Prophylactic and therapeutic efficacy of poly(I,C)-LC against Rift Valley fever virus infection in mice.

The prophylactic and therapeutic efficacy of polyriboinosinic-polyribocytidylic acid stabilized with poly-L-lysine and carboxymethylcellulose [poly(I,C)-LC] was evaluated in female Swiss Webster mice against a lethal infection of Rift Valley fever virus (RVFV). Prophylactically, the best effect was obtained with 2-3 doses of 1-20 micrograms poly(I,C)-LC, which fully protected the mice in a schedule-dependent fashion against an LD100 RVFV challenge. Multiple intermittent therapeutic administration of 20 micrograms poly(I,C)-LC 24 or 48 h after infection protected 100% and 50% of the mice, respe…

Interepidemic Rift Valley fever in East Africa: the recent risk landscape and projected impacts of global change.

Rift Valley fever (RVF) is a zoonotic disease that causes sporadic, multi-country epidemics. However, there is limited understanding of RVF virus circulation during interepidemic periods and the potential impacts of global change on interepidemic RVF. To address these knowledge gaps, we built a predictive model using recent interepidemic RVF outbreak data from Kenya, Tanzania and Uganda. We then projected interepidemic RVF risk for three future time periods (2021-2040, 2041-2060, 2061-2080) under three global change scenarios representing different trajectories for climate and human population…

Spatiotemporal patterns of Rift Valley fever virus in Africa: a retrospective genomic epidemiology and phylodynamic modelling study.

BACKGROUND: Rift Valley fever virus (RVFV) is a mosquito-borne zoonotic pathogen causing outbreaks in humans and ruminants across Africa and the Arabian Peninsula. Originally restricted to the Great Rift Valley, RVFV has expanded geographically, prompting its classification by WHO as a pathogen of pandemic potential. We investigated the evolutionary and spatial dynamics of RVFV across Africa. METHODS: We used genomic data generated at the International Livestock Research Institute Nairobi genomic laboratory (BioProject PRJNA1106221) and combined with publicly available datasets retrieved from…

Biochemical Basis for LRP1 Interaction With Rift Valley Fever Virus Glycoprotein and Its Role in Viral Entry.

The LDL receptor related protein 1 (LRP1) is a host entry factor for Rift Valley fever virus (RVFV), a negative sense RNA virus in the Bunyaviricetes class within the Phenuiviridae family. Previous studies revealed that RVFV glycoproteins (Gn and Gc; GnGc) primarily interact with LRP1 clusters II (CLII) and IV (CLIV) to promote viral entry. However, the mechanism of RVFV GnGc binding to LRP1 is not fully characterized. In this study, we have successfully mapped the roles of individual LRP1 ligand binding regions, known as CR domains, and identified CR25 to be sufficient for high affinity RVFV …

Co-Infection of Culex tarsalis Mosquitoes with Rift Valley Fever Phlebovirus Strains Results in Efficient Viral Reassortment.

Rift Valley fever phlebovirus (RVFV) is a zoonotic mosquito-borne pathogen endemic to sub-Saharan Africa and the Arabian Peninsula which causes Rift Valley fever in ruminant livestock and humans. Co-infection with divergent viral strains can produce reassortment among the L, S, and M segments of the RVFV genome. Reassortment events can produce novel genotypes with altered virulence, transmission dynamics, and/or mosquito host range. This can have severe implications in areas where RVFV is endemic and convolutes our ability to anticipate transmission and circulation in novel geographic regions.…

Atypical hyperendemicity of Rift Valley fever in Southwestern Uganda associated with the rapidly evolving lineage C viruses.

INTRODUCTION: Recent Rift Valley fever (RVF) epidemiology in eastern Africa region is characterized by widening geographic range and increasing frequency of small disease clusters. Here we conducted studies in southwestern (SW) Uganda region that has since 2016 reported increasing RVF activities. METHODS: A 22-month long hospital-based study in three districts of SW Uganda targeting patients with acute febrile illness (AFI) or unexplained bleeding was followed by a cross-sectional population-based human-animal survey. We then estimated RVFV force of infection (FOI) and yearly cases using the …

Rift Valley Fever Outbreak Investigation Associated with a Dairy Farm Abortion Storm, Mbarara District, Western Uganda, 2023.

In Africa, Rift Valley Fever poses a substantial risk to animal health, and human cases occur after contact with infected animals or their tissues. RVF has re-emerged in Uganda after nearly five decades, with multiple outbreaks recorded since 2016. We investigated a unique RVF outbreak associated with an animal abortion storm of 30 events and human cases on a dairy farm in Mbarara District, Western Uganda, in February 2023. Genomic analysis was performed, comparing animal and human RVF viruses (RVFV) circulating in the region. A cluster of thirteen human RVF cases and nine PCR-positive animals…

Limiting viral replication in hematopoietic cells delays Rift Valley fever virus disease progression in C57BL/6 mice.

Rift Valley fever virus (RVFV) causes mild to severe disease in livestock and humans. It was first identified in 1931 during an epizootic in Kenya and has spread across Africa and into the Middle East. Hematopoietic cells are one of the major targets of RVFV in vivo; however, their contribution to RVFV pathogenesis remains poorly understood. To address this, we generated a recombinant miRNA-targeted virus, RVFVmiR-142, to limit viral replication in hematopoietic cells and evaluated the consequences on RVFV disease manifestations in C57BL/6 mice. MicroRNAs are evolutionarily conserved non-codin…

LRP1 Interacts with the Rift Valley Fever Virus Glycoprotein Gn via a Calcium-Dependent Multivalent Electrostatic Mechanism.

The Rift Valley fever virus (RVFV) is a highly pathogenic, mosquito-borne zoonotic virus that poses a significant risk to livestock, human health, and global public health security. Although RVFV is classified by the World Health Organization (WHO) as a priority pathogen with epidemic potential, no licensed vaccines or effective antiviral therapies are currently available. A limited understanding of the molecular mechanisms of RVFV entry has hindered therapeutic development. Here, we elucidate the molecular basis by which the RVFV envelope glycoprotein Gn recognizes its receptor, low-density l…

Ability of a dynamical climate sensitive disease model to reproduce historical Rift Valley Fever outbreaks over Africa.

Rift Valley Fever (RVF) is a zoonosis transmitted by Aedes and Culex mosquitoes, and is considered a priority pathogen by the WHO. RVF epidemics mostly occur in Africa and can decimate livestock herds, causing significant economic losses and posing health risks for humans. RVF transmission is associated with the occurrence of El Niño events that cause floods in eastern Africa and favour the emergence of mosquitoes in wetlands. Different risk models have been developed to forecast RVF transmission risk but very few studies have validated models at pan-African scale. This study aims to validate …

IFITMs from Naturally Infected Animal Species Exhibit Distinct Restriction Capacities against Toscana and Rift Valley Fever Viruses.

Rift Valley Fever virus (RVFV) and Toscana virus (TOSV) are two pathogenic arthropod-borne viruses responsible for zoonotic infections in both humans and animals; as such, they represent a growing threat to public and veterinary health. Interferon-induced transmembrane (IFITM) proteins are broad inhibitors of a large panel of viruses belonging to various families and genera. However, little is known on the interplay between RVFV, TOSV, and the IFITM proteins derived from their naturally infected host species. In this study, we investigated the ability of human, bovine, and camel IFITMs to rest…

Climate Conditions During a Rift Valley Fever Post-epizootic Period in Free State, South Africa, 2014-2019.

Rift Valley fever virus (RVFV) activity in Southern Africa tends to occur during periods of sustained elevated rainfall, cooler than normal conditions, and abundant vegetation cover creating ideal conditions for the increase and propagation of populations of RVFV mosquito vectors. These climatic and ecological conditions are modulated by large-scale tropical-wide El Niño-Southern Oscillation (ENSO) phenomena. The aim of this 5-year study was to investigate climatic conditions during Rift Valley fever "post-epizootic" period in Free State province of the Republic of South Africa, which historic…

Seroprevalence and Associated Risk Factors of Rift Valley Fever in Livestock from Three Ecological Zones of Malawi.

The epidemiology of Rift Valley fever (RVF) is poorly understood in Malawi. Here, a cross-sectional study was conducted (March-June 2020) to investigate the seroprevalence and potential risk factors of RVF virus (RVFV) in cattle, goats, and sheep in three ecological zones of Malawi. A total of 1523 serum samples were tested for anti-RVFV IgG and IgM antibodies by ELISA. Additionally, a questionnaire survey was used to assess potential RVF risk factors. The overall seroprevalence was 17.14% (261/1523; 95% CI = 15.33-19.11) for individual livestock and 33.24% (120/361; 95% CI = 28.18-38.11) for …

Leveraging livestock movements to urban slaughterhouses for wide-spread Rift Valley fever virus surveillance in Western Kenya.

Rift Valley fever virus (RVFV) is an economically devastating, zoonotic arbovirus endemic across Africa with potential to cause severe disease in livestock and humans. Viral spread is primarily driven by movement of domestic ruminants and there is a high potential for transboundary spread. Despite influx of livestock to urban areas in response to the high demand for meat and animal products, RVFV has not been detected in any urban center. The objectives of this study were to determine the feasibility of assessing risk of RVFV introduction to urban Kisumu, Kenya, by testing slaughtered livestoc…

Detection of Rift Valley Fever Virus RNA in Formalin-Fixed Mosquitoes by In Situ Hybridization (RNAscope®).

Rift Valley fever virus (RVFV) causes a zoonotic mosquito-borne haemorrhagic disease that emerges to produce rapid large-scale outbreaks in livestock within sub-Saharan Africa. A range of mosquito species in Africa have been shown to transmit RVFV, and recent studies have assessed whether temperate mosquito species are also capable of transmission. In order to support vector competence studies, the ability to visualize virus localization in mosquito cells and tissue would enhance the understanding of the infection process within the mosquito body. Here, the application of in situ hybridization…

Risk assessment for the Rift Valley fever occurrence in China: Special concern in south-west border areas.

Rift Valley fever (RVF) is a mosquito-borne zoonotic disease. Since its first outbreak in 1930, RVF epidemics have caused huge economic losses and public health impacts in Africa. In 2000, RVF became a disease of global concern as it spread to the Arabian Peninsula. In our study, a Geographic Information System-based risk assessment for the occurrence of Rift Valley fever in China was established by means of ecological niche modelling. Based on occurrence records (RVF records from FAO EMPRES-i, vector records from literatures and GBIF) and high-resolution environmental layers, the prediction m…
