Memory formation and storage rely on multiple interconnected brain areas, the contribution of which varies during memory consolidation. The medial prefrontal cortex, in particular the prelimbic cortex (PL), was traditionally found to be involved in remote memory storage, but recent evidence points toward its implication in early consolidation as well. Nevertheless, the inputs to the PL governing these dynamics remain unknown. Here, we first performed a brain-wide, rabies-based retrograde tracing screen of PL engram cells activated during contextual fear memory formation in male mice to identif

Memories of negative experiences exert important control of behavior in the face of actual or anticipated threat. Sometimes, however, this control extends to non-threatening situations, a phenomenon known as overgeneralization of negative memories. Overgeneralization is a reliable cognitive phenotype of major depressive disorder, generalized anxiety disorder, and post-traumatic stress disorder. We therefore sought to develop an animal model to study stress-induced generalization of negative memories (SIG) and determine its dependence on the episodic-like memory circuit. We found that male and

Sensory-driven decisions are formed by accumulating information over time. Although parietal cortex activity is thought to represent accumulated evidence for sensory-based decisions, recent perturbation studies in rodents and non-human primates have challenged the hypothesis that these representations actually influence behavior. Here, we asked whether the parietal cortex integrates acoustic features from auditory cortical inputs during a perceptual decision-making task. If so, we predicted that selective inactivation of this projection should impair subjects' ability to accumulate sensory evi

Circulating branched-chain amino acids (BCAAs) are elevated in obesity and diabetes, and recent studies support a causal role for BCAAs in insulin resistance and defective glycemic control. The physiological mechanisms underlying BCAA regulation are poorly understood. Here we show that insulin signaling in the mediobasal hypothalamus (MBH) of rats is mandatory for lowering plasma BCAAs, most probably by inducing hepatic BCAA catabolism. Insulin receptor deletion only in agouti-related protein (AgRP)-expressing neurons (AgRP neurons) in the MBH impaired hepatic BCAA breakdown and suppression of

Extracellular adenosine, a danger signal, can cause hypothermia. We generated mice lacking neuronal adenosine A1 receptors (A1AR, encoded by the Adora1 gene) to examine the contribution of these receptors to hypothermia. Intracerebroventricular injection of the selective A1AR agonist (Cl-ENBA, 5'-chloro-5'-deoxy-N6-endo-norbornyladenosine) produced hypothermia, which was reduced in mice with deletion of A1AR in neurons. A non-brain penetrant A1AR agonist [SPA, N6-(p-sulfophenyl) adenosine] also caused hypothermia, in wild type but not mice lacking neuronal A1AR, suggesting that peripheral neur

The brain circuits and synaptic processes that underlie alcohol addiction are currently the subject of intensive research. Here we focus on hippocampal circuitry and show that chemogenetic inhibition of dentate gyrus (DG) during presentation of alcohol-associated cues has long-lasting effects on mice behavior. DG inhibition enhances alcohol seeking and drinking, suggesting that DG regulates addiction-related behaviors. To test this hypothesis, we perform whole-cell patch-clamp recordings from the granule cells of DG and look for electrophysiological correlates of alcohol addiction. We observe

Pain is often described as a "biopsychosocial" process, yet social influences on pain and underlying neural mechanisms are only now receiving significant experimental attention. Expression of pain by one individual can be communicated to nearby individuals by auditory, visual, and olfactory cues. Conversely, the perception of another's pain can lead to physiological and behavioral changes in the observer, which can include induction of hyperalgesia in "bystanders" exposed to "primary" conspecifics in which hyperalgesia has been induced directly. The current studies were designed to investigate

Inspiratory accessory respiratory muscles (ARMs) enhance ventilation when demands are high, such as during exercise and/or pathological conditions. Despite progressive degeneration of phrenic motor neurons innervating the diaphragm, amyotrophic lateral sclerosis (ALS) patients and rodent models are able to maintain ventilation at early stages of disease. In order to assess the contribution of ARMs to respiratory compensation in ALS, we examined the activity of ARMs and ventilation throughout disease progression in SOD1G93A ALS model mice at rest using a combination of electromyography and unre

BACKGROUND: Intraneuronal accumulation of hyperphosphorylated tau is a hallmark of Alzheimer's disease (AD). Given the significant correlation between tau pathology and memory loss in AD patients, identifying vulnerable brain regions, particularly susceptible neuron types in these regions, will advance our understanding of AD onset and shed light on therapeutic strategies to manage its progression.
METHODS: Immunofluorescent staining was employed to identify the brain regions and neuron types vulnerable to tau pathology in AD. A combination of chemogenetics, electrophysiological recording, in…

To probe the potential casual effects of increased PV activity during whisker trimming, we used chemogenetics to acutely manipulate the activity of PV cells and found that modulating PV cell activity strongly affects sensory evoked responses in local Pyr and PV cells, as well as Somatostatin (SST) interneurons. In particular, increased PV cell activity stro …

Rotator cuff injury (RCI) is frequently accompanied by psychological comorbidities, particularly anxiety-related symptoms, which predict poor functional recovery, yet the central mechanisms linking affective disturbance to impaired tendon-bone healing remain unclear. Here, by integrating clinical neuroimaging, imaging transcriptomics, and mechanistic animal experiments, we identify hypothalamic paraventricular nucleus (PVN) oxytocinergic (OXT) neurons as a critical central node coordinating emotional regulation and bone-tendon interface (BTI) repair. In patients with RCI, resting-state fMRI (r

Chronic post-surgical pain (CPSP) in rats is characterized by persistent mechanical allodynia and spinal neuronal hypersensitivity. Astrocyte-derived L-lactate, a key modulator of neuronal excitability and synaptic plasticity, was herein investigated for its role in CPSP development following skin/muscle incision and retraction (SMIR). SMIR triggered long-lasting mechanical allodynia, concomitantly with astrocyte activation and elevated L-lactate levels in the spinal dorsal horn. Blockage of glycogenolysis by 4-dideoxy-1,4-imino-D-arabinitol (DAB), inhibition of carbonic anhydrase (CA) by acet

Dysregulated interactions between stress-sensitive neural circuits and immune signaling contribute to depression. We investigated whether the septohippocampal cholinergic pathway mediates the behavioral and neuroimmune effects of transcutaneous auricular vagus nerve stimulation (taVNS). Male mice were studied using chronic unpredictable mild stress (CUMS) and lipopolysaccharide (LPS) challenge models. Depression-related behaviors were assessed using the sucrose preference, forced swim, tail suspension, open field, and elevated plus maze tests. Cholinergic markers, hippocampal cytokines, microg

Food-related stimuli can promote feeding behaviors independent of metabolic need. Cue-potentiated feeding (CPF) studies in rodents offer the potential to reveal the psychobiological mechanisms underlying learned overeating behaviors. We examined whether lateral hypothalamic area (LHA) cells expressing the feeding signal Melanin Concentrating Hormone (MCH) are sufficient and necessary for CPF. Tg(Pmch-Cre) mice received bilateral infusion of the Cre-dependent inhibitory DREADD (hM4Di), or optrode placement together with the excitatory opsin ChR2. Mice received food deprivation by restricting ac

Short-term declarative memory impairment, encompassing deficits in spatial working memory and object recognition, represents a prevalent non-motor symptom of Parkinson's disease (PD). Despite its clinical significance, the neural circuit mechanisms underlying this cognitive dysfunction remain poorly understood. Here, we identify a previously underappreciated dopaminergic projection from the locus coeruleus (LC) to the retrosplenial cortex (RSC) as a critical regulator of short-term declarative memory. Using optogenetic activation of LC dopaminergic neurons, we evoked dopamine release in the RS

The precise regulation of synaptic plasticity by local redox states represents a fundamental, yet poorly understood, dimension of neural circuit control. Traditional models of oxidative stress often invoke global damage, failing to resolve how specific neuronal circuits utilize hydrogen peroxide (H2O2) for signaling. Here, we developed a chemogenetic strategy to interrogate this question, targeting the expression of D-amino acid oxidase (DAAO) for the spatially and temporally controlled production of H2O2 within distinct neuronal subpopulations of the medial septal area (MSA). Contrary to indu

The nucleus accumbens (NAc) is a critical node in the neural circuitry underlying reward and motivated behavior, including hedonic feeding, and its dysfunction is implicated in maladaptive behaviors in numerous psychiatric disorders. Medium spiny neurons (MSNs) in the NAc are predominantly categorized into dopamine 1 receptor-expressing (D1-MSNs) and dopamine 2 receptor-expressing (D2-MSNs) subtypes, which are thought to exert distinct and sometimes opposing roles in reward-related processes. Here, we used optogenetic, chemogenetic, and fiber photometry approaches in Cre driver mouse lines to

Sleep disturbances are increasingly recognized as a lifestyle factor for abnormal pain perception. Recent studies highlight the critical role of gut microbiota in maintaining physiological balance and reveal a bidirectional relationship between microbiota alterations, sleep disorders, and pathological pain. This study investigates the potential role of microbiota in linking sleep deprivation to abnormal pain. Using a chronic sleep deprivation (CSD) model in mice, we observed significantly lower pain thresholds compared to controls. Brain-wide functional ultrasound imaging revealed reduced cere
