Projections from the central amygdaloid nucleus to the precuneiform nucleus in the mouse.

The mouse precuneiform nucleus has been proposed as the midbrain locomotion center, a function ascribed to its caudal neighbor, cuneiform nucleus, in the rat, cat and other species. The present study investigated the projections from the central amygdaloid nucleus to the precuneiform nucleus in the mouse using retrograde tracer injections (fluoro-gold) into the precuneiform nucleus and anterograde tracer injections (biotinylated dextran amine) into the central amygdaloid nucleus. The entire central amygdaloid nucleus except the rostral pole had retrogradely labeled neurons, especially in the m

Effects of visceral inputs on the processing of labyrinthine signals by the inferior and caudal medial vestibular nuclei: ramifications for the production of motion sickness.

Neurons located in the caudal aspect of the vestibular nucleus complex have been shown to receive visceral inputs and project to brainstem regions that participate in generating emesis, such as nucleus tractus solitarius and the "vomiting region" in the lateral tegmental field (LTF). Consequently, it has been hypothesized that neurons in the caudal vestibular nuclei participate in triggering motion sickness and that visceral inputs to the vestibular nucleus complex can affect motion sickness susceptibility. To obtain supporting evidence for this hypothesis, we determined the effects of intraga

Common theme for drugs effective in overactive bladder treatment: inhibition of afferent signaling from the bladder.

The overactive bladder syndrome and detrusor overactivity are conditions that can have major effects on quality of life and social functioning. Antimuscarinic drugs are still first-line treatment. These drugs often have good initial response rates, but adverse effects and decreasing efficacy cause long-term compliance problems, and alternatives are needed. The recognition of the functional contribution of the urothelium/suburothelium, the autonomous detrusor muscle activity during bladder filling and the diversity of nerve transmitters involved has sparked interest in both peripheral and centr

Cystitis, co-morbid disorders and associated epithelial dysfunction.

Interstitial cystitis (painful bladder syndrome / interstitial cystitis; PBS/IC) is a persistent pain syndrome affecting the urinary bladder with symptoms including urinary frequency, bladder pain and nocturia.(–6) Various animal models have been studied, most of which mimic some aspect of the human condition of interest to the investigator(s). This review will provide examples of various animal models including those incorporating chronic stress, thought to produce features that share similarities to that of PBS/IC patients, whose symptoms are often exacerbated by various stressors. Suppression of bladder overactivity by activation of somatic afferent nerves in the foot.

OBJECTIVE: To investigate the possibility of suppressing bladder overactivity by electrical activation of somatic afferent nerves in the foot. MATERIALS AND METHODS: Cats with an intact spinal cord were studied under α-chloralose anaesthesia. Bladder pressure was recorded via a urethral catheter. Foot stimulation was applied via surface pad electrodes attached to the skin of the front or hind foot. RESULTS: Reflex micturition was inhibited by electrical stimulation of the hind foot at either low (5 Hz) or high (20 Hz) frequencies, but stimulation of the front foot was ineffective. The mean (

Physiological mechanisms of neuropathic pain: the orofacial region.

Neuropathic pain in the orofacial region is the clinical manifestation of trigeminal nerve injury following oral surgeries such as tooth extraction, dental implantation or tooth pulp treatment. Normally non-noxious touching of the facial skin or oral mucosa elicits strong pain named allodynia, and normally noxious stimulation causes intolerable pain named hyperalgesia in the trigeminal neuropathic pain patients. Although the mechanisms underlying trigeminal neuropathic pain have been studied by many researchers, the detailed mechanisms are still unknown. In this chapter, we are focusing on tri

The cortical representation of sensory inputs arising from bone.

In the present study, we show that sensory information from bone reaches the discriminative areas of the somatosensory cortices by electrically stimulating the nerve to the cat humerus and recording evoked potentials on the surface of the primary (SI) and secondary (SII) somatosensory cortex. The SI focus was located over the rostral part of the postcruciate cortex, caudal to the lateral aspect of the cruciate sulcus. The SII focus was identified on the anterior ectosylvian gyrus, lateral to the suprasylvian sulcus. These foci were located adjacent to, or within areas that responded to stimula

Differential projections from the vestibular nuclei to the flocculus and uvula-nodulus in pigeons (Columba livia).

The pigeon vestibulocerebellum is divided into two regions based on the responses of Purkinje cells to optic flow stimuli: the uvula-nodulus responds best to self-translation, and the flocculus responds best to self-rotation. We used retrograde tracing to determine whether the flocculus and uvula-nodulus receive differential mossy fiber input from the vestibular and cerebellar nuclei. From retrograde injections into the both the flocculus and uvula-nodulus, numerous cells were found in the superior vestibular nucleus (VeS), the cerebellovestibular process (pcv), the descending vestibular nucle

Response monitoring without sensory feedback.

OBJECTIVE: The elicitation of an evoked potential, the 'error negativity' (Ne) when subjects commit errors in speeded tasks, is often taken as an index of response monitoring processes. The presence of a Ne-like wave on purely correct trials challenges the current conceptions about the nature of such a monitoring system. Here, we evaluate the possibility that the Ne-like wave on correct trials is merely due to reafferences, and at the same time, we test directly the general opinion according to which the Ne is generated by an internal signal. METHODS: We studied the presence of a Ne-like wave

Four response stages of capsaicin-sensitive primary afferent neurons to capsaicin and its analog: gastric acid secretion, gastric mucosal damage and protection.

BACKGROUND: Dinutuximab is a monoclonal antibody that targets the GD2 antigen used in the treatment of high-risk neuroblastoma. Dinutuximab-associated rhombencephalitis and myelitis is a rare, steroid-responsive, serious, but reversible pathology. To date, three transverse myelitis cases and one rhombencephalitis case due to dinutuximab have already been reported. Moreover, a recently published article identified five inflammatory CNS demyelination cases (four myelitis and one rhombencephalitis). We present a 5-year-old patient with rhombencephalitis and myelitis following dinutuximab-beta tre

The ascending serotonergic system in the hamster: comparison with projections of the dorsal and median raphe nuclei.

The ascending serotonergic projections are derived largely from the midbrain median and dorsal raphe nuclei, and contribute to the regulation of many behavioral and physiological systems. Serotonergic innervation of the hamster circadian system has been shown to be substantially different from earlier results obtained with other methods and species. The present study was conducted to determine whether similar differences are observed in other brain regions. Ascending projections from the hamster dorsal or median raphe were identified using an anterograde tracer, Phaseolus vulgans leucoagglutin

Afferent projections to the cingulate cortex in albino rats: a study with a retrograde labeling method using fluoro-gold.

We studied the neuronal populations that project their axons to the cingulate cortex in albino rats using the retrograde fluorescent dye of 4% Fluoro-Gold injected into the anterior, middle and posterior portions of the cingulate cortex. The result showed that the following ipsilateral structures are sending fibers to these three portions: the prefrontal cortex, frontoparietal motor cortex, indusium griseum, dorsal endopiriform nucleus, lateral part of medial mammillary nucleus, nuclei of diagonal band of Broca, anterior pretectum, anterior part of caudate-putamen, hippocampal formation, anter

Development and regulation of dendritic stratification in retinal ganglion cells by glutamate-mediated afferent activity.

In the mature retina, the dendrites of retinal ganglion cells (RGCs) are segregated into either ON or OFF sublaminae of the inner plexiform layer (IPL), but early in development the dendritic processes of these cells are multistratified, ramifying throughout the IPL. We examined the time course of dendritic stratification in developing beta cells, the largest class of ganglion cells in the cat retina, by retrograde labeling of fixed tissue with Dil. Dendritic stratification begins in the central and peripheral retina by embryonic day 50, about 2 weeks before birth and is not fully completed un

Neuronal activity in normal and deafferented forelimb somatosensory cortex of the awake cat.

Three hundred and seventy-three neurons were recorded from the forelimb representation in the primary somatosensory cortex of unanesthetized, quietly resting adult cats. Of these, 177 were studied from 2 days to 3 weeks after transection of the radial, median and ulnar nerves. Following deafferentation the proportion of cells without receptive fields increased from 24 to 82%, however, the average rate of spontaneous activity did not change nor did the probability of encountering a neuron with a receptive field as a function of depth. Receptive field sizes increased dramatically following deaff

Effect of abdominal compression on diaphragmatic tendon organ activity.

Previous studies suggest that afferents in the diaphragm participate in the reflex reduction in phrenic nerve efferent activation when the length of the diaphragm is increased by abdominal compression. The present study determined the response of tendon organ afferents in the diaphragm to increases in abdominal pressure. Five cats were anesthetized with thiopental sodium (60 mg/kg ip to induce, supplemented intravenously). Extracellular recordings from nine individual tendon organ afferents were made from right cervical dorsal root ganglia 5 and 6. Right crural electromyographic activity was r

Special considerations in orofacial pain.

A pathologic dental condition is by far the most common cause of orofacial pain. There are, however, certain patients who possess and exhibit myriad signs and symptoms that mimic pathologic dental conditions but do not have a dental cause. These patients pose a diagnostic dilemma for the dental practitioner. Unfortunately, there are many documented cases of misdiagnosis and mismanagement in patients with deafferentiation pain, neuropathic pain, trigeminal neuralgia, and temporal arteritis. These orofacial pain disorders warrant special consideration for the dental practitioner and are reviewed

Effects of denervation on the glomus cell membrane.

Trophic influences of the carotid nerve (CN) on carotid body (CB) glomus cells were studied by comparing the membrane potential (Em), input resistance (Ro) and voltage noise (Erms) of normal and 3-31-day denervated cells had more negative Ems. Higher Ros were recorded at 3 and 6 days. Erms sharply increased at 3 days, returned to normal at 6-15 days and was below normal at 31 days. A transmitter (ACh) and NaCN, producing histotoxic anoxia, were used for stimulation. These substances either depolarized or hyperpolarized innervated cells and increased or decreased voltage noise. Denervation sele

Disrupted eyelid conditioning in a patient with damage to cerebellar afferents.

A 54-year-old woman with damage to cerebellar circuitry resulting from a cerebrovascular accident underwent classical conditioning of the eye-blink response to a tone conditioned stimulus and an air-puff unconditioned stimulus. In contrast to 5 age-matched controls who readily acquired the conditioned response (CR), emitting a mean of 56.7 CRs over 70 trials, the patient emitted only 6 CRs in 100 trials and never emitted 2 consecutive CRs. There were no differences in spontaneous blink rate, sensitivity to the air puff, or sensitivity to the tone between the experimental subject and the contro

Behavioral evidence and supporting electrophysiological observations for electroreception in the blind cave salamander, Proteus anguinus (Urodela).

Conditioning experiments revealed that Proteus perceives a back-and-forth moving (approximately 1 Hz) direct-current field and its polarity. Minimum behavioral thresholds occurred at a current density of 0.15 microA/cm2, corresponding to a voltage gradient of 0.5 mV/cm. Recordings from afferent nerve fibers showed that ampullary electroreceptors in Proteus respond as do other nonteleost receptors, i.e. with an increase in discharge rate to cathodal current and a decrease to anodal current (threshold: approximately 1 mV/cm).
