Melatonin signal transduction in the goldfish, Carassius auratus.

Generation and reception of melatonin signals in the goldfish, Carassius auratus, are reviewed. The photoreceptive pineal gland of the goldfish generates circulating melatonin rhythms according to a given photoperiod under light-dark cycles and in a circadian manner under continuous dark conditions. Melatonin is also produced in the retina in a similar fashion. Melatonin produced in the pineal gland and retina is considered to act as internal zeitgeber in the brain and retina, respectively, controlling various physiological events via specific melatonin binding sites that are coupled with G pr…

Aromatase activity in the brain of the three-spined stickleback, Gasterosteus aculeatus. II. Effects of castration in winter.

Non-breeding stickleback males were gonadectomized or sham-operated in winter. Treatment with long photoperiod at high temperature stimulated sexual maturation in control fish. Brains were freeze-sectioned and punches were taken with hollow needles from different diencephalic regions and the punches incubated with [19-3H]-androstenedione. After extraction of steroids with CH2Cl2 tritium activity remaining in the water phase was measured. This gives a measure of aromatization as tritiated water and formic acid is formed when [19-3H]-androstenedione is aromatized to estrogens. Aromatase activity…

Modification of the beta-adrenergic sensitivity of rat submandibular gland by constant light.

Rats exposed for 2, 5, 10, 20, 35 and 50 days to constant light (CL) showed beta-adrenergic desensitization of the submandibular gland as indicated by dose-response curves to 1.5, 2.0, 3.0, 10.0, 30.0 and 50.0 micrograms/kg isoproterenol. The phenomenon, evident with 1.5, 2.0 and 50.0 micrograms/kg after 2 days at CL, was more intense as time of exposure increased to reach values of about 50% those for controls after 20 days. After 35 and 50 days, desensitization showed some reversion, but the secretory responses were still lower than for controls maintained under a typical photoperiod. In the…

Melatonin directly resets the rat suprachiasmatic circadian clock in vitro.

The environmental photoperiod regulates the synthesis of melatonin by the pineal gland, which in turn induces daily and seasonal adjustments in behavioral and physiological state. The mechanisms by which melatonin mediates these effects are not known, but accumulating data suggest that melatonin modulates a circadian biological clock, either directly or indirectly via neural inputs. The hypothesis that melatonin acts directly at the level of the suprachiasmatic nucleus (SCN), a central mammalian circadian pacemaker, was tested in a rat brain slice preparation maintained in vitro for 2-3 days. …

Melatonin for the treatment of spinal cord injury.

Spinal cord injury (SCI) from trauma or disease severely impairs sensory and motor function. Neurorehabilitation after SCI is a complex medical process that focuses on improving neurologic function and repairing damaged connections in the central nervous system. An increasing number of preclinical studies suggest that melatonin may be useful for the treatment of SCI. Melatonin is an indolamine that is primarily secreted by the pineal gland and known to be regulated by photoperiodicity. However, it is also a versatile hormone with antioxidative, antiapoptotic, neuroprotective, and anti-inflamma…

Current knowledge on the melatonin system in teleost fish.

Melatonin is a much conserved feature in vertebrates that plays a central role in the entrainment of daily and annual physiological rhythms. Investigations aiming at understanding how melatonin mediates the effects of photoperiod on crucial functions and behaviors have been very active in the last decades, particularly in mammals. In fish a clear-cut picture is still missing. Here we review the available data on (i) the sites of melatonin production in fish, (ii) the mechanisms that control its daily and annual rhythms of production and (iii) the characterization of its different receptor subt…

Melatonin and pregnancy in the human.

The purpose of this systematic review is to access the current state of knowledge concerning the role for melatonin in human pregnancy. Melatonin is a neuroendocrine hormone secreted nightly by pineal gland and regulates biological rhythms. The nighttime serum concentration of melatonin shows an incremental change toward the end of pregnancy. This small lipophilic indoleamine crosses the placenta freely without being altered. Maternal melatonin enters the fetal circulation with ease providing photoperiodic information to the fetus. Melatonin works in a variety of ways as a circadian rhythm mod…

The chronobiology and neurobiology of winter seasonal affective disorder.

This review summarizes research on the chronobiology and neurobiology of winter seasonal affective disorder (SAD), a recurrent subtype of depression characterized by a predictable onset in the fall/winter months and spontaneous remission in the spring/summer period. Chronobiological mechanisms related to circadian rhythms, melatonin, and photoperiodism play a significant role in many cases of SAD, and treatment of SAD can be optimized by considering individual differences in key chronobiological markers. Converging evidence also points to a role for the major monoamine neurotransmitters seroto…

Fattening responses to daily intervals of heat exposure in the Syrian hamster.

Warm ambient temperature (38 degrees C) provided daily for one hr induced time-dependent changes in body weight and fat stores in the Syrian hamster. In animals held on 10-hr daily photoperiods and room temperature (23 degrees C), daily one-hour thermopulses at 8 and 20 hr after light onset stimulated increases in body weights and indices of body fat storage. Abdominal fat pad weights of these groups were twice those of untreated controls after 17 and 28 days of thermoperiodic treatments. On the other hand, daily thermopulses were completely ineffective at 0 and 16 hr after light onset. These …

CmPP6 and CmMIEL1 interact with CmBBX7 to regulate flowering time in chrysanthemum.

Flowering is a crucial process in the growth and development of plants, playing an essential role in their life cycles. Thus, research into the regulation of flowering time holds significant importance. While CmBBX7 has been identified as a flowering activator regulated by photoperiod, the molecular mechanisms underlying its regulation at the protein level remain unclear. In this study, we utilized yeast two-hybrid screening to identify two proteins that interact with CmBBX7: the phosphatase CmPP6 and the E3 ubiquitin ligase CmMIEL1. Ubiquitination assays confirmed that CmMIEL1 promotes the de…

BrDDB1A promoted flowering via age pathway in Chinese cabbage.

The transition from vegetative to reproductive growth, marked by flowering, is a pivotal developmental switch in plants, whereby the age pathway serves as an important regulatory mechanism governing this process. In this study, we screened a late-flowering mutant from EMS mutagenesis in Chinese cabbage. The late-flowering phenotype was unaffected by photoperiod, vernalization and GA3 treatments, and obvious changes occurred at the transcriptional levels of age flowering pathway-related genes, which indicated that the mutant trait was mediated by the age pathway. Causal gene BrDDB1A, involved i…

The suprachiasmatic nuclei as a seasonal clock.

In mammals, the suprachiasmatic nucleus (SCN) contains a central clock that synchronizes daily (i.e., 24-h) rhythms in physiology and behavior. SCN neurons are cell-autonomous oscillators that act synchronously to produce a coherent circadian rhythm. In addition, the SCN helps regulate seasonal rhythmicity. Photic information is perceived by the SCN and transmitted to the pineal gland, where it regulates melatonin production. Within the SCN, adaptations to changing photoperiod are reflected in changes in neurotransmitters and clock gene expression, resulting in waveform changes in rhythmic ele…

Melatonin biosynthesis in the thymus of humans and rats.

Melatonin is an indoleamine widely distributed in the evolution that shows a great functional versatility, playing an important role as a transmitter of photoperiodic information and exhibiting antioxidant, oncostatic, anti-aging and immunomodulatory properties. In vertebrates, this molecule is produced by the pineal gland and other extrapineal sites. The present study was carried out to investigate the presence of melatonin in thymus and the possibility of an endogenous melatonin synthesis in this organ, in which T cells are matured. In this work, we demonstrate in humans and rats that thymus…

Melatonin and the pineal gland: influence on mammalian seasonal and circadian physiology.

The pineal hormone melatonin is secreted with a marked circadian rhythm. Normally, maximum production occurs during the dark phase of the day and the duration of secretion reflects the duration of the night. The changing profile of secretion as a function of daylength conveys photoperiodic information for the organization of seasonal rhythms in mammals. The role of melatonin in mammalian circadian physiology is less clear. However, exogenous melatonin can phase shift, and in some cases entrain, circadian rhythms in rodents and humans. It can also lower body temperature and induce transient sle…

Melatonin, immune modulation and aging.

Melatonin is a hormone secreted by the pineal gland in response to photoperiods and influences many important biological processes. For one, Melatonin has been shown to produce resistance to cancer and infectious diseases in aged animals. Studies in animals have demonstrated melatonin-related mechanisms of action on immunoregulation. Additionally, melatonin has been successfully used in humans, along with interleukin-2, as a treatment of solid tumors. In vivo and in vitro studies show melatonin enhances both natural and acquired immunity in animals. Despite all of this intriguing evidence, mel…
