Characterization of sugar binding by osteoclast inhibitory lectin.

Osteoclast inhibitory lectin (OCIL) is a membrane-bound C-type lectin that blocks osteoclast differentiation and, via binding to its cognate receptor NKRP1D, inhibits natural killer cell-mediated cytotoxicity. OCIL is a member of the natural killer cell receptor C-type lectin group that includes CD69 and NKRP1D. We investigated carbohydrate binding of soluble recombinant human and mouse OCIL in enzyme-linked immunosorbent assay-based assays. OCIL bound immobilized high molecular weight sulfated glycosaminoglycans, including fucoidan, lambda-carrageenan, and dextran sulfate, but not unsulfated

Effects of different intercropping patterns on tea rhizosphere environment and leaf quality.

Intercropping can improve tea soils and leaf quality, but partner-specific responses remain unclear. We compared four systems-Paris polyphylla-tea (CT), leguminous green manure-tea (LF), Pleurotus ostreatus-tea (CJ), and Polygonatum kingianum-tea (HJ)-with matched monoculture controls. Tea and soil traits were integrated with high-throughput sequencing, LEfSe, PICRUSt2 functional prediction, and correlation analyses. LF significantly increased soil available and total phosphorus, enriched the potentially beneficial fungi Mortierella and Trichoderma in the tea rhizosphere, and enhanced the pred

Effects of Foliar Calcium and Magnesium Application on Peach (Prunus persica L.) Nutrition Status and Fruit Quality Formation.

Peach fruit quality-including sweetness, firmness, and nutritional value-is strongly determined by mineral nutrition, yet limited soil availability of calcium (Ca) and magnesium (Mg) under production activities often constrains high-quality fruit formation in juicy peach production. This study aimed to evaluate the effects of foliar application of Ca and Mg on leaf nutrient status and fruit quality in peach (Prunus persica L. Batsch). Seven treatments were established, including 0.1% (T1), 0.2% (T2), and 0.3% (T3) CaCl2 solutions, 0.1% (T4), 0.2% (T5), and 0.3% (T6) MgSO4 solutions, and a wate

Metataxonomic, chemical, and sensory aspects of cocoa fermentation using single hybrids and combinations of hybrids and their effects on chocolate quality.

Extensive research has been conducted on cocoa fermentation in single hybrids. However, comparisons between hybrid combinations and single hybrids are limited. This study investigated cacao fermentation using a hybrid combination (PS 1319, PH 16, BN 34, CEPEC 2002, SJ02, CCN 10, and Ipiranga) and SJ02 hybrid alone. Microbial diversity was determined by next-generation sequencing (NGS), organic acids, carbohydrates, and alcohols by HPLC, volatile compounds by GC/MS, and the sensory analysis by acceptance test and Check-All-That-Apply (CATA). The NGS analysis identified 64 fungal species and 60

Goose surfactant protein A inhibits the growth of avian pathogenic Escherichia coli via an aggregation-dependent mechanism that decreases motility and increases membrane permeability.

Pulmonary collectins have been reported to bind carbohydrates on pathogens and inhibit infection by agglutination, neutralization, and opsonization. In this study, surfactant protein A (SP-A) was identified from goose lung and characterized at expression- and agglutination-functional levels. The deduced amino acid sequence of goose surfactant protein A (gSP-A) has two characteristic structures: a shorter, collagen-like region and a carbohydrate recognition domain. The latter contains two conserved motifs in its Ca2+-binding site: EPN (Glu-Pro-Asn) and WND (Trp-Asn-Asp). Expression analysis usi

Evolution of vancomycin-resistant Enterococcus faecium during colonization and infection in immunocompromised pediatric patients.

Patients with hematological malignancies or undergoing hematopoietic stem cell transplantation are vulnerable to colonization and infection with multidrug-resistant organisms, including vancomycin-resistant Enterococcus faecium (VREfm). Over a 10-y period, we collected and sequenced the genomes of 110 VREfm isolates from gastrointestinal and blood cultures of 24 pediatric patients undergoing chemotherapy or hematopoietic stem cell transplantation for hematological malignancy at St. Jude Children's Research Hospital. We used patient-specific reference genomes to identify variants that arose ove

Multiple phenotypic changes in mice after knockout of the B3gnt5 gene, encoding Lc3 synthase--a key enzyme in lacto-neolacto ganglioside synthesis.

BACKGROUND: Ganglioside biosynthesis occurs through a multi-enzymatic pathway which at the lactosylceramide step is branched into several biosynthetic series. Lc3 synthase utilizes a variety of galactose-terminated glycolipids as acceptors by establishing a glycosidic bond in the beta-1,3-linkage to GlcNaAc to extend the lacto- and neolacto-series gangliosides. In order to examine the lacto-series ganglioside functions in mice, we used gene knockout technology to generate Lc3 synthase gene B3gnt5-deficient mice by two different strategies and compared the phenotypes of the two null mouse group

Probing of the receptor-binding sites of the H1 and H3 influenza A and influenza B virus hemagglutinins by synthetic and natural sialosides.

To compare features of the receptor-binding sites (RBSs) of different influenza virus hemagglutinins (HA), binding of a number of synthetic sialic acid (SA) analogs and natural sialosides by a panel of about 30 human influenza A and B virus strains was studied in a competitive ligand binding assay. For all the viruses tested, the N-acetyl group of Neu5Ac, as well as the natural orientation of the carboxylic group at C2 and the hydroxylic group at C4, was essential for binding. Significant type- and subtype-specific differences were observed in virus recognition of asialic parts of sialosides.

Genomic analysis of differentially expressed genes in liver and biliary epithelial cells of patients with primary biliary cirrhosis.

The characterization of differentially expressed genes provides a powerful tool for identifying molecules that may be involved in the pathogenesis of disease. We have used two independent techniques to identify overexpressed transcripts in bile duct cells and in liver from patients with primary biliary cirrhosis (PBC). In the first method, we used suppressive subtractive hybridization to compare mRNA from isolated PBC bile duct epithelial cells (BECs) to normal BECs and identified 71 clones as transcribed at higher levels in PBC-BECs. Amongst these clones, 62/71 had matches in a non-redundant

Associations between sodium intake and gut microbiota composition.

INTRODUCTION: High salt intake is associated with adverse health outcomes. Emerging evidence highlights the importance of the gut microbiome in human health, but large-scale human data on salt intake and the microbiome are limited. We examined the cross-sectional association between estimated 24-h sodium excretion (est24hNa) and the gut microbiome in a population-based cohort. METHODS: We included 9,220 participants from the Swedish SCAPIS cohort with available shotgun metagenomic sequencing of fecal samples and urine analyses. We estimated the 24-h sodium excretion using the Kawasaki formula

Tea Tree Essential Oil Enriches Lactic Acid Bacteria and Improves Paper Mulberry Silage Quality.

Paper mulberry (Broussonetia papyrifera) is a high-protein forage, but its high buffering capacity and low water-soluble carbohydrate (WSC) content hinder silage fermentation. This study evaluated the effect of tea tree (Melaleuca alternifolia) essential oil (TTO) as a natural additive for paper mulberry silage. TTO was added at 0 (CK), 500 (CSD), and 1000 mg/kg (CSG) fresh weight, with samples collected on days 7, 15, 30, and 90. After 90 days, CSD had the lowest pH and the highest lactic acid content. NH3-N/TN was significantly lower than in CK (p < 0.05), and the retention of dry matter, cr

On the evolution of Neurospora crassa ACW-1 and NCW-3, proteins with different cell wall interaction mechanisms.

The fungal cell wall is populated with cell wall-resident glycoproteins (CWPs), mostly uncharacterized, that show an atypical evolutionary behavior. Most CWPs are glycosylphosphatidylinositol (GPI) proteins, followed by proteins with internal repeats (PIR), and non-covalently attached proteins that harbor carbohydrate binding domains (CBM). Several structural CWPs are initially bound to the same wall carbohydrates, either covalently or non-covalently. However, it is not yet clear whether they are subjected to the same evolutionary constraints. In Neurospora crassa, CWPs ACW-1 (NCU08936) and NC

Microscale analyses of the effect of arginine on oral biofilms.

OBJECTIVES: Arginine is a caries-preventive agent that modulates the microbial composition of dental biofilms and raises pH via bacterial ammonia production. The effect of arginine on biofilm pH has only been measured in bulk, and it is unknown how treatment affects microscale pH developments inside dental biofilms. Moreover, little is known about arginine's impact on the biofilm matrix. This study investigated the effects of arginine on biofilm pH, matrix architecture and microbial composition in a saliva-derived biofilm model. METHODS: 72-h biofilms were grown with or without sucrose, and a

The Mucosally-Adherent Rectal Microbiota Contains Features Unique to Alcohol-Related Cirrhosis.

Most studies examining correlations between the gut microbiota and disease states focus on fecal samples due to ease of collection, yet there are distinct differences when compared to samples collected from the colonic mucosa. Although fecal microbiota has been reported to be altered in cirrhosis, correlation with mucosal microbiota characterized via rectal swab has not been previously described in this patient population. We conducted a cross-sectional analysis using 39 stool and 39 rectal swabs from adult patients with cirrhosis of different etiologies and performed shotgun metagenomic seque

Comparative characterization of bacterial communities in geese fed all-grass or high-grain diets.

BACKGROUND: Gut microbial composition is dependent on diet. Geese are herbivores and can digest crude fibre, but the relationship between composition of the microbiota and a fibre-rich diet in geese is not well understood. RESULTS: Here, caecal and faecal samples were collected simultaneously from all-grass-fed geese and high-grain-fed geese and the hypervariable V3-V4 regions of the bacterial 16S rRNA gene were sequenced. The results was identified that high-grass-fed geese possessed significantly higher alpha diversity both in caecum and faeces compared with that in all-grain-fed geese. In

Pathogenesis of shigella diarrhea. XI. Isolation of a shigella toxin-binding glycolipid from rabbit jejunum and HeLa cells and its identification as globotriaosylceramide.

A glycolipid that specifically binds shigella toxin was isolated from both HeLa cells and rabbit jejunal mucosa and identified as globotriaosylceramide (Gb3) by its identical mobility on HPTLC to authentic erythrocyte Gb3. Toxin also bound to a band tentatively identified as alpha-hydroxylated Gb3. In addition, toxin bound to P1 antigen present in group B human erythrocyte glycolipid extracts. The common feature of the three binding glycolipids is a terminal Gal alpha 1----4Gal disaccharide linked beta 1----4 to either Glc or GlcNAc. Globoisotriaosylceramide, which differs from Gb3 only in pos

COOH-terminal propeptides of the major human procollagens. Structural, functional and genetic comparisons.

The sequences of the carboxy-terminal extensions (COOH-propeptides) of at least one chain of all of the major human procollagens have only recently been deduced, and include those of the interstitial (alpha 1(I), alpha 2(I), alpha 1(II), alpha 1(III)), basement membrane (alpha 1(IV)) and pericellular (alpha 2(V)) procollagens. Comparisons of DNA and protein sequences, corresponding to these COOH-propeptides domains, established the early divergence of the basement membrane alpha 1(IV) COOH-propeptide from the corresponding sequences of the interstitial and pericellular procollagens.
