High SIGLEC10-Expressing Tumor-Associated Macrophages Participate in Remodeling the Tumor Immune Microenvironment and Predict Poor Prognosis in Colorectal Cancer.

BACKGROUND: Sialic acid-binding immunoglobulin-like lectin 10 (SIGLEC10), a member of the SIGLEC family, is selectively expressed on multiple immune cell subsets and plays a key role in immune regulation through the recognition of sialylated ligands. Recent studies suggest that SIGLEC10 functions as an emerging immune checkpoint that contributes to the regulation of tumor-associated macrophages (TAMs) within the tumor microenvironment (TME). METHODS: Immunohistochemistry was performed to assess SIGLEC10 expression in a CRC cohort comprising 202 patients, and the association between SIGLEC10 e…

BTG2-deficient mast cells remodel the tumor and tumor-draining lymph node microenvironment leading to chemotherapy resistance in breast cancer.

BACKGROUND: Breast cancer is currently the most frequently diagnosed malignancy worldwide, with chemotherapy resistance being a major contri … l. CONCLUSIONS: This study highlights the immunoregulatory role of mast cells in the breast cancer tumor microenvironment and establishes a link between BTG2 expression in mast cells and neoadjuvant chemotherapy response. These findings provide a foundational basis for preserving functional lymph nodes and optimizing combined radiotherapy treatment strategies.

Molecular, metabolic, and histological subtypes of pancreatic ductal adenocarcinoma and its tumor microenvironment: Insights into tumor heterogeneity and clinical implications.

Pancreatic ductal adenocarcinoma (PDAC) is a highly heterogeneous cancer with poor prognosis and limited therapeutic options. Bulk transcriptomic profiling has identified two major gene expression-based molecular subtypes: classical/progenitor and basal-like/squamous/quasimesenchymal. These subtypes differ in biological characteristics, differentiation status, drug sensitivity, and clinical outcomes. Advances in single-cell and spatial transcriptomics have further revealed intermediate/hybrid states, as well as distinct subtypes within the tumor microenvironment. These technologies have also u…

Targeting tumor-associated macrophage polarization with traditional Chinese medicine active ingredients: Dual reversal of chemoresistance and immunosuppression in tumor microenvironment.

Chemotherapy resistance and immunosuppression are major causes of tumor treatment failure. The polarization state of tumor-associated macrophages (TAMs) is a central regulatory hub for both processes. Traditional Chinese medicine (TCM) has the characteristics of multi-component, multi-target, and multi-pathway. It regulating M1/M2 polarization is promising due to the high plasticity of TAMs. This review comprehensively explores the anti-tumor effects of TCM active components through multiple targets such as metabolic reprogramming. The mechanism includes regulating TAM's polarization, reversin

Tumor Budding in Colorectal Carcinoma Showing a Paradoxical Mitotic Index (Via PHH3) With Possible Association to the Tumor Stromal Microenvironment.

BACKGROUND: Colorectal carcinomas (CC) are one of the most commonly diagnosed malignancies. Tumor budding (the histologic process of dissociation that occurs at the invasive margin of colorectal cancer), has significant prognostic implications, in that higher tumor budding is associated with adverse histopathologic and clinical outcomes. Because of this prognostic significance, more research is needed to further understand the pathologic and immunohistochemical (IHC) associations pertaining to this important prognostic variable. In this study, we will further evaluate selective clinopathologic

IDO-inhibitor potentiated immunogenic chemotherapy abolishes primary tumor growth and eradicates metastatic lesions by targeting distinct compartments within tumor microenvironment.

Immunogenic chemotherapy (IC) is a type of chemotherapy where certain chemodrugs induce immunogenic cancer cell death (ICD), which in turn arouses T cell antitumor immunity. However, IC concurrently upregulates a key immune suppressor, indoleamine-2,3-dioxygenase (IDO), in both cancer cells and immune cells. IDO-mediated immunosuppression significantly offsets IC's therapeutic benefits in cancer patients, suggesting a necessity of combination with IDO inhibitors. Here, we report an enzyme-, pH-, and redox-triple-sensitive nanosystem using mesoporous silica nanoparticles (MSNs) as a core encaps

Cross talk between follicular Th cells and tumor cells in human follicular lymphoma promotes immune evasion in the tumor microenvironment.

The microenvironment of human follicular lymphoma (FL), an incurable B cell non-Hodgkin's lymphoma, is thought to play a major role in its p … n in a feed-forward cycle. Thus, TFH appear to play a major role in generating an immunosuppressive tumor microenvironment that promotes immune escape and tumor survival and growth. Our results provide novel insights into the cross talk among TFH, tumor cells, and Tregs in FL, and offer potential targets for development of therapeutic strategies to overcome immune evasi …

Tumor-specific in situ IL-12 expression combined with αPD-L1 suppresses melanoma growth by remodeling the tumor immune microenvironment.

Interleukin-12 (IL-12) potently activates antitumor immune responses and compensates for the challenge of insufficient T cell activation and infiltration faced in immune checkpoint therapy. However, its clinical application is limited by severe systemic toxicity and the upregulation of PD-L1 on tumor cells during treatment. Here, we developed iLANpTyr-IL-12, a lipid-assisted nanoparticle system loaded with tyrosinase (Tyr) promoter-driven IL-12 plasmids, designed to achieve specific and sustained IL-12 expression exclusively in melanoma cells. The secreted IL-12 promoted dendritic cell maturat

Triple-remodeling of tumor microenvironment through hyaluronidase-assisted folate-targeted lipid nanoparticle-mediated siVEGF/siPD-L1 for enhanced tumor immunotherapy.

BACKGROUND: In tumor therapy, traditional single-treatment methods often fail to achieve ideal results because of problems like the complex tumor microenvironment, immune escape mechanisms, and abnormal blood vessel formation. Although there are now several combined treatment strategies, challenges remain in precise drug delivery and achieving multi - target synergistic effects. METHODS: This study aims to create an efficient combined anti-tumor lipid nanoparticle (LNP) delivery system. We added DMG-PEG2000-FA to the four-component lipid nanoparticle system and used microfluidic technology to …

Prostate tumor immune microenvironment changes following immunotherapy shared by patients who developed anti-tumor response or immune-related adverse events.

Checkpoint inhibitors targeting PD-1/PD-L1 have revolutionized cancer immunotherapy, yet their efficacy in prostate cancer has been limited. We previously reported a clinical trial (NCT02499835) evaluating PD-1 blockade combined with an anti-tumor DNA vaccine, pTVG-HP (encoding prostatic acid phosphatase), in patients with metastatic castration-resistant prostate cancer. Decreases in serum prostate-specific antigen (PSA) levels were observed in 35% of patients, and 42% of patients developed immune-related adverse events (irAE). In long-term follow-up, any decrease in PSA or the development of

Dynamic interactions in the tumor niche: how the cross-talk between CAFs and the tumor microenvironment impacts resistance to therapy.

The tumor microenvironment (TME) is a complex ecosystem of cells, signaling molecules, and extracellular matrix components that profoundly influence cancer progression. Among the key players in the TME, cancer-associated fibroblasts (CAFs) have gained increasing attention for their diverse and influential roles. CAFs are activated fibroblasts found abundantly within the TME of various cancer types. CAFs contribute significantly to tumor progression by promoting angiogenesis, remodeling the extracellular matrix, and modulating immune cell infiltration. In order to influence the microenvironment…

Cancer stem cells as the source of tumor associated myoepithelial cells in the tumor microenvironment developing ductal carcinoma in situ.

The heterogeneous cell population in the stromal microenvironment is considered to be attributed to the multiple sources from which the cells originate. Tumor associated myoepithelial cells (TAMEs) are one of the most important populations in the tumor microenvironment (TME) especially in breast cancer. On the other hand, cancer stem cells (CSCs) have previously been described to be the origin of tumor-associated cellular components in the TME. We prepared a cancer stem cell model converting mouse-induced pluripotent stem cells (miPSCs) in the presence of conditioned medium of breast cancer ce…

Targeting Intercellular Communication in the Bone Microenvironment to Prevent Disseminated Tumor Cell Escape from Dormancy and Bone Metastatic Tumor Growth.

Metastasis to the bone is a common feature of many cancers including those of the breast, prostate, lung, thyroid and kidney. Once tumors metastasize to the bone, they are essentially incurable. Bone metastasis is a complex process involving not only intravasation of tumor cells from the primary tumor into circulation, but extravasation from circulation into the bone where they meet an environment that is generally suppressive of their growth. The bone microenvironment can inhibit the growth of disseminated tumor cells (DTC) by inducing dormancy of the DTC directly and later on following forma

Targeting immunosuppressive tumor microenvironment to combat immunotherapy resistance: From mechanistic insights to therapeutic strategies.

With advances in deciphering the intricate interplay between tumors and the immune system, tumor immunotherapy has flourished significantly and has revolutionized conventional tumor therapies, owing to its durable immunological memory and favorable safety profile. Nevertheless, immunotherapy is not currently beneficial for all patients, largely due to the immunosuppressive tumor microenvironment (TME). Considerable efforts have been devoted to bolstering the efficacy of immunotherapy by remodeling the TME. Effective remodeling of the TME and the associated reversal of immunosuppressive conditi…
