[Participation of metabolic systems of carotid chemoreceptors in their effector regulation].

Electrical stimulation of the rat sinus nerve altered fluorescence of restored pyridin nucleotides and oxidative flavoproteids in the carotid body cells due to enhancement of cellular respiration. This was followed by an increase of cGMP content in the carotid body. The greatest shifts of these parameters occurred in square-wave stimulation with 20/sec frequency. The effector influences on the carotid body metabolism are most effectively blocked with pentamine and seem to be of cholinergic nature, the transmitter effect of acetylcholine being modulated with enkephalins. The cGMP system may ser

Effect of anoxic radiosensitizers on cellular and mitochondrial oxygen consumption and respiration control ratio.

Misonidazole increased oxygen consumption of Ehrlich ascites cells and reversed the blockage of oxidative phosphorylation induced by amytal and KCN. It alos increased oxygen consumption of mitochondria in the presence of alpha-ketoglutarate substrate, but decreased the respiratory control ratio. Cellular oxygen consumption decreased, but mitochondrial consumption was increased by metronidazole, and the respiratory control ratio was not affected by this substance. It was concluded that misonidazole influences respiration in a similar way to uncouplers, and the interaction site for both substanc

Predictors of hyperlactataemia among children presenting with malaria in a low transmission area in The Gambia.

BACKGROUND: Hyperlactataemia and metabolic acidosis are important risk factors for malaria death, but measuring lactate at the point of care is not financially viable in many resource-poor settings. This study aimed to identify combinations of routinely available parameters that could identify children at high risk of hyperlactataemia. METHODS: Using data from a study of Gambian children aged six months to 16 years with severe or uncomplicated malaria, logistic regression modelling with a forward stepwise model selection process was used to develop a predictive model for hyperlactataemia from

The Interleukin-2-mTORc1 Kinase Axis Defines the Signaling, Differentiation, and Metabolism of T Helper 1 and Follicular B Helper T Cells.

The differentiation of CD4(+) helper T cell subsets with diverse effector functions is accompanied by changes in metabolism required to meet their bioenergetic demands. We find that follicular B helper T (Tfh) cells exhibited less proliferation, glycolysis, and mitochondrial respiration, accompanied by reduced mTOR kinase activity compared to T helper 1 (Th1) cells in response to acute viral infection. IL-2-mediated activation of the Akt kinase and mTORc1 signaling was both necessary and sufficient to shift differentiation away from Tfh cells, instead promoting that of Th1 cells. These finding

Extracellular vesicles from long COVID patients promote RUNX2-mediated cellular stress via dysregulated miR-204 and p53 pathway activation.

BACKGROUND: Subjects with Long COVID, also known as post-acute sequelae of SARS-CoV-2 infection (PASC), experience a wide range of symptoms, including fatigue and respiratory disturbances, affecting their quality of life. Despite the increasing prevalence of Long COVID, the underlying pathogenic mechanisms remain poorly understood. Extracellular vesicles (EVs) are known to be involved in various processes, such as tissue repair and the transmission of viral particles. However, the specific characteristics and functional roles of EVs derived- from patients with Long COVID (LC-EVs) are poorly ch

A screening model to predict microalgae biomass growth in photobioreactors and raceway ponds.

A microalgae biomass growth model was developed for screening novel strains for their potential to exhibit high biomass productivities under nutrient-replete conditions in photobioreactors or outdoor ponds. Growth is modeled by first estimating the light attenuation by biomass according to Beer-Lambert's Law, and then calculating the specific growth rate in discretized culture volume slices that receive declining light intensities due to attenuation. The model uses only two physical and two species-specific biological input parameters, all of which are relatively easy to determine: incident li

MIZ-1 controls transcriptional programs required for BCR signaling, actin dynamics, and naïve B cell survival.

B cell receptor (BCR) signaling is required for the regulation of normal B lymphocyte development. However, many B cell lymphoma subtypes employ constitutive BCR signaling for transformation, survival, and growth. Here we report that mice expressing a POZ domain-deficient MIZ-1 (MIZ-1ΔPOZ), a well-known binding partner of the oncogene MYC, exhibit reduced surface receptor expression on naïve B cells and impaired BCR signaling. Transcriptomic and ChIP-seq analyses revealed that, in addition to autophagy related genes, MIZ-1 directly regulates genes involved in BCR signal transduction and actin

PM2.5 induced lung injury through upregulating ROS-dependent NLRP3 Inflammasome-Mediated Pyroptosis.

The main cause of air pollution is PM2.5, which directly causes lung injury through respiration. Oxidative stress and inflammation are considered to be the key mechanism of cell damage. Pyroptosis is a process of the programmed death of inflammatory cells and as a dangerous endogenous signal, it is widely involved in different inflammatory diseases. However, few studies have been conducted on PM2.5 exposure and cell pyroptosis. In this study, we aimed to investigate the effect of PM2.5 on apoptosis, pyroptosis and cell cycle arrest regulated by reactive oxygen species production. Balb/c mice w

Trypanosoma brucei: differentiation of in vitro-grown bloodstream trypomastigotes into procyclic forms.

Trypanosoma brucei strain 366D trypomastigotes grown at 37 degrees C in the presence of a human fibroblast cell line formed foci underneath the feeder cells whereas trypanosomes grown in the presence of a human epithelial cell line grew only in the culture supernatant. A culture system was developed to study the differentiation of bloodstream trypomastigotes grown in the epithelial cell system into procyclic trypomastigotes at 27 degrees C. The morphological differentiation into the procyclic form was complete by 48 h. Cell division did not occur until 30-40 h after transfer to 27 degrees C. V

The purple membrane of Halobacterium halobium: a new system for light energy conversion.

Patches of a distinctly different structure from the rest of the cell membrane occur in Halobacterium halobium. The isolated patches are called the purple membrane. It derives its colour from a retinal-protein complex, bacteriorhodopsin, which is the only protein species in this membrane serving a phototransducing function. Light energy is converted by a photochemical cycle going on continuously under illumination and accompanied by a cyclic release and uptake of protons. In the intact cell, this cycle operates as a vectorial process and therefore builds up an electrochemical gradient across t

Transcriptional changes related to metabolic and iron acquisition strategies of avian pathogenic Escherichia coli are associated with embryonic survival during yolk sac infection.

INTRODUCTION: Avian pathogenic Escherichia coli (APEC) causes significant economic losses in poultry production, yet the molecular mechanisms underlying divergent infection outcomes remain poorly defined. This study aimed to elucidate the in vivo transcriptional profiles in APEC from infected chicken embryos compared to in vitro grown culture. METHODS: Chicken embryos were experimentally infected with an APEC isolate. Subsequently, RNA sequencing was performed on APEC isolated from the yolk of experimentally infected embryos that either died or survived the infection, alongside in vitro grown

NLRX1 Prevents M2 Macrophage Polarization and Excessive Renal Fibrosis in Chronic Obstructive Nephropathy.

BACKGROUND: Chronic kidney disease often leads to kidney dysfunction due to renal fibrosis, regardless of the initial cause of kidney damage. Macrophages are crucial players in the progression of renal fibrosis as they stimulate inflammation, activate fibroblasts, and contribute to extracellular matrix deposition, influenced by their metabolic state. Nucleotide-binding domain and LRR-containing protein X (NLRX1) is an innate immune receptor independent of inflammasomes and is found in mitochondria, and it plays a role in immune responses and cell metabolism. The specific impact of NLRX1 on mac

Redox Cycling Dioxonaphthoimidazoliums Disrupt Iron Homeostasis in Mycobacterium bovis Bacillus Calmette-Guérin.

The dioxonaphthoimidazolium scaffold is a novel, highly bactericidal redox cycling antituberculosis chemotype that is reliant on the respiratory enzyme Type II NADH dehydrogenase (NDH2) for the generation of reactive oxygen species (ROS). Here, we employed Mycobacterium bovis Bacillus Calmette-Guérin (M. bovis BCG) reporter strains to show that ROS generated by the redox cycler SA23 simulated an iron deficient state in the bacteria, which led to a compensatory increase in the expression of the iron acquisition mbtB gene while collaterally reducing the expression of the iron storage bfrB gene.

Microbial metabolite Enterobactin impairs mitochondrial respiration and alleviates colitis.

BACKGROUND AND AIMS: Host-microbe interactions critically shape cellular metabolism and immune responses. The bacterial siderophore enterobactin (Ent) is known for pilfering iron from the host, but recent evidence suggests that it may also deliver iron to host mitochondria. Its impact on mitochondrial respiration, however, remains poorly understood. Here, we assess the interplay among Ent, iron, and lipocalin-2 (Lcn2, which sequesters Ent) on mitochondrial function. We also examine the effects of the 2, 3-dihydroxybenzoic acid (2, 3-DHBA), the monomeric derivative of Ent, and the mammalian sid

Selective Metal Chelation by a Thiosemicarbazone Derivative Interferes with Mitochondrial Respiration and Ribosome Biogenesis in Candida albicans.

Metal chelation is generally considered as a promising antifungal approach but its specific mechanisms are unclear. Here, we identify 13 thiosemicarbazone derivatives that exert broad-spectrum antifungal activity with potency comparable or superior to that of fluconazole in vitro by screening a small compound library comprising 89 thiosemicarbazone derivatives as iron chelators. Among the hits, 19ak exhibits minimal cytotoxicity and potent activity against either azole-sensitive or azole-resistant fungal pathogens. Mechanism investigations reveal that 19ak inhibits mitochondrial respiration ma

Infection-Induced Intestinal Dysbiosis Is Mediated by Macrophage Activation and Nitrate Production.

Oral infection of C57BL/6J mice with Toxoplasma gondii results in a marked bacterial dysbiosis and the development of severe pathology in the distal small intestine that is dependent on CD4+ T cells and interferon gamma (IFN-γ). This dysbiosis and bacterial translocation contribute to the development of ileal pathology, but the factors that support the bloom of bacterial pathobionts are unclear. The use of microbial community profiling and shotgun metagenomics revealed that Toxoplasma infection induces a dysbiosis dominated by Enterobacteriaceae and an increased potential for nitrate respirati

31P NMR studies of intact anchorage-dependent mouse embryo fibroblasts.

31P NMR spectra of "normal" and x-ray-transformed anchorage-dependent mouse embryo fibroblasts (C3H/10T 1/2) at 145.7 MHz and 37 degrees C were obtained by using a solenoid coil probe with a sensitivity approximately 2.6 times higher than that of the standard Helmholtz coil probe. The cells were grown as monolayers on the surface of microcarrier beads. Results show that these cells have low levels of NMR-detectable ADP, are highly impermeable to protons, are capable of maintaining high ATP levels in the absence of respiration, and, when ATP generation is blocked, ultimately hydrolyze their exi

Single-Cell RNA-Seq Analysis of Olfactory Mucosal Cells of Alzheimer's Disease Patients.

Olfaction is orchestrated by olfactory mucosal cells located in the upper nasal cavity. Olfactory dysfunction manifests early in several neurodegenerative disorders including Alzheimer's disease, however, disease-related alterations to the olfactory mucosal cells remain poorly described. The aim of this study was to evaluate the olfactory mucosa differences between cognitively healthy individuals and Alzheimer's disease patients. We report increased amyloid-beta secretion in Alzheimer's disease olfactory mucosal cells and detail cell-type-specific gene expression patterns, unveiling 240 differ

Ciclopirox activates PERK-dependent endoplasmic reticulum stress to drive cell death in colorectal cancer.

Ciclopirox (CPX) modulates multiple cellular pathways involved in the growth of a variety of tumor cell types. However, the effects of CPX on colorectal cancer (CRC) and the underlying mechanisms for its antitumor activity remain unclear. Herein, we report that CPX exhibited strong antitumorigenic properties in CRC by inducing cell cycle arrest, repressing cell migration, and invasion by affecting N-cadherin, Snail, E-cadherin, MMP-2, and MMP-9 expression, and disruption of cellular bioenergetics contributed to CPX-associated inhibition of cell growth, migration, and invasion. Interestingly, C

MGMT deficiency augments STING-mediated inflammatory responses accompanied by metabolic alterations in macrophages.

The cGAS-STING pathway senses cytosolic DNA derived from both pathogens and host cells and plays a central role in innate immune responses. O6-methylguanine-DNA methyltransferase (MGMT) is a DNA repair enzyme that removes alkylation-induced DNA lesions and modulates macrophage inflammatory responses. Here, we investigated the role of MGMT in macrophage responses to STING activation. Bone marrow-derived macrophages (BMMs) from Lyz2ΔMgmt mice produced higher levels of IL6, TNFα, and IFNβ following stimulation with the STING agonist DMXAA, accompanied by increased phosphorylation of TBK1 and IRF3
