[Changes in biological properties of botulinum neurotoxin a induced by chemical modification of its molecule by tryptophan and tyrosine].

Using spectrophotometric titration of botulinic neurotoxin A by N-bromosuccinimide, the oxidation of one tryptophane residue was shown to induce an almost complete detoxication of the toxic protein. The conformation of the toxin molecule remained thereby unchanged, as well as the precipitation capacity of the modified toxin after oxidation of two tryptophane residues. The toxin with three or more modified tryptophane residues did not produce precipitation bands with antiserum against original toxin. Nitration of the tyrosine residues in the neurotoxin molecule with tetranitromethane gradually

Interactions between Aβ and mutated Tau lead to polymorphism and induce aggregation of Aβ-mutated tau oligomeric complexes.

One of the main hallmarks of the fronto-temporal dementia with Parkinsonism linked to chromosome 17 (FTDP-17) is the accumulation of neurofibrillary tangles in the brain as an outcome of the aggregation of mutated tau protein. This process occurs due to a number of genetic mutations in the MAPT gene. One of these mutations is the ∆K280 mutation in the tau R2 repeat domain, which promotes the aggregation vis-à-vis that for the wild-type tau. Experimental studies have shown that in Alzheimer's disease Aβ peptide forms aggregates both with itself and with wild-type tau. By analogy, in FTDP-17, it

Calcium binds to leptospiral immunoglobulin-like protein, LigB, and modulates fibronectin binding.

Pathogenic Leptospira spp. express immunoglobulin-like proteins, LigA and LigB, which serve as adhesins to bind to extracellular matrices and mediate their attachment on host cells. However, nothing is known about the mechanism by which these proteins are involved in pathogenesis. We demonstrate that LigBCen2 binds Ca(2+), as evidenced by inductively coupled plasma optical emission spectrometry, energy dispersive spectrometry, (45)Ca overlay, and mass spectrometry, although there is no known motif for Ca(2+) binding. LigBCen2 binds four Ca(2+) as determined by matrix-assisted laser desorption

Further studies on the soluble form (gs) of rabies virus glycoprotein (g): molecular structure of gs protein and possible mechanism of the shedding.

In this study, we investigated the antigenic structures and maturation of some C-terminal-deficient derivatives of rabies virus glycoprotein (G). The Gs protein, a soluble form of G protein shed from infected cells, displayed antigenicity to most of our conformational epitope-specific anti-G mAbs, but took the 1-30-44 epitope-deficient conformation (termed G(C) form). (The 1-30-44 epitope was acid-sensitive and dependent on two separate regions, the Lys-202-containing and Asn-336-containing regions; Kankanamge et al., Microbiol. Immunol., 47: 507-519). Intact G proteins took the 1-30-44 epitop

Zika Virus-Like Particles Bearing a Covalent Dimer of Envelope Protein Protect Mice from Lethal Challenge.

Zika virus (ZIKV) envelope (E) protein is the major target of neutralizing antibodies in infected hosts and thus represents a candidate of interest for vaccine design. However, a major concern in the development of vaccines against ZIKV and the related dengue virus is the induction of cross-reactive poorly neutralizing antibodies that can cause antibody-dependent enhancement (ADE) of infection. This risk necessitates particular care in vaccine design. Specifically, the engineered immunogens should have their cross-reactive epitopes masked, and they should be optimized for eliciting virus-speci

Elevated levels of p53 protein in the neutrophils and monocytes of a patient with chronic idiopathic thrombocytopenic purpura or possible early myelodysplasia?

A 68 year old female who presented with long-term thrombocytopenia was clinically diagnosed as having chronic idiopathic thrombocytopenia purpura (ITP). Increased levels of the tumour suppressor p53 protein were detected by immunohistochemistry in the neutrophils and some monocytes of the peripheral blood preparation using the antibody DO-1, recognizing mutant and wild type p53 protein conformations. However, no positive staining in the peripheral blood samples from 41 myelodysplasias (MDS) and six normal individuals was observed. Single-stranded conformational polymorphism analysis performed …

GeoEPred: A multimodal structure-aware geometric deep learning framework for Gram-negative bacterial secreted effector prediction with sequence semantics.

Accurate prediction of effector proteins secreted by Gram-negative bacteria is important for elucidating bacterial pathogenic mechanisms and developing precise anti-infective strategies. Although existing methods have benefited from the strong sequence feature extraction capacity of pretrained protein language models, reliance on linear sequence information alone often fails to fully capture the three-dimensional conformational signals required for virulence functions. Meanwhile, conventional structure-based methods are limited by the scarcity of experimentally resolved protein structures. To

Phenylketonuria as a protein misfolding disease: The mutation pG46S in phenylalanine hydroxylase promotes self-association and fibril formation.

The missense mutation pG46S in the regulatory (R) domain of human phenylalanine hydroxylase (hPAH), associated with a severe form of phenylketonuria, generates a misfolded protein which is rapidly degraded on expression in HEK293 cells. When overexpressed as a MBP-G46S fusion protein, soluble and fully active tetrameric/dimeric forms are assembled and recovered in a metastable conformational state. When MBP is cleaved off, G46S undergoes a conformational change and self-associates with a lag phase and an autocatalytic growth phase (tetramers≫dimers), as determined by light scattering. The self

Wavelength-dependent conformational changes in collagen after mid-infrared laser ablation of cornea.

We ablated porcine corneas with a free electron laser tuned to either 2.77 or 6.45 microm, two matched wavelengths that predominantly target water and protein, respectively. The ejected nonvolatile debris and the crater left behind were examined by circular dichroism, Raman spectroscopy, and scanning electron microscopy to characterize the postablation conformation of collagen proteins. We found near-complete unfolding of collagen secondary and tertiary structure at either ablating wavelength. On the other hand, we found excess fibril swelling and evidence for excess cis-hydroxyproline in the

Proton nuclear magnetic resonance studies on the variant-3 neurotoxin from Centruroides sculpturatus Ewing: sequential assignment of resonances.

We report the sequential assignment of resonances to specific residues in the proton nuclear magnetic resonance spectrum of the variant-3 neurotoxin from the scorpion Centruroides sculpturatus Ewing (range southwestern U.S.A.). A combination of two-dimensional NMR experiments such as 2D-COSY, 2D-NOESY, and single- and double-RELAY coherence transfer spectroscopy has been employed on samples of the protein dissolved in D2O and in H2O for assignment purposes. These studies provide a basis for the determination of the solution-phase conformation of this protein and for undertaking detailed struct

A way to probe the microenvironment of free sulfhydryls in intact proteins with a series of monofunctional organic mercurials.

Monofunctional organic mercurials (MFOHgs) of different sizes and electronic properties were used to evaluate the accessibility of SH groups in intact proteins. The change in conformation of proteins, such as BLG and OVA, when treated with MFOHgs of increasing size was used as a measure of the accessibility of SH groups contained within these proteins.

The MODNAP benchmark set for modeling ribose-based modified nucleic acid and nucleic acid-protein complexes: a case study with AlphaFold 3.

Modified nucleic acids play important roles in biology and biotechnology. However, their chemical and structural diversity as well as effects on nucleic acid structure and intermolecular interactions remain incompletely characterized. The performance of structure prediction and co-folding algorithms for modeling modified nucleic acids and nucleic acid-protein complexes has also not been evaluated. Here, we systematically analyze ribose-based modified nucleotides present in structures deposited in the Protein Data Bank. Modified nucleotides are primarily localized to the nucleobase, where they

Host Determinants of Prion Strain Diversity Independent of Prion Protein Genotype.

UNLABELLED: Phenotypic diversity in prion diseases can be specified by prion strains in which biological traits are propagated through an epigenetic mechanism mediated by distinct PrP(Sc) conformations. We investigated the role of host-dependent factors on phenotypic diversity of chronic wasting disease (CWD) in different host species that express the same prion protein gene (Prnp). Two CWD strains that have distinct biological, biochemical, and pathological features were identified in transgenic mice that express the Syrian golden hamster (SGH) Prnp. The CKY strain of CWD had a shorter incuba

Spectrophotometric and kinetic studies on normal and thyrotoxic cardiac myosins.

Kinetic and microcalorimetric methods were used to study the interaction of thyrotoxic (V1) and normal adult cardiac myosin (V3) with ATP. It was shown that the overall enthalpy and entropy change was larger in the interaction of ATP with the thyrotoxic than with the normal myosin. There is evidence that the observed enthalpy changes were generated by protein conformational changes connected to the sequential destabilization and restabilization of the prevalent, energetically favored conformation of the myosin molecule. The V1-ATP interaction created more entropy than the V3-ATP interaction, a…

Harnessing an RNA-mediated chaperone for the assembly of influenza hemagglutinin in an immunologically relevant conformation.

A novel protein-folding function of RNA has been recognized, which can outperform previously known molecular chaperone proteins. The RNA as a molecular chaperone (chaperna) activity is intrinsic to some ribozymes and is operational during viral infections. Our purpose was to test whether influenza hemagglutinin (HA) can be assembled in a soluble, trimeric, and immunologically activating conformation by means of an RNA molecular chaperone (chaperna) activity. An RNA-interacting domain (RID) from the host being immunized was selected as a docking tag for RNA binding, which served as a transducer

The snRNP 15.5K protein folds its cognate K-turn RNA: a combined theoretical and biochemical study.

The human 15.5K protein binds to the 5' stem-loop of U4 snRNA, promotes the assembly of the spliceosomal U4/U6 snRNP, and is required for the recruitment of the 61K protein and the 20/60/90K protein complex to the U4 snRNA. In the crystallographic structure of the 15.5K-U4 snRNA complex, the conformation of the RNA corresponds to the family of kink-turn (K-turn) structural motifs. We simulated the complex and the free RNA, showing how the protein binding and the intrinsic flexibility contribute to the RNA folding process. We found that the RNA is significantly more flexible in the absence of t

Imidazole-[1,5‑a]‑Pyridines Can Occupy the EGFR Allostery with a Strong Polar Interaction.

The imidazole-[1,5-a]-pyridine derivatives were recently synthesized and showed remarkable bioactivity against three cancer cell lines, but an understanding of their activities is still missing. To prompt a detailed investigation into their molecular binding mechanisms, we carried out a comprehensive computational workflow for structure-based drug design. The protocol encompasses (i) ligand polarization using quantum chemical calculations, (ii) docking algorithms to generate initial protein-ligand conformations, (iii) molecular dynamics simulations to evaluate ligand diffusion within the prote
