Investigating Leptospira dynamics in a multi-host community using an agent-based modelling approach.

Leptospirosis, a neglected bacterial zoonosis, is a global public health issue disproportionately affecting impoverished communities such as urban slums in the developing world. A variety of animal species, including peridomestic rodents and dogs, can be infected with different strains of leptospirosis. Humans contract leptospirosis via exposure to water or soil contaminated with the urine of infected animals. Due to the unavailability of safe and effective vaccines, preventive strategies mainly focus on minimizing human exposure to contaminated environment. In marginalized communities, this a

From pathogenesis to non-medical therapy: A review of exploring sensory and multi-sensory stimulation as a non-invasive treatment approach for Alzheimer's disease.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder, marked by the accumulation of amyloid-beta plaques, tau pathology, and synaptic dysfunction, ultimately leading to memory loss, cognitive decline, neuropsychiatric symptoms and neuronal degeneration that impairs daily functioning. Despite the availability of pharmacological treatments developed to slow down symptom progression, their clinical efficacy remains uncertain, and they are frequently associated with adverse side effects, underscoring the urgent demand for safer and more effective therapeutic alternatives. Non-pharm

The Aqueous Extract of Dacryodes edulis (Burseraceae) Leaves Inhibits Cell Proliferation Induced by Estradiol on the Uterus and Vagina of Ovariectomized Female Wistar Rats.

Proliferation is a cellular process strongly linked to the genesis of cancer. Natural substances with antiproliferative activities are currently potential alternatives in the treatment of cancers. Dacryodes edulis, for instance, is a medicinal plant traditionally used in the treatment of cancer. Scientific studies have reported the antioxidant activity of this plant. In addition, the presence of prostate cancer chemopreventive polyphenols was reported in D. edulis extracts. Therefore, this study was aimed to evaluate the effects of the aqueous extract of D. edulis leaves on cell proliferation

Reprogramming of pig dermal fibroblast into insulin secreting cells by a brief exposure to 5-aza-cytidine.

Large animal models provide useful data for pre-clinical research including regenerative medicine. However whereas the derivation of tissue specific stem cells has been successful. pluripotent stem cells so far have been difficult to obtain in these species. A possible alternative could be direct reprogramming but this has only been described in mouse and human. We have recently described an alternative method for reprogramming human somatic cells based on a brief demethylation step immediately followed by an induction protocol. Aim of the present paper was to determine whether this method is

Potential impact of wildlife on the tropical bont tick eradication program in the Caribbean.

Wildlife are hosts for A. variegatum throughout its range in Africa, and have been demonstrated to serve as hosts for larvae and nymphs on several islands in the Caribbean. Studies conducted in the Caribbean have indicated that most larvae and nymphs and all adults of the tick feed on livestock; therefore, eradication can be approached through the treatment of livestock. However, since small numbers of animals such as the mongoose are infested with larvae and nymphs, these animals may represent a short-term alternative host system. Such infestations potentially could result in a presence of sm

A murine model of herpes simplex virus-induced ocular disease for antiviral drug testing.

Previous studies to determine the efficacy of new antiviral compounds in treating HSV ocular infections have used rabbit models. However, rabbits are expensive to purchase and maintain, and require the use of substantial amounts of test compounds. We have used the currently licensed antiviral trifluorothymidine in a murine model of HSV-induced ocular infection to demonstrate that the less expensive murine model can be used for the in vivo evaluation of potentially useful antiviral compounds. Treatment with TFT reduced the severity of blepharitis, vascularization of the cornea, stromal keratiti

Dynamic neuroreceptor positron emission tomography in non-anesthetized rats using point source based motion correction: A feasibility study with [11C]ABP688.

To prevent motion artifacts in small animal positron emission tomography (PET), animals are routinely scanned under anesthesia or physical restraint. Both may potentially alter metabolism and neurochemistry. This study investigates the feasibility of fully awake acquisition and subsequent absolute quantification of dynamic brain PET data via pharmacokinetic modelling in moving rats using the glutamate 5 receptor radioligand [11C]ABP688 and point source based motion correction. Five male rats underwent three dynamic [11C]ABP688 PET scans: two test-retest awake PET scans and one scan under anest

Heat stress on cattle embryo: gene regulation and adaptation.

Global warming has been affecting animal husbandry and farming production worldwide via changes in organisms and their habitats. In the tropics, these conditions are adverse for agriculture and animal production in some areas, due to high temperatures and relative humidity, affecting competitiveness related to economic activities. These environments have deteriorated livestock production, due to periods of drought, reduction in forage quality and heat stress, eliciting negative effects on reproduction, weight gain, and reduced meat and milk production. However, the use of animals adapted to tr

Feasibility of pig and human-derived aortic valve interstitial cells seeding on fixative-free decellularized animal pericardium.

Glutaraldehyde-fixed pericardium of animal origin is the elective material for the fabrication of bio-prosthetic valves for surgical replacement of insufficient/stenotic cardiac valves. However, the pericardial tissue employed to this aim undergoes severe calcification due to chronic inflammation resulting from a non-complete immunological compatibility of the animal-derived pericardial tissue resulting from failure to remove animal-derived xeno-antigens. In the mid/long-term, this leads to structural deterioration, mechanical failure, and prosthesis leaflets rupture, with consequent need for

A nucleoprotein-enriched diet suppresses dopaminergic neuronal cell loss and motor deficit in mice with MPTP-induced Parkinson's disease.

Parkinson's disease (PD) is an obstinate progressive neurodegenerative disease and characterized by locomotor impairment and dopaminergic neuronal degeneration in the substantia nigra pars compacta (SNc). We examined in here the dietary effect of nucleoprotein (NP) extracted from salmon soft roe on 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-injected PD-like mice model to prevent the symptom as an alternative medicine. Male C57/BL6 mice were given either an artificially modified NP-free diet (NF) or NF supplied with 1.2% NP for 1 week. Then, mice were injected intraperitoneally four ti

Inactivation of glucocorticoid receptor in noradrenergic system influences anxiety- and depressive-like behavior in mice.

The aim of this study was to investigate whether conditional inactivation of the glucocorticoid receptors (GRs) in noradrenergic neurons affects animal behavior in mice. Selective ablation of GRs in the noradrenergic system was achieved using the Cre/loxP approach. We crossed transgenic mice expressing the Cre recombinase under the dopamine beta-hydroxylase (DBH) promoter with animals harboring the floxed GR gene. The resulting GR(DBHCre) mutant mice exhibited no alterations in terms of normal cage behavior, weight gain, spatial memory or spontaneous locomotor activity, regardless of gender. T

Comparative accuracy evaluation of patient-specific 3D-printed guide and neuronavigation for ventriculoperitoneal shunt in dogs: a dual-phase ex vivo and in vivo study.

BACKGROUND: Ventriculoperitoneal shunting (VPS) is an effective treatment for canine hydrocephalus, but complications related to ventricular catheter (VC) misplacement remain a concern. Although neuronavigation improves accuracy, its cost and complexity limit veterinary use. Patient-specific 3D-printed guiding systems (PS-3DGS) offer a potential alternative. OBJECTIVES: To compare the accuracy and feasibility of PS-3DGS compared to electromagnetic neuronavigation for VC placement in 3D-printed canine cranio-ventricular models (CVMs) and Beagle dogs. ANIMALS AND STUDY DESIGN: Ten 3D-printed C

A mobile, high-throughput semi-automated system for testing cognition in large non-primate animal models of Huntington disease.

BACKGROUND: For reasons of cost and ethical concerns, models of neurodegenerative disorders such as Huntington disease (HD) are currently being developed in farm animals, as an alternative to non-human primates. Developing reliable methods of testing cognitive function is essential to determining the usefulness of such models. Nevertheless, cognitive testing of farm animal species presents a unique set of challenges. The primary aims of this study were to develop and validate a mobile operant system suitable for high throughput cognitive testing of sheep. NEW METHOD: We designed a semi-automa

Validation crisis in animal models of drug addiction: beyond non-disordered drug use toward drug addiction.

In standard drug self-administration settings, animals have no choice than drug use. As a result, serious doubt exists about the interpretation of drug use in experimental animals. Is it symptomatic of an underlying addiction state or merely an expectable response to lack of choice? This incertitude in turn casts a shadow over many behavioral and neurobiological changes that have been well documented in animals following extended drug self-administration. Do they reflect pathological dysfunctions or normal neurobiological adaptations? Here I address these questions by focusing on intravenous c

Skin sensitisation testing--new perspectives and recommendations.

Various methodological aspects of skin sensitisation testing have been explored, particularly in the context of animal welfare considerations and reliability and sensitivity of test methods. Recommendations are made for the conduct of current and proposed OECD skin sensitisation tests with respect to appropriate test configurations for the purposes of hazard identification and labelling, and the requirement for positive controls. Specifically, the following aspects of guinea pig sensitisation test methods have been addressed: (1) the number of test and control animals required; (2) the option

Oral reference dose (RfD) derivation for five bisphenol A alternatives integrating BMD and NOAEL/LOAEL approaches.

Bisphenols (BPs) are widely used in plastic manufacturing, food packaging, and other industrial applications, with bisphenol A (BPA) being one of the most extensively produced chemicals globally. However, due to its endocrine-disrupting properties, BPA has been linked to reproductive abnormalities, metabolic disorders, neurodevelopmental impairments, and other adverse health effects, leading to regulatory restrictions. These restrictions have resulted in increased usage of BPA alternatives and thus exposure to the alternatives, whose toxicity thresholds remain insufficiently characterized. To

Comprehensive overview of alternative medicine in the treatment of ankylosing spondylitis: Symptoms, pathogenesis, diagnosis and treatments (Review).

Ankylosing spondylitis (AS) is an autoimmune disease characterized by chronic inflammation in the sacroiliac joints and cartilage, resulting in symptoms such as back pain, morning stiffness, sacroiliitis, and extra-articular manifestations. Several drugs reduce the inflammatory response, such as nonsteroidal anti-inflammatory drugs, and disease-modifying antirheumatic drugs. However, the adverse effects and high costs associated with drug therapy highlight the need for complementary treatments. In the present review, significant alternative and complementary remedies for the treatment of AS ar
