The primary target in multiple sclerosis (MS) is believed to be either myelin itself (myelinopathy) or the myelin-forming cell, the oligodendrocyte (oligodendrogliopathy). Although axonal injury occurs in MS, it is regarded as a secondary event to the myelin damage. Here, the lesion develops from myelin (outside) to the axon (inside) (Outside-In model). Recently, gray matter lesions and axonal injury in normal-appearing white matter have also been reported in MS. This raises two questions. 1) Is axonal injury exclusively secondary to myelin damage or from a direct insult to the axon or neurons

The paravermal cerebellar cortex contains three spatially separate zones (the C1, C3 and Y zones) which form a functionally coupled system involved in the control of voluntary limb movements. A series of 'modules' has been postulated, each defined by a set of olivary neurons with similar receptive fields, the cortical microzones innervated by these neurons and the group of deep cerebellar nuclear neurons upon which the microzones converge. A key feature of this modular organization is a correspondence between cortical input and output, irrespective of the zonal identity of the microzone. This

Manganese-enhanced magnetic resonance imaging (MEMRI) was used to investigate retrograde axonal tracing in the rat sciatic nerve model to assess its potential to examine peripheral nerve injury. The right sciatic nerve was exposed and crushed. After each recovery period, the distal part of the right sciatic nerve was injected with manganese (400 mM, 15 microl). After allowing 3 days for manganese transport the animals were subsequently scanned to visualize the sciatic nerve and its corresponding spinal cord and dorsal root ganglia with T1-weighted MRI. Thirty-four animals were randomly divided

Retinal ganglion cell death is the final common pathway of virtually all diseases of the optic nerve, including glaucomatous optic neuropathy. In recent years it has been shown that retinal ganglion cells die after axonal injury via a programmed cell death process called apoptosis. The dynamics of retinal ganglion cell death reflect the timing and degree of the axonal injury, rather than its nature. For example, whether mediated by ischemia (corresponding to abnormalities of peripapillary circulation) or compression (e.g., changes in retrograde transport caused by increased intraocular pressur

The paranodal junctions flank mature nodes of Ranvier and provide a barrier between ion channels at the nodes and juxtaparanodes. These junctions also promote node assembly and maintenance by mechanisms that are poorly understood. Here, we examine their role in the accumulation of NF186, a key adhesion molecule of PNS and CNS nodes. We previously showed that NF186 is initially targeted/accumulates via its ectodomain to forming PNS (hemi)nodes by diffusion trapping, whereas it is later targeted to mature nodes by a transport-dependent mechanism mediated by its cytoplasmic segment. BACKGROUND: Ion and water transport in the central nervous system (CNS) is governed by tightly coupled mechanisms involving electrodiffusion, osmotic pressure, and fluid convection. Disruptions to these processes are implicated in pathological conditions. Understanding the coordinated roles of glial cells and perivascular spaces in regulating ionic and fluid homeostasis is essential for interpreting neural function and dysfunction.
METHODS: We developed a multicompartment model of the optic nerve incorporating axons, glial cells, extracellular space (ECS), and three perivascular compartments

Progressive supranuclear palsy (PSP) is an adult-onset neurodegenerative disorder characterized by postural instability, ocular motor dysfunction, akinesia, and cognitive impairment. The accumulation of tau protein disrupts microtubule dynamics and axonal transport, leading to neurodegeneration in subcortical structures such as the basal ganglia and brainstem. These effects result in the clinical features of motor dysfunction and cognitive impairment associated with PSP. Dementia in patients with PSP is common but may be difficult to diagnose. This narrative review describes the cognitive decl…

KIF5C encodes a protein belonging to the kinesin family and involved in cellular transport. Variants in KIF5C were first associated a decade ago with microcephaly and malformations of cortical development, with a phenotypic spectrum ranging from polymicrogyria to pachygyria. Currently, eight patients have been reported so far. Here we describe a new paediatric patient carrying the recurrent p.(Glu237Lys) KIF5C variant associated with a distinctive neuroradiological pattern of abnormal posterior course of the corticospinal tract at the level of the pons with a thickened anterior component of th

The past few years have seen the identification of dozens of genes with causal roles in motor neuron diseases (MNDs), particularly for amyotrophic lateral sclerosis and hereditary spastic paraplegia. Although many additional MND genes remain to be identified, the accumulated genetic evidence has already provided new insights into MND pathogenesis, which adds to the well-established involvement of superoxide dismutase 1 (SOD1) mutations. The pathways that have been recently implicated include those that affect RNA processing, axonal transport and mitochondrial function. The functional classes o…

After gene mutations of SOD1 were found in familial amyotrophic lateral sclerosis (ALS) in 1993, many studies have elucidated pathogenesis of this progressive motor neuron disease. Among them, oxidative stress, impaired axonal transport, imbalance of survival & death signals, organellic stress (for mitochondria, endoplasmic reticulum and proteasome) are the most important with linking each other through energy failure within the motor neuron. New therapeutic approaches have also been tried, such as free radical scavenger edaravone, a continuous intra-thecal injection of neurotrophic factor IGF…

Infections with defined Herpesviruses, such as Pseudorabies virus (PRV) and Varicella zoster virus (VZV) can cause neuropathic itch, referred to as "mad itch" in multiple species. The underlying mechanisms involved in neuropathic "mad itch" are poorly understood. Here, we show that PRV infections hijack the RNA helicase DDX3X in sensory neurons to facilitate anterograde transport of the virus along axons. PRV induces re-localization of DDX3X from the cell body to the axons which ultimately leads to death of the infected sensory neurons. Inducible genetic ablation of Ddx3x in sensory neurons re

Activating PI3K/AKT/mTOR signaling pathway via deleting phosphatase and tensin homolog (PTEN) has been confirmed to enhance intrinsic growth capacity of neurons to facilitate the axons regeneration of central nervous system after injury. Considering conditional gene deletion is currently not available in clinical practice, we exploited capsid residue tyrosine 444 to phenylalanine mutated single-stranded adeno-associated virus serotype 2 (AAV2) as a vector delivering short hairpin RNA to silence PTEN to promote retinal ganglion cells (RGCs) survival and axons regeneration in adult rat optic ner

Amyotrophic lateral sclerosis (ALS) is an adult-onset motor neuron disease that has been associated with a diverse array of genetic changes. Prominent among these are mutations in RNA-binding proteins (RBPs) or repeat expansions that give rise to toxic RNA species. RBPs are additionally central components of pathologic aggregates that constitute a disease hallmark, suggesting that dysregulation of RNA metabolism underlies disease progression. In the context of neuronal physiology, transport of RNAs and localized RNA translation in axons are fundamental to neuronal survival and function. Severa

Transient receptor potential melastatin 8 (TRPM8) is activated by innocuous cool and noxious cold and plays a crucial role in cold-induced acute pain and pain hypersensitivity. To help understand the mechanism of TRPM8-mediated cold perception under normal and pathologic conditions, we used light microscopic immunohistochemistry and Western blot analysis in mice expressing a genetically encoded axonal tracer in TRPM8-positive (+) neurons. We investigated the coexpression of TRPM8 and vesicular glutamate transporter 1 (VGLUT1) and VGLUT2 in the trigeminal ganglion (TG) and the dental pulp befor

BACKGROUND AND AIM: Changes in neural plasticity crucially modulate functional recovery after central nerve injury. To describe morphological peripheral nerve injury-related changes, we comprehensively elucidated post-sciatic nerve transection (post-SNT) plasticity changes in the lumbar spinal corticospinal tract (CST), motor neurons, and cholinergic interneurons (CINs).
METHODS: In a rat SNT model, we compared post-SNT changes in lumbar spinal cord plasticity at 2, 4, and 6 weeks with those in the Sham group. Using neural tracers and immunohistochemistry, we labeled and analyzed the CST axon

BACKGROUND: Glaucoma is a complex neurodegeneration and a leading cause of blindness worldwide. Current therapeutic strategies, which are all directed towards lowering the intraocular pressure (IOP), do not stop progression of the disease. We have demonstrated that recombinant adeno-associated virus (rAAV) gene delivery of a form of erythropoietin with attenuated erythropoietic activity (EpoR76E) can preserve retinal ganglion cells, their axons, and vision without decreasing IOP. The goal of this study was to determine if modulation of neuroinflammation or oxidative stress played a role in the

Several in vitro studies indicate that the extracellular matrix (ECM) glycoprotein laminin can promote neurite outgrowth from CNS (central nervous system) neurons. Laminin has been detected immunohistochemically in astrocytes in the embryonic but not the uninjured adult mammalian CNS. In the injured adult CNS, it is found in some reactive astrocytes located near the site of CNS lesions. In the present study, we have attempted to examine the relationship between these laminin+ astrocytes and the axonal sprouting that occurs after CNS injuries. This was studied in the intracranially transected a

Neurons are highly specialized for the processing and transmission of electrical signals and use cytoskeleton-based motor proteins to transport different vesicles and cellular materials. Abnormalities in intracellular transport are thought to be a critical factor in the degeneration and death of neurons in both the central and peripheral nervous systems. Several recent studies describe disruptive mutations in the minus-end-directed microtubule motor cytoplasmic dynein that are directly linked to human motor neuropathies, such as SMA (spinal muscular atrophy) and axonal CMT (Charcot-Marie-Tooth
