Lower Serum Magnesium Concentrations are associated With Specific Heavy Drinking Markers, Pro-Inflammatory Response and Early-Stage Alcohol-associated Liver Injury§.

AIM: Chronic heavy alcohol intake frequently causes liver inflammation/injury, and altered mineral metabolism may be involved in this liver pathology. In this study, we evaluated the association of heavy drinking, changes in serum magnesium levels and biochemical evidence of liver injury in alcohol-use-disorder (AUD) patients who had no clinical signs or symptoms of liver injury. We also aimed to identify any sex-based differences in patients with mild or no biochemical evidence of liver injury induced by heavy drinking. METHODS: 114 heavy drinking alcohol-dependent (AD) female and male patie

Identifying "Hitting Bottom" Among Individuals with Alcohol Problems: Development and Evaluation of the Noteworthy Aspects of Drinking Important to Recovery (NADIR).

BACKGROUND: Alcohol use disorder (AUD) is a costly public health concern; yet, many individuals with AUD never receive formal treatment. Prior studies have identified that "hitting bottom" may be an important factor in seeking treatment for AUD) and the notion that "hitting bottom" is necessary for recovery is commonly portrayed in the popular media. Yet, "hitting bottom" has never been formally operationalized. OBJECTIVES: The present article aimed to operationalize "hitting bottom." METHODS: A multiphase process was used to develop a measure of hitting bottom among individuals experiencing

Salidroside Regulates Inflammatory Response in Raw 264.7 Macrophages via TLR4/TAK1 and Ameliorates Inflammation in Alcohol Binge Drinking-Induced Liver Injury.

The current study was designed to investigate the anti-inflammatory effect of salidroside (SDS) and the underlying mechanism by using lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages in vitro and a mouse model of binge drinking-induced liver injury in vivo. SDS downregulated protein expression of toll-like receptor 4 (TLR4) and CD14. SDS inhibited LPS-triggered phosphorylation of LPS-activated kinase 1 (TAK1), p38, c-Jun terminal kinase (JNK), and extracellular signal-regulated kinase (ERK). Degradation of IκB-α and nuclear translocation of nuclear factor (NF)-κB were effectively bloc

The startle reflex in alcohol-dependent patients: changes after cognitive-behavioral therapy and predictive validity for drinking behavior. A pilot study.

BACKGROUND: Previous studies demonstrated an attenuation of the affect-modulated startle reflex when alcohol-dependent patients were viewing alcohol-associated pictures. This indicates an appetitive valence of these stimuli. We used the affect-modulated startle reflex to assess the effects of behavioral treatment on the emotional processing of alcohol-associated stimuli. Further, we examined whether the affect-modulated startle reflex is a predictor of treatment success. METHODS: Forty-three alcohol-dependent patients (21 females, mean age 45.67 years, SD 9.45) were recruited consecutively fr

Does treatment status matter? An examination of daily approach and avoidance inclinations predicting drinking in individuals diagnosed with alcohol use disorder.

AIMS: Traditional craving models do not assess competing desires and account for craving as a function of attempts to change problematic drinking behaviors, thereby limiting the utility of the information obtained. The Ambivalence Model of Craving defines craving as the coactivation of both desires to consume (approach) and to avoid consuming alcohol (avoidance). The current study examined the daily associations between approach/avoidance, and drinking in those diagnosed with alcohol use disorder (AUD), including how treatment status moderated such relationships. METHOD: Individuals diagnosed

Comparative analysis of test tube and volumetric drinking monitor methods in voluntary ethanol consumption in female mice for prenatal alcohol exposure.

OBJECTIVE: We directly compared a low-tech test tube (TT) method and a high-tech volumetric drinking monitor (VDM) to assess their impact on female mice alcohol drinking. METHOD: We adapted a voluntary drinking paradigm described by Brady et al. (2012) for both TT and VDM. Eight mice were assigned to each method, and we measured their drinking volume and blood alcohol concentrations (BACs), comparing values using t-tests. RESULTS: After 2 weeks of drinking alcohol BACs were not significantly different between the VDM (82 ± 21 mg/dL) vs.TT (73 ± 23 mg/dL) groups [t(14) = 0.27, p = 0.7935]. Ca…

Associations between ZIP code-level alcohol outlet density and binge drinking among people who inject drugs in 22 US metropolitan areas.

BACKGROUND: Although alcohol outlet density (AOD) is associated with drinking behaviors in the general population, little evidence exists about this relationship among people who inject drugs (PWID). Establishing this connection is particularly important, as alcohol use exacerbates the risk of opioid overdose. This study investigated 1) the association between ZIP code-level AOD and binge drinking among PWID who use opioids, and 2) the potential moderating effect of race/ethnicity on the AOD-binge drinking relationship. METHODS: This analysis linked 2018 National HIV Behavioral Surveillance (

Exploration of the multidisciplinary integration teaching model in forensic science: A case study of death by high fall after drinking alcohol.

Undergraduate education in forensic science plays a critical role in cultivating highly qualified forensic professionals with comprehensive analytical skills and practical competency. However, traditional teaching methods often prove insufficient when complex cases involving the intersection of multiple disciplines are encountered, and they frequently fail to meet the demands of practical teaching. This teaching practice is mainly for 4th-year undergraduate forensic science students and aims to explore a multidisciplinary integrated teaching model in forensic science on the basis of a complex

Genetically modeled GLP1R and GIPR agonism reduce binge drinking and alcohol-associated phenotypes: a multi-ancestry drug-target Mendelian randomization study.

Pharmacological modulation of glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP) through dual GIP/GLP-1 receptor agonists, commonly used for diabetes and obesity, shows promise in reducing alcohol consumption. We applied drug-target Mendelian randomization (MR) using genetic variation at these loci to assess their long-term effects on problematic alcohol use (PAU), binge drinking, alcohol misuse classifications, liver health, and other substance use behaviors. Genetic proxies for lowered BMI, modeling the appetite-suppressing and weight-reducing effects of v

Comparative Analysis of Test Tube and Volumetric Drinking Monitor Methods in Voluntary Ethanol Consumption in Female Mice for Prenatal Alcohol Exposure.

OBJECTIVE: Fetal alcohol spectrum disorders affect approximately 1 in 20 school age children in the United States of America. To study fetal alcohol spectrum disorders, mouse models are commonly used. Of the many approaches of gestational exposure, voluntary drinking paradigms represent the most similar mechanism of drinking as human exposure. These exposures can be done through low-tech solutions such as test tubes (TT), or more high-tech methods such as a volumetric drinking monitor (VDM). Here were compare the TT method and the VDM directly, to evaluate their effect on female mouse drinking

Adherence to alcohol consumption-related recommendations and predictors of heavy episodic drinking among patients with NCDs during the COVID-19 pandemic.

INTRODUCTION: Managing non-communicable diseases (NCDs) requires adherence to lifestyle recommendations like a healthy diet, regular exercise, smoking cessation, and limiting alcohol intake. The COVID-19 pandemic introduced barriers to maintaining these habits, including limited healthcare access, increased stress, and reduced physical activity. This study assessed adherence to lifestyle recommendations, with a focus on heavy episodic drinking (HED), among NCD patients during the pandemic in Arba Minch, Ethiopia, to identify areas for public health intervention. METHOD: A cross-sectional stud

GHSR blockade, but not reduction of peripherally circulating ghrelin via β1-adrenergic receptor antagonism, decreases binge-like alcohol drinking in mice.

Alcohol use disorder (AUD) and binge drinking are highly prevalent public health issues. The stomach-derived peptide ghrelin, and its receptor, the growth hormone secretagogue receptor (GHSR), both of which are expressed in the brain and periphery, are implicated in alcohol-related outcomes. We previously found that systemic and central administration of GHSR antagonists reduced binge-like alcohol drinking, whereas a ghrelin vaccine did not. Thus, we hypothesized that central GHSR drives binge-like alcohol drinking independently of peripheral ghrelin. To investigate this hypothesis, we antagon…
