Regulators of Ca(2+) signaling in mast cells: potential targets for treatment of mast cell-related diseases?

A calcium signal is essential for degranulation, generation of eicosanoids and optimal production of cytokines in mast cells in response to … iseases although studies in other types of cells have exposed genetic defects that lead to aberrant calcium signaling in immune diseases. Pharmacologic agents that inhibit or activate the regulatory components of calcium signaling in mast cells are also discussed along with the prospects for development of novel SOCE inhibitors that may prove beneficial in the trea …

Matrix stiffness induces Ca2+-DCLK1-PIP5K1A mechanotransduction as a context-specific amplifier in pancreatic cancer progression and chemotherapy resistance.

Alterations in extracellular matrix (ECM) architecture and stiffness are hallmarks of aggressive pancreatic cancer progression. However, the mechanisms by which ECM biomechanical properties regulate malignant biological behavior remain unknown. Here, we reveal that calmodulin-dependent protein kinase doublecortin-like kinase 1 (DCLK1) integrates biomechanical signaling and promotes pancreatic cancer cell progression. DCLK1 expression and activation are selectively induced under conditions of high biomechanical stress mediated through the piezo-type mechanosensitive ion channel component 1 (PIE

Memantine modulates TRPM3-associated glutamatergic signaling via vGLUT2 in a mouse model of obsessive-compulsive disorder.

Obsessive-compulsive disorder (OCD) is a chronic neuropsychiatric disorder involving neuroinflammation, glutamatergic dysregulation, and altered calcium signaling. Our recent work identified transient receptor potential melastatin 3 (TRPM3) channels as key contributors to OCD pathogenesis via modulation of neuroinflammation and calcium homeostasis. Notably, calcium permeable N-Methyl-D-aspartate (NMDA) receptor-mediated excitotoxicity is implicated in OCD through its impact on synaptic plasticity and neuronal survival in the medial prefrontal cortex (mPFC), a critical region for cognitive flex…

The pancreas misled: signals to pancreatitis.

Acute pancreatitis is an increasingly common and sometimes severe disease for which there is little specific therapy. Chronic pancreatitis is a common and grossly debilitating sequel that is largely irreversible, whatever treatment is adopted. In the face of these burdens, the absence of specific treatments is a spur to research. The acinar cell is the primary target of injury from alcohol metabolites, bile, hyperlipidaemia, hyperstimulation and other causes. These induce abnormal, prolonged, global, cytosolic calcium signals, the prevention of which also prevents premature digestive enzyme ac

Insulin Growth Factor 1 affects glutamate receptor activity differently in primary cultures of neocortical versus hippocampal neurons.

Insulin-like growth factor-1 (IGF-1) plays a critical role in neuronal signaling. Disrupted insulin/IGF-1 signaling is implicated in Alzheimer's disease, among other conditions, yet its specific influence on glutamate receptor-mediated calcium responses remains unclear. We examined the impacts of IGF-1 on glutamate receptor function in primary rat neurons monitored for intraneuronal calcium following stimulation with glutamate, AMPA, or NMDA/glycine. Pharmacological blockers (CNQX for AMPA receptors, APV for NMDA receptors, and nimodipine for L-type calcium channels) were applied to define rec

Extracellular calcium-sensing receptors in fishes.

The extracellular calcium-sensing receptor (CaSR) in fishes, like the CaSRs of tetrapod vertebrates, is a dimeric seven transmembrane, G protein-coupled receptor. The receptor is expressed on the plasma membranes of a variety of tissues and cells where it functions as a sensor of extracellular calcium concentration ([Ca(2+)](o)) in the physiological range. In the context of systemic calcium homeostasis, CaSR expressed in endocrine tissues that secrete calciotropic and other hormones (pituitary gland and corpuscles of Stannius) may play a central role in global integrative signaling, whereas re

Cancer-bone microenvironmental interactions promotes STAT3 signaling.

Prostate cancer (PCa) patients' mortality is mainly attributed to complications caused by metastasis of the tumor cells to organs critical for survival, such as bone. We hypothesized that PCa cell-bone interactions would promote paracrine signaling. A panel of PCa cell lines were cocultured with hydroxyapatite ([HA]; inorganic component of bone) of different densities. Conditioned media (CM) was collected and analyzed for calcium levels and effect on paracrine signaling, cell migration, and viability in vitro and in vivo. Our results showed that calcium levels were elevated in CM from cancer c

Cutaneous TRPV4 Potentiates the Afferent Transmission of Electroacupuncture Stimulation.

Electroacupuncture stimulation (ES) exerts therapeutic effects across various conditions, primarily by activating peripheral sensory nerves. The mechanosensitive ion channel transient receptor potential vanilloid 4 (TRPV4), expressed in peripheral non-neuronal cells, represents a potential molecular sensor for ES, yet its specific contribution to ES signal transmission remains unclear. In this study, we combined in vivo calcium imaging in transgenic mice with pharmacological interventions to investigate how cutaneous TRPV4 modulates afferent signaling at the lumbar 4 dorsal root ganglion (DRG)

Calcium measurement in living filamentous fungi expressing codon-optimized aequorin.

Calcium signalling is little understood in filamentous fungi largely because easy and routine methods for calcium measurement in living hyphae have previously been unavailable. We have developed the recombinant aequorin method for this purpose. High levels of aequorin expression were obtained in Neurospora crassa, Aspergillus niger and Aspergillus awamori by codon optimization of the aequorin gene. Three external stimuli (mechanical perturbation, hypo-osmotic shock and high external calcium) were found transiently to increase [Ca(2+)](c). Each of the calcium signatures associated with these ph

Impairment of calcium mobilization in phagocytic cells in glycogen storage disease type 1b.

Patients with glycogen storage disease (GSD) type 1b, in contrast to patients with GSD 1a, are susceptible to recurrent bacterial infections suggesting defective phagocytic function. We have demonstrated a selective defect in respiratory burst activity but not in degranulation by phagocytic cells in GSD 1b but not in GSD 1a. The respiratory burst abnormality in phagocytic cells from GSD 1b patients was associated with impaired calcium mobilization, whereas these processes were normal in GSD 1a patients. Therefore, the alteration in calcium mobilization was an indication of a signalling defect

Complement C3 inhibition restores myasthenia gravis AChR antibody-mediated muscle pathophysiology.

BACKGROUND: In the autoimmune disorder myasthenia gravis (MG), the nicotinic acetylcholine receptors (nAChRs) are the primary targets of pathogenic antibodies. While MG mechanisms have been extensively studied in animal models, functional insights into how antibody binding disrupts nAChR-dependent calcium signaling and the effects of complement inhibition in human muscle cells remain limited. METHODS: We used real-time live-cell calcium imaging to assess the effects of cholinergic stimulation or inhibition on human muscle cells with pharmacological agents, sera from patients with acetylcholin…

Two-pore channels (TPCs) acts as a hub for excitation-contraction coupling, metabolism and cardiac hypertrophy signalling.

Ca2+ signaling is essential for cardiac contractility and excitability in heart function and remodeling. Intriguingly, little is known about the role of a new family of ion channels, the endo-lysosomal non-selective cation "two-pore channel" (TPCs) in heart function. Here we have used double TPC knock-out mice for the 1 and 2 isoforms of TPCs (Tpcn1/2-/-) and evaluated their cardiac function. Doppler-echocardiography unveils altered left ventricular (LV) systolic function associated with a LV relaxation impairment. In cardiomyocytes isolated from Tpcn1/2-/- mice, we observed a reduction in the

Voltage-Gated Calcium Channels in Nonexcitable Tissues.

The identification of a gain-of-function mutation in CACNA1C as the cause of Timothy syndrome, a rare disorder characterized by cardiac arrhythmias and syndactyly, highlighted roles for the L-type voltage-gated Ca2+ channel CaV1.2 in nonexcitable cells. Previous studies in cells and animal models had suggested that several voltage-gated Ca2+ channels (VGCCs) regulated critical signaling events in various cell types that are not expected to support action potentials, but definitive data were lacking. VGCCs occupy a special position among ion channels, uniquely able to translate membrane excitab

Molecular mechanism of bitter taste receptor agonist-mediated relaxation of airway smooth muscle.

G-protein-coupled receptors (GPCRs) belonging to the type 2 taste receptors (TAS2Rs) family are predominantly present in taste cells to allow the perception of bitter-tasting compounds. TAS2Rs have also been shown to be expressed in human airway smooth muscle (ASM), and TAS2R agonists relax ASM cells and bronchodilate airways despite elevating intracellular calcium. This calcium "paradox" (calcium mediates contraction by pro-contractile Gq-coupled GPCRs) and the mechanisms by which TAS2R agonists relax ASM remain poorly understood. To gain insight into pro-relaxant mechanisms effected by TAS2R

Cardiac resynchronization therapy restores sympathovagal balance in the failing heart by differential remodeling of cholinergic signaling.

RATIONALE: Cardiac resynchronization therapy (CRT) is the only heart failure (HF) therapy documented to improve left ventricular function and reduce mortality. The underlying mechanisms are incompletely understood. Although β-adrenergic signaling has been studied extensively, the effect of CRT on cholinergic signaling is unexplored. OBJECTIVE: We hypothesized that remodeling of cholinergic signaling plays an important role in the aberrant calcium signaling and depressed contractile and β-adrenergic responsiveness in dyssynchronous HF that are restored by CRT. METHODS AND RESULTS: Canine tach…

EP2 receptor activates dual G protein signaling pathways that mediate contrasting proinflammatory and relaxatory responses in term pregnant human myometrium.

Prostaglandin (PG) E2 (PGE(2)) plays a central role in the regulation of smooth muscle contractions. Classically, PGE(2) stimulates contract … ne-associated PGE synthase-1, and PGE(2). This was entirely dependent on EP2-mediated activation of calcium signaling. Both calcium signaling and up-regulation of PGHS-2 were insensitive to the Gαi inhibitor pertussis toxin but inhibited by small interfering RNA knockdown of Gαq/11. There were no differences in EP2 mRNA or protein levels between upper or lower segm …
