experimental drugs. (2) Complex changes in channel gating suggest mechanisms in which drug affinity depends on channel state. (3) Here, we use the effects of external TEA+, two local anaesthetics (lidocaine and bupivacaine), and phenol on rat brain Kv1.1 K+ channels expressed in Xenopus oocytes to illustrate three mechanisms. (4) The open state has the highest affinity in the local anaesthetic model but the lowest in the phenol model, and while local anaesthetics simply block the open channel, phenol can produce a conduct…

Treatment of external urethral sphincter hypertonicity by pudendal nerve block using phenol solution in patients with spinal cord injury.

We report our experience utilizing the technique of phenol block of the pudendal nerve in the treatment of voiding dysfunction due to hypertonicity of the external urethral sphincter. We have performed 13 pudendal nerve blocks using a 7% phenol solution in seven patients with spinal cord injury who could not obtain relaxation of the external urethral sphincter with a large postvoid urine residual (150 ml to 600 ml) despite large doses of antispasticity drugs and intermittent catheterisations over three weeks. These drugs were discontinued at least 48 hours before this procedure. The efficacy o…

[Effect of synthetic phenol antioxidants on the parietal pH value of the gastrointestinal tract in intact and vagotomized rats].

The influence of synthetic phenol antioxidants dibunol and BH-3 on the value of parietal pH has been studied in the stomach, duodenum, jejunum, ileum, caecum and rectum of intact and vagotomized rats 1, 7, 14, 30 and 60 days after treatment. It was shown that vagotomy leads to an increase in the stomach pH and to a decrease in pH in other parts of the gastrointestinal tract. Maximum BH-3 doses lead to pH increase in the stomach of intact rats. Chronic administration of therapeutic drug doses does not change pH value in control and vagotomized animals.

2,6-Di-tert-butyl-4-(2'-thenoyl)phenol(R-830): a novel nonsteroidal anti-inflammatory agent with antioxidant properties.

R-830, a di-tert-butylphenol, has been shown to be anti-inflammatory in a number of animal models. These include conventional systems such as carrageenan-induced edema and adjuvant arthritis of the rat and ultraviolet-induced erythema in the guinea pig in which the acidic nonsteroidal anti-inflammatory drugs (e.g., indomethacin) are effective. The anti-inflammatory activity of R-830 has also been demonstrated in other models (e.g., graft vs. host reaction and reversed passive cutaneous Arthus reaction in the rat, contact sensitivity in the mouse) in which the acidic nonsteroidal drugs are not …

Poly(phenolic) deposition is a prerequisite to poly(aliphatic) deposition for functional potato suberin.

Suberin deposition in specialized cells is a critical aspect of plant protection, providing both water retention and pathogen protection functions. Tissue specific suberization during plant development is mirrored by its wound induction in cells that do not normally suberize and provides a robust experimental system to investigate the process. Suberized cells are characterized by a cell wall-associated lignin-like poly(phenolic) domain, and an ester-linked poly(aliphatic) domain, with the latter often laid down in distinct lamellar bands between the cell wall and plasma membrane. Previous work…

Soluble Mn(III) drives the oxidative polymerization of phenolics into toxic dissolved oligomers.

Phenolic compounds are widespread aromatic contaminants in aquatic environments and are of particular concern before disinfection because of their toxicity and potential contribution to disinfection byproduct formation. Although manganese-based oxidation processes are widely applied to remove phenolic contaminants, reliance on parent-compound disappearance alone may overlook toxic transformation products generated during oxidation. Herein, we demonstrated that ligand-stabilized soluble Mn(III), a reactive intermediate in Mn redox cycling, redirects phenolic transformation from apparent removal…

Biological evidence versus structural bias - phenolic Mannich bases in medicinal chemistry.

Phenolic Mannich bases (pMb's) are a distinct subclass of C-Mannich bases in which aminomethylation occurs at an aromatic carbon ortho or para to a phenolic hydroxyl group, typically combining a basic amine with a redox-active phenolic unit. The amine can enhance aqueous solubility, while intramolecular hydrogen bonding between the phenolic proton and the tertiary amine may modulate conformational bias, apparent polarity, and membrane permeability, with potential consequences for pharmacokinetics. At the same time, the phenolic Mannich bases motif is frequently flagged by PAINS filters due to …

Palladium-catalyzed dual dearomatization of indoles and phenols: access to indolinone-fused polycycles.

A palladium-catalyzed intermolecular dual dearomatization reaction that combines indole and phenol substrates in a single operation has been developed. The cascade proceeds through an intramolecular Heck cyclization of the indole moiety to generate a benzylic palladium intermediate, which then coordinates with an alkyne and triggers the dearomatization of the phenol component. This divergent protocol enables rapid assembly of indolinone-fused polycycles bearing contiguous stereocenters as well as spirocyclic frameworks, significantly expanding the structural complexity accessible from simple p…

Apple phenolic compounds enhance dental remineralization in human: a multifactorial in situ study.

INTRODUCTION: Excessive consumption of sugar-sweetened beverages is a major contributor to dental caries and a global public health concern due to its negative effects on oral health. Phenolic compounds found in edible plants have shown promising effects on caries prevention. Several studies indicate that these bioactive substances can inhibit caries formation through various mechanisms. However, studies which consider the complex interplay of the various factors are missing. In particular, human studies on the caries reducing effects of phenolic compounds in the co-presence of fermentable sug…

Oral Administration of Metal-Phenolic-Inspired Curcumin Self-Assemblies for Colitis Alleviation.

Natural bioactive compounds hold promise for ulcerative colitis (UC) but are limited by their poor bioavailability and efficacy. Herein, we propose an interdisciplinary strategy termed "natural bioactive compound-nanotechnology-multipronged modulation" to develop an oral, biocompatible nanoparticle system (CTAl NPs) via a self-assembly strategy. Curcumin (Cur) was successfully encapsulated within a tannic acid (TA) and Al3+ coordination network through hydrogen bonding, π-π stacking, and coordination interactions. The resulting CTAl NPs confer therapeutic benefits through three synergistic mec

Exogenous tryptophan and taurine enhance phenolic metabolism and antioxidant defenses in walnut kernels.

Amino acids are key signaling molecules in plant stress responses. Fresh walnuts are rich in nutrients but face significant nutrient loss, enzymatic browning, and a decline in antioxidant capacity during postharvest storage. This study evaluated exogenous tryptophan and taurine in quality maintenance of fresh intact walnut (Juglans regia L. cv. 'Niangqing'), focusing on antioxidant capacity and phenolic compounds accumulation. Exogenous tryptophan and taurine were applied individually or in combination, effectively maintaining walnut appearance, delaying browning, and extending the shelf life.…

Efficient hydrogenolysis of lignocellulose into phenolic monomers over a CuO/m-ZrO2 catalyst.

The catalytic reduction of lignin into bio-chemicals is desirable yet remains a challenge due to its heterogeneity and complexes. Herein, a copper oxide supported monoclinic ZrO2 (CuO/m-ZrO2) catalyst is fabricated for efficient reductive catalytic fractionation (RCF) of softwood lignin, affording a yield of monophenols up to 18.7 wt% with a high selectivity (82.3%) for 4-n-propanol guaiacol. Some key parameters, such as β-O-4' models, solvent, mass ratio, reaction temperature and time, are systematically investigated to reveal their effects on the yields of monomers. Notably, mechanistic stud…
