The yeast, the antifungal, and the wardrobe: a journey into antifungal resistance mechanisms of Candida tropicalis.

Candida tropicalis is a prominent non-Candida albicans Candida species involved in cases of candidemia, mainly causing infections in patients in intensive care units and (or) those presenting neutropenia. In recent years, several studies have reported an increase in the recovery rates of azole-resistant C. tropicalis isolates. Understanding C. tropicalis resistance is of great importance, since resistant strains are implicated in persistent or recurrent and breakthrough infections. In this review, we address the main mechanisms underlying C. tropicalis resistance to the major antifungal classe

Loss of FZO1 gene results in changes of cell dynamics in fission yeast.

Mitochondrial fission and fusion dynamics are critical cellular processes, and abnormalities in these processes are associated with severe human disorders, such as Beckwith‑Wiedemann syndrome, neurodegenerative diseases, Charcot‑Marie‑Tooth disease type 6, multiple symmetric lipomatosis and microcephaly. Fuzzy onions protein 1 (Fzo1p) regulates mitochondrial outer membrane fusion. In the present study, Schizosaccharomyces pombe (S. pombe) was used to explore the effect of FZO1 gene deletion on cell dynamics in mitosis. The mitochondrial morphology results showed that the mitochondria appeared

Yeast communities of primary and secondary peat swamp forests in southern Thailand.

Khanthuli peat swamp forest (PSF) is one of a few fertile peat swamp forests that remain in Thailand. It is composed of primary PSF and some areas which have been degraded to secondary PSF due to drought, wildfires and land conversion, which have resulted in a decrease in peat layers and change in the species of the plant community. In this study, diversity of yeasts in peat from both primary and secondary PSF areas of the Khanthuli PSF was determined based on culture-dependent approaches, using dilution plate and enrichment techniques. A total of 66 yeast isolates were identified by the analy…

Recent trends in molecular diagnostics of yeast infections: from PCR to NGS.

The incidence of opportunistic yeast infections in humans has been increasing over recent years. These infections are difficult to treat and diagnose, in part due to the large number and broad diversity of species that can underlie the infection. In addition, resistance to one or several antifungal drugs in infecting strains is increasingly being reported, severely limiting therapeutic options and showcasing the need for rapid detection of the infecting agent and its drug susceptibility profile. Current methods for species and resistance identification lack satisfactory sensitivity and specifi

Genome-Wide Response to Drugs and Stress in the Pathogenic Yeast Candida glabrata.

Candida glabrata is the second most common cause of candidemia worldwide and its prevalence has continuously increased over the last decades. C. glabrata infections are especially worrisome in immunocompromised patients, resulting in serious systemic infections, associated to high mortality rates. Intrinsic resistance to azole antifungals, widely used drugs in the clinical setting, and the ability to efficiently colonize the human host and medical devices, withstanding stress imposed by the immune system, are thought to underlie the emergence of C. glabrata. There is a clear clinical need to u

Cryptococcal Titan Cells: When Yeast Cells Are All Grown up.

Cryptococcus neoformans is a human pathogenic yeast that causes hundreds of thousands of deaths worldwide among susceptible individuals, in particular, HIV+ patients. This yeast has developed several adaptation mechanisms that allow replication within the host. During decades, this yeast has been well known for a very peculiar and unique structure that contributes to virulence, a complex polysaccharide capsule that surrounds the cell wall. In contrast to other fungal pathogens, such as Candida albicans or Aspergillus fumigatus, the role of morphological transitions has not been studied in the

Effect of enzymatically hydrolyzed yeast on health and performance of transition dairy cattle.

The objective of this study was to evaluate the effect of supplementing enzymatically hydrolyzed yeast (EHY; Celmanax Arm and Hammer Animal Nutrition, Princeton, NJ) on transition dairy cattle. Forty multiparous Holstein cows were blocked by predicted transmitting ability and randomly assigned to 1 of 2 treatments (EHY, n = 20; or control, CON, n = 20) from 21 d before expected calving to 60 d postpartum. The EHY cows received 56 and 28 g/d in close-up and lactating diets, respectively. Dry matter intake, health events, milk production parameters, feed efficiency, colostrum quality, reproducti

Cationic Amphiphiles Induce Macromolecule Denaturation and Organelle Decomposition in Pathogenic Yeast.

Cationic amphiphiles are a large and diverse class of antimicrobial agents. Although their mode of action is not fully resolved, it is generally accepted that these antimicrobials perturb the structural integrity of the plasma membrane leading to the microbial cell disruption. Here we report on the development of inherently fluorescent antifungal cationic amphiphiles and on the study of their effects on cells of Candida, one of the most common fungal pathogens in humans. Fluorescent images of Candida yeast cells that express a fluorescent reporter protein revealed that the cationic amphiphiles

The RHEB-mTOR axis regulates expression of Tf2 transposons in fission yeast.

The human TSC2 gene, mutations in which predispose individuals to the disease tuberous sclerosis complex (TSC), encodes a GTPase-activating protein for the GTPase RHEB. Loss of TSC2 results in constitutive activation of RHEB and its target mammalian target of rapamycin (mTOR). We have previously reported that fission yeast (Schizosaccharomyces pombe) Tf2 retrotransposons (hereafter Tf2s) are abnormally induced upon nitrogen starvation in cells lacking the tsc2+ gene (Δtsc2), a homolog of the human TSC2 gene, and in cells with a dominant-active mutation in the fission yeast RHEB GTPase (rhb1-DA

Rapid host strain improvement by in vivo rearrangement of a synthetic yeast chromosome.

Synthetic biology tools, such as modular parts and combinatorial DNA assembly, are routinely used to optimise the productivity of heterologous metabolic pathways for biosynthesis or substrate utilisation, yet it is well established that host strain background is just as important for determining productivity. Here we report that in vivo combinatorial genomic rearrangement of Saccharomyces cerevisiae yeast with a synthetic chromosome V can rapidly generate new, improved host strains with genetic backgrounds favourable to diverse heterologous pathways, including those for violacein and penicilli

Water quality and antifungal susceptibility of opportunistic yeast pathogens from rivers.

Yeasts from water sources have been associated with diseases ranging from superficial mucosal infections to life threatening diseases. The aim of this study was to determine the water quality as well as diversity and antifungal susceptibility of yeasts from two rivers. Yeast levels and physico-chemical parameter data were analyzed by principal component analysis to determine correlations between physico-chemical data and yeast levels. Yeast morphotypes were identified by biochemical tests and 26S rRNA gene sequencing. Disk diffusion antifungal susceptibility tests were conducted. Physico-chemi…

Iron toxicity in yeast: transcriptional regulation of the vacuolar iron importer Ccc1.

All eukaryotes require the transition metal, iron, a redox active element that is an essential cofactor in many metabolic pathways, as well as an oxygen carrier. Iron can also react to generate oxygen radicals such as hydroxyl radicals and superoxide anions, which are highly toxic to cells. Therefore, organisms have developed intricate mechanisms to acquire iron as well as to protect themselves from the toxic effects of excess iron. In fungi and plants, iron is stored in the vacuole as a protective mechanism against iron toxicity. Iron storage in the vacuole is mediated predominantly by the va

Isolation and characterization of sake yeast mutants with enhanced isoamyl acetate productivity.

Isoamyl acetate is an important flavor compound in sake. However, production of isoamyl acetate by Saccharomyces cerevisiae is significantly reduced during sake brewing with rice that has a high polishing ratio, because unsaturated fatty acids derived from the outer layer of rice repress the expression of ATF1, which encodes an alcohol acetyl transferase. Yeast mutants capable of relieving this repression would allow the brewing of rice with high polishing ratios, improving the diversity of taste and flavor of sake. Atf1p is also believed to contribute to biological membrane homeostasis. We is

Effects of live yeast supplementation on lipopolysaccharide-induced inflammatory responses in broilers.

The effects of supplemental live yeast (LY) on inflammatory responses in broilers challenged with lipopolysaccharide (LPS) were investigated. One-day-old broilers were randomly divided into two treatment groups with two subgroups of each (8 replicate pens; 10 birds/pen) and were fed a basal diet without or with 0.5 g/kg of LY (Saccharomyces cerevisiae NCYC 47Hr+, 1.0 × 1010 CFU/g). Birds from each subgroup of the two treatment groups were intra-abdominally injected with LPS (1.5 mg/kg of BW) or saline at 21, 23, 25, and 27 d of age. Samples were obtained after 8 h of the first injection (d 21)

A conserved phosphatase destroys toxic glycolytic side products in mammals and yeast.

Metabolic enzymes are very specific. However, most of them show weak side activities toward compounds that are structurally related to their physiological substrates, thereby producing side products that may be toxic. In some cases, 'metabolite repair enzymes' eliminating side products have been identified. We show that mammalian glyceraldehyde 3-phosphate dehydrogenase and pyruvate kinase, two core glycolytic enzymes, produce 4-phosphoerythronate and 2-phospho-L-lactate, respectively. 4-Phosphoerythronate strongly inhibits an enzyme of the pentose phosphate pathway, whereas 2-phospho-L-lactat

The crystal structure of yeast mitochondrial ThrRS in complex with the canonical threonine tRNA.

In mitochondria of Saccharomyces cerevisiae, a single aminoacyl-tRNA synthetase (aaRS), MST1, aminoacylates two isoacceptor tRNAs, tRNA1(Thr) and tRNA2(Thr), that harbor anticodon loops of different size and sequence. As a result of this promiscuity, reassignment of the CUN codon box from leucine to threonine is facilitated. However, the mechanism by which a single aaRS binds distinct anticodon loops with high specificity is not well understood. Herein, we present the crystal structure of MST1 in complex with the canonical tRNA2(Thr) and non-hydrolyzable analog of threonyl adenylate. Our struc

RBE of α-Particles for Delayed Production of Colonies by Irradiated Yeast Cells.

The effect of delayed colony appearance by irradiated cells exemplifying genetic instability is confirmed to be more expressed for diploid wild-type yeast cells characterized by a sigmoidal shape of the survival curve and capable of recovery from radiation damage in contrast to isogenic haploid strain incapable of diploid-specific recovery and characterized by the exponential form of the survival curve. The dependence of the delayed appearance of colonies by diploid yeast cells on the dose of ionizing radiation shows more pronounced manifestation after the action of α-particles (RBE = 4.2 ± 0.

Plug-and-Play Benzylisoquinoline Alkaloid Biosynthetic Gene Discovery in Engineered Yeast.

Benzylisoquinoline alkaloid (BIA) metabolism has been the focus of a considerable research effort over the past half-century, primarily because of the pharmaceutical importance of several compounds produced by opium poppy (Papaver somniferum). Advancements in genomics technologies have substantially accelerated the rate of gene discovery over the past decade, such that most biosynthetic enzymes involved in the formation of the major alkaloids of opium poppy have now been isolated and partially characterized. Not unexpectedly, the availability of all perceived biosynthetic genes has facilitated
