Bacterial meningitis in children under 15 years of age in Nepal.

BACKGROUND: Bacterial meningitis in children is a life-threatening problem resulting in severe morbidity and mortality. For the prompt initiation of antibacterial therapy, rapid and reliable diagnostic methods are of utmost importance. Therefore, this study was designed to find out the rate of bacterial pathogens of meningitis from suspected cases by performing conventional methods and latex agglutination. METHODS: A descriptive type of study was carried out from May 2012 to April 2013. Cerebrospinal fluid (CSF) specimens from 252 suspected cases of meningitis were subjected for Gram staining

Comparison of Buffered, Acidified Plate Antigen to Standard Serologic Tests for the Detection of Serum Antibodies to Brucella abortus in Elk (Cervus canadensis).

Brucellosis (caused by the bacterium Brucella abortus) is a zoonotic disease endemic in wild elk (Cervus canadensis) of the Greater Yellowstone Ecosystem, US. Because livestock and humans working with elk or livestock are at risk, validated tests to detect the B. abortus antibody in elk are needed. Using the κ-statistic, we evaluated the buffered, acidified plate antigen (BAPA) assay for agreement with the results of the four serologic tests (card test [card], complement fixation test [CF], rivanol precipitation plate agglutination test [RIV], standard plate agglutination test [SPT]) that are

An evaluation of three serological tests for antibody to Brucella suis in pigs.

The Rose Bengal Plate Agglutination test (RBT), the complement fixation text (CFT) and the tube agglutination test (TAT) were applied to serums from 345 feral and 80 domestic pigs sampled at slaughter. At least 2 of the 3 serological tests were applied to each serum. Tissues from all pigs were cultured for Brucella suis and the degree of culture effort was categorised from 1 to 4 in decreasing order. Fifty-eight feral and 35 domestic pigs were culture-positive. A greater proportion of culture-positive pigs was obtained for category 1 and 2 culture effort. Tissues yielding B. suis most often we

Fragment D-dimer levels: an objective marker of vaso-occlusive crisis and other complications of sickle cell disease.

Although abnormalities in coagulation tests have been reported during vaso-occlusive crises in patients with sickle cell disease, objective, readily performed laboratory tests that document the occurrence of this complication have not been available. We examined the relationship between fibrin D-dimer levels and the occurrence of complications in patients with sickle cell disease, using a commercially available latex bead agglutination assay. The patients were either asymptomatic, hospitalized for vaso-occlusive crisis, or had other complications of sickle cell disease including leg ulcers, ch

[Diagnostic research on leptospirosis in the Milanese territory].

168 serum specimens have been tested for Leptospirosis in the years from 1980 to 1983. The identification procedure was a macroscopic plate agglutination screening test with formalin fied antigens and a confirmatory microscopic agglutination-lysis test of live antigens of actively growing cultures of different pathogenic serotypes propagated in a fluid medium. 34 serum specimens (23.8%) relevant to 18 cases were positive. L. icterohaemorrhagiae was positive in 12 cases (63.1%), L. pomona in 2 cases (10.5%), L. canicola in 1 case (5.3%), 1 case was positive for both L. grippotyphosa and L. bata

Immunoreactivity in pulmonary echinococcosis. A comparative study of immunodiagnostic tests.

A comparison was made of the diagnostic value of five immunological tests-complement fixation (CF), latex agglutination (LA), bentonite flocculation (BF), passive haemagglutination (PHA), and intradermal (ID)-in patients operated on for pulmonary echinococcosis. The sensitivity of all five tests was significantly lower in the patients with pulmonary echinococcosis than in a comparable group with liver echinococcosis. Some 252 patients with miscellaneous nonhydatid diseases and parasitic infections were also tested; nonspecific reactions were highest in the ID and CF tests and lowest in the LA

First record of the presence of antibodies against Leptospira in Lagostomus maximus, and Leptospira weillii serogroup Celledoni in Chaetophractus villosus, Argentina.

Leptospirosis is a zoonosis with a worldwide distribution, which is transmitted through direct contact with infected animals or by contact with contaminated water or soil. Objectives was to examine the presence of antibodies against 23 serogroups of Leptospira in Lagostomus maximus and Chaetopractus villosus in La Pampa province, Argentina. Ninety-four serum samples from L. maximus (80) and C. villosus (14) were taken. The samples were analysed by microscopic agglutination test (MAT). Only 5/94 (5.3 %) serum samples presented antibodies against Leptospira sp. Three (21.4 %) serum samples from

Use of thiol reagents to disperse cold autoagglutination.

Thiol reagents dithiothreitol (DTT) and 2-mercaptoethanol (2-ME) are sulfhydryl reagents that can be used to disperse cold autoagglutinins coating red blood cells (RBCs). DTT and 2-ME are primarily used when warm washing of the coated RBCs fails to successfully disperse the cold autoantibody. Using a weak concentration of DTT or 2-ME, the cold IgM agglutinin can be removed from the coated RBCs without disrupting the IgG or complement coating the RBCs. The treated RBCs can be used for ABO typing, antigen typing, or the direct antiglobulin test.

Diagnostic method-based underestimation of leptospirosis in clinical and research settings; an experience from a large prospective study in a high endemic setting.

BACKGROUND: Leptospirosis has globally significant human mortality and morbidity, yet estimating the clinical and public health burden of leptospirosis is challenging because timely diagnosis remains limited. The goal of the present study was to evaluate leptospirosis undercounting by current standard methods in both clinical and epidemiological study settings. METHODOLOGY/PRINCIPAL FINDINGS: A prospective hospital-based study was conducted in multiple hospitals in Sri Lanka from 2016 to 2019. Culture, whole blood, and urine samples were collected from clinically suspected leptospirosis cases

Seroepidemiological Analysis of Leptospiral infection using MAT in Stray Dogs in Alborz, Iran.

Leptospirosis is a zoonotic disease with global importance, and the animals are the source of transmission of this disease through shedding in their urine. Accordingly, it is essential to conduct epidemiological studies of leptospirosis in order to diagnose this disease in dogs and reduce the risk of transmission to humans. This study aimed to perform a seroepidemiological analysis of Leptospiral infection in stray dogs in Alborz, Iran, using the Microscopic Agglutination Test (MAT). In total, 110 blood samples were collected from stray dogs to detect the antibodies against leptospira interrog

Lethal Outcome of Leptospirosis in Southern Russia: Characterization of Leptospira Interrogans Isolated from a Deсeased Teenager.

This article describes a lethal case of leptospirosis that occurred in Southern Russia. The Leptospira strain was isolated and characterized using a microscopic agglutination test, MALDI-TOF mass spectrometry, targeted PCR, and high-throughput sequencing. We show that molecular and mass-spectrometry methods can be an alternative to conventional methods of leptospirosis diagnostics and Leptospira study, which require highly qualified staff and can be performed only at specialized laboratories. We also report the first whole genome of L. interrogans isolated in Russia.

Leptospira infections in freshwater fish in Morogoro Tanzania: a hidden public health threat.

Leptospirosis caused by spirochete bacterium of genus Leptospira affects humans and animals worldwide. Rodents are major reservoirs of leptospires whereas wetland and aquatic migratory birds also carry and transmit leptospires. Leptospirosis studies in fish are lacking in African countries despite favourable environment and abundant reservoirs, which can spread leptospires into aquatic habitats and infect fish. The objectives of this study were to determine presence of Leptospira in fish; the prevalent Leptospira serovars and whether are related to serovars reported in animals; and to ascertai
