Intercellular communication via exosomes: A new paradigm in the pathophysiology of neurodegenerative disorders.

Neurodegenerative disorders are one of the leading causes of death and disability and pose a great economic burden on healthcare systems. Generally, these neurodegenerative disorders have a progressive deterioration in neural function and structure, and deposition of misfolded proteins commonly occurs, such as amyloid-β in AD and α-synuclein in PD. However, there exists a special class of exosomes, which acts like a transmitter and enhances communication between cells. The present review discusses the significant role of exosomes in neurodegenerative diseases, with a focus on Amyotrophic later

Exploring the Spatial Limits of Extracellular Vesicles-Mediated Intercellular Communication.

Extracellular vesicles (EVs) are biological nanovectors that retain molecular signatures of their cells of origin and mediate intercellular communication, resulting in ideal platforms for the development of diagnostic tools and bio-inspired drug delivery technologies. Despite their potential, the mechanisms underlying EV release, uptake and distribution remain unclear. Here, we leverage high-resolution live-cell imaging, quantitative analytical methods and in vivo mouse models to define the major determinants of EV diffusion. Our findings reveal that cell density plays a crucial role in EV dis

Impact of nanoparticle properties on immune cell interactions in the lymph node.

The lymphatic system plays an important role in health and many diseases, such as cancer, autoimmune, cardiovascular, metabolic, hepatic, viral, and other infectious diseases. The lymphatic system is, therefore, an important treatment target site for a range of diseases. Lymph nodes (LNs), rich in T cells, B cells, dendritic cells, and macrophages, are also primary sites of action for vaccines and immunotherapies. Promoting the delivery of therapeutics and vaccines to LNs can, therefore, enhance treatment efficacy and facilitate avoidance of off-target side effects by enabling a reduction in t

Ruminating on rudeness: Exploring veterinarians' experiences of client incivility.

BACKGROUND: Interactions with clients have been identified as a key potential stressor within veterinary practice. However, there is a lack of research investigating the experience, and impact, of specific behaviours, such as incivility. The current study aimed to address this literature gap by investigating veterinarian perceptions of client rudeness. METHOD: Telephone interviews (n = 18) were used to gather qualitative data regarding veterinarian perceptions of client rudeness. This encompassed description of an uncivil interaction, potential causes of incivility, the impact on the veterina

A Purified Platelet-Derived Exosome Product for Chronic Wound Healing: A Novel Therapeutic Strategy and Next-Generation Delivery Platform.

Chronic wounds remain a major unmet clinical challenge, often failing to progress to normal healing due to persistent inflammation, impaired angiogenesis, and cellular senescence. Exosomes have recently been investigated as promising acellular therapeutics capable of restoring intercellular communication and promoting tissue regeneration. Among these, the Purified Exosome Product (PEP) represents a next-generation, platelet-derived exosome formulation manufactured under Good Manufacturing Practice (GMP) conditions with high purity, stability, and reproducibility. This review summarizes the cur

Mommy issues: how maternal diet shapes offspring mitochondrial physiology.

Exposure to early life stressors, including maternal obesity and the consumption of a calorically dense diet, during critical windows of fetal development is associated with an elevated and earlier risk of developing childhood obesity, type 2 diabetes, and other cardiometabolic diseases. As mitochondria lie at the intersection of energy metabolism and cellular stress response, alteration in their function is a likely mediator of the observed effects of maternal diet quality on offspring cardiometabolic health. This review summarizes the effect of a maternal high-calorie diet on offspring mitoc

The effects of oxytocin on social behavior and eye gaze: Insights from dog-human partnership.

Oxytocin (Oxt) plays a crucial role in forming stable social bonds and promoting social interactions in both humans and animals. During the long history of domestication, dogs have evolved advanced social communication, empathy, and a remarkable ability to perceive and respond to human emotions. Given the effective and intimate social interactions between dogs and humans, the laboratory Beagle dogs have been utilized to model mental disorders such as autism spectrum disorder (ASD). Indeed, ASD-associated Shank3 mutant dogs show impaired social interaction with both conspecifics and heterospeci

Structural basis of sex pheromone detection in aphids.

Sex pheromones play a central role in regulating animal behavior and reproduction. In insects, these signals are perceived through specialized odorant receptors (ORs) that mediate species-specific communication and safeguard genetic integrity. However, the structural basis of sex pheromone detection remains largely unresolved. Here, we identified two ORs in the pea aphid Acyrthosiphon pisum, along with the conserved OR co-receptor (Orco), which together mediate recognition of the pheromone components nepetalactone and nepetalactol. Functional assays demonstrated that ApOR21-Orco and ApOR22-Orc

The role of exosomes in ankylosing spondylitis: from biological features and functional cargo to clinical applications.

Exosomes are small extracellular vesicles secreted by nearly all cell types and widely distributed in body fluids. They not only mediate intercellular material transfer but also play an important role in the regulation of immune pathways. Given their diverse biological functions, studies investigating the regulatory roles of exosomes in ankylosing spondylitis (AS) are receiving increasing attention. The functions of exosomes in AS largely depend on their bioactive cargo, including microRNAs (miRNAs), circular RNAs (circRNAs), long non-coding RNAs (lncRNAs), proteins, and other molecules. In ad

Heartbeats and hidden codes: Decoding cardiac aging with multi-omics and computational approaches.

Aging is a major risk factor for cardiovascular disease, yet the mechanisms driving age-related cardiac decline remain incompletely defined. Although traditional omics approaches have identified global signatures of inflammation and metabolic shifts, bulk analyses often mask cell-type-specific heterogeneity. Recent single-cell multi-omics innovations address this gap by enabling simultaneous profiling of gene expression and chromatin accessibility. These high-resolution analyses reveal that fibroblasts and macrophages exhibit particularly pronounced age-associated remodeling, marked by heighte

Developmental Disturbances in Animal Models of Autism Spectrum Disorder.

Although the early signs of autism spectrum disorder (ASD) are widely studied, the significant ambiguity and heterogeneity in symptoms require the comparison of available models, approaches, and the search for common denominators and key indicators. Early ASD symptoms in animal models include impaired somatic development (e.g., delayed eye opening), alterations in primitive motor reflexes, disrupted sensory function as well as communication deficits, such as reduced ultrasonic vocalization. This review aims to summarize early ASD-related symptoms based on studies involving transgenic or neurot

Research Priorities for Zoonotic and Pandemic Influenza Vaccines: Evidence and Recommendations from the WHO Public Health Research Agenda for Influenza (2024 Update).

Zoonotic influenza viruses, including highly pathogenic avian influenza and swine-origin variants, continue to cause sporadic human infections with, in some cases, high case fatality rates and potential for sustained human-to-human transmission. The COVID-19 pandemic underscored both the possibilities of rapid vaccine innovation and the persistent challenges in equitable access and public trust. This paper synthesizes the vaccine-related priorities from the 2024 update of the World Health Organization Public Health Research Agenda for Influenza, integrating evidence from systematic literature

ECM-binding properties of extracellular vesicles: advanced delivery strategies for therapeutic applications in bone and joint diseases.

Extracellular vesicles (EVs) and the extracellular matrix (ECM) are essential in maintaining bone and joint health by facilitating intercellular communication, regulating tissue processes and providing structural support. EVs with a large surface area carry diverse biomolecules to steer the function of cells in their surroundings. To understand how EVs localize to specific sites, we here review the available knowledge on EV surface biomolecules and their interactions with ECM components that are crucial for regulating bone remodeling, cartilage maintenance, and immune responses, playing roles

DNA methylation of the endothelin receptor B makes blue fish yellow.

Animal pigmentation is essential for concealment and communication in nature. Though selection acts on the phenotypes of an organism, these traits are often resolved to genes that shape pigmentation in populations. In contrast, plasticity in coloration can occur within individuals in response to ambient environmental changes that act through integrative processes. It is well known that some animals often change color to match their visual ecology. The cichlid fish, Astatotilapia burtoni, can switch between robust blue and yellow body morphs in the lab and the wild. Different colors result from

Ethical issues in Nipah virus control and research: addressing a neglected disease.

Nipah virus is a priority pathogen that is receiving increasing attention among scientists and in work on epidemic preparedness. Despite this trend, there has been almost no bioethical work examining ethical considerations surrounding the epidemiology, prevention, and treatment of Nipah virus or research that has already begun into animal and human vaccines. In this paper, we advance the case for further work on Nipah virus disease in public health ethics due to the distinct issues it raises concerning communication about the modes of transmission, the burdens of public health surveillance, th

Adhesive anti-fibrotic interfaces on diverse organs.

Implanted biomaterials and devices face compromised functionality and efficacy in the long term owing to foreign body reactions and subsequent formation of fibrous capsules at the implant-tissue interfaces1-4. Here we demonstrate that an adhesive implant-tissue interface can mitigate fibrous capsule formation in diverse animal models, including rats, mice, humanized mice and pigs, by reducing the level of infiltration of inflammatory cells into the adhesive implant-tissue interface compared to the non-adhesive implant-tissue interface. Histological analysis shows that the adhesive implant-tiss
