Occurrence and molecular characterization of Cryptosporidium and Giardia in captive wild ruminants in Belgium.

Both Cryptosporidium and Giardia are frequently found in the stool of domestic ruminants, especially young animals. Wild ruminants are also host to these protozoa, but the prevalence of these parasites in both free-ranging and captive nondomesticated ruminants needs to be further investigated. Moreover, the role of wild ruminants serving as reservoirs for these zoonotic parasites remains unclear. Therefore, a cross-sectional survey was conducted to estimate the occurrence of Cryptosporidium and Giardia in captive wild ruminants younger than 6 mo and to determine the potential of these animals …

Molecular identification, genetic diversity and distribution of Theileria and Babesia species infecting small ruminants.

Detection and identification of Theileria and Babesia species in 920 apparently healthy small ruminants in eastern Turkey, as well as parasite genetic diversity, was investigated using a specifically designed reverse line blot (RLB) assay. The hypervariable V4 region of the 18S ribosomal RNA (rRNA) gene was amplified and hybridized to a membrane onto which catchall and species-specific oligonucleotide probes were covalently linked. Three Theileria and one Babesia genotype were identified. Comparison of the Theileria genotypes revealed 93.6-96.2% similarity among their 18S rRNA genes. Two Theil…

Distribution of brucellosis among small ruminants in the pastoral region of Afar, eastern Ethiopia.

A cross-sectional study was conducted in the pastoral region of Afar, in eastern and central Ethiopia, to determine the distribution of brucellosis in small ruminants. Between December 2005 and June 2006, 1,568 serum samples were taken: 563 samples from sheep and 1,005 from goats. One hundred and forty-seven of these (9.4%) tested positive using the Rose Bengal plate test (RBPT), and 76 (4.8%) also tested positive by the complement fixation test (CFT). Brucellosis was detected in all five administrative zones of the region. The difference in prevalence (P) among the zones was not statistically…

Spiroplasma spp. from transmissible spongiform encephalopathy brains or ticks induce spongiform encephalopathy in ruminants.

Spiroplasma, small motile wall-less bacteria, are linked by molecular and serological studies to the transmissible spongiform encephalopathies (TSEs), which include scrapie in sheep, chronic wasting disease (CWD) in deer and Creutzfeldt-Jakob disease in humans. In this study, two experiments were undertaken to determine the role of spiroplasma in the pathogenesis of TSE. In experiment 1, Spiroplasma mirum, a rabbit tick isolate that had previously been shown to experimentally induce spongiform encephalopathy in rodents, was inoculated intracranially (IC) into ruminants. S. mirum-inoculated dee…

A review of dicrocoeliosis of ruminants including recent advances in the diagnosis and treatment.

Despite its widespread presence among grazing ruminants, dicrocoeliosis, also known as "small liver fluke" disease, is poorly known and often underestimated by researchers and practitioners in many countries. This is primarily due to the multiple parasitic infections which affect ruminant livestock and mask the pathology of dicrocoeliosis, to the difficulties in diagnosing it with coprological techniques and, finally, to the few effective drugs found. Furthermore, the biological cycle of Dicrocoelium, which requires a snail and an ant as intermediate hosts, and the high number of ecological an…

Cathepsin L proteinases as vaccines against infection with Fasciola hepatica (liver fluke) in ruminants.

Fasciolosis, caused by trematodes of the genus Fasciola, is one of the most important diseases of farmed ruminants in temperate and tropical zones. The appearance of Fasciola hepatica populations that are resistant to common flukicidal drugs means that new methods of treatment will soon be required. The future prospect for the development of anti-liver fluke vaccines is optimistic and given their consumer acceptability and environmental friendliness, offer the best way forward. Cathepsin L proteases (Fhe CL 1 and Fhe CL 2), secreted by liver flukes at all stages of their development in the mam…

Tick-borne diseases in ruminants of Central and Southern Italy: epidemiology and case reports.

Sera and blood from cattle and sheep were examined for the presence of Babesia and Theileria spp by microscopy and serology at the Parasitology Department of the Istituto Zooprofilattico Sperimentale of Abruzzo and Molise (IZSAM). Of the 47 bovine herds (323 animals) tested, 15 were found positive for Babesia bigemina and 1 for Babesia bovis. Two outbreaks occurred, one caused by B. bigemina and one by B. bovis. The B. bigemina outbreak occurred in Abruzzo and has been followed for two years. The isolate of B. bigemina was very pathogenic leading to the death of two cows out of 57. The vector

Visual-evoked potentials and electroretinograms in ruminants with thiamine-responsive polioencephalomalacia or suspected listeriosis.

Electrodiagnostic visual testing (electroretinogram [ERG] and visual-evoked potential [VEP]) was performed on 5 ruminants (3 lambs, 1 kid, and 1 steer) with thiamine-responsive polioencephalomalacia (PEM) and on 2 sheep with listeriosis. The lambs and kid had typical clinical signs of PEM, especially blindness. In these animals, the ERG was normal but the VEP was abnormal. Follow-up recordings in the kid and 1 lamb indicated an improvement in VEP recordings accompanying a gradual return of vision after thiamine treatment. Possible subtle changes in VEP peak latencies could not be assessed beca…

The redox potential, rH and pH values in the gastrointestinal tract of small ruminants.

Oxidation-reduction potentials (Eh), pH and rH in the gastrointestinal tract were measured in six goats and two sheep fed on ground barley and hay, with or without the addition of urea. Each ration was supplied for three weeks. The animals were slaughtered after morning feeding, the contents of relevant parts of the gastrointestinal tract were sampled, pH and Eh values were measured and rH values calculated. The range of the oxidation-reduction potential was rather extensive, from -300 to +186 mV. The variability of rH values was smaller, being between 4.6 and 12.9, except for three values. Th

Lactogenic hormones: binding sites, mammary growth, secretory cell differentiation, and milk biosynthesis in ruminants.

Roles of the lactogenic hormones prolactin and placental lactogen in mammary development in ruminants were reviewed. In contrast with other ruminants, failure to detect lactogenic activity in the serum of pregnant cows (in excess of that attributed to prolactin) suggests that placental lactogen may have little direct effect on mammary growth or lactogenesis. However, replacement and ablation experiments using ergocryptine provide definitive evidence that increased periparturient secretion of prolactin is necessary for maximal milk production in cattle. Quantitative microscopy indicates a relat…

The copper-molybdenum antagonism in ruminants. I. The formation of thiomolybdates in animal rumen.

The formation of thiomolybdates, MoOxS4--x2--(x = 0, 1, 2, or 3), from molybdate and sulphide salts in aqueous media has been studied under conditions which simulate the fluid phase in the rumen. The influences of the sulphide:molybdenum ratio, pH and phosphate levels on the nature of the species formed were investigated. The thiomolybdates, in particular the MoS42-- ion, have been implicated as the active intermediates in the widespread molybdenum induced copper deficiency that affects ruminants. The results presented here show that, under physiological conditions, di- and trithiomolybdates w…

Cerebrocortical necrosis in ruminants: effect of thiaminase type 1-producing Clostridium sporogenes in lambs.

Large numbers of orally inoculated thiaminase type 1-producing Clostridium sporogenes failed to establish in the alimentary tract of two conventionally born lambs. Conversely, when similar inoculations were given to two gnotobiotic lambs, large populations of Cl sporogenes established in their rumens and correspondingly high levels of thiaminase were produced. No clinical symptoms of thiamine deficiency or cerebrocortical necrosis were seen despite the presence of high levels of thiaminase in the rumen of one of the gnotobiotic lambs for a period of 86 days.

Migration inhibition studies on peripheral leukocytes and lymph node cells from ruminants with fascioliasis.

Delayed type hypersensitivity against antigens of Fasciola hepatica has been repeatedly documented in infected hosts. Evidence has been presented to suggest that the delayed reactivity may develop earlier in the regional lymph nodes of the parasitized organ than in other lymph nodes of the body (Soulsby 1971).

Associative effects of external buffers and plant secondary metabolites on ruminal fermentation during an induced subacute ruminal acidosis challenge in beef steers.

Subacute ruminal acidosis (SARA) is a prevalent problem in cattle consuming highly fermentable diets. It was hypothesized that a novel feed additive (BRIC) containing alkaline salts and traces of plant secondary metabolites may modulate ruminal fermentation. The experiment evaluated four treatments during an induced SARA protocol. Treatments were (DM basis): 1) sodium bicarbonate at 1.2% (BICARB), 2) a commercial buffer at 1.2% (BRIC), 3) a blend of tannins and saponins at 0.15% (BXS), and 4) a combination of 1.2% BRIC plus 0.15% BXS (BRIC-BXS). Eight ruminally cannulated steers were used in a

The effects of antibiotic-free supplementation on the ruminal pH variability and methane emissions of beef cattle under the challenge of subacute ruminal acidosis (SARA).

Subacute ruminal acidosis (SARA) in feedlot cattle during the feed transition to grain-based diets is a significant constraint to animal health and productivity. This experiment assessed an antibiotic-free supplement (ProTect®) effects on ruminal pH variability and methane (CH4) emissions of cattle during the challenge of SARA. Ten 18-month-old Angus steers (472 ± 4.8 kg) were randomly allocated into monensin (n = 5) and ProTect® groups (n = 5) and progressively introduced to grain diets incorporating monensin or ProTect® for 36 days of the experiment [starter (7 days; 45% grain), T1 (7 days;

Don't look back in anger: neural correlates of reappraisal, analytical rumination, and angry rumination during recall of an anger-inducing autobiographical memory.

Despite the enormous costs associated with unrestrained anger, little is known about the neural mechanisms underlying anger regulation. Behavioral evidence supports the effectiveness of reappraisal in reducing anger, and demonstrates that rumination typically maintains or augments anger. To further understand the effects of different anger regulation strategies, during functional magnetic resonance imaging 21 healthy male and female undergraduates recalled an anger-inducing autobiographical memory. They then engaged in three counterbalanced anger regulation strategies: reappraisal, analytical

Reconceptualizing co-rumination: A novel theoretical and multidisciplinary perspective.

Co-rumination is a social emotion regulation strategy characterized by extensive and exhaustive discussions of stressors, problems, and negative emotions with another person. While research establishing the costs and benefits associated with co-rumination has been formative, the focus on explaining heightened internalizing symptoms and increased relationship quality (i.e., the "tradeoff hypothesis") in mostly adolescent friendships needs to be expanded. Through a social psychological lens, we pave a way forward by offering a new theoretical conceptualization of co-rumination that emphasizes th

Evolutionary and epidemiological considerations for anthelmintic treatments in ruminant livestock.

Anthelmintic drug resistance is a concern for the sustained control of gastrointestinal nematodes (GINs) in ruminant livestock globally. Evolutionary-epidemiological modelling, which considers both parasite dynamics and resistance dynamics in response to interventions, can be useful in determining which anthelmintic resistance management (ARM) strategies may be effective without compromising parasite control. We address two key questions in ARM. First, how to improve the measurement of AR in populations. Second, identifying effective ARM strategies to slow the spread of AR while maintaining ef
