Reduction of Cr(VI) by hydrazine in solution saturated with KHCO3.

Cr(VI) that exists in many industrial wastes is considered highly toxic. One of the most common ways to dispose of these wastes is to reduce Cr(VI) to Cr(III), which is less toxic and can be easily removed or fixed. Reduction of Cr(VI) by hydrazine in aqueous solutions saturated with potassium bicarbonate was studied in this paper. The content of Cr(VI), molar ratio of hydrazine to Cr(VI) and reaction temperature were 200-1000 ppm, 2:1-10:1 and 25-70 degrees C, respectively. Results showed that the overall reaction was third-order, i.e. first-order with respect to hydrazine and second-order wi

Hyperpolarization following activation of K+ channels by excitatory postsynaptic potentials.

We have postulated that an excitatory postsynaptic potential (e.p.s.p.) may open voltage-sensitive K+ ('M') channels, in an appropriate depolarizing range, and that this could alter the e.p.s.p. waveform. Consequently, the fast e.p.s.p. in neurones of sympathetic ganglia, elicited by a nicotinic action of acetylcholine (ACh), could be followed by a hyperpolarization, produced by the opening of M channels during the depolarizing e.p.s.p. and their subsequent slow closure (time constant-150 mg). This introduces the concept that transmitter-induced p.s.ps may trigger voltage-sensitive conductance

Compound heterozygous KCTD7 variants in progressive myoclonus epilepsy.

KCTD7 is a member of the potassium channel tetramerization domain-containing protein family and has been associated with progressive myoclonic epilepsy (PME), characterized by myoclonus, epilepsy, and neurological deterioration. Here we report four affected individuals from two unrelated families in which we identified KCTD7 compound heterozygous single nucleotide variants through exome sequencing. RNAseq was used to detect a non-annotated splicing junction created by a synonymous variant in the second family. Whole-cell patch-clamp analysis of neuroblastoma cells overexpressing the patients'

Ammonium and Potassium Salts of a Hexacoordinate Phosphorus(V) Anion Featuring P-O and P-C Bonds.

Ammonium and potassium salts featuring the chiral mer-[P(C6H4CO2)3]-, mer-[1]-, have been isolated. Specifically, treating phosphorane P(C6H4CO2)2(C6H4CO2H), 2, with N-containing bases [Nbase = PhNMe2, PhNH2, pyridine (py), isoquinoline, (-)-brucine, N( n-C8H17)3] afforded ammonium salts [NbaseH]+- mer-[1]-. Each compound was fully characterized spectroscopically, and four were subjected to X-ray crystallographic analysis. Salts were isolated with the racemic mixture of the mer-[1]- anion except in the case of [(-)-brucineH]-Λ- mer-[1], for which the crystal analyzed was enantiomerically pure.

X-ray Structure and Properties of the Ferrous Octaethylporphyrin Nitroxyl Complex.

The preparation and characterization of the iron octaethylporphyrin nitroxyl ion, [Fe(OEP)(NO)(-)], is reported. The complex was synthesized by the one-electron reduction of Fe(OEP)(NO) using anthracenide as the reducing agent. The compound was isolated as the potassium (2.2.2)cryptand salt. The anion was characterized using X-ray analysis with visible and infrared spectroscopy. The spectral features of the iron nitroxyl complex were consistent with previous literature reports. The important structural changes upon reduction were a significant decrease in the Fe-N-O bond angle from 142° to 127

Muscle channelopathies and electrophysiological approach.

Myotonic syndromes and periodic paralyses are rare disorders of skeletal muscle characterized mainly by muscle stiffness or episodic attacks of weakness. Familial forms are caused by mutation in genes coding for skeletal muscle voltage ionic channels. Familial periodic paralysis and nondystrophic myotonias are disorders of skeletal muscle excitability caused by mutations in genes coding for voltage-gated ion channels. These diseases are characterized by episodic failure of motor activity due to muscle weakness (paralysis) or stiffness (myotonia). Clinical studies have identified two forms of p

Anthrax lethal toxin-induced inflammasome formation and caspase-1 activation are late events dependent on ion fluxes and the proteasome.

Anthrax lethal toxin (LT) is cytotoxic to macrophages from certain inbred mouse strains. The gene controlling macrophage susceptibility to LT is Nalp1b. Nalp1b forms part of the inflammasome, a multiprotein complex involved in caspase-1 activation and release of interleukin (IL)-1beta and IL-18. We confirm the role of caspase-1 in LT-mediated death by showing that caspase inhibitors differentially protected cells against LT, with the degree of protection corresponding to each compound's ability to inhibit caspase-1. Caspase-1 activation and cytokine processing and release were late events inhi

Nerve excitability changes in chronic renal failure indicate membrane depolarization due to hyperkalaemia.

Multiple nerve excitability measurements were used to investigate axonal membrane properties in patients with chronic renal failure (CRF). Nine patients were studied during routine haemodialysis therapy. The median nerve was stimulated at the wrist and compound muscle action potentials recorded from abductor pollicis brevis. Stimulus-response behaviour, strength-duration time constant, threshold electrotonus, current-threshold relationship and recovery cycle (refractoriness, superexcitability and late subexcitability) were recorded using a recently described protocol. In six patients, sequenti

Interaction between cicletanine and the eicosanoid system in human subcutaneous resistance arteries.

In human subcutaneous resistance arteries in-vitro, the relaxation produced by the novel furopyridine compound cicletanine (30 microMs) was inhibited by 51% (P less than 0.0001) in the presence of 20 microMs of the cyclo-oxygenase inhibitor indomethacin. Maximal cicletanine-induced relaxation was reduced by both the cyclo-oxygenase inhibitor mefenamic acid and the relatively specific blocker of prostacyclin synthetase, tranylcypromine by 55% (P less than 0.0005) and 43% (P less than 0.01), respectively. The potassium channel blocker, glibenclamide (100 microMs), did not affect the relaxation p

The use of plasma aldosterone and urinary sodium to potassium ratio as translatable quantitative biomarkers of mineralocorticoid receptor antagonism.

BACKGROUND: Accumulating evidence supports the role of the mineralocorticoid receptor (MR) in the pathogenesis of diabetic nephropathy. These findings have generated renewed interest in novel MR antagonists with improved selectivity against other nuclear hormone receptors and a potentially reduced risk of hyperkalemia. Characterization of novel MR antagonists warrants establishing translatable biomarkers of activity at the MR receptor. We assessed the translatability of urinary sodium to potassium ratio (Na+/K+) and plasma aldosterone as biomarkers of MR antagonism using eplerenone (Inspra®),

Evaluation of the nutritional value and active compounds of commercial and non-commercial oilseed meals.

Cold pressing is one of the most common methods of extracting oil from seeds, generating by-products in the form of meals that are often discarded as waste. Given the large quantities of these meals and the presence of valuable compounds, a thorough investigation of their nutritional value and potential applications is essential. The aim of this study was to evaluate and compare the residual oil, total phenol and flavonoid content, phenolic compounds, antioxidant activity, as well as the ash, carbohydrate, protein, and mineral nutrient content of 15 commercial and non-commercial oilseed meals.

A Transcriptomic and Metabolomic Study on the Biosynthesis of Iridoids in Phlomoides rotata from the Qinghai-Tibet Plateau.

Phlomoides rotata is a traditional Chinese herbal medicine that grows in the Qinghai-Tibet Plateau region at a 3100-5000 m altitude. Iridoid compounds are the main active compounds of the P. rotata used as medical ingredients and display anti-inflammatory, analgesic, and hepatoprotective properties. To better understand the biological mechanisms of iridoid compounds in this species, we performed a comprehensive analysis of the transcriptome and metabolome of P. rotata leaves from four different regions (3540-4270 m). Global metabolome profiling detected 575 metabolites, and 455 differentially

A Four-Electron-Transfer Robust Small-Molecule Organic Anode for Low-Strain Potassium Storage.

Potassium-ion batteries (PIBs) are promising for large-scale energy storage owing to abundant potassium resources. However, their development is constrained by the lack of high-capacity and durable anodes capable of accommodating the large-radius K+. Herein, a small-molecule organic compound, 2,2'-bipyridine-4,4'-dicarboxylic acid (BDA), is presented as a promising anode for PIBs. The cooperative multielectron redox activity of the carbonyl and imine moieties affords a high reversible capacity, while the conjugated framework and dense molecular packing strengthen the structural stability. The advantage of a two-stage nitrification method for fertilizer recovery from human urine.

Recycling nutrients (nitrogen, phosphorus, and potassium) from human urine can potentially offset more than 13% of global agricultural fertilizer demand. Biological nitrification is a promising method for converting volatile ammonia in high-strength human urine into stable ammonium nitrate (a typical fertilizer), but it is usually terminated in the intermediate production of nitrite due to the inhibition of nitrite-oxidizing bacteria by free nitrous acid (FNA). This study aimed to develop a stable nitrification process in a unique two-stage bioreactor by removing critical barriers associated w

The effect of soil environmental factors on the yield and quality of Pueraria lobata.

Pueraria lobata is a typical medicinal and edible plant with great market value and demand, thus exploring the relationship between soil environmental factors and the yield and quality of Pueraria lobata is of great significance for its high-value cultivation. In this study, using the Guige 1 variety (Pueraria montana var. Thomsonii) selected by our research group as the material to compare the effects of five soil types, endophytes in three parts of Pueraria lobata and two fertilizers on its yield and quality. The results showed that the comprehensive evaluation effect of five soil types on t

Suppression of the excitability of rat nociceptive secondary sensory neurons following local administration of the Phytochemical, (-)-Epigallocatechin-3-gallate.

The phytochemical, polyphenolic compound, (-)-epigallocatechin-3-gallate (EGCG), is the main catechin found in green tea. Although a modulatory effect of EGCG on voltage-gated sodium and potassium channels has been reported in excitable tissues, the in vivo effect of EGCG on the excitability of nociceptive sensory neurons remains to be determined. Our aim was to investigate whether local administration of EGCG to rats attenuates the excitability of nociceptive spinal trigeminal nucleus caudalis (SpVc) neurons in response to mechanical stimulation in vivo. Extracellular single unit recordings w

Iptakalim alleviates synaptic damages via targeting mitochondrial ATP-sensitive potassium channel in depression.

Synaptic plasticity damages play a crucial role in the onset and development of depression, especially in the hippocampus, which is more susceptible to stress and the most frequently studied brain region in depression. And, mitochondria have a major function in executing the complex processes of neurotransmission and plasticity. We have previously demonstrated that Iptakalim (Ipt), a new ATP-sensitive potassium (K-ATP) channel opener, could improve the depressive-like behavior in mice. But the underlying mechanisms are not well understood. The present study demonstrated that Ipt reversed depre

A critical role for the inward rectifying potassium channel Kir7.1 in oligodendrocytes of the mouse optic nerve.

Inward rectifying potassium channels (Kir) are a large family of ion channels that play key roles in ion homeostasis in oligodendrocytes, the myelinating cells of the central nervous system (CNS). Prominent expression of Kir4.1 has been indicated in oligodendrocytes, but the extent of expression of other Kir subtypes is unclear. Here, we used qRT-PCR to determine expression of Kir channel transcripts in the mouse optic nerve, a white matter tract comprising myelinated axons and the glia that support them. A novel finding was the high relative expression of Kir7.1, comparable to that of Kir4.1,

The Novel Inodilator ORM-3819 Relaxes Isolated Porcine Coronary Arteries: Role of Voltage-Gated Potassium Channel Activation.

Relaxation and changes in the transmembrane potential of vascular smooth muscle induced by ORM-3819, a novel inodilating compound, were investigated in isolated porcine coronary arteries. Isometric tone was studied on arterial rings precontracted by KCl (30 mM), and resting membrane potential was investigated by a conventional microelectrode technique. ORM-3819 in the concentration range 0.38-230.6 µM evoked concentration-dependent relaxation with a maximum value of 58.1% and an effective concentration of the relaxing substance that caused 50% of maximum relaxation of 72.2 µM. Nitrile-synthesizing enzyme: Screening, purification and characterization.

Cyanide is known as a toxic compound for almost all living organisms. We have searched for cyanide-resistant bacteria from the soil and stock culture collection of our laboratory, and have found the existence of a lot of microorganisms grown on culture media containing 10 mM potassium cyanide. Almost all of these cyanide-resistant bacteria were found to show β-cyano-L-alanine (β-CNAla) synthetic activity. β-CNAla synthase is known to catalyze nitrile synthesis: the formation of β-CNAla from potassium cyanide and O-acetyl-L-serine or L-cysteine. We found that some microorganisms were able to de
