Behavioral effects of prenatal d-amphetamine in rats: a parallel trial to the Collaborative Behavioral Teratology Study.

Pregnant Sprague-Dawley rats treated with 0, 0.5 or 2.0 mg/kg of d-amphetamine sulphate on days 12-15 of gestation or left untreated as part of the NCTR Collaborative Behavioral Teratology Study were assigned to either the Collaborative Study or Cincinnati Test protocol after birth. Offspring assigned to the Cincinnati test system were evaluated for growth, mortality, incisor eruption, eye opening, vaginal patency, surface righting, negative geotaxis, pivoting, auditory startle, olfactory orientation, swimming ontogeny, figure-8 activity, and complex water maze (Biel) problem solving. Amphetam…

Anorexic and behavioural effects of a new imidazo-isoindole derivative (mazindol) in comparison with d-amphetamine in the rat.

The effect of an imidazo-isoindole derivative (mazindol) upon motor and feeding activity and two different avoidance behaviours have been compared with those of d-amphetamine in rats. It was found that both drugs depress feeding activity in a dose-related manner and increase the motor activity of the animals. However, the ratio of the lowest motility-increasing dose and the lowest appetite suppressant dose is 0.5 for d-amphetamine and 2 for mazindol. Mazindol, like d-amphetamine, caused a stereotyped behaviour in rats when given in very high doses, but the range between the anorexic and the st…

Effects of chronic d-amphetamine on social behavior of the rat: implications for an animal model of paranoid schizophrenia.

Hooded rats in a social colony were given increasing daily doses of d-amphetamine up to 8 mg/kg. Time-lapse 16 mm cinematographically recorded behavior was analyzed for the following: grooming, feeding, sex, sleeping, resting, sterotypy, agonistic behavior, muricidal activity, and the location and movement of each rat. Subordiant rats receiving d-amphetamine actively withdrew from social interactions by retreating to strategically defensible locations in the environment. They remained hypervigilant of other rats and overreacted to their approaches by either fleeing or by defensively rearing an…

Optimal drug, dose and duration of antipsychotic therapy following amphetamine and cannabis-induced psychosis: A systematic review.

BACKGROUND: Amphetamine- and cannabis-induced psychoses are common clinical presentations that are difficult to differentiate from primary psychotic disorders. However, there are limited guidelines around the appropriate use of antipsychotic medications to treat these disorders. This review examined the literature considering the optimal drug, dose and duration of antipsychotic therapy to treat and prevent relapse of amphetamine- and cannabis-induced psychoses. METHODS: A systematic review was conducted to identify original empirical studies considering antipsychotic drugs for the management …

Chronic manganese exposure disrupts dopamine biosynthesis and amphetamine response in a mouse model of altered dopamine transport.

The capacity for excess manganese to alter dopaminergic systems resulting in neurobehavioral deficits is clear, yet far less is known the im … differed according to age and sex. Female mice also exhibited a decrease in activity in response to amphetamine compared to control-fed mice (70 ppm) littermates independent of the decreased response observed in DATT356M/+ mice. Males showed a more robust effect of genotype suppression of the amphetamine response but no further effect of manganese. Behavioral …

Occipital lobe to femoral blood ratios of amphetamine, methamphetamine, cocaine, benzoylecgonine, cocaethylene, and fentanyl in postmortem cases.

Interpretation of postmortem toxicology results is complicated by postmortem redistribution (PMR), which can alter drug concentrations between tissues and blood (BL). This study evaluates brain (BR) to BL concentration ratios for amphetamine (AMP), methamphetamine (MAMP), cocaine (COC), benzoylecgonine (BE), cocaethylene (CE), and fentanyl (FENT) using postmortem casework data to evaluate the utility of BR tissue as an alternative matrix. BR:BL ratios varied widely across analytes: AMP (2.2-5.8), BE (0.2-2.1), CE (1.2-5.1), COC (0.1-5.8), MAMP (2.2-7.3), and FENT (1.1-24.8). Lipophilic compoun…

Detection of 3,4-Methylene Dioxy Amphetamine in Urine by Magnetically Improved Surface-Enhanced Raman Scattering Sensing Strategy.

Abuse of illicit drugs has become a major issue of global concern. As a synthetic amphetamine analog, 3,4-Methylene Dioxy Amphetamine (MDA) causes serotonergic neurotoxicity, posing a serious risk to human health. In this work, a two-dimensional substrate of ITO/Au is fabricated by transferring Au nanoparticle film onto indium-tin oxide glass (ITO). By magnetic inducing assembly of Fe3O4@Au onto ITO/Au, a sandwich-based, surface-enhanced Raman scattering (SERS) detection strategy is designed. Through the use of an external magnet, the MDA is retained in the region of hot spots formed between F…

Cannabidiol prevents amphetamine relapse and modulates D1- and D2-receptor levels in mesocorticolimbic brain areas of rats.

Amphetamine (AMPH) is an addictive psychostimulant highly used worldwide and its consumption is related to neurotoxic effects. Currently, there is no pharmacotherapy approved for treating AMPH or other psychostimulant drug addiction. Different studies have shown promising properties of cannabidiol (CBD) for treating many neurological and psychiatric diseases, and recently, CBD is being considered a potential strategy for the treatment of drug addiction disorders. Thus, we investigated possible CBD beneficial effects on relapse symptoms following AMPH re-exposure considering drug relapse is the…

Association study of GABA system genes polymorphisms with amphetamine-induced psychotic disorder in a Han Chinese population.

GABA system genes have been implicated in neurotrophy and neurogenesis, which play pivotal roles in an individual's variation in vulnerability to amphetamine addiction or amphetamine-induced psychosis (AIP). We hypothesized that common genetic variants in the GABA system genes may be associated with amphetamine-induced psychotic disorder. In our study, thirty-six single nucleotide polymorphisms (SNPs) within the GABA system genes were genotyped in 400 amphetamine-induced psychotic disorder patients and 400 amphetamine use disorders patients (AUP) (not including those categorized as psychosis) …

Signaling Mechanisms in the Nitric Oxide Donor- and Amphetamine-Induced Dopamine Release in Mesencephalic Primary Cultured Neurons.

Previous research has shown that nitric oxide (NO) synthase inhibitors prevent rodents' sensorimotor gating impairments induced by dopamine releasing drugs, such as amphetamine (Amph) and methylphenidate. The mechanisms of this effect have not been entirely understood. In the present work, we investigated some possible mechanisms by which the NO donor, NOC-12 (3-ethyl-3-(ethylaminoethyl)-1-hydroxy-2-oxo-1-triazene), influence spontaneous and Amph-induced dopamine release, using rat mesencephalic primary cultured neurons preparations. Our results showed that NOC-12 increased dopamine release in…
