Salubrinal improves mechanical properties of the femur in osteogenesis imperfecta mice.

Salubrinal is an agent that reduces the stress to the endoplasmic reticulum by inhibiting de-phosphorylation of eukaryotic translation initiation factor 2 alpha (eIF2α). We and others have previously shown that the elevated phosphorylation of eIF2α stimulates bone formation and attenuates bone resorption. In this study, we applied salubrinal to a mouse model of osteogenesis imperfecta (Oim), and examined whether it would improve Oim's mechanical property. We conducted in vitro experiments using RAW264.7 pre-osteoclasts and bone marrow derived cells (BMDCs), and performed in vivo administration

Efficient construction of rAAV-based gene targeting vectors by Golden Gate cloning.

The recombinant adeno-associated virus (rAAV) has proven to be an efficient and attractive tool for targeted genome engineering. Here we present a novel method employing the Golden Gate cloning strategy for fast and efficient construction of rAAV-based gene knockout or single-nucleotide knockin vectors. Two vectors, pGolden-Neo and pGolden-Hyg, were generated as common assembling modules to confer antibiotic resistance to the targeting vector. To validate the method, we then generated two rAAV-based targeting vectors: pAAV-pTP53-KO and pAAV-hTau(P301L)-KI. Furthermore, we generated a pGolden-A

Agrobacterium tumefaciens-mediated genetic transformation of haptophytes (Isochrysis species).

Isochrysis galbana and Isochrysis sp. are economically important microalgae from the division of haptophytes. Here, we report Agrobacterium-mediated stable DNA transfer into their nuclear genomes. Initial studies were performed to standardize co-cultivation media and determine the sensitivity of the microalgae to selective agents. Up to 1 mg/ml of the antibiotic hygromycin did not inhibit growth, whereas both the haptophytes bleached in artificial seawater (ASW) medium containing micromolar concentrations of the herbicide norflurazon. Co-cultivation of Isochrysis sp. and I. galbana with Agroba

Effect of ultraviolet filters on skin superoxide dismutase activity in hairless mice after a single dose of ultraviolet radiation.

Organic sunscreens may decrease their protective capability and also behave as photo-oxidants upon ultraviolet radiation (UVR) exposure. The present study investigated the effect of a cream gel formulation containing the UV filters benzophenone-3, octyl methoxycinnamate, and octyl salicylate on skin superoxide dismutase (SOD) after a single dose of UVR (2.87 J/cm(2)). The retention of these UV filters was first evaluated in vivo using hairless mice to guarantee the presence of the filters in the skin layers at the moment of irradiation. The in vivo effect of the UV filters on skin SOD was then

Prunella vulgaris extract and rosmarinic acid suppress lipopolysaccharide-induced alteration in human gingival fibroblasts.

Periodontitis is a chronic disease associated with inflammation of the tooth-supporting tissues. The inflammation is initiated by a group of gram-negative anaerobic bacteria. These express a number of irritating factors including a lipopolysaccharide (LPS), which plays a key role in periodontal disease development. Plant extracts with anti-inflammatory and anti-microbial properties have been shown to inhibit bacterial plaque formation and thus prevent chronic gingivitis. In this study we tested effects of Prunella vulgaris L. extract (PVE; 5, 10, 25microg/ml) and its component rosmarinic acid

Effect of opioid receptor antagonists on hypothalamic-pituitary-adrenal activity in rhesus monkeys.

Some opioid antagonists increase the release of adrenocorticotropic hormone (ACTH) and cortisol in humans and, therefore, may indicate that endogenous opioids modulate hypothalamic-pituitary-adrenal axis activity. The type of opioid receptor that may be related to these endocrine effects is unknown. The purpose of this experiment was to evaluate the ability of different opioid antagonists to increase ACTH and cortisol plasma levels in rhesus monkeys. Eight monkeys received intramuscular injections of various antagonists: 0.0032-1.0 mg/kg naltrexone, 0.1-3.2 mg/kg naltrindole (delta-selective),

Differential protection by two sunscreens from UV radiation-induced immunosuppression.

A controversy has arisen concerning the ability of sunscreens to protect mice from the immunosuppressive effects of UV radiation. We have assessed the photoprotection in hairless mice of two sun protection factor (SPF)15 sunscreens containing different UVB (280-320-nm) absorbers, namely, octyl-N-dimethyl-p-aminobenzoate (o-PABA) or 2-ethylhexyl-p-methoxycinnamate (2-EHMC). Following three minimum erythemal exposures to UV radiation, both systemic suppression of contact hypersensitivity to 2,4-dinitrofluorobenzene and induction of susceptibility to transplanted UV radiation-induced tumor cells

ABA-cytokinin crosstalk regulate drought tolerance in Ziziphus jujuba var. spinosa through the phenylpropanoid pathway: insights from physiological and multi-omics integration.

Drought stress is a major constraint in jujube production. Although abscisic acid (ABA) and cytokinin exert opposing effects on plant growth, recent findings revealed that their combined application enhanced drought tolerance in jujube; however, the underlying mechanism remains elusive. In this study, three exogenous treatments were applied to sour jujube seedlings under drought stress: 5 mg L-1 ABA, a cytokinin mixture of 50 mg L-1 6-benzylaminopurine (6-BA) and 100 mg L-1 kinetin (KT), and a combined ABA and cytokinins solution. Transcriptomic and metabolomic analyses were integrated to eluc

Comparative In Vitro Analysis of Five Starch Sources: Nutrient Release Patterns and Functional Effects in a Simulated Gastrointestinal Environment.

Corn, paddy, wheat, sorghum, and cassava serve as the primary energy sources in both human and animal diets. This study aimed to evaluate their nutrient release patterns in a simulated gastrointestinal environment and to assess the in vitro biological activity of the metabolites produced during digestion. The results showed that wheat exhibited the highest dry matter degradation in the stomach-jejunum-ileum digestion stage, while wheat and paddy showed the highest crude protein degradation compared with the other starch sources. In addition, wheat had a higher total free sugar concentration th

Comparing and Optimizing Tissue Clearing Protocols for Cochlear Immunostaining and 3D Reconstruction by Light-Sheet Microscopy.

PURPOSE: Current standard methods for immunohistochemical analysis of the inner ear rely on cryosectioning of the intact cochlea or dissection of the organ of Corti. Cryosections provide only limited structural information and do not allow quantification of hair cell numbers per basilar membrane length, whereas dissections require substantial technical expertise and often result in damage to the basal cochlear turns. Tissue clearing has largely overcome these limitations and has been widely applied to organs such as the brain, heart, liver, and kidney. However, despite several reports describi
