Root-Apex Proton Fluxes at the Centre of Soil-Stress Acclimation.

Proton (H+) fluxes in plant roots play critical roles in maintaining root growth and facilitating plant responses to multiple soil stresses, including fluctuations in nutrient supply, salt infiltration, and water stress. Soil mining for nutrients and water, rates of nutrient uptake, and the modulation of cell expansion all depend on the regulation of root H+ fluxes, particularly at the root apex, mediated primarily by the activity of plasma membrane (PM) H+-ATPases. Here, we summarize recent findings on the regulatory mechanisms of H+ fluxes at the root apex under three abiotic stress conditio…

Salmonella persistence in soil depends on reciprocal interactions with indigenous microorganisms.

Fresh fruits and vegetables have numerous benefits to human health. Unfortunately, their consumption is increasingly associated with food-borne diseases, Salmonella enterica being their most frequent cause in Europe. Agricultural soils were postulated as reservoir of human pathogens, contributing to the contamination of crops during the growing period. Since the competition with the indigenous soil microbiota for colonization sites plays a major role in the success of invading species, we hypothesized that reduced diversity will enhance the chance of Salmonella to successfully establish in agr…

Paracoccidioides spp. in Soil from the Pampa Biome in Southern Brazil.

High rates of paracoccidioidomycosis disease and infection have been recently reported in southern Brazil. However, the knowledgment about the ecoepidemiology of Paracoccidioides spp. in the region is limited, therefore, this study aimed to evaluate the presence of Paracoccidioides spp. DNA in soil samples from a rural area in Southern Rio Grande do Sul, Brazil. Thirty pools of soil samples from Bagé, RS (31º19'53"S 54º06'25"W) were submitted to physicochemical analysis, and to fungal DNA extraction by Norgen Biotek® Kit (Thorold, Canada), followed by Nested PCR technique with ITS4 and ITS5 as…

Reduced colonization by soil invertebrates to irradiated decomposing wood in Chernobyl.

Soil is inhabited by a range of microbes, invertebrates and vertebrates that disintegrate and decompose dead wood and leaf litter. These communities can be perturbed by ionizing radiation from natural radiation sources or from radiation originating from nuclear accidents such as those at Chernobyl, Fukushima and Three Mile Island. We used experimental manipulations of wood quality due to differences in exposure to ionizing radiation among tree trunks and ambient radiation levels of the soil to test the hypothesis that radioactively contaminated wood would result in a negative correlation betwe…

Soil dissolved organic matter affects mercury immobilization by biogenic selenium nanoparticles.

Molecular weight (MW) heterogeneity is a fundamental property of dissolved organic matter (DOM) in soil, which has been demonstrated to influence the binding behaviour between DOM and engineered nanoparticles. In the present study, DOM, extracted from black soil, was dialyzed into four fractions: above 10,000 Da, 3500-10,000 Da, 1000-3500 Da and 100-1000 Da. Homoaggregation and fluorescence quenching titration of selenium nanoparticles (SeNPs) was examined in the presence of the different DOM fractions, as well as the consequences for immobilization of elemental mercury. It was found that the …

Composting To Inactivate Foodborne Pathogens for Crop Soil Application: A Review.

Compost is organic material that has been degraded into a nutrient-stabilized humus-like substance through intense microbial activity, which can provide essential plant nutrients (nitrogen, phosphorus) to aid in the growth of fruits and vegetables. Compost can be generated from animal waste feedstocks; these can contain human pathogens, which can be inactivated through the heat and microbial competition promoted during the composting process. Outbreaks of infections caused by bacterial pathogens such as Escherichia coli O157:H7, Salmonella, and Listeria monocytogenes on fruit and vegetable com

Characterising variation in wheat traits under hostile soil conditions in India.

Intensive crop breeding has increased wheat yields and production in India. Wheat improvement in India typically involves selecting yield and component traits under non-hostile soil conditions at regional scales. The aim of this study is to quantify G*E interactions on yield and component traits to further explore site-specific trait selection for hostile soils. Field experiments were conducted at six sites (pH range 4.5-9.5) in 2013-14 and 2014-15, in three agro-climatic regions of India. At each site, yield and component traits were measured on 36 genotypes, representing elite varieties from…

Nitrate capture and slow release in biochar amended compost and soil.

Slow release of nitrate by charred organic matter used as a soil amendment (i.e. biochar) was recently suggested as potential mechanism of nutrient delivery to plants which may explain some agronomic benefits of biochar. So far, isolated soil-aged and composted biochar particles were shown to release considerable amounts of nitrate only in extended (>1 h) extractions ("slow release"). In this study, we quantified nitrate and ammonium release by biochar-amended soil and compost during up to 167 h of repeated extractions in up to six consecutive steps to determine the effect of biochar on the ov…

Mercury fractionation in soil and sediment samples using thermo-desorption method.

Mercury is one of the most hazardous elements. Its transformation and transport in the environment strongly depend on its chemical form. The information about Hg form is also important for understanding the bioavailability as well as toxicity of this element. There are many methods for Hg speciation but most of them are expensive and time consuming. Therefore the aim of this research was to develop a simple thermo-desorption method for mercury fractionation in soil and sediment samples using a direct mercury analyser. The DMA-80 direct mercury analyser (Milestone, Italy) was chosen for this pu…

Importance of organic amendment characteristics on bioremediation of PAH-contaminated soil.

This study investigates the importance of the organic matter characteristics of several organic amendments (i.e., buffalo manure, food and kitchen waste, fruit and vegetables waste, and activated sewage sludge) and their influence in the bioremediation of a polycyclic aromatic hydrocarbons (PAH)-contaminated soil. The removal of low molecular weights (LMW) and high molecular weights (HMW) PAHs was monitored in four bioremediation reactors and used as an indicator of the role of organic amendments in contaminant removal. The total initial concentration of LMW PAHs was 234 mg kg(-1) soil (dry we…

Influence of commercial formulation on leaching of four pesticides through soil.

Studies with small soil columns (2cm i.d.×5.4cm depth) compared leaching of four pesticides added either as technical material or as commercial formulations. Pesticides were selected to give a gradient of solubility in water between 7 and 93mgL-1, comprising azoxystrobin (emulsifiable concentrate, EC, and suspension concentrate, SC), cyproconazole (SC), propyzamide (SC) and triadimenol (EC). Columns of sandy loam soil were leached with 6 pore volumes of 0.01M CaCl2 either 1 or 7days after treatment. Separate experiments evaluated leaching of triadimenol to full breakthrough following addition …

Hepatic fibrosis and the microenvironment: fertile soil for hepatocellular carcinoma development.

Hepatocellular carcinoma is an emerging worldwide health threat that has few curative treatment options and poor overall survival. Progressive hepatic fibrosis is a common pathway for all forms of chronic liver disease and is closely linked epidemiologically to hepatocellular carcinoma risk. However, the molecular events that predispose a fibrotic liver to cancer development remain elusive. Nonetheless, a permissive hepatic microenvironment provides fertile soil for transition of damaged hepatocytes into hepatocellular carcinoma. Key predisposing features include alterations in the extracellul…

Availability of different phosphorus forms in agricultural soil to Microcystis aeruginosa.

We investigated the availability of different forms of particulate soil phosphorus (P) to Microcystis aeruginosa by sequential extraction and bioassay. We cultured M. aeruginosa in media containing, as the sole source of P, soils sequentially extracted with 1 M NH4Cl, 0.11 M bicarbonate dithionite, 1 M NaOH, and 0.5 M HCl. Analyses of chlorophyll-a, particulate organic carbon, and particulate organic nitrogen showed that M. aeruginosa could utilize some of the P remaining in the soil after each extraction. Alkaline phosphatase (AP) assays of sequentially extracted soils showed distinct pattern…

Trace metals in soil and vegetables and associated health risk assessment.

The objective of this study was to assess the contamination level of trace metals in soil and vegetables and health risk to the urban population in Bangladesh. The range of Cr, Ni, Cu, As, Cd, and Pb in agricultural soils was 158-1160, 104-443, 157-519, 41-93, 3.9-13, and 84-574 mg/kg, respectively. Sequential extraction tests revealed that the studied metals were predominantly associated with the residual fraction, followed by the organically bound phase. Concerning Cu, Ni, Cd, and Pb in vegetables, more than 50 % samples exceeded the FAO/WHO recommended permissible limits. Considering the tr…

Sporothrix schenckii sensu stricto isolated from soil in an armadillo's burrow.

Sporotrichosis is a polymorphic disease of man and animals caused by traumatic implantation of propagules into the skin and subcutaneous tissue. Pathogenic species includes S. brasiliensis, S. schenckii, S. globosa and S. luriei. The disease is remarkable for its occurrence as sapronoses and/or zoonosis outbreaks in tropical and subtropical areas; although, the ecology of the clinical clade is still puzzling. Here, we describe an anamorphic Sporothrix strain isolated from soil in an armadillo's burrow, which was located in a hyper endemic area of Paracoccidioidomycosis in Brazil. This isolate …

Forest calcium depletion and biotic retention along a soil nitrogen gradient.

High nitrogen (N) accumulation in terrestrial ecosystems can shift patterns of nutrient limitation and deficiency beyond N toward other nutrients, most notably phosphorus (P) and base cations (calcium [Ca], magnesium [Mg], and potassium [K]). We examined how naturally high N accumulation from a legacy of symbiotic N fixation shaped P and base cation cycling across a gradient of nine temperate conifer forests in the Oregon Coast Range. We were particularly interested in whether long-term legacies of symbiotic N fixation promoted coupled N and organic P accumulation in soils, and whether biotic …

Modelling the geographical distribution of soil-transmitted helminth infections in Bolivia.

BACKGROUND: The prevalence of infection with the three common soil-transmitted helminths (i.e. Ascaris lumbricoides, Trichuris trichiura, and hookworm) in Bolivia is among the highest in Latin America. However, the spatial distribution and burden of soil-transmitted helminthiasis are poorly documented. METHODS: We analysed historical survey data using Bayesian geostatistical models to identify determinants of the distribution of soil-transmitted helminth infections, predict the geographical distribution of infection risk, and assess treatment needs and costs in the frame of preventive chemoth…
